TY - JOUR. T1 - Comparative effects of somatomedin C and insulin on the metabolism and growth of cultured human fibroblasts. AU - Conover, Cheryl A.. AU - Hintz, Raymond L.. AU - Rosenfeld, Ron G.. PY - 1985/1. Y1 - 1985/1. N2 - At concentrations of 25 ng/ml in serum‐free medium, somatomedin C (SM‐C) and insulin stimulated 3H‐thymidine incorporation in adult human fibroblasts 4‐ and 1.5‐fold, respectively. The presence of 0.25% human hypopituitary serum (HHS), which by itself had little effect, enhanced the mitogenicity of both SM‐C and insulin. Furthermore, 10−7 M dexamethasone dramatically potentiated SM‐C stimulation (70‐fold) and insulin stimulation (28‐fold) of 3H‐thymidine incorporation. With dexamethasone and 0.25% HHS, significant stimulation of DNA synthesis was seen at 2.5 ng/ml for both SM‐C and insulin. The effects of SM‐C and insulin on 3H‐thymidine incorporation were additive. These 3H‐thymidine incorporation results were clearly supported by cell ...

Loss of muscle bulk happens early in the course of CHF (15)and the development of cachexia is an adverse prognostic feature (16). The origin of the weight loss is not clear, but recent studies have drawn attention to the potential importance of neurohormonal and immune activation. Tumor necrosis factor is elevated in CHF (5,6); there is an increase in the ratio between circulating levels of catabolic and anabolic steroids (3), and an insulin-resistant state develops (17,18). The resting metabolic rate of patients with heart failure is increased (19)and resting leg oxygen consumption has been shown to be increased (8).. Growth hormone is secreted by the anterior pituitary, and mediates its major metabolic effects through activation of somatomedins, predominantly IGF-I (20,21). Levels of GH can be greatly elevated in untreated heart failure (9)and we have found elevated levels in patients with cardiac cachexia (4). Paradoxically, perhaps, there is some evidence for a beneficial effect in patients ...

Two types of receptors for insulin-like growth factors (IGFs) were characterized in glioma cell lines established from different human brain tumors of glial origin (astrocytoma grades III and IV) by competitive binding assay, affinity labeling, and protein phosphorylation. Type I IGF receptor is a heterodimer composed of α-subunits (Mr 130,000), which bind IGF I and II with equal affinity, and of β-subunits (Mr 98,000), which show tyrosine kinase activity and autophosphorylation stimulated by IGF I and II with equal potency. The type II IGF binding site is a monomer (Mr 250,000) which binds IGF II with 10 times higher affinity than IGF I. The cellular concentration of type II IGF binding site is about 2- to 5-fold higher than the amount of type I IGF receptor. The characteristics of the two types of IGF receptors in human glioma cell lines are similar to those described recently in fetal rat astrocytes. In contrast the type I IGF receptor in glioma cells is different from that studied ...
Background: The maternal insulin-like growth factor (IGF) system is considered to be involved in fetal growth regulation. However, available data linking this system to fetal growth are contradictory and incomplete. Aims: To measure components of the IGF system before, during and after pregnancy in healthy women and to relate these results, and their changes during pregnancy, to fetal weight (gestational week 31) and birth weight. Methods: Serum concentrations of IGF-I, IGF-II, IGF-binding protein (IGFBP)-1, IGFBP-3 and IGFBP-3 protease activity were assessed in 23 women before conception, at weeks 8, 14, 20, 32 and 35 of pregnancy and 2 weeks postpartum. The data were analyzed using simple and multiple linear regression. Results: One third of the variability in fetal weight was explained by IGF-I in combination with IGFBP-3 protease activity, both assessed at gestational week 32 (p = 0.013). Birth weight was negatively correlated (r = -0.43 to -0.59) with IGFBP-1 at gestational week 20 (p = 0

Normal tissue growth involves a complex interaction of cell proliferation, hypertrophy, migration, differentiation and senescence which proceeds within strict temporal and anatomical constraints. While classical endocrine hormones such as growth hormone and thyroxine can influence the overall rate of growth and maturation they do not contribute to the fundamental intercellular signalling within tissues that underlies these processes. This appears to be the primary role of peptide growth factors. During the rapid development of the embryo and fetus peptide growth factors are associated with tissue induction, the clonal growth of stem-cell populations, and the appearance of differentiated function in the presumptive organs. In post-natal life the actions of growth factors are less obvious although notable exceptions demonstrate that the same biological pathways exist; the best studied examples are the role of somatomedins during longitudinal skeletal growth and the coordinated actions of several growth
Caloric restriction (CR) inhibits tumorigenesis in rodents. To understand the basis for this effect the binding of insulin, insulin-like growth factor I/somatomedin C (IGF-I/Sm-C), insulin-like growth factor II/multiplication stimulating activity (IGF-II/MSA), and epidermal growth factor were examined to membrane preparations of 7,12-dimethylbenz(a)anthracene-induced mammary adenocarcinomas and several normal tissues from female Sprague-Dawley rats. Animals were fed ad libitum (AL) or 25% and 40% calorically restricted diets. Large, palpable (LP) and small, ≤ 100 mg, nonpalpable (SNP) tumors were evaluated. Growth factor binding to tumors was differentially affected by CR. IGF-I/Sm-C binding was comparable for AL-LP, AL-SNP, and 25% CR-LP tumors, but elevated in 25% CR-SNP tumors. Scatchard analysis revealed high and low affinity IGF-I/Sm-C binding sites, with AL-SNP and 25% CR-SNP tumors exhibiting similar levels of high affinity sites and at a greater concentration than AL-LP and 25% CR-LP ...
Insulin-like Growth Factor II (IGF-II) analogues in which at least one of R37 and R38 is replaced with another amino add residue, the most preferred being IGF-II R37Q R38Q, can readily be produced in E. coli, unlike natural IGF-II, which is cleaved on secretion. The analogues retain activity on the type I and type II IGF receptors but have lower affinity for the insulin receptor; they are therefore more specific in their action.

Insulin-like growth factor 1 (IGF-1), also called somatomedin C, is a protein that in humans is encoded by the IGF1 gene.IGF-1 has also been referred to as a sulfation factor and its effects were termed nonsuppressible insulin-like activity (NSILA) in the 1970s. IGF-1 is a hormone similar in molecular structure to insulin. It plays an important role in childhood growth and continues to have anabolic effects in adults. A synthetic analog of IGF-1, mecasermin, is used for the treatment of growth failure. IGF-1 consists of 70 amino acids in a single chain with three intramolecular disulfide bridges. IGF-1 has a molecular weight of 7,649 Daltons ...
Studies have been conducted on fibromyalgia patients that have monitored their brain, hormonal, and metabolic activity. In these studies, the patients monitored activity have shown a rather large number of abnormalities. When brain scans of patients diagnosed with fibromyalgia the tests revealed that the patients had reduced blood flow to specific regions of their brains.. The specific regions of the brain were those that were in control of pain sensation to the body. The brain system known as the hypothalamus-pituitary-adrenal gland axis, a system that controls some very important functions, including depression, growth, stress response, and sleep, is particularly interesting to researchers of fibromyalgia.. A research target that has seen lots of attention lately is the somatomedin C hormone, also known as the insulin-like growth factor in laymans terms. This hormone is produced by the pituitary gland in the human brain while the body is in deep sleep. Its primary responsibility is to ...

The insulin‐like growth factor (IGF)‐system includes insulin‐like growth factors I and II (IGF‐I and IGF‐II) along with the type I (IGF‐1R) and type II (IGF2R) cell‐surface receptors, the insulin receptor (IR) and circulating IGF‐binding proteins (IGFBPs) (Denley et al, 2005). The biological actions of the IGFs are mediated by IGF‐1R and IR, leading to cell growth, differentiation and survival. Their distribution and activity is controlled via high‐affinity association with IGFBPs, and the binding sites on IGFs for IGFBPs have been delineated in detail by structural studies (Headey et al, 2004; Carrick et al, 2005; Sitar et al, 2006). In mammals, the activity of IGF‐II (but not IGF‐I) is further moderated by IGF2R, which sequesters IGF‐II for internalization and degradation. IGF2R is classed as a growth inhibitor, with loss of function causing increased growth (Foulstone et al, 2005). In line with this, Igf2r is a putative tumour suppressor gene and mutations have been ...

Abstract: The Insulin-like growth factor-1 (IGF-1) system is dynamic and complex, involving many binding proteins, binding-proteinrelated proteases, and receptors. It has emerged in time as a powerful defence to life processes of many cytotypes, tissues and systems. Mainly in body metabolism, diabetes and cardiovascular system, but also in brain and kidney, IGF-1 plays a key role in maintaining homeostasis, increasing progenitor cell potential, and improving physiologic performance both in rest and stress conditions. Its vasculoprotective and insulin ...
7.1. Insulin-like growth factors (IGF-I & IGF-II). Of the growth factors, those with the most potent effects on growing skeletal tissue are the IGFs, previously known as somatomedins. IGFs are synthesised in the liver and circulate bound to carrier proteins (Froesch et al., 1985). The major factors regulating IGF concentrations in serum are growth hormone, nutritional intake and thyroid hormones, the latter being necessary for growth hormone secretion. The traditional view was that growth hormone acted indirectly on the growth plate via IGF-I, a potent mitogen for growth plate chondrocytes. However, there is increasing evidence that growth hormone has direct effects on the growth plate (Tripper et al., 1989). (This close relationship between circulating IGFs and growth hormone will be discussed in more detail in the chapter covering the hormonal regulation of growth.). In addition to having effects on the growth plate chondrocytes, locally synthesised and circulating IGFs retained in bone matrix ...

Protein, glucose, and calcium metabolism were studied using stable labeled tracer infusions of L-[1-13C]leucine, [6,6-2H2]glucose, and 42Ca and 44Ca; substrate oxidation rates were assessed using indirect calorimetry; muscle strength was determined by isokinetic and isometric dynamometry of the anterior quadriceps, as well as growth factors, hormones, glucose, and lipid concentrations in plasma before and after 8 weeks of rhIGF-I (60 microg/kg, sc, twice daily), followed by 4 weeks of washout, then 8 weeks ofrhGH (12.5 microg/kg-day, sc); the ...
Purpose: We demonstrated that IGFBP-3 stimulates hematopoietic stem cells (HSC) to differentiate into endothelial cells, form capillaries, and stabilize the vasculature (Chang, et al, PNAS 2007). Local IGFBP- 3 production is increased by hypoxia and facilitates the homing of HSC to areas of injury. In the circulation, IGFBP-3 is bound to HDL. In this study, we investigated the signaling pathways responsible for the robust migratory effects of IGFBP-3.. Methods: The effects of IGFBP-3 on NO generation in human vascular precursors (CD 34+, CD14−), human lung microvascular endothelial cells, and human umbilical vein endothelial cells were examined using DAF-FM fluorescence. Western analysis was use for detection of eNOS and vasodilator-stimulated phosphoprotein (VASP), which redistributes to lamellipodia forming an active motor complex that supports motility and is phosphorylated in response to NO. Localization of VASP was performed by immunohistochemistry. SK-1 was assessed following IGFBP-3 ...

There are several limitations in this study. First, to reduce cost, we used a candidate gene approach and subjectively selected 25 SNPs from 7 IGF axis genes for study. The coverage and information of our SNP list highly depended on the data from the HapMap project. Furthermore, other types of genetic polymorphisms, such as copy number variants, 90 may be responsible for the underlying association with AMD risk. Therefore, false-negative findings are possible. On the other hand, because of the small sample size for geographic atrophy (Table 5), false-positive findings are also possible. In addition, we used Bonferronis correction for our hypothesis testing on the 25 SNPs. Furthermore, multiple tests were performed for each SNP. This may also increase the likelihood of false-positive findings. Although rs2872060 is implicated in AMD risk, no study has related it to major systemic diseases or mortality. However, polymorphisms in IGF axis genes are related to risk for some diseases and aging, and ...

Ligand binding properties of five single amino acid substituted variants (V11A, D12A, Q15A, Q15E, and F16A) of human insulin-like growth factor I (IGF-I) were analyzed with respect to their binding affinities and binding kinetics to recombinant IGF binding protein 1 (IGFBP-1) and a soluble form of the IGF type I receptor (sIGF-I(R)), respectively. Side chains of the substituted residues are all predicted to be the most surface exposed in the alpha-helical portion of the B-region of the IGF-I molecule. The IGF-I variants were produced as fusion proteins to a IgG(Fc) binding protein domain, Z. Ligand binding kinetic rates were determined using BIAcore biosensor interaction analysis technology. All IGF-I variants showed altered binding affinities to both IGFBP- I and sIGF-I(R). Secondary structure content of the IGF-I variants was estimated using far-UV circular dichroism spectroscopy, followed by variable selection secondary structure calculations. The amount of calculated alpha-helicity is ...

Author(s): Adams, Gregory R | Abstract: Similar to bone, skeletal muscle responds and adapts to changes in loading state via mechanisms that appear to be intrinsic to the muscle. One of the mechanisms modulating skeletal muscle adaptation it thought to involve the autocrine and/or paracrine production of insulinlike growth factor-I. This brief review outlines components of the insulinlike growth factor-I system as it relates to skeletal muscle and provides the rationale for the theory that insulinlike growth factor-I is involved with muscle adaptation.
Objective To retrospectively validate the WINROP (weight, insulinlike growth factor I, neonatal, retinopathy of prematurity [ROP]) algorithm in a Brazilian population. WINROP aims to predict ROP and is based on longitudinal weight measurements from birth until postmenstrual age 36 weeks. WINROP has predicted 100% of severe ROP in 3 neonatal intensive care unit settings in the United States and Sweden.. Methods In children admitted to the neonatal intensive care unit at Hospital de Clínicas de Porto Alegre, Porto Alegre, Brazil, from April 2002 to October 2008, weight measurements had been recorded once a week for children screened for ROP, 366 of whom had a gestational age of 32 weeks or less. The participating children had a median gestational age of 30 weeks (range, 24-32 weeks) at birth and their median birth weight was 1215 g (range, 505-2000 g).. Results For 192 of 366 children (53%), no alarm or low-risk alarm after postmenstrual age 32 weeks occurred. Of these, 190 of 192 did not develop ...

The interactions of insulin-like growth factors (IGFs) with the type I IGF receptor are modulated by a family of high-affinity IGF binding proteins (IGFBPs). One of these, IGFBP2, demonstrates a striking spatiotemporal relationship with IGF-I during cerebellar and retinal development. IGF-I mRNA is transiently expressed in large projection neurons--cerebellar Purkinje and retinal ganglion cells--while IGFBP2 mRNA is selectively expressed by contiguous neuroglia--Bergmann glia in the cerebellum and Muller cells and astrocytes of the nerve fiber layer in the retina. IGF-I and IGFBP2 gene expression is not only neuroanatomically coordinated but also temporally synchronized, peaking together during the postnatal maturation of these structures. This pattern of IGF system expression suggests that IGFBP2 is closely related to IGF-Is action in the developing nervous system.. ...

Evolutionarily conserved pathways are emerging as the underlying mechanistic explanation for the relationships observed between maintenance of genomic integrity, control of repair and cell death, and cancer and ageing. From work in C. elegans, the central role of the insulin/insulin‐like growth factor 1 (IGF1) pathway has become apparent. Long‐lived daf‐2 (insulin/IGF1 receptor) mutants counteract tumour growth and promote longevity in C. elegans by triggering apoptosis through the Forkhead box, class O (FOXO) transcription factor DAF‐16; their murine and human homologues are also known to influence tumorigenesis. The transcriptional targets of DAF‐16 were recently found to be involved in extending longevity and include many that influence p53‐dependent apoptosis (Pinkston‐Gosse & Kenyon, 2007). A loss of p53 results in increased cancer incidence in mice and humans; mice that constitutively overexpress p53 are cancer resistant but short lived (Tyner et al, 2002). A large proportion ...
IDDM2-encoded predisposition to type 1 diabetes has recently been mapped to the minisatellite or variable number of tandem repeat (VNTR) locus upstream of the insulin and insulin-like growth factor II genes on human chromosome 11p15.5. In a UK case-control study (n=228 sporadic diabetics; n=441 healthy controls), we show here that the genotype homozygous for VNTR class I alleles is predisposing to disease (RR=2.68), and VNTR class III alleles are dominantly protective (RR=0.37). In 722 diabetic families from the UK (n=356), USA (n=173), Denmark (n=55) and Sardinia (n=138), we have analysed the transmission of class I alleles to diabetic offspring from class I/III heterozygous parents. We confirm that in families from the USA, class I alleles are transmitted preferentially from fathers. However, in family data sets from the UK, Denmark and Sardinia, the reverse is true and maternal transmission is stronger. Furthermore, in the UK family data set, the difference between maternal and paternal ...
