Seeds of Arabidopsis contain ~40% of triacylglycerol. It is converted to sugar to support post germination growth. We identified an Arabidopsis T-DNA knockout mutant that was sugar dependent during early seedling establishment. Our study showed that the β-oxidation process involved in catabolising the free fatty acids released from the seed triacylglycerol was impaired in this mutant. This mutant was confirmed to be transcriptional null for the Protein Acyl Transferase 15, AtPAT15 (At5g04270), one of the 24 protein acyl transferases in Arabidopsis. Although it is the shortest AtPAT15 contains the signature Asp-His-His-Cys cysteine rich domain which is essential for the enzyme activity of this family of proteins. The function of AtPAT15 was validated because it rescued the growth defect of the yeast protein acyl transferase mutant akr1 and it was also auto-acylated in vitro. Transient expression of AtPAT15 in Arabidopsis and tobacco localized AtPAT15 in the Golgi apparatus. Taken together, our ...

The Oryza sativa L. F8 population derived from a cross between salt tolerance cv. Ahlemi Tarom and salt sensitive cv. Neda was used in the study. Germinated seeds floated on water for 3 d, and after were transferred to float on Yoshidas nutrient solution for 11 d. two weeks after sowing, the seedling was transferred to nutrient solution containing 51.19 mM NaCl (electrical conductivity 6 dSm-1) for 7 d, then NaCl concentration was increased to 163.8 mM (12 dSm-1) for further 7 d. After this period, the traits were measured. The linkage map was performed using F8 populations, 40 SSR markers, 16 ISSR markers (76 alleles), 2 IRAP markers (7 alleles) and iPBS marker (3 alleles). The map length was 1419 cM with an average distance of 13.07 cM between the 2 adjacent markers. The QTL analysis showed that a total of 73 QTLs were identified that controlled 20 traits under normal and stress conditions. Among the QTLs, qCHLN-8, qSLN-8, qWLN-3, qWLN-9, qLAN-3, qLAN-8 and qLAN-9, qRFWN-1, qRFWN-3b and qRFWN-8, qFBN

Rice crop is sensitive to cold stress at seedling stage. A panel of population representing 304 shortlisted germplasm lines was studied for seedling stage chilling tolerance in indica rice. Six phenotypic classes were exposed to six low temperature stress regimes under control phenotyping facility to investigate response pattern. A panel of sixty six genotypes representing all phenotypic classes was used for ensuring genetic diversity, population structure and association mapping for the trait using 58 simple sequence repeat (SSR) and 2 direct trait linked markers. A moderate level of genetic diversity was detected in the panel population for the trait. Deviation of Hardy-Weinbergs expectation was detected in the studied population using Wrights F statistic. The panel showed 30% variation among population and 70% among individuals. The entire population was categorized into three sub-populations through STRUCTURE analysis. This revealed tolerance for the trait had a common primary ancestor for each
I was recently re-reading the ca. 4th-century BC collection of advice and admonitions attributed to the Chinese philosopher Mencius. When I came across the following famous passage, parenthood made me see it in a new light:. You must not be like the man from Sung. There was a man from Sung who pulled at his seedlings because he was worried about their failure to grow. Having done so, he went on his way home, not realizing what he had done. I am worn out today, said he to his family. I have been helping the seedlings to grow. His son rushed out to take a look and there the seedlings were, all shrivelled up. There are few in the world who can resist the urge to help their seedlings grow. There are some who leave the seedlings unattended, thinking that nothing they can do will be of any use. They are the people who do not even bother to weed. There are others who help the seedlings grow. They are the people who pull at them. Not only do they fail to help them but they do the seedlings positive ...

A genetic program that in sunflower seeds is activated by Heat Shock transcription Factor A9 (HaHSFA9) has been analyzed in transgenic tobacco seedlings. The ectopic overexpression of the HSFA9 program protected photosynthetic membranes, which resisted extreme dehydration and oxidative stress conditions. In contrast, heat acclimation of seedlings induced thermotolerance but not resistance to the harsh stress conditions employed. The HSFA9 program was found to include the expression of plastidial small Heat Shock Proteins that accumulate only at lower abundance in heat-stressed vegetative organs. Photosystem II (PSII) maximum quantum yield was higher for transgenic seedlings than for non-transgenic seedlings, after either stress treatment. Furthermore, protection of both PSII and Photosystem I (PSI) membrane protein complexes was observed in the transgenic seedlings, leading to their survival after the stress treatments. It was also shown that the plastidial D1 protein, a labile component of the PSII
This year, because of rising input costs, more plug growers are focusing efforts on getting maximum yields from every plug tray. Buying good seed, sowing it accurately and providing good germination conditions are just half the battle. There is a considerable difference between germination and usable seedlings. Losing seedlings after germination is finished costs a lot more money! If you are having problems consistently with losing seedlings of certain crops, then you are doing something consistently wrong. However, if you lose seedlings every once in a while and dont understand why, then you need to look at the influences of your growing environment and culture on your techniques.. Plug growers around the country lose seedlings after germination for a number of reasons, including tip or leaf burn, tip abortion and stunted seedlings. These problems dont show up all the time, but they do rear their ugly heads every year. Understanding which crops are sensitive to these problems, when they are ...
Salt stress is one of the major abiotic stress which severely limits plant growth and reduces crop productivity across the world. In the present study, the effects of exogenous pyridoxal-5-phosphate (vitamin B6, VB6) on seedling growth and development of wheat under salt stress were investigated. The results showed that exogenous application of pyridoxal-5-phosphate (VB6) significantly increased the RWC, biomass, the concentration of photosynthetic pigments, proline, the activities of superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), together with decreasing the content of Malondiadehyde (MDA) and hydrogen peroxide (H2O2) in wheat leaves under salt stress. Meanwhile, the transcript level of P5CR, P5CS, SOD, TaSOS1 and TaSOS4 were also up-regulated after treatment with pyridoxal-5-phosphate. VB6 acts as a signal in regulating the activities of plant antioxidant enzymes and SOS pathway to improve resistance to salt stress. The current study results may give an insight into the ...

Environmental stresses frequently affect plant growth and development, and many genes have been found to be induced by unfavorable environmental conditions. Here, we reported the biological functions of TaNAC2D, a stress-related NAC (NAM, ATAF, and CUC) gene from wheat. TaNAC2D showed transcriptional activator activity in yeast. TaNAC2D-GFP fusion protein was localized in the nucleus of wheat mesophyll protoplasts. TaNAC2D transcript abundance was significantly induced by NaCl, PEG6000, and abscisic acid (ABA) at seedling stage, and repressed by NaCl and PEG6000 at mature plant stage. When TaNAC2D was introduced into Arabidopsis, the 35-day-old soil-grown TaNAC2D-overexpression (TaNAC2D-OX) plants displayed slower stomatal closure, higher water loss rate, and more sensitivity to salt and drought stresses compared with WT plants. In contrast, TaNAC2D-OX seedlings, grown on 1/2 MS medium supplemented with different concentrations of NaCl, Mannitol, and MV, had enhanced tolerances to salt, osmotic and

Light signals are essential environmental cues for seedlings to elaborate their developmental program during the transition from heterotrophy to autotrophy. The effects of light on seedling development can be observed at many levels, including gene expression, cell differentiation, and plant morphology. During seedling development, higher plants, such as Arabidopsis, can adopt two contrasting development schemes: skotomorphogenesis, which takes place in the dark, and photomorphogenesis, which occurs in the light (Kendrick and Kronenberg, 1994). Dark-grown seedlings undergoing skotomorphogenesis exhibit etiolated morphologies, including long hypocotyls, apical hooks, and closed and undeveloped cotyledons. The genes required for chloroplast development and photosynthesis are expressed at low or undetectable levels. When exposed to light, seedlings undergo photomorphogenesis and exhibit deetiolated morphologies, such as short hypocotyls and open and developed cotyledons. Expression of genes ...

Rhynchosporium secalis can overcome a single resistance gene of barley in a relatively short period of time. Novel genes and quantitative trait locis (QTLs) are therefore vital to control scald in barley. A population of 220 double haploid lines was developed from a cross of Vlamingh and WABAR2147, where Vlamingh showed adult plant resistance (APR) and WABAR2147 showed seedling resistance to a group of isolates. The population was tested for APR to scald under natural infection in two consecutive seasons in addition to a seedling screen with three isolates. One single gene was mapped to chromosome 6H based on the seedling test, and two QTLs (QSc.VlWa.4H and QSc.VlWa.6H) were mapped to chromosomes 4H and 6H based on APR. Epistatic interaction was observed between the two QTLs, and environment/QTL interaction was only observed for QSc.VlWa.6H which co-segregated with the seedling resistance gene and contributed to basal resistance against scald during whole growth stages. QSc.VlWa.4H explained ...
1. Alpine and Arctic treeline expansion depends on establishment of tree seedlings beyond the current treeline, which is expected to occur with climate warming. However, treelines often fail to respond to higher temperatures, and it is therefore likely that other environmental factors are important for seedling establishment.. 2. We aimed to analyse our current understanding of how temperature and a range of other environmental drivers affect tree seedling establishment at the alpine and Arctic treelines world-wide and to assess the relative importance of temperature compared with other factors and how they interact.. 3. We collected 366 observations from 76 experimental and observational papers for a qualitative analysis of the role of a wide range of environmental factors on tree seed germination, tree seedling growth, survival and natural occurrence. For a subset of these studies, where the experimental design allowed, we conducted formal meta-analyses to reveal if there were global drivers ...
During the younger stages of a plants growth it is called a seedling.. Once a seed sprouts and its roots take hold, it will grow into a baby plant. Up until the moment that it needs transplanting we refer to it as a seedling.. View more information about seedlings and the seedling stage here:. ...
Weed plants need light throughout their life cycle, but during the seedling stage not really so much. And if you wonder how much light is ideal, we recommend to err on the side of caution. Its hardly the best option to keep your cannabis seedlings under HPS grow lights. Use weaker lights, instead, like CFLs, T5s, or LED lights. This will not only allow you to save on electricity, but spare your sprouts from shock. And if you insist on using HPS or other high intensity discharge (HID) lamps, raise them high, probably as high as your grow room allows. Still, under light of this type, seedlings can show signs of stress, slow down their growth, and demonstrate various worrying symptoms. In this case, though it may seem counterintuitive to you, youll see better results if you temporarily keep your weed in the window - until its mature enough to handle strong light.. Another question is when to put weed seedling under light. Some growers do it even before their sprouts emerge from the medium. ...
We can measure the useful light to a plant (PAR - photosynthetically active radiation, or the parts of light that the plant can use to photosynthesize) in micromoles per meter square per second (micromoles/sec-m2) . The goal here is to deliver 120-150 micromoles/sec-m2 of PAR to your seedlings.. Some people will use a PAR meter to figure out how much PAR theyre getting to their seedlings, and will adjust the distance of their lights based on that measurement. Dont worry- you dont need one for this if youre using LED bars or fluorescent bars. Just remember the following distances.. LED bars (like Philips): 8-12 inches away from seedlings.. T5 fluorescents: 5-6 inches away from seedlings.. ...

No matter what medium is used for starting seeds, it should drain well and have a fairly low pH. Mixes with a pH of 6.4 or less are not as susceptible to damping off as those with a higher pH.. To prevent damping off, tap water should not be used on seedlings. Tap water can be alkaline in many cases and it will gradually increase the pH in the planting medium. Instead, use distilled water. If you know that your water is alkaline, you may also add a tablespoon of white vinegar to a gallon of filtered tap water and use that for watering your seedlings.. The planting medium should be kept moist but should never be soggy. Damping off fungi thrive in conditions that are too wet. Ideally, the medium should be watered from the bottom, rather than the top. To water from the bottom, simply place the flats or pots of seedlings in a shallow container of water until the planting medium has absorbed enough water to moisten it. Do not allow the flats or pots to remain in the water until the medium becomes ...

Not really. That would encourage the plants to get leggy. Long spindly stems. The same happens when seedlings are put in a window; they try to stretch up to the sun. Called heliotrophic. They also bend out toward the light. Florescent lights are cooler with full spectrum light and you put the bulbs within an inch of the top of the plant. It does not burn the leaves and makes the seedling stems compact and strong. A cheap shop light will do the trick so no need to buy an expensive lighting system. As the plants grow make it so the top of the plant is about an inch below the bulb. Mine are in a basement so I raise the light. But lowering the plant does the same thing ...

The protocol has four sub-protocols, which are about the measurement of malondialdehyde, chlorophyll proline, soluble sugar, and glutathione content, respectively, in Arabidopsis seedling by using spectrophotometer. These methods are simple, effective and reproducible, which will help the researchers who are not familiar with these approaches, quickly get reliable results.
I am very worried for my seedlings. The seedlings are way too long and leggy. The conditions are the same as last year - except that I started the seeds in March, and here it is already May. So, I set up two grow lights and put a heating pad under the tray. I pulled out the really leggy seedlings that look seriously doomed and replanted -- fingers crossed ...
Leggy seedlings need more light. Place your source of artificial light only about 2" from the tops of the plants and they will become stout and hardy instead of thin and spindly. You can deeply plant your tomatoes but the cabbages should be replanted at the same soil level as they were growing in the container. More light will help the plants stop reaching up and continuing to grow leggy. Both basil and parsley should be thinned so there is an inch between the little plants. Start out by clipping off the obviously overcrowded plants, saving the most robust specimens. Then thin them to within a quarter of an inch; a week or so later thin them to within a half inch, and by the time they have developed 2 sets of leaves you should thin them to about an inch apart. At this time they can be divided and transplanted into larger pots. Handle the seedlings by the leaves, not the stems - if a leaf is crushed the plant can regrow it but it the stem is crushed, the plant will die. Seedlings dont mind being ...

Nows the time to get your seedlings going. Heres how with a quick n easy way thatll turn household trash into garden treasure: When you reach the end of a roll of foil, dont throw the cardboard tube away. Instead, turn it into a bunch of seed-starting pots. Just cut the tube into pieces about 3 inches long, and wrap aluminum foil around the outside of each piece to keep the cardboard from falling apart when it gets wet. Pack the little pots closely together on a waterproof tray or shallow pan, add seed-starting mix, and sow seeds to your hearts content. Then come transplant time, remove the foil, and plant your seedlings, pots and all. Now youre seedlings will be rarin for spring!. While were at it, lets talk about getting a jump start on spring skin care. If March came in like a lion in your neck of the woods, then you know those bone-chilling winds sure can take a toll. So heres a simple facial formula that will slough off dead skin cells, refine pores, and even out your skin tone ...

I have since started some new seedlings and am learning more each time. One seemingly obvious point i have picked up, but really neglected in my first lot - is some advice I found online. One gentleman said that he finds watering the seedlings from below in the tray rather than spraying them from top nearly always works so much better. So I have placed an egg carton inside a shallow metal tray, (you can use the lid of the egg carton, but mine had holes) and filled that with water. I also put a small hole in the bottom of the egg carton sections before putting the soil in so the water could go directly in. The egg carton itself is cardboard and will act as a wick soaking up the water. I then just loosely placed plastic wrap over the top. Already I can see the difference in two days. The soil has not yet dried out, the carton is soaked. Before the soil was drying out in a matter of hours. I am also having some success with my heirloom beetroot and cucumber, each with a new seedling appearing. I ...

The Lincoln Highway was the first paved highway in the United States to run coast-to-coast. It was the first of a myriad of named highways that served the motoring public in the early days of interstate travel. When the Lincoln Highway was being built, most of the road was paved with gravel or macadam, if it was paved at all. In much of Iowa, the Lincoln Highway was a route of a wheel-sucking gumbo of mud after a rainfall. The Portland Cement Association members and Lincoln Highway Association provided states with enough paving material to build one mile of demonstration highway, known as "Seedling Miles" that were out in the country, to show motorists what a good road could be like. Building the Seedling Miles out in the country was a stoke of genius, because a family might climb into the car for a Sunday drive, with father determined to see the Seedling Mile. The trip there, over bad roads, made a brilliant contrast to the one mile of concrete paving, adding another voice of support to the ...
