Previous reports showed that the Steroidal Glycoalkaloid Solamargine inhibited proliferation of non-melanoma skin cancer cells. However, Solamargine was not tested systematically on different types of melanoma cells and was not simultaneously tested on normal cells either. In this study we aimed to investigate the effect of Solamargine and the mechanism involved in inhibiting the growth of different types of melanoma cells. Solamargine effect was tested on normal cells and on another three melanoma cell lines. Vertical growth phase metastatic and primary melanoma cell lines WM239 and WM115, respectively and the radial growth phase benign melanoma cells WM35 were used. The half inhibitory concentration IC50 of Solamargine was determined using Alamarblue assay. The cellular and subcellular changes were assessed using light and Transmission Electron Microscope, respectively. The percentage of cells undergoing apoptosis and necrosis were measured using Flow cytometry. The different protein expression was
Solanum aculeatissimum Jacq. () is a perennial undershrub (30 to 90 cm in height) and tropical America is its place of origin. It grows wild throughout the world, and is used to treat bronchitis and rheumatism in China.. Three alkaloids, solanine (), solasonine (), and β-solamardine () have been isolated from the acid methanolic extracts of the fruits (). The fruit, Kikania lei (Hawaii), is cooked and eaten. Solanum aculeatissimum also contains two new steroidal glycosides, aculeatisides A () and B () in the roots (). The yields of solasodine and aculeatiside B are surprisingly high (3.8 and 3.0% dry weight, respectively). Solasodine (), an aglycon of solasonine and β-solamargine and nuatigenin (), an aglycon of aculeatiside A and B are potentially useful precursors for the manufacture of steroidal hormones and pharmaceuticals, since nuatigenin and solasodine can be converted to the pregnane derivatives () as well as diosgenin (). Thus the potential utilization of the sapogenin nuatigenin has ...
In this work, confocal Raman imaging was used to study the formation of lipid droplets (LDs) in vitro in a single endothelial cell upon incubation with polyunsaturated fatty acids (10 or 25 μM) including arachidonic acid (AA) and its deuterated analog (AA-d8), eicosapentaenoic acid (EPA) and docosahexaenoic
Plant-derived BEC with its main component solamargine possesses anticancer activities via its effect on a variety of biological pathways in a wide range of human cancer cells. High cure rates with BEC therapy have been obtained in animals with deadly cancers, and, in humans with terminal cancers promising results have been reported. At a clinical level, optimal concentrations of BEC have been established in a topical cream formulation Curaderm, for effective removal of skin cancers, but optimal concentrations of BEC have not been reported for other cancers. The objective of this study was to determine whether combination therapy of Cisplatin with BEC would result in synergism using cure rates as end points. BEC on its own cures Sarcoma 180 in mice, whereas, Cisplatin on its own has no effect on Sarcoma 180 activity. A combination of BEC and Cisplatin shows synergism, resulting in higher cure rates than BEC and Cisplatin at comparable individual concentrations.
Skin cancer that is non-melanoma can be easily and more importantly effectively be treated with BEC5 which is also branded as Curaderm. BEC5 Curaderm cream is particularly powerful when spread over Basal cell carcinomas. Consisting of a specially purified plant extract means BEC5 Curaderm is successful on Basal cell carcinomas ( BCC ) or Squamous cell carcinomas ( SCC ) which are the most widespread forms of skin cancer. Curaderm cream applications are also effective on benign tumours such as age spots, Keratoses, sun spots and Keratocanthmoas. Taken from Solanum Sodomaeum this is a plant that is discovered in Australasia. This plant includes an important extract - Solasodine Glycosides. This is an active ingredient that is also discovered in smaller quantities in the eggplant and aubergine salosodine Glycosides and has been established as being very effective for treating cancerous skin conditions. As a result it is incorporated into BEC5 curaderm cream. BEC5 curaderm cream is a renowned sun ...

Background: Wnt11 is involved in many developmental and cellular processes. We have previously reported that Wnt11 overexpressing (MSCWnt11) significantly improved cardiac function and increased angiogenesis. Here, we aimed to elucidate how Wnt11 delivered by MSC increase blood vessel formation.. Methods and Results: An acute myocardial infarction (MI) model in SD rats was developed by ligation of the left anterior descending (LAD) coronary artery. Conditioned medium (CdM) collected from MSCWnt11 (CdMWnt11) was injected immediately into the ischemic border area. Rats treated with CdMWnt11 showed a significantly improved cardiac function. Furthermore, CdMWnt11 increased blood vessel density and regional blood flow in ischemic myocardium. Fluorescent immunostaining showed that both vWF-positive single endothelial cell numbers and microvessel numbers were significantly increased in CdMWnt11 treated myocardium. In vitro studies showed that CdMWnt11 significantly increased number and length of ...

Upon entering our digestive tract, anti-nutrients irritate the intestinal tract resulting in intestinal permeability - in other words, wheat (breads, pasta, pizza, cereal) containing gliadin and glutenin, potatoes containing glycoalkaloids and saponins, and legumes containing saponins and lectins - all of those foods irritate the lining of the gastrointestinal tract, breaking down the the natural barrier of the intestinal wall. This breakdown of the intestinal wall is sometimes referred to as leaky gut syndrome. Intestinal permeability is associated with chronic low-grade inflammation (resulting in diverticulitis), inappropriate immune responses (allergies), and autoimmune diseases (such as celiac disease, inflammatory bowel disease (IBS), rheumatoid arthritis, Type 1 diabetes, and Multiple Sclerosis).. For the record, sweet potatoes and yams do not contain saponins or glycoalkaloids.. ...
Steroidal glycoalkaloids (SGAs) are plant secondary metabolites known to be toxic to animals and humans and that have putative roles in defense against pests. The proposed mechanisms of SGA toxicity are sterol-mediated disruption of membranes and inhibition of cholinesterase activity in neurons. It has been suggested that phytopathogenic microorganisms can overcome SGA toxicity by enzymatic deglycosylation of SGAs. Here, we have explored SGA-mediated toxicity toward the invasive oomycete Phytophthora infestans, the causative agent of the late blight disease in potato and tomato, as well as the potential for SGA deglycosylation by this species. Our growth studies indicate that solanidine, the nonglycosylated precursor of the potato SGAs a-chaconine and a-solanine, has a greater physiological impact than its glycosylated forms. All of these compounds were incorporated into the mycelium, but only solanidine could strongly inhibit the growth of P. infestans in liquid culture. Genes encoding several ...

Steroidal glycoalkaloids (SGAs) are toxic specialized metabolites that are found in the Solanaceae. Potato (Solanum tuberosum) contains the SGAs α-solanine and α-chaconine, while tomato (Solanum lycopersicum) contains α-tomatine, all of which are biosynthesized from cholesterol. However, although two cytochrome P450 monooxygenases that catalyze the 22- and 26-hydroxylation of cholesterol have been identified, the 16-hydroxylase remains unknown. Feeding with deuterium-labeled cholesterol indicated that the 16α- and 16β-hydrogen atoms of cholesterol were eliminated to form α-solanine and α-chaconine in potato, while only the 16α-hydrogen atom was eliminated in α-tomatine biosynthesis, suggesting that a single oxidation at C-16 takes place during tomato SGA biosynthesis while a two-step oxidation occurs in potato ...
Citation: N/A Interpretive Summary: Technical Abstract: Colorado potato beetle (CPB) is a destructive pest of the cultivated potato. Certain glycoalkaloids in potato leaves are effective deterrents to this insect; however, in tubers these compounds can be toxic to humans. Leptines are foliar specific glycoalkaloids produced by the related species, Solanum chacoense. We are studying the inheritance of leptine production in segregating F-1 and F-2 populations derived from two S. chacoense accessions, 55-1 and 55-3, which are (respectively) high and low leptine producers. The F-1 segregates 1:1 for high (,70% of total glycoalkaloids) and low (,20% of TGA) leptine content. Segregation data from the F-1 and F-2 populations suggest a two-gene model for leptine production: a dominant repressor and a recessive inducer. Using two bulked DNA samples composed of high and low-leptine individuals from the F-1 population, we are using various molecular markers (RAPDs, SSRs, DS-PCR, and AFLPs) to search for ...

The mean duration of intravenous therapy use was 8.81 days in continuous and intermittent infusion (61.5%), in 20G catheter (39.3%), inserted in the dorsal hand vein arc (36.9 %), with mean time of usage of 68.4 hours. The type of infusion (p=0.044) and the presence of chronic disease (p=0.005) and infection (p=0.007) affected the development of phlebitis.. CONCLUSION: There was a high frequency of phlebitis in the sample, being influenced by concomitant use of continuous and intermittent infusion of drugs and solutions, and more frequent in patients with chronic diseases and ...
We report, for the first time, the remarkable efficacy of uttroside B, a potent saponin from Solanum nigrum Linn, against liver cancer. The compound has been isolated and characterized from the leaves of Solanum nigrum Linn, a plant widely used in traditional medicine and is a rich resource of several anticancer molecules. Uttroside B, that comprises of β-D-glucopyranosyl unit at C-26 of the furostanol and β-lycotetraosyl unit at C-3, is ten times more cytotoxic to the liver cancer cell line, HepG2 (IC50: 0.5 μM) than sorafenib (IC50: 5.8 μM), the only FDA-approved drug for liver cancer. Moreover, it induces cytotoxicity in all liver cancer cell lines, irrespective of their HBV status, while being non-toxic to normal immortalized hepatocytes. It induces apoptosis in HepG2 cells by down-regulating mainly the activation of MAPK and mTOR pathways. The drastic reduction in HepG2-xenograft tumor size achieved by uttroside B in NOD-SCID mice and substantiation of its biological safety through both acute

Belladonna is a perennial herb that is native to Europe and Asia Minor, but is now grown quite often in the United States, Europe, and India. While growing in the wild, which Belladonna commonly does, a slight dose can be fatal. The nickname "deadly nightshade" is a good clue of its potency. When Belladonna was first used it was for cosmetic purposes. Women felt that if they used it to dilate their pupils, they would look more sexy and alluring. This explains the name Belladonna, which means "beautiful lady" in Italian ...
Background: Advanced glycation end products (AGEs) signal through the receptor for AGE (RAGE), which can lead to hepatic fibrosis in hyperglycemia and hyperlipidemia. We investigated the inhibitory effect of aqueous extracts from Solanum nigrum (AESN) on AGEs-induced RAGE signaling and activation of hepatic stellate cells (HSCs) and hyperglycemia induced by high-fat diet with ethanol. Methods: An animal model was used to evaluate the anti-hepatic fibrosis activity of AESN in rats fed a high-fat diet (HFD; 30%) with ethanol (10%). Male Wistar rats (4 weeks of age) were randomly divided into four groups (n = 6): (1) control (basal diet); (2) HFD (30%) + ethanol (10%) (HFD/ethanol); (3) HFD/ethanol + AESN (100 mg/kg, oral administration); and (4) HFD/ethanol + pioglitazone (10 mg/kg, oral administration) and treated with HFD for 6 months in the presence or absence of 10% ethanol in dietary water. Results: We found that AESN improved insulin resistance and hyperinsulinemia, and downregulated lipogenesis via

An alkaloid, originally from Atropa belladonna, but found in other plants, mainly SOLANACEAE. Hyoscyamine is the 3(S)-endo isomer of atropine. Atropine, a naturally occurring belladonna alkaloid, is a racemic mixture of equal parts of d- and l-hyoscyamine, whose activity is due almost entirely to the levo isomer of the drug. Atropine is commonly classified as an anticholinergic or antiparasympathetic (parasympatholytic) drug. More precisely, however, it is termed an antimuscarinic agent since it antagonizes the muscarine-like actions of acetylcholine and other choline esters. Adequate doses of atropine abolish various types of reflex vagal cardiac slowing or asystole. The drug also prevents or abolishes bradycardia or asystole produced by injection of choline esters, anticholinesterase agents or other parasympathomimetic drugs, and cardiac arrest produced by stimulation of the vagus. Atropine may also lessen the degree of partial heart block when vagal activity is an etiologic factor. Atropine ...

Often some of the most deadly substances were once used for cosmetic purposes-that is, before their lethal qualities became obvious. An example of such commercial carelessness was thallium being marketed as a depilatory agent. It effectively removed unwanted hair, but it had a rather severe side effect of killing the person in the process. Belladonna,…
