The Grooved Pegboard is a manipulative dexterity test consisting of 25 holes with randomly positioned slots. Pegs with a key along one side must be rotated to match the hole before they can be inserted, requiring more complex visual-motor coordination than most pegboard tests.
Transcranial anodal stimulation (tDCS) improves manual dexterity in healthy old adults. The underlying changes in finger force behavior for this improved dexterity have not been reported. Here, we investigated the effects of tDCS (20-min) over primary motor cortex (M1) combined with repeated practice on the Grooved pegboard test (tDCS+MP) on the fingertip forces applied to an object during grasp and manipulation. Eight right-handed able-bodied individuals (60-85 years) participated in a sham-controlled, single-blinded study. Each participant received anodal and sham intervention in two sessions at least 5-day apart. Before and after intervention, they performed a key-slot task that required inserting a slot on an object onto a stationary bar, an isometric force production task using a pinch grip, and the Grooved pegboard test. Anodal relative to sham tDCS+MP allowed participants to better retain the improved performance on the pegboard test. For the isometric task, anodal tDCS+MP significantly ...

In the present study we investigated the membrane events and the ionic processes which mediate the stimulatory effect of ouabain on the release of endogenous dopamine (DA) and previously taken-up (3H)DA release from rat hypothalamic tuberoinfundibular dopaminergic (TIDA) neurons. Ouabain (0.1-1 mM) dose-dependently stimulated endogenous DA and newly taken-up (3H)DA release. This effect was counteracted partially by nomifensine (10 microM). Removal of Ca++ ions from the extracellular space in the presence of the Ca++-chelator ethylene glycol bis(beta-aminoethyl ether)-N,N-tetraacetic acid prevented completely ouabain-elicited (3H)DA release. Lanthanum (1 mM) and cobalt (2 mM), two inorganic Ca++-entry blockers, were able to inhibit this stimulatory effect, whereas verapamil (10 microM) and nitrendipine (50 microM), two organic antagonists of the voltage-operated channel for Ca++ ions, failed to affect ouabain-induced (3H)DA release. By contrast, adriamycin (100-300 microM), a putative inhibitor ...
The pegboard consists of a board with two parallel rows with 25 holes into which cylindrical metal pegs are placed by the examinee. The test involves a total of four trials.[5] To begin, there is a brief practice. The subsets for preferred, non-preferred, and both hands require the patient to place the pins in the holes as quickly as possible, with the score being the number of pins placed in 30 seconds. ...
Giving birth triggers a wide repertoire of physiological and behavioural changes in the mother to enable her to feed and care for her offspring. These changes require coordination and are often orchestrated from the central nervous system, through as of yet poorly understood mechanisms. A neuronal population with a central role in puerperal changes is the tuberoinfundibular dopamine (TIDA) neurons that control release of the pituitary hormone, prolactin, which triggers key maternal adaptations, including lactation and maternal care. Here, we used Ca2+ imaging on mice from both sexes and whole-cell recordings on female mouse TIDA neurons in vitro to examine if they adapt their cellular and network activity according to reproductive state. In the high-prolactin state of lactation, TIDA neurons shift to faster membrane potential oscillations, a reconfiguration that reverses upon weaning. During the estrous cycle, however, which includes a brief, but pronounced, prolactin peak, oscillation frequency ...
PD is associated with alterations of gut motor functions (Cloud and Greene, 2011; Pfeiffer, 2011; Pellegrini et al., 2015), which have been proposed to result both from an early impairment of the ENS and as a consequence of central nigrostriatal degeneration associated with dopaminergic denervation. In this context, our purpose was to evaluate the impact of nigrostriatal dopaminergic denervation on the patterns of colonic motility and related cholinergic control. Indeed, current data on the abnormalities of colonic cholinergic neuromotor control in PD are scarce and inconsistent. Thus, we aimed at characterizing the alterations occurring in the 6-OHDA model by a multidisciplinary functional, molecular, and morphologic approach. Overall, our results provide convincing evidence that the induction of nigrostriatal denervation, which reflects one of the main pathologic hallmarks of PD, is associated with significant alterations of colonic excitatory cholinergic neurotransmission, resulting in ...
Although most research in PD has a focus on presynaptic dopaminergic denervation, new lessons may be learned by exploring to what extent alterations in non-motor and non-dopaminergic systems may play a role in the balance impairment in this disorder. This project is designed to evaluate striatal pre-synaptic dopaminergic, cortical, thalamic, and brainstem cholinergic, cognitive, and oculomotor mechanisms underlying the risk of falling and imbalance in PD. The proposed study will exploit advances in our center in the development of dynamic biomathematical modeling of electronic platform data, application of specialized cognitive and oculomotor assessments, and positron emission tomography (PET). This novel multi-system approach holds promise for understanding mechanisms of falling in PD beyond pre-synaptic dopaminergic nigrostriatal denervation ...

The aim of this study was to determine whether tactile spatial resolution and digital dexterity performance are associated during childhood. A total of 104 healthy children (52 boys and 52 girls) were assessed. The tactile spatial resolution threshold was determined using an enlarged set of JVP domes with a procedure adapted for children. Digital dexterity was tested with the Purdue Pegboard Test. While tactile spatial resolution and digital dexterity improved with age following a parallel evolution, no correlation was found between the scores of the children for both tests. We suggest that central maturational processes are responsible for a global refinement of sensory and motor function between the ages of 4 and 10 years, reaching the range of adult values at this age. ...

We learn new motor tasks by trial and error, repeating what works best and avoiding past mistakes. To repeat what works best we must register a satisfactory outcome, and in a study [1] we showed the existence of an evoked activity in the basal ganglia that correlates with accuracy of task performance and is associated with reiteration of successful motor parameters in subsequent movements. Here we report evidence that the signaling of positive trial outcome relies on dopaminergic input to the basal ganglia, by recording from the subthalamic nucleus (STN) in patients with nigrostriatal denervation due to Parkinsons Disease (PD) who have undergone functional neurosurgery. Correlations between subthalamic evoked activities and trial accuracy were weak and behavioral performance remained poor while patients were untreated; however, both improved after the dopamine prodrug levodopa was re-introduced. The results suggest that the midbrain dopaminergic system may be important, not only in signaling explicit

N2 - BACKGROUND: Craniopharyngiomas are slow-growing, locally invasive intracranial tumors that can generate considerable morbidity, and recurrences are often difficult to manage. Because reliable morphologic criteria for accurately predicting the clinical outcome of these tumors are lacking, we evaluated the growth potential of craniopharyngiomas by measuring their proliferative activity based on MIB-1 immunostaining for the Ki-67 antigen, which is expressed during all phases of the cell cycle except G0. METHODS: Paraffin sections from 37 cases of craniopharyngiomas were immunostained with the monoclonal antibody MIB-1, and a labeling index was derived in each case from an the with the ...

Olivopontocerebellar atrophy (OPCA) refers to a group of ataxias characterized by progressive neurological degeneration affecting the cerebellum, the pons and the inferior olives.
Four types of hereditary OPCA have disappeared from medical nomenclature, as they have been found to be the same as an already classified form of spinocerebellar atrophy. Type 2, autosomal recessive and Type 5 are still classified as olivopontocerebellar atrophy, though when their genetic associations are identified they may be renamed or combined with other conditions. ...
Olivopontocerebellar atrophy (OPCA) is a neurodegenerative syndrome characterized by prominent cerebellar and extrapyramidal signs, dysarthria, and dysphagia. Those who study OPCA quickly learn that it is not a single entity, and that its nosology can be confusing.
Multiple system atrophy (MSA) is a progressive neurodegenerative disorder characterised by a variable combination of autonomic failure, levodopa-unresponsive parkinsonism, cerebellar ataxia and pyramidal symptoms. The pathological hallmark is the oligodendrocytic glial cytoplasmic inclusion (GCI) consisting of α-synuclein; therefore, MSA is included in the category of α-synucleinopathies. MSA has been divided into two clinicopathological subtypes: MSA with predominant parkinsonism and MSA with predominant cerebellar ataxia, which generally correlate with striatonigral degeneration and olivopontocerebellar atrophy, respectively. It is increasingly recognised, however, that clinical and pathological features of MSA are broader than previously considered.. In this review, we aim to describe recent advances in neuropathology of MSA from a review of the literature and from information derived from review of nearly 200 definite MSA cases in the Mayo Clinic Brain Bank. In light of these new ...
Abstract: Differences between dyslexics and controls in the unimanual and bimanual conditions of the peg placement section of the Purdue Pegboard Test were examined. Twenty-three disabled and twenty-three normal readers were studied. The groups were carefully screened on a neuropsychological battery. The disabled readers were comprised of a relatively homogeneous language-disordered subgroup exhibiting deficits in naming. Significant Group X Condition interactions were obtained for both raw and percentile scores and indicated that disabled readers performed worse than controls in the unimanual compared to bimanual conditions. The dyslexics performed particularly poorly compared with controls on the left hand condition. The implications of these data for hypotheses which argue for left hemisphere dysfunction, as well as those which posit interhemispheric transfer deficits in reading disabled children, are discussed. ...
Several studies indicate the functional importance of the motor cortex for higher cognition, language and semantic processing, and place the neural substrate of these processes in sensorimotor action-perception circuits linking motor, sensory and perisylvian language regions. Interestingly, in individuals with autism spectrum disorder (ASD), semantic processing of action and emotion words seems to be impaired and is associated with hypoactivity of the motor cortex during semantic processing. In this study, the relationship between semantic processing, fine motor skills and clinical symptoms was investigated in 19 individuals with ASD and 22 typically-developing matched controls. Participants completed two semantic decision tasks involving words from different semantic categories, a test of alexithymia (the Toronto Alexithymia Scale), and a test of fine motor skills (the Purdue Pegboard Test). A significant Group × Word Category interaction in accuracy (p

A major challenge to FTD/PSP doctors and researchers is why the same underlying cause of disease (say a gene mutation) can cause very different symptoms in two people, even in the same family. Conversely, two very different underlying illnesses can cause symptoms that are identical. Understanding the connection between the underlying disease and an individuals symptoms needs a map of clinicopathological correlations. These rely on the generous gift of brain donation, and the experience of pathologists at the Cambridge Brain Bank. It is a very personal decision, whether to donate ones brain after death to research, but it is a topic we typically discuss with patients and families many years ahead. The Cambridge Brain Bank plays an important part in proving the value of new tests for diagnosis like PET scanning, blood tests, and psychology tests.. [Back to top]. ...
In the present work, we studied the prognostic significance in breast cancer patients of DNA amplification at eight genes or loci, AIB1 (20q11), CCND1 (11q13), EMS1 (11q13), ERBB2 (17q12-q21), FGFR1 (8p12), MDM2 (12q13), MYC (8q24), and RMC20C001 (20q13), and of mutations of p53. These genetic anomalies were selected because of their frequent occurrence in sporadic breast tumors and because of the existence of clinicopathological correlations (4 , 8) . The rational of our study was to test whether genetic anomalies that affect different genes or loci bore prognostic significance in different patient subsets. Furthermore, we were interested in verifying whether specific combinations of genetic alterations were associated with a worsened course of the disease.. Of the nine parameters studied, six (FGFR1, CCND1, EMS1, RMC20C001, MDM2, and p53 mutations) correlated with bad prognosis in either the univariate or multivariate analysis on the whole population of patients. Using a stepwise analysis, we ...

You can try to use them all willy nilly but to do the best possible job, you have to be clear on how and when, or if, each tool is used. There are also some you cant use without resulting
Olivopontocerebellar atrophy (OPA) is an uncommon but serious neurological disorder. It causes nerve tissue degeneration and atrophy in the brain. Doctors believe that OPA is similar to a multiple system atrophy (MSA) disorder. Different MSA disorders occur in different sites within the brain.. OPA shares many symptoms with MSA disorders. One common symptom is ataxia. Ataxia is a difficulty in controlling your muscle movements for gait. Diagnosing OPA can be challenging because so many of the symptoms mirror those of MSA disorders. Neurological disorders also share symptoms with OPA. For example, Parkinsons disease can look similar to OPA. Some of the shared symptoms include tremors and balance problems. Diagnostic imaging tests help neurologists look for areas of damage and diagnose disorders.. Theres no cure for OPA. Doctors are able to offer treatment that helps patients live as long as possible. The life expectancy for people with OPA differs because brains degenerate at different ...
Degenerative brain disease. Coloured single photon emission computed tomography (SPECT) scans of axial sections through the brain of a patient with striatonigral degeneration (SND). SND is a neurodegenerative disease, similar to Parkinsons, that may be present in multisystem atrophy. The scans show areas of neuronal loss (blue and black) at the front of the brain (top). This leads to the symptoms, which include; muscular rigidity, unstable posture, urinary problems, tremor and depression. There is no cure and drugs only alleviate symptoms in the short term. SPECT scans use gamma cameras to detect radioactive tracers, injected into the blood, that accumulate in areas of high metabolic activity. - Stock Image M260/0329
