Sporothrix schenckii is a fungus that can be found world-wide in the environment. The species is present in soil as well as in and on living and decomposing plant material such as peat moss. It can infect humans as well as animals and is the causative agent of sporotrichosis, commonly known as rose handlers disease. The most common route of infection is the introduction of spores to the body through a cut or puncture wound in the skin. Infection commonly occurs in otherwise healthy individuals but is rarely life-threatening and can be treated with antifungals. In the environment it is found growing as filamentous hyphae. In host tissue it is found as a yeast. The transition between the hyphal and yeast forms is temperature dependent making S. schenckii a thermally dimorphic fungus. Sporothrix schenckii sensu stricto and S. brasiliensis are usually associated to sporotrichosis, a subcutaneous mycosis worldwide distributed. Comparative analyses between these two species indicate they contain genetic and physiological differences that are likely to impact the interaction with host cells. Here, we study the composition of the cell wall from conidia, yeast-like cells and germlings of both species and found they contained the same sugar composition. The carbohydrate proportion in the S. schenckii sensu stricto wall was similar across the three cell morphologies, with exception in the chitin content, which was significantly different in the three morphologies. The cell wall from germlings showed lower rhamnose content and higher glucose levels than other cell morphologies. In S. brasiliensis, the wall sugars were constant in the three morphologies, but glucose was lower in yeast-like cells. A severe case of S. brasiliensis disseminated sporotrichosis in a young female HIV-positive patient from Rio de Janeiro State in Brazil is presented. The severity of the case is shown by the isolation of the strain S. brasiliensis from cultures of the skin, sputum, blood and central nervous system, and the probable fungal osteomyelitis present radiologically. Virulence studies in a mouse infection model have shown that S. brasiliensis is significantly more lethal and results in higher fungal burdens compared to S. schenckii as well as other examined Sporothrix spp. Recently this species was also associated to aggressive and atypical cases [10,19].. A total of 4860 animals were screened 2570(52.88%) were goats and 2290(47.12%) were sheep. The fungi associated with the infections were identified. Of the 2570 and 2290 of goats and sheep, 80(3.11%) and 144(6.29%) had fungal lesions respectively. Fifty soil samples from the environment were collected for fungal analysis and 31 nomads were checked for fungal lesions. Antifungal biogram and animal pathogenicity studies were also done. Prevalence of fungal infections was higher on the animals from farms than those at the markets. Infection was more prevalent in animals between 13-24 months of age. The glaborous skin was mostly affected (37.5%) in the goats, while in the sheep, the face was affected most (62.5%). Crude extracts and fractions of five species of Polygala - P. campestris, P. cyparissias, P. paniculata, P. pulchella and P. sabulosa - were investigated for their in vitro antifungal activity against opportunistic Candida species, Cryptococcus gattii and Sporothrix schenckii with bioautographic and microdilution assays. In the bioautographic assays, the major extracts were active against the fungi tested. A review of the clinical features of seven patients with sporotrichosis arthritis showed that six had joint infection without previous skin or lung involvement and that one with myelofibrosis had joint and skin infection. The average time from onset of joint symptoms to diagnosis was 25 months, resulting in joint damage that required arthrodesis in four patients. Tissue from open synovial biopsy was superior to synovial fluid for obtaining a positive culture; concomitant synovial fluid and synovial tissue cultures were superior to either one alone. Granulomatous inflammation was seen in synovial tissue in six patients biopsied. Amphotericin B with surgical debridement of the affected joint was successful treatment in four patients. Although an uncommon cause of joint disease, sporotrichosis arthritis may go unrecognized and mimic other forms of arthritis, resulting in irreparable damage in an otherwise curable form of arthritis. ...

To the editor: In their excellent article on septic bursitis in the prepatellar and olecranon bursae, Ho, Tice, and Kaplan (1) reported 24 culture-positive cases of septic bursitis, 22 with Staphylococcus aureus and two with group A, beta-hemolytic streptococci, and at least two culture-negative cases in patients taking antibiotics. This distribution is corroborated by their review of the literature and a subsequent report (2). The production of cellulolytic enzymes by the rot fungus Stereum sanguinolentum has been studied on different carbon sources. When powdered cellulose or carboxymethyl cellulose was used as the carbon source, high yields of extracellular cellulolytic enzymes were obtained, but there was no production of these enzymes with cellobiose as the carbon source. The heterogeneities in the protein structure of the enzymes were studied by the isoelectric focusing method. Also, the separation of two cellulase peaks from culture solutions of the rot fungus Stereum sanguinolentum has been demonstrated on a DEAE-Sephadex A-50 column. After dialysis, when the carbohydrate content of both the enzymes decreased to zero, the mixed enzymes gave one homogeneous cellulase peak upon rechromatography on a DEAE-Sephadex A-50 column as well as on column electrophoresis. The results strongly indicate that S. sanguinolentum only excretes one cellulase enzyme in a culture solution with powdered cellulose as the carbon source.

The taxonomic history of Ceratocystis, a genus in the Ceratocystidaceae, has been beset with questions and debate. This is due to many of the commonly used species recognition concepts (e.g., morphological and biological species concepts) providing different bases for interpretation of taxonomic boundaries. Species delineation in Ceratocystis primarily relied on genealogical concordance phylogenetic species recognition (GCPSR) using multiple standard molecular markers. Questions have arisen regarding the utility of these markers e.g., ITS, BT and TEF1-α due to evidence of intragenomic variation in the ITS, as well as genealogical incongruence, especially for isolates residing in a group referred to as the Latin-American clade (LAC) of the species. This study applied a phylogenomics approach to investigate the extent of phylogenetic incongruence in Ceratocystis. The patient stayed 12 days at the hospital, and was submitted to several radiological and laboratory tests. Computerized axial tomography and resonance magnetic imaging were normal. The enzyme-linked immunosorbent assay (ELISA) for HIV 1 and 2 was repeatedly negative. Other two samples of CSF were collected and cultures to bacteria and fungi were negative, slide latex agglutination (SLA) tests to cryptococcal antigen, Haemophilus influenza, Neisseria meningitidis, Streptococcus pneumoniae were negative. The patient was followed on consultations and five CSF examinations were done during a follow up period of a month, when the diagnosis was finally established. In the first CSF sample, a polymerase chain reaction (PCR) to Mycobacterium tuberculosis was negative. This article reviews updated treatment guidelines, which state that itraconazole has become the preferred therapy for most forms of infection. Amphotericin B remains the mainstay of treatment for severe cases, but lipid formulations are now preferred because of their more favorable toxicity profile. The repeatability and the operator-based reproducibility of the Real-Time PCR was demonstrated. Background: This is a retrospective, analytic observational study where we describe cases of sporotrichosis and mycetoma from Acapulco General Hospital and Community Dermatology Mexico C.A. over 25 years. Analysis of environmental features that favour the development of such diseases has been made, as well as the limitations in the study and treatment of such diseases in resource poor settings. Methods: We reviewed the information on 76 sporotrichosis and 113 mycetoma patients out of a total of 14,000 consultations at Acapulco General Hospital and from Community Dermatology clinics. We analysed the epidemiological and mycological characteristics and the investigations used for diagnosis such as direct examination, culture, intradermal test reactions, and biopsy. This guy is named Usin. I found him in front of a local super market last December. Hes a fighter, when I found him he was hungry, smelly and full of ring worms. After a couple of weeks, we also found out that he is infected with Sporotrichosis. Hes been on treatment for about 6 months.
