Both live and reactivated sea urchin sperm flagella were broken by passage through a pipette. Distal fragments stopped, but basal fragments continued to beat. Intact flagella were about 48 micrometer long; basal fragments were about 4-20 micrometer long. Beat periods of tritonated fragments were 11% shorter than those of unbroken controls, possibly reflecting decreased viscous loading; beat periods of live fragments were 18% shorter than those of unbroken controls, possibly reflecting reduced rates of ATP consumption as well as decreased viscous loading. The undulations of basal fragments were compared with those of the basal regions of unbroken flagella, using the patterns of development of the radii and angles of basal bends. Methods and results We conducted genetic analyses using whole-exome sequencing in 50 Han Chinese probands with MMAF. Two homozygous stop-gain variants (c.910C,T (p.Arg304*) and c.3400delA (p.Ile1134Serfs*13)) of the SPEF2 (sperm flagellar 2) gene were identified in two unrelated consanguineous families. Our goal was to determine the direction of force generation of the inner dynein arms in flagellar axonemes. We developed an efficient means of extracting the outer row of dynein arms in demembranated sperm tail axonemes, leaving the inner row of dynein arms structurally and functionally intact. Sperm tail axonemes depleted of outer arms beat at half the beat frequency of sperm tails with intact arms over a wide range of ATP concentrations. The isolated, outer arm-depleted axonemes were induced to undergo microtubule sliding in the presence of ATP and trypsin. I have run into what seems to be somewhat of a common problem however. I am getting some pretty noticeable hum. I only have two sources coming from my Decware input selector box into the amp. One is a Denon turntable and the other is an Oppo 105 BDP. When I remove the interconnects from the Oppo the hum is eliminated. All of my components are run through a low dollar surge protector which is going into a standard wall socket. I have tried switching plugs around. Moving the Oppo/Amp off of the surge protector individually and plugging them directly into the wall with no change in the hum (didnt really expect to make a difference). I dont have the budget to have a special circuit created just for this room or the buy any exotic type of power conditioner. Any suggestions as to a reasonable remedy would be welcomed. I have a Rachel amp and have read in the forum that their may be a difference in the grounding on these compared to other Decware amps. I am no engineer and dont know if this actually ...
