Staphylococcus haemolyticus - Wikipedia
The ability to adhere to medical devices and subsequently form biofilms is a major virulence factor associated with S. haemolyticus.[3][5][13][14] Biofilm formation increases antibiotic resistance[5][13][14] and often leads to persistent infections.[15][16] S. haemolyticus biofilms are not polysaccharide intercellular adhesin (PIA) dependent, and the lack of the ica operon (the gene cluster that encodes the production of PIA) can be used to distinguish S. haemolyticus isolates from other CoNS species.[3][12][14] Biofilm formation is influenced by a variety of factors including carbohydrates, proteins, and extracellular DNA. Detachment assays with NaIO4, proteinase K, or DNase result in 38%, 98%, and 100% detachment, respectively. The high level of detachment associated with DNase treatment has led several authors to suggest a cell-to-surface and/or cell-to-cell adhesion function for extracellular DNA. Biofilm formation also appears to be influenced by the presence of glucose and NaCl. Biofilm ...
An electron microscopic study of clinical and laboratory-derived strains of teicoplanin-resistant Staphylococcus haemolyticus |...
Staphylococcal resistance to glycopeptides (which involves more teicoplanin than vancomycin) is uncommon and largely confined to Staphylococcus haemolyticus, an emerging nosocomial pathogen with a tendency to develop antibiotic resistance. In this study, six S. haemolyticus strains, including two isogenic pairs of teicoplanin-susceptible/-resistant strains and two resistant clinical isolates, were used in a morphologic and morphometric electron microscope investigation. Cells from both clinical and laboratory-derived teicoplanin-resistant strains exhibited abnormally roughened, irregular outlines when observed by transmission electron microscopy. However, no significant differences in cell wall thickness resulted from morphometric analysis when the susceptible/resistant cells of the two isogenic pairs were compared. By scanning electron microscopy, an abnormally roughened, blistered surface was associated with teicoplanin-resistant cocci. A certain variability was noted between strains, not ...
Antimicrobial metallic copper surfaces kill Staphylococcus haemolytic by Christophe Espirito Santo, Davide Quaranta et al.
Recently, copper (Cu) in its metallic form has regained interest for its antimicrobial properties. Use of metallic Cu surfaces in worldwide hospital trials resulted in remarkable reductions in surface contaminations. Yet, our understanding of why microbes are killed upon contact to the metal is still limited and different modes of action have been proposed. This knowledge, however, is crucial for sustained use of such surfaces in hospitals and other hygiene-sensitive areas. Here, we report on the molecular mechanisms by which the Gram-positive Staphylococcus haemolyticus is inactivated by metallic Cu. Staphylococcus haemolyticus was killed within minutes on Cu but not on stainless steel demonstrating the antimicrobial efficacy of metallic Cu. Inductively coupled plasma mass spectroscopy (ICP-MS) analysis and in vivo staining with Coppersensor-1 indicated that cells accumulated large amounts of Cu ions from metallic Cu surfaces contributing to lethal damage. Mutation rates of Cu- or steel-exposed cells

Serious proliferation of multiresistant Staphylococcus in intensive care units
Specialist physician Christina Agvald-Öhman studied the infection route of bacteria strains at a Swedish intensive care unit for her thesis. The results of her work show that the spread of infection between patients is surprisingly high. Between 70 and 80 per cent of the patients who were in care at the unit for three to five days were involved in the spread of infection. Infections are mainly spread via staff and equipment, which could be avoided if the hygiene rules were properly followed, says Dr Agvald-Öhman. By far the most important rule, which is easily forgotten, is to wash your hands properly after contact with each patient. The bacteria strain under study was a type of Staphylococcus normally found on the skin of healthy people, but which can easily establish itself in the respiratory passages of ill people; it is also one of the most common causes of hospital-related blood poisoning. Dr Agvald-Öhman believes that other types of bacteria are transmitted in a similar ...

AU - Archer, Gordon L.. PY - 1998/2. Y1 - 1998/2. N2 - mecA, the gene that mediates methicillin resistance, and its accompanying mec locus DNA, insert near the gyrA gene in Staphylococcus aureus. To investigate whether there is a similar relationship between mecA and gyrA in coagulase-negative staphylococci (CNS), mecA- and gyrA-specific DNA fragments were used to probe methicillin-resistant isolates of Staphylococcus epidermidis (MRSE) (n = 11) and Staphylococcus haemolyticus (MRSH) (n = 11). The gyrA probe hybridized to the same SmaI DNA fragment as the mecA probe in all strains tested. However, since the size of the SmaI fragments containing meca and gyrA varied from 73 to 600 kb, the distance between the two genes was determined more precisely. Cloned mecA or gyrA fragments plus vector sequences each ...

Isolation and characterisation of a 17-kDa staphylococcal heparin-binding protein with broad specificity
article{e1ea31f7-ded7-4c2d-b186-c463a49831f9, abstract = {A previous study reported the ability of staphylococci to bind heparin and heparin-dependent host growth factors. The present study isolated and identified heparin- and basic fibroblast growth factor (bFGF)-binding surface components of S. epidermidis strain RP12 and S. haemolyticus strain SM 131. The staphylococcal heparin-binding component(s) were purified by affinity chromatography on heparin-Sepharose and a major heparin-binding protein, here designated HBP, was identified by immunoblot in these two coagulase-negative staphylococcal (CNS) species. The HBP was shown to be acidic with an approximate pI of 4.6 and a molecular mass around 17 kDa. The binding of heparin to HBP was inhibited by heparin, fucoidan, pentosan polysulphate and various other sulphated polysaccharides, but not by non-sulphated compounds. However, the purified HBP from both S. epidermidis and S. haemolyticus revealed broad specificity, and also bound bFGF, ...

The discovery of a new species exhibiting more effective antibacterial properties is necessary because of the demand on Lavandula species, which continues to increase in a variety of industries. Lavandula pubescens might be a good alternative, as it exhibits strong antibacterial activity. In this study, the chemical composition of the essential oils from different organs (flowers, leaves, stems, and roots) of L. pubescens was identified using gas chromatography-mass spectrometry. Furthermore, the antimicrobial activities of different solvent extracts (methanol, ethanol, diethyl ether, hexane, and ethyl acetate) and different organ (flower, leaf, stem, and root) extracts of L. pubescens were evaluated. Only the ethyl acetate extracts of L. pubescens exhibited antibacterial activity against all bacterial strains tested, including Staphylococcus haemolyticus, Escherichia coli (KF 918342), Aeromonas hydrophila (KCTC 12487), E. coli (ATCC 35150), Cronobacter sakazakii (ATCC 29544), and Aeromonas salmonicida

Aim: This Swedish study determined which species of coagulase-negative staphylococci (CoNS) were found in neonatal blood cultures and whether they included Staphylococcus capitis clones with decreased susceptibility to vancomycin. Methods: CoNS isolates (n = 332) from neonatal blood cultures collected at orebro University Hospital during 1987-2014 were identified to species level with matrix-assisted laser desorption/ionisation time-of-flight mass spectrometry (MALDI-TOF MS). The antibiotic susceptibility pattern of S. capitis isolates was determined by the disc diffusion test and Etest, and the presence of heterogeneous glycopeptide-intermediate S. capitis (hGISC) was evaluated. Results: Staphylococcus epidermidis (67.4%), Staphylococcus haemolyticus (10.5%) and S. capitis (9.6%) were the most common CoNS species. Of the S. capitis isolates, 75% were methicillin-resistant and 44% were multidrug-resistant. No isolate showed decreased susceptibility to vancomycin, but at least 59% displayed the ...

PURPOSE: To evaluate the fluoroquinolone susceptibilities of ocular isolate coagulase-negative staphylococci (CoNS), identified at the Microbiology Laboratory - UNIFESP. DESIGN: Experimental laboratory investigation. METHODS: The minimum inhibitory concentrations (MICs) of 21 strains of methicillin-resistant coagulase-negative staphylococci (MRCoNS) and 22 methicillin-sensitive coagulase-negative staphylococci (MSCoNS) to ciprofloxacin, ofloxacin, gatifloxacin and moxifloxacin were determined, using the E-test method standardized by the Clinical and Laboratory Standards Institute (CLSI/NCCLS). RESULTS: The MIC90s (µg/ml) for the second generation of tested fluoroquinolones were higher than the fourth generation, especially for the methicillin-resistant coagulase-negative staphylococci group. CONCLUSION: Our results indicate that methicillin-sensitive coagulase-negative staphylococci are more susceptible to quinolones than are methicillin-resistant coagulase-negative staphylococci and that ...
Rapid colonization with methicillin-resistant coagulase-negative staphylococci after surgery - Zurich Open Repository and...
BACKGROUND: Antimicrobial resistance may compromise the efficacy of antibiotic prophylaxis before surgery. The aim of this study was to measure susceptibility and clonal distribution of coagulase-negative staphylococci (CoNS) colonizing the skin around the surgery access site before and after the procedure. METHODS: From March to September 2004, a series of 140 patients undergoing elective major abdominal surgery were screened for CoNS colonization at admission and 5 days after surgery. All isolates were tested for antibiotic susceptibility and genotyped by pulsed-field gel electrophoresis (PFGE). RESULTS: Colonization rates with CoNS at admission and after surgery were 85% and 55%, respectively. The methicillin-resistant CoNS rate increased from 20% at admission to 47% after surgery (P = 0.001). The PFGE pattern after surgery revealed more patients colonized with identical clones: 8/140 patients (8/119 strains) and 26/140 patients (26/77 strains), respectively (P , 0.001). CONCLUSIONS: Our ...
Genomic analysis of an emerging multiresistant Staphylococcus aureus strain rapidly spreading in cystic fibrosis patients...
Staphylococcus aureus is a major human pathogen responsible for a variety of nosocomial and community-acquired infections. Recent reports show that the prevalence of Methicillin-Resistant S. aureus (MRSA) infections in cystic fibrosis (CF) patients is increasing. In 2006 in Marseille, France, we have detected an atypical MRSA strain with a specific antibiotic susceptibility profile and a unique growth phenotype. Because of the clinical importance of the spread of such strain among CF patients we decided to sequence the genome of one representative isolate (strain CF-Marseille) to compare this to the published genome sequences. We also conducted a retrospective epidemiological analysis on all S. aureus isolated from 2002 to 2007 in CF patients from our institution. CF-Marseille is multidrug resistant, has a hetero-Glycopeptide-Intermediate resistance S. aureus phenotype, grows on Cepacia agar with intense orange pigmentation and has a thickened cell wall. Tomos is the Greek word for cut or section, and tomography is a technique for digitally cutting a specimen open using X-rays to reveal its interior details. The Panoramic x-ray is an example of one such slice. It is the simplest form of tomogram in medicine, because it does not attempt to show any volumetric, or 3-D information. It produces a two dimensional image of a single slice along the mandibular arch.. The x-ray beam is fan shaped, with the fan unfolded in the vertical dimension. This beam is projected through a vertical slit close to the patient as the machine slowly revolves around his or her head. The beam and the x-ray cassette revolve around the patients head in the same direction (clockwise when viewed from the top), while the cassette itself translates in the opposite direction. Patente US4038709 - Dual hydraulic hospital bed - Google Patentes
An improved dual hydraulic hospital bed having hydraulically actuated support means for controlling the movement of the bed frame. A dual mode hydraulic pump actuates the support means and controls the movement of additional hydraulic means to control the position of the bed frame and a support frame and thus the position of the upper body and legs of a patient. The dual mode hydraulic pump is driven by a foot petal or compressed air from a compressed air source, such as the compressed air wall outlets found in most hospital rooms. Preferably, the improved dual hydraulic hospital bed includes a bidirectional spring loaded cassette holder for positioning an X-ray cassette below any of the radiolucent panels affixed to the supporting frame to facilitate the taking of X-rays.
