In the present study, we used phage display to develop an engineered antibody domain, called SD1, recognizing an epitope at the C-terminus of mesothelin. This epitope does not overlap with any previously developed mesothelin therapeutic antibodies in preclinical or clinical development. SD1-hFc shows strong CDC and ADCC activity against mesothelin-expressing cancer cells in vitro and causes growth inhibition of human tumor xenografts in nude mice in vivo. Our results suggest that the SD1 single-domain antibody represents a new class of antimesothelin mAbs and is a promising therapeutic candidate for novel mesothelin-targeted therapy.. We designed a vector for the bacterial expression of recombinant antibodies fused to a double tag composed of 6xHis and the EPEA amino acid sequence. EPEA sequence (C tag) is tightly bound by a commercial antibody when expressed at the C-term end of a polypeptide. The antigen is released in the presence of 2 M MgCl2. Consequently, constructs fused to the 6xHis-C tags can be purified by two successive and orthogonal affinity steps. Single-domain antibodies were produced either in the periplasmic or in the cytoplasmic space of E. coli. Herein, we report the creation of a library of novel nanobody capture ligands to detect p24, one of the earliest markers of HIV infection. We demonstrate that these nanobodies, one tenth the size of conventional antibodies, exhibit high sensitivity and broad specificity to global HIV-1 subtypes. Biophysical characterization indicates strong 690 pM binding constants and fast kinetic on-rates, 1 to 2 orders of magnitude better than monoclonal antibody comparators. A crystal structure of the lead nanobody and p24 was obtained and used alongside molecular dynamics simulations to elucidate the molecular basis of these enhanced performance characteristics. Hello, I have a lot of problems with the preview in adobe premiere pro and quicktime when it comes to color grading. These problems I experienced on different computer running Mac os x and different monitors calibrated or not. The issue is that the footage (h.264 / AVCHD) no matter if it´s from a 5d or a sony a7s looks alway more saturated and more punchy in the preview of premiere then after exporting and opening in quicktime.
