Phospholipid patterns of 15 representative strains of the genus Amycolatopsis were recorded by two-dimensional thin-layer chromatography. The structure analysis of the isolated phospholipids was verified by fast atom bombardment-mass spectroscopy. The positive- and negative-ion spectra of the partially purified phospholipid fractions qualitatively reflect their distinctive composition. All strains contained diphosphatidylglycerol, phosphatidylglycerol, and phosphatidylinositol. Two different types of phosphatidylethanolamine and phosphatidylmethylethanolamine were detected, viz., compounds with or without hydroxy fatty acids. These phospholipid patterns underline the integrity of the genus. Fast atom bombardment-mass spectrometry analysis of phospholipid patterns may serve as an aid for differentiation of bacterial species.

Both one- and two-dimensional NMR techniques were applied. 13C NMR sub-spectra of the CHn (n=0...3) groups, obtained by the Distortionless Enhancement by Polarisation Transfer (DEPT) technique, were used for the elucidation of the qualitative and quantitative composition of humic and fulvic acids in the soils. The chemical structure of SOM was further analysed at the molecular level through Fast Atom Bombardment Mass Spectrometry (FABMS) and Gas Chromatography-Mass Spectrometry (GC/MS). A novel glutathione-conjugated metabolite of morphine has been isolated from the bile of guinea pigs given morphine. The metabolite was separated by preparative HPLC on a reverse phase column (YMC-GEL C18) using methanol/water (1:4, v/v) as eluate and purified by HPLC on another reverse phase column (mu-Bondapak phenyl) using water/acetonitrile/trimethylamine/acetic acid (150:3:2:1, v/v) as a mobile phase. As a simple model of ultraviolet (UV)-induced protein denaturation, Amyloid P hexapeptide (APH, Phe-Thr-Leu-Cys-Phe-Arg) was exposed to UVA irradiation. A new product was isolated by HPLC and identified by fast atom bombardment mass spectrometry (FAB-MS) as the disulfide dimer. Thus, UVA induced disulfide bond formation. We also examined the effect of replacing the Cys residue of APH with other amino acid residues (methionine, tryptophan, tyrosine and histidine) that might be susceptible to oxidative damage. In the case of [Trp4]-APH, the amount of the analog was decresed at 40 J/cm2 or higher. When APH and [Trp4]-APH were mixed and UVA-irradiated, the combination afforded a greater number of products than the sum of those obtained when they were separately irradiated. A radioimmunoassay, immunohistochemical techniques, and high pressure liquid chromatography (HPLC) methods have been developed for the study of the pentapeptide proctolin in the lobster Homarus americanus. Proctolin-like immunoreactivity is present in nearly every portion of the lobster nervous system; immunoreactivity is found in the brain, in each of the ganglia and connectives of the ventral nerve cord, and in many of the nerve roots that emerge from the cord. The greatest amounts are found in the pericardial organs, which are well known neurosecretory structures, and these structures have been selected for more detailed study. The immunoreactive material in the pericardial organs appears to be authentic proctolin. This material co-migrates with synthetic proctolin in two HPLC systems. The amino acid sequence of human alpha-fetoprotein, a 67-kDa protein present in mammalian embryonic serum, was verified by fast atom bombardment mass spectrometric (FAB/MS) analyses of three different enzymatic digests of the protein. Human alpha-fetoprotein obtained from a large-scale cell culture was digested with trypsin and V-8 protease either separately on two different samples or combined on the same one. The V-8 protease digest of the protein was partially fractionated by HPLC; the other samples were directly analyzed by FAB/MS without previous purification steps. About 90% of the alpha-fetoprotein amino acid sequence was verified by mass spectrometric analysis; this also confirmed that the cell-derived protein is identical with the hepatoma-derived protein. FAB analysis revealed that the N terminus of the mature protein is arginine rather than threonine, with the threonine occupying the second position. This dissertation explores innovations in index-based risk transfer products (IBRTPs) as a means to address an important insurance market failure that leaves many poor and vulnerable populations exposed to considerable uninsured risk. IBRTPs can address problems of covariate risk, asymmetric information and high transaction costs that have precluded the emergence of formal insurance market in low-income areas, where uninsured risk remains a leading cause of persistent poverty. A brief introductory chapter situates this dissertation in the broader, emergent literature on IBRTPs. The second chapter explains how the strong relation between widespread human suffering and weather shocks creates an opportunity to develop famine indexed weather derivatives to finance improved emergency response to humanitarian crises.
