British Library EThOS: Physiological studies on the growth and development of the carrot, Daucus carota L.
This thesis is concerned with a study of the external, environmental conditions on the growth and development of the carrot Daucus carota L., as well as the carotene content, morphogenetic and other effects of defoliation or extirpation of the apex. Auxanometer records of root growth in different environments were also obtained. Experiments have also been carried out to simulate environmental and other effects by means of external application of known plant growth substances, in an attempt to explain morphogenetic changes in terms of internal plant regulation. Briefly, the results of daylength/temperature experiments on tops and roots have indicated that maximum total plant growth results at relatively warm temperatures, but under these conditions the leaf fraction is favoured; lone-day treatment results in similar effects. Root and top temperatures had striking differential effects. Both can be simulated by gibberellic acid application which is not dependent on the site of treatment. At lower ...

A fusion protein linking a truncated form of diphtheria toxin (DT(388)) to human interleukin-3 (DT(388)IL3) kills malignant progenitors from some patients with acute myeloid leukemia (AML) while sparing normal progenitors. This study evaluated two variants of DT(388)IL3 with increased affinity for the IL-3 receptor (IL-3R) for their cytotoxicity to AML progenitors and determined the ability of quantitative reverse transcription-PCR assessment of expression of the IL-3R subunits to predict the effectiveness of wild-type DT(388)IL3 and its variants. Both the IL-3 deletion variant (Delta125-133) and the amino acid substitution variant (K116W) showed enhanced toxicity against AML colony-forming cells (AML-CFC; but not normal CFC) compared with wild-type DT(388)IL3 with the K116W variant achieving ,90% AML-CFC kill with 17 of 23 patient samples. This variant was also more effective against AML cells engrafting in nonobese diabetic severe combined immunodeficient mice. There was a significant ...

Effects of Bioactive Compounds from Carrots (Daucus carota L.), Polyacetylenes, Beta-Carotene and Lutein on Human Lymphoid...
New therapies for leukaemia are urgently needed. Carrots have been suggested as a potential treatment for leukaemia in traditional medicine and have previously been studied in other contexts as potential sources of anticancer agents. Indicating that carrots may contain bioactive compounds, which may show potential in leukaemia therapies. This study investigated the effects of five fractions from carrot juice extract (CJE) on human lymphoid leukaemia cell lines, together with five purified bioactive compounds found in Daucus carota L, including: three polyacetylenes (falcarinol, falcarindiol and falcarindiol-3-acetate) and two carotenoids (beta-carotene and lutein). Their effects on induction of apoptosis using Annexin V/PI and Caspase 3 activity assays analysed via flow cytometry and inhibition of cellular proliferation using Cell Titer Glo assay and cell cycle analysis were investigated. Treatment of all three lymphoid leukaemia cell lines with the fraction from carrot extracts which contained ...

Development of oral pharmaceutical formulation of standardized crude ethanolic extract of Atractylodes lancea (Thunb) DC.
Cholangiocarcinoma (CCA), the adenocarcinoma of the biliary duct, is commonly reported in Asia with the highest incidence in northeastern Thailand. Chemotherapy of this type of cancer is limited due to the lack of effective chemotherapeutic drugs. A series of previous studies support further research and development of Atractylodes lancea (Thunb) DC. (AL) as a potential candidate for the treatment of CCA as a crude ethanolic extract. In the present study, we aimed to develop an oral pharmaceutical formulation (capsule) of the standardized AL crude ethanolic extract for further clinical development in patients with CCA. Major steps included macroscopic and microscopic authentication of the AL rhizomes, preparation of standardized AL extract, preparation of oral pharmaceutical formulation (capsule) of the standardized AL extract, quantitative and qualitative analysis of the marker compound (atractylodin) in the formulated AL extract, evaluation of contaminations of heavy metals, pesticides ...
Variation for Salinity Tolerance During Seed Germination in Diverse Carrot [Daucus carota (L.)] Germplasm in: HortScience...
Salinity stress is considered one of the most important abiotic factors that limits the productivity of crop plants (Flowers and Yeo, 1995), and the estimated global cost due to salinity is more than $12 billion annually (Qadir et al., 2008). Annually, ≈10 million hectares of land are becoming salinized to a point where the land can no longer sustain adequate crop production. This is due to the extensive use of irrigation and high rates of evapotranspiration, which result in increased salt accumulating in the soil (Rozema and Flowers, 2008). Most crops, including cultivated carrot (Daucus carota var. sativus), are categorized as glycophytic plants. The growth of glycophytes is greatly reduced in saline soils because they lack physiological mechanisms such as salt glands and bladders that allow halophytes, which are salt-loving plants, to thrive in high salinity (Flowers et al., 2010). One approach to combating the negative effects of salinity stress in glycophytic crops is identifying new ...
Influence of some thermal treatments on carotenoids content of carrots (Daucus carota L.)-Diana Alexandra Cosoreci, Mirela ...
Through this work we followed the influence of some thermal treatments on the carotenoids content of carrots. Carrots were analyzed fresh and after blanching, boiling, freezing raw and freezing blanching prior. Carotenoids content of the samples was determined spectrophotometrically. As raw material were used carrots from the domestic market, Flam variety. The highest carotenoid content was recorded for raw carrot (82.716 mg / g). By boiling for 20 minutes, the content of carotenoids in carrot remains only 28% (23.209 mg /g). Frozen carrot also lose the contents of carotenoids, leaving only 33.8% (27.961 mg /g). The lowest content of carotenoids(20.196 g/g) was found in carrots blanched and then frozen for 2 months at -18˚C ...

Thirty two male albino rats weighing 110±10 g were divided into four groups, the first group received the basal diet only and served as (negative control), the second group received the hypercholesterolemic diet and served as positive control, the other groups received hypercholesterolemic diet supplemented with 10%, 20% CaPP for six weeks. The obtained results revealed that groups supplemented with 10% and 20% CaPP significantly decrease total lipid, total cholesterol, triglycerides, low density lipoprotein cholesterol, liver enzymes: alanine aminotransferase, aspartate aminotransferase compared to positive and negative groups. Organs weight, body weight gain significantly decreased compared with positive control. Moreover dietary carrot pomace powder can used to reduce the body ...
Temporal and spatial expression of terpene synthase genes associated with agarwood formation in Aquilaria malaccensis Lam | New...
The diseased wood, agarwood, from the tropical tree taxa Aquilaria, is famed for its unique fragrance and medicinal values, mainly due to its richness in secondary metabolites such as the sesquiterpenes. The presence of sesquiterpenes in high numbers and amounts correlates with agarwood of high quality. To understand the synthesis of this important compound, we cloned two candidate genes in the terpenoid synthesis pathway from Aquilaria malaccensis Lam., a major agarwood tree species. The genes encoding sesquiterpene synthase (AmSesTPS1) and δ-guaiene synthase (AmGuaiS1) were successfully cloned from callus RNA, using specific primers derived from transcriptomic data, in a reverse transcription PCR reaction. The full-length complementary DNA (cDNA) sequence of AmSesTPS1 was 1632 bp encoding for 544 amino acids, and AmGuaiS1 was 1644 bp encoding for 547 amino acids. Sequence alignment analysis showed that AmSesTPS1 shared between 99 to 100 % identity with sesquiterpene synthase from Aquilaria sinensis

Cox Inhibitors as Potential Chemotherapic Drugs for Mesothelioma: Ingenta Connect
Malignant mesothelioma (MM) is an uncommon neoplasm that arises from the cells lining the body cavities, in particular the pleural and peritoneal cavities. The treatment of MM is a major challenge with frustrating results for clinicians and patients alike. Despite the adoption of newly developed radiotherapic and chemotherapic regimens, the prognosis remains dismal and only modest improvements have been obtained thus far. In this scenario, a better comprehension of the molecular patterns is of paramount importance. Cyclooxygenases catalyze the rate limiting step in the production of prostanoids. Accumulating data demonstrate that overexpression of these enzymes, and in particular of cyclooxygenases- 2, promotes multiple events involved in tumorigenesis; in addition, numerous studies show that inhibition of cyclooxygenases- 2 can delay or prevent certain forms of cancer. Aim of this review is to discuss the current state of the art in mesothelioma, with a particular emphasis on the recent ...
