The Pan-Provincial Vaccine Enterprise, Inc. (PREVENT) and Vaxent announced Tuesday that they have launched their Phase 1 clinical trial for StreptAnova, a vaccine engineered to protect people against Group A streptococcal (GAS) infections.
The broodstock were injected with the vaccine at a dose of 108 CFU per fish at second gonadal maturity until spawning. Challenge tests were carried out on the offspring at 5, 10, 15 and 20 days after hatching using five different strains, both individually and in combination. The researchers, who were mainly based at Nanjing Agricultural University, observed immunological parameters in broodstock, eggs and larvae and relative per cent survival (RPS) of larvae after being challenged with pathogenic S. agalactiae. The results showed that the leukocytes, phagocytic activity, respiratory burst, lysozyme activity and antibody levels of vaccinated broodstock were at a higher level compared with unvaccinated broodstock. The high level of the lysozyme activity, antibody levels and recombination activating gene 1 (RAG1) were also observed in eggs and larvae from vaccinated broodstock. Larvae produced from vaccinated broodstock when challenged with pathogenic S. agalactiaehad RPS values over 50 percent ...

The incorporation of lipid moieties into synthetic peptide vaccines has been demonstrated to self-adjuvant otherwise poorly immunogenic peptides, whereas carbohydrates have emerged to be advantageous carriers for assembling these peptides. With the advent of an efficient native chemical ligation method, which is compatible with both peptides and carbohydrates, we have developed highly pure self-adjuvanting tetravalent group A streptococcal vaccine candidates assembled on carbohydrate templates. The utility of chemoselective ligation has overcome difficulties in the synthesis and purification of branched high molecular weight peptides. Circular dichroism measurements provided the evidence of alpha-helix formation of the assembled peptide epitopes, which may have impact on their immunogenicity. (C) 2009 Elsevier Ltd. Synthesis of four glycolipids with different number of lauroyl groups on glucose or cellobiose as scaffolds is described. Their immunological evaluations either admixed with or covalently linked to J8, a peptide antigen derived from the C-terminus of the antiphagocytic M-protein of group A streptococcus, are also investigated. Administration of mixtures of J8 and glycolipids to B10BR (H-2k) mice induced low-levels of J8-specific IgG antibodies. While glycolipopeptides, in which J8 was covalently linked to the synthetic glycolipids, demonstrated high-levels of antibody titers comparable with the co-administration of these glycolipopeptides with complete Freunds adjuvant, suggesting clearly the strong potency of the synthesized glycolipids as self-adjuvanting moieties ...

MEMPHIS-A strep throat vaccine developed at the University of Tennessee medical school might be the first tested on humans, a UT-Memphis professor who helped create it said Tuesday.. Dr. James Dale said the vaccine protects laboratory animals from streptococci bacteria without causing dangerous side effects in the animals immune systems, as other experimental vaccines have done.. It is the first treatment safe enough to be considered for clinical trials, Dale said.. He is working with ID Vaccine Corp. of Vancouver, British Columbia, and the National Institutes of Health for FDA approval in 1998 to test humans, perhaps as early as next summer. Initially, about 30 people would be vaccinated, he said.. Dale said other experimental streptococci vaccines have caused rheumatic fever in animals, stimulating the animals immune system to attack and damage the heart, liver and other tissue.. The safety of these vaccines depends on eliminating these harmful effects, Dale said. In our tests, immunized ...
Streptococcus pyogenes (group A Streptococcus (GAS)) causes ∼700 million human infections each year, resulting in over 500,000 deaths. Since NES provides the intracellular access by opening nanosize pores on the cell membrane, the process should be largely cargo agnostic. Here, we show that NES not only delivers protein peptides but also allows intracellular delivery and gene editing of Cas9-ribonucleoproteins (RNPs). CRISPR technology (38) has had a profound impact on various research and medical applications, including identification of gene mutations of specific disease, development of disease models, and gene therapy (39). However, intracellular delivery of Cas9 RNPs for gene editing remains a challenge (40). We programmed the Cas9 RNPs to knock out PPIB, a house keeping gene that encoded an enzyme catalyzing the cis-tran isomerization and regulating protein folding and maturation. The purified recombinant Streptococcus pyogenes Cas9 (SpyCas9) protein carried two nuclear localization signal (NLS) sequences at its C terminus, followed by a GFP protein containing another NLS at C terminus (with a total molecular weight of 187 ...

N2 - Streptococcus agalactiae (group B Streptococcus, GBS) is a leading cause of severe invasive disease in neonate, elderly, and immunocompromised patients worldwide. Despite recent advances in the diagnosis and intrapartum antibiotic prophylaxis (IAP) of GBS infections, it remains one of the most common causes of neonatal morbidity and mortality, causing serious infections. Furthermore, recent studies reported an increasing number of GBS infections in pregnant women and elderly. Although IAP is effective, it has several limitations, including increasing antimicrobial resistance and late GBS infection after negative antenatal screening. Maternal immunization is the most promising and effective countermeasure against GBS infection in neonates. However, no vaccine is available to date, but two types of ...
Group A Streptococcus (GAS) is an important human pathogen responsible for superficial infections of the skin and throat and serious invasive infections including necrotizing fasciitis and streptococcal toxic shock syndrome. Repeated mild infections can also lead to long-term autoimmune complications. Disease and economic burden data support the rationale for the development of a vaccine against GAS. One of the several hurdles in GAS vaccine development is the variability of protection observed in different animal models, particularly in mice. We have used a non-human primate (NHP) model for pharyngeal infection to assess GAS vaccine protective efficacy. The NHP model was validated using full-M1 protein as a vaccine antigen and used to evaluate the protective efficacy of an experimental vaccine (Combo#5) containing streptolysin O, IL-8 protease, C5a peptidase, arginine deiminase and trigger factor. All antigens were adjuvanted with Alum and NHPs were immunised via intramuscular injection on ...
In this paper, we describe a murine model that mimics natural S. pneumoniae pathogenesis involving the transition of S. pneumoniae from commensal to pathogen during a viral coinfection caused by PR8 influenza virus. We show the influence of an increasing influenza viral load on increased bacterial densities and define quantitatively the NP thresholds of two diverse S. pneumoniae STs (6A and 8) associated with invasive infections. We show that vaccination with PhtD helps prevent the S. pneumoniae NP density from reaching a pathogenic threshold during an influenza virus coinfection. Although, PCV13 vaccination led to a greater reduction of S. pneumoniae NP density, the outcome vis-à-vis prevention of invasive S. pneumoniae pathogenesis did not differ significantly between PhtD- and PCV13-vaccinated mice. We further show that while CD4+ T cells represent the main protective correlate in PhtD-vaccinated adult mice, protection in PCV13-vaccinated mice is CD4+ T cell independent. Infant mice require ...

TY - JOUR. T1 - The alpha C protein mediates internalization of group B Streptococcus within human cervical epithelial cells. AU - Bolduc, G. R.. AU - Baron, M. J.. AU - Gravekamp, C.. AU - Lachenauer, C. S.. AU - Madoff, L. C.. PY - 2002/11/1. Y1 - 2002/11/1. N2 - Group B Streptococcus (GBS) is the leading cause of bacterial chorioamnionitis and neonatal pneumonia, sepsis, and meningitis. Deletion of the alpha C protein gene (bca) attenuates the virulence of GBS in an animal model; significant survival differences in the first 24 h of infection suggest a pathogenic role for the alpha C protein early in the infection process. We examined the role of alpha C protein in the association between GBS and mucosal surfaces using a human cervical epithelial cell line, ME180. Fluorescent and confocal microscopy and flow cytometry demonstrated that 9-repeat alpha C protein binds to the surface of ME180 cells. Isolated N-terminal region of this protein also binds to these cells and competitively inhibits ...
Streptococcus suis and group B Streptococcus (GBS) are encapsulated streptococci causing septicemia and meningitis. Antibodies (Abs) against capsular polysaccharides (CPSs) have a crucial protective role, but the structure/composition of the CPS, including the presence of sialic acid, may interfere with the generation of anti-CPS Ab responses. We investigated the features of the CPS-specific Ab response directed against S. suis serotypes 2 and 14 and GBS serotypes III and V after infection or immunization with purified native or desialylated CPSs in mice. Whereas S. suis-infected mice developed a very low/undetectable CPS-specific IgM response, significant anti-CPS IgM titers were measured in GBS-infected animals (especially for type III GBS). No isotype switching was detected in S. suis- or GBS-infected mice. While the expression of sialic acid was essential for the immunogenicity of purified GBS type III CPS, this sugar was not responsible for the inability of purified S. suis types 2, 14 and GBS type
Strangles (equine distemper) is a contagious upper respiratory tract infection of horses and other equines caused by a gram-positive bacterium, Streptococcus equi. As a result, the lymph nodes swell, compressing the pharynx, larynx and trachea and can cause airway obstruction leading to death, hence the name Strangles. Strangles is enzootic in domesticated horses worldwide. The contagious nature of the infection has at times led to limitations on sporting events. The disease is spread by an infected horse when nasal discharge or pus from the draining lymph nodes contaminate pastures, feed troughs, brushes, bedding, tack etc. Equines of any age may contract the disease, although younger and elderly equines are more susceptible. Young equines may lack immunity to the disease because they have not had prior exposure. Geriatric equines may have a weaker immune system. A horse with strangles will typically develop abscesses in the lymph nodes of the head and neck causing coughing fits and difficulty ...
Infection with group A streptococcus (GAS) may result in a number of clinical conditions, including the potentially life-threatening postinfectious sequelae of rheumatic fever and rheumatic heart disease. As part of the search for a vaccine to prevent GAS infection, a conformationally constrained and minimally conserved peptide, J14, from the M protein of GAS has been defined. In the present study, J14 was formulated with bacterial outer membrane proteins (proteosomes) and then intranasally administered to outbred mice without additional adjuvant. Such immunization led to high titers of J14-specific serum immunoglobulin (Ig) G and mucosal IgA. After upper respiratory tract GAS challenge, immunized mice demonstrated increased survival and reduced GAS colonization of the ...
The revised 2010 GBS American Academy of Pediatrics guidelines for neonatal management were designed to broaden the scope to include all neonates, to increase the clarity of the recommendations, and to decrease unnecessary laboratory evaluations and empirical antibiotics for infants at low risk. Although this strategy will never prevent all infections, the revised guidelines should result in a further decrease in cases of perinatal GBS disease. The management of neonates continues to be based on clinical signs, the presence of maternal risk factors for GBS neonatal disease, and the likely efficacy of IAP (or maternal antimicrobial treatment in the case of clinical or occult chorioamnionitis) in preventing early-onset disease. The revised infant management algorithm (Fig 1) is derived from recent data summarized in the published CDC document regarding the epidemiology of GBS disease and the usefulness of a limited evaluation of well-appearing neonates.. All newborn infants with signs suggestive ...
A veterinarian can make a preliminary diagnosis based on the symptoms (see above). The initial diagnosis can be confirmed by taking a sample of the nasal fluids (from the back of the horses naval cavity - nasopharynx) or abscess fluid (if the disease has progressed to that stage) and sending it into a laboratory which can examine the fluid for presence of the strangles bacteria. In 2008, a blood test for strangles was developed in the UK to allow quick and early testing.. If you suspect a horse has strangles, it should be immediately isolated from other horses (to prevent the spread of the disease). The disease is highly contagious, so a responsible horse owner will take immediate action to prevent other horses from being affected, including both his/her own horses and horses with other owners... You should have a veterinarian examine the horse as soon as possible:. ...
Estimates of disease burden and data on the sources of invasive postpartum group A streptococcus (GAS) infections will help guide public health action. Active, population-based surveillance was conducted in 9 regions from 1995 through 2000. A case of GAS infection was defined as isolation of GAS from a sterile site in a resident of a surveillance area who was pregnant or in the postpartum period. Census and live birth data were used to calculate rates. Eighty-seven cases of postpartum GAS infection (2.2% of 3957 invasive GAS infections) occurred at 3%-8% of hospitals annually. We estimate that 220 cases occurred annually in the United States. Two or more cases were noted during 6 months at 8 hospitals, during 1 year at 13 hospitals, and during 2 years at 16 hospitals. Cases due to identical emm types clustered more frequently than expected by chance. Although postpartum GAS infections are rare, the clustering of infections due to identical strains suggests that some invasive cases may have a ...
Non-antigen-specific stimulatory cancer immunotherapies are commonly complicated by off-target effects. Antigen-specific immunotherapy, combining viral tumor antigen or personalized neoepitopes with immune targeting, offers a solution. However, the lack of flexible systems targeting tumor antigens to cross-presenting dendritic cells (DCs) limits clinical development. Although antigen-anti-Clec9A mAb conjugates target cross-presenting DCs, adjuvant must be codelivered for cytotoxic T lymphocyte (CTL) induction. We functionalized tailored nanoemulsions encapsulating tumor antigens to target Clec9A (Clec9A-TNE). Clec9A-TNE encapsulating OVA antigen targeted and activated cross-presenting DCs without additional adjuvant, promoting antigen-specific CD4+ and CD8+ T cell proliferation and CTL and antibody responses. OVA-Clec9A-TNE-induced DC activation required CD4 and CD8 epitopes, CD40, and IFN-α. Clec9A-TNE encapsulating HPV E6/E7 significantly suppressed HPV-associated tumor growth, while ...

Puppy strangles comes up suddenly manifesting as marked swelling of the muzzle, eyelids, and face in a puppy between the ages of 3 weeks and four months of age. The inner surface of the ear flaps are commonly involved as well and occasionally lesions in other body areas occur. Fever, joint swelling, and loss of appetite occur in approximately 25% of patients. Despite the extreme inflammation and secondary infections the heart of treatment for this condition is immune-suppression. High doses of corticosteroids such as prednisone are needed as this condition appears to be an immune-mediated disease.
