Vancomycin-resistant Staphylococcus hominis endophthalmitis following cataract surgery Jun Yeon Won,1 Moosang Kim21Department of Otolaryngology, School of Medicine, Kangwon National University, Chuncheon, Korea; 2Department of Ophthalmology, School of Medicine, Kangwon National University, Chuncheon, KoreaAbstract: We report a case of acute postoperative endophthalmitis caused by vancomycin-resistant Staphylococcus hominis, treated at our hospital. An 80-year-old male presented 2 days after uncomplicated phacoemulsification and posterior chamber intraocular lens implantation, with a 24-hour history of progressive visual loss and redness in the operated (right) eye. On examination, best corrected visual acuity was counting fingers. Anterior segment examination revealed conjunctival injection, chemosis, corneal edema, and hypopyon. B-scan ultrasonography showed vitreous opacification, but no retinal detachment. Acute postoperative endophthalmitis was diagnosed. We performed vitrectomy with vancomycin in
Staphylococcus hominis is a coagulase-negative member of the bacterial genus Staphylococcus, consisting of Gram-positive, spherical cells in clusters. It occurs very commonly as a harmless commensal on human and animal skin and is known for producing thioalcohol compounds that contribute to body odour. Like many other coagulase-negative staphylococci, S. hominis may occasionally cause infection in patients whose immune systems are compromised, for example by chemotherapy or predisposing illness. Colonies of S. hominis are small, usually 1-2 mm in diameter after 24 hours incubation at 35 °C, and white or tan in colour. Occasionally, strains are resistant to novobiocin and may be confused with other resistant species (e.g. S. saprophyticus). It is one of only two species of Staphylococcus to display sensitivity to desferrioxamine, the other being S. epidermidis. Unlike S. epidermidis, S. hominis produces acid from trehalose, so the two tests together serve to identify the species. Numerous ...
Results of blood cultures at that time showed the presence of the local external contamination, outside the biological vaccine, in particular bacteria, which was consistent with the clinical pictures of hospitalized children. Microbiology studies conducted on children reported the finding of Staphylococcus hominis, a type of bacterium commonly found in the skin of people. These findings rule out other bacteria, such as those found in the gastrointestinal tract and airways. Molecular studies demonstrated that isolated Staphylococcus hominis in different patients was the same, namely that the bacteria came from a single source of contamination. ...
The microbiome can promote or disrupt human health by influencing both adaptive and innate immune functions. We tested whether bacteria that normally reside on human skin participate in host defense by killing Staphylococcus aureus, a pathogen commonly found in patients with atopic dermatitis (AD) and an important factor that exacerbates this disease. High-throughput screening for antimicrobial activity against S. aureus was performed on isolates of coagulase-negative Staphylococcus (CoNS) collected from the skin of healthy and AD subjects. CoNS strains with antimicrobial activity were common on the normal population but rare on AD subjects. A low frequency of strains with antimicrobial activity correlated with colonization by S. aureus. The antimicrobial activity was identified as previously unknown antimicrobial peptides (AMPs) produced by CoNS species including Staphylococcus epidermidis and Staphylococcus hominis. These AMPs were strain-specific, highly potent, selectively killed S. aureus, ...
Staphylococcus cohnii is a Gram-positive, coagulase-negative member of the bacterial genus Staphylococcus consisting of clustered cocci. The species commonly lives on human skin; clinical isolates have shown high levels of antibiotic resistance. A strain of S. cohnii was found to contain a mobile genetic element very similar to the staphylococcal cassette chromosome encoding methicillin resistance element seen in Staphylococcus aureus. Background: Permafrost preserves a variety of viable ancient microorganisms. Some of them can be cultivated after being kept at subzero temperatures for thousands or even millions of years. Objective: To cultivate bacterial strains from permafrost. Design: We isolated and cultivated two bacterial strains from permafrost that was obtained at Mammoth Mountain in Siberia and attributed to the Middle Miocene. Bacterial genomic DNA was sequenced with 40-60× coverage and high-quality contigs were assembled. The first strain was assigned to Staphylococcus warneri species (designated MMP1) and the second one to Staphylococcus hominis species (designated MMP2), based on the classification of 16S ribosomal RNA genes and genomic sequences ...

A rapid quantitative real-time PCR for detection of M. hominis from cervical swab samples was developed. To our knowledge it is the first LightCycler PCR protocol where quantification of M. hominis is combined with melting curve assay. The LightCycler PCR reproducibility was able to detect down to ten copies/reaction of the genomic PG21 DNA. All other 20 M. hominis isolates were positive in the assay. Additionally, isolates 132 and 4712, which based on melting temperature belong to the two other groups than PG21, were used to document detection limits of M. hominis isolates. Similarly to PG21, the detection limits were 5 copies/μl, equals to 10 copies per reaction for both isolates.. The average efficiency was high, but small differences were seen. This can be caused by different factors such as presence of inhibitors in the sample and treatment of the sample.. The artificially constructed samples consisting of PG21 DNA and coloured BEa and SP-4 media showed that these media inhibited the PCR ...

Abstract (EN): Modifying the surface properties of the stainless steel used in heat exchangers to decrease its tendency for the building-up of milk deposits seems to be a promising strategy to reduce fouling during heat treatment in the dairy industry. In this work, several modified stainless steel surfaces, obtained by reactive magnetic sputtering, were used as fouling supports for a milk-simulating mineral solution under constant conditions of temperature, pH and calcium concentration. The aim of the work was to quantify deposition and removal parameters, based on the influence of surface energy properties on the type of deposit. The fouling process was characterized by the surface reaction coefficient of the ions present in solution, as well as the adhesion coefficients of the particles formed in the bulk. An insight on the mechanisms of deposition and removal processes was achieved, as well as by the dependence between those two processes and the electron-donor component (gamma(-)) of the ...

Methicillin-resistant staphylococci were tested for susceptibility to cephradine, cephaloridine, cephalexin, and cefuroxime and 30 degree C and 37 degree C on ordinary media and on media of enhanced osmotic strength. The coagulase-negative strains were divided into Staphylococcus epidermis and Staphylococcus hominis. Generally the number of susceptible strains decreased with low incubation temperature and osmotic support. When Staphylococcus aureus was tested against cephalexin and cefuroxime, most were susceptible at 37 degree C whereas all were resistant at 30 degree C. A similar trend was found in the case of cephradine, where it was accentuated by osmotic support. While almost all strains of Staph. epidermidis were sensitive to all four drugs under all test conditions, the susceptibility of Staph. hominis was generally decreased by either lower incubation temperature or osmotic support or both.. ...

Posted on January 18, 2009 Comments (0) Bacteria continue to gain resistance to commonly used antibiotics. In this weeks JBC, one potential new antibotic has been found in the tiny freshwater animal Hydra.. The protein identified by Joachim Grötzinger, Thomas Bosch and colleagues at the University of Kiel (Germany), hydramacin-1, is unusual (and also clinically valuable) as it shares virtually no similarity with any other known antibacterial proteins except for two antimicrobials found in another ancient animal, the leech.. Hydramacin proved to be extremely effective though; in a series of laboratory experiments, this protein could kill a wide range of both Gram-positive and Gram-negative bacteria, including clinically-isolated drug-resistant strains like Klebsiella oxytoca (a common cause of nosocomial infections). Hydramacin works by sticking to the bacterial surface, promoting the clumping of nearby bacteria, then disrupting the bacterial membrane.. Grötzinger and his team also ...

A recently described cytotoxic monoclonal antibody (mAb 1D5) raised against Blastocystis hominis isolate B, was tested for reactivity with 13 different isolates of Blastocystis. The isolates used were previously isolated from humans, rats and reptiles and were maintained as axenised cultures throughout the course of this stady. Five B. hominis isolates (B, C, E, G and H) were found to react with mAb 1D5 in immunoblotting studies and the indirect fluorescence antibody test. The pattern of fluorescence observed for all five isolates was diffuse and patchy. Immunoblotting studies revealed that mAb 1D5 reacted with a 29-30-kDa protein found in all five isolates. Results of a cytotoxic assay showed that the mAb exhibited a complement-independent cytotoxic effect on all the exposed isolates. Microscopic observations showed differences in morphology between the Blastocystis cells exposed and unexposed to mAb. Acridine orange staining performed on both exposed and unexposed cells showed similar internal ...

TY - JOUR. T1 - Determination of the chromosomal relationship between mecA and gyrA in methicillin-resistant coagulase-negative staphylococci. AU - Fey, Paul D.. AU - Climo, Michael W.. AU - Archer, Gordon L.. PY - 1998/2. Y1 - 1998/2. N2 - mecA, the gene that mediates methicillin resistance, and its accompanying mec locus DNA, insert near the gyrA gene in Staphylococcus aureus. To investigate whether there is a similar relationship between mecA and gyrA in coagulase-negative staphylococci (CNS), mecA- and gyrA-specific DNA fragments were used to probe methicillin-resistant isolates of Staphylococcus epidermidis (MRSE) (n = 11) and Staphylococcus haemolyticus (MRSH) (n = 11). The gyrA probe hybridized to the same SmaI DNA fragment as the mecA probe in all strains tested. However, since the size of the SmaI fragments containing meca and gyrA varied from 73 to 600 kb, the distance between the two genes was determined more precisely. Cloned mecA or gyrA fragments plus vector sequences each ...

So here is a big question. If there is such a wide diversity of properties in different biofilms, how can we expect to find characteristics that apply to all biofilms? We are glad you ask. What has been discovered is that in spite of their wide diversity, biofilms do seem to have some common attributes, such as their ability to grow on virtually any surface, how they attach to a surface, their mode of growth, their ability to spread, how they are nourished, how they maintain themselves as a colony, and so forth.. For example, the image at the right shows pitting and corrosion of a stainless steel surface. This was caused by a biofilm, whose presence influenced how and how fast minerals were deposited on the surface. This, in turn, modified the electrochemical properties of the stainless steel, which caused the pitting corrosion of this seemingly impervious metal.. Can we extend what we learn about this kind of biofilm to other sorts of biofilms, such as plaque on teeth? Apparently so, as ...

PURPOSE: To evaluate the fluoroquinolone susceptibilities of ocular isolate coagulase-negative staphylococci (CoNS), identified at the Microbiology Laboratory - UNIFESP. DESIGN: Experimental laboratory investigation. METHODS: The minimum inhibitory concentrations (MICs) of 21 strains of methicillin-resistant coagulase-negative staphylococci (MRCoNS) and 22 methicillin-sensitive coagulase-negative staphylococci (MSCoNS) to ciprofloxacin, ofloxacin, gatifloxacin and moxifloxacin were determined, using the E-test method standardized by the Clinical and Laboratory Standards Institute (CLSI/NCCLS). RESULTS: The MIC90s (µg/ml) for the second generation of tested fluoroquinolones were higher than the fourth generation, especially for the methicillin-resistant coagulase-negative staphylococci group. CONCLUSION: Our results indicate that methicillin-sensitive coagulase-negative staphylococci are more susceptible to quinolones than are methicillin-resistant coagulase-negative staphylococci and that ...
BACKGROUND: Antimicrobial resistance may compromise the efficacy of antibiotic prophylaxis before surgery. The aim of this study was to measure susceptibility and clonal distribution of coagulase-negative staphylococci (CoNS) colonizing the skin around the surgery access site before and after the procedure. METHODS: From March to September 2004, a series of 140 patients undergoing elective major abdominal surgery were screened for CoNS colonization at admission and 5 days after surgery. All isolates were tested for antibiotic susceptibility and genotyped by pulsed-field gel electrophoresis (PFGE). RESULTS: Colonization rates with CoNS at admission and after surgery were 85% and 55%, respectively. The methicillin-resistant CoNS rate increased from 20% at admission to 47% after surgery (P = 0.001). The PFGE pattern after surgery revealed more patients colonized with identical clones: 8/140 patients (8/119 strains) and 26/140 patients (26/77 strains), respectively (P , 0.001). CONCLUSIONS: Our ...
This article summarizes the results obtained studying the interaction between polonium and stainless steel 316L in different atmospheres by the thermochromatography method. This issue is particularly important in terms of licensing of the MYRRHA prototype ADS reactor, where highly radiotoxic α-emitting polonium isotopes are produced in lead-bismuth eutectic used as both spallation target material and coolant, while stainless steel 316L is going to be the main structural material. The polonium adsorption enthalpy on stainless steel 316L in inert atmosphere was measured as -147±6 kJ mol-1. The findings obtained using reducing atmospheres show that the affinity of polonium for stainless steel 316L increases with the metallic character of the stainless steel surface. The presence of moisture in inert and reducing atmosphere does not significantly influence the polonium adsorption. Oxidizing atmospheres induce the formation of polonium dioxide which has a stronger interaction with the stainless ...
