Often prior protein separation of complex protein mixtures by 2D-PAGE is needed, requiring more time and expertise than instructors of large laboratory classes can devote. We have developed an experimental module for our Biochemistry Laboratory course that engages students in MS-based protein identification following protein separation by one-dimensional SDS-PAGE, a technique that is usually taught in this type of course. The module is based on soybean seed storage proteins, a relatively simple mixture of proteins present in high levels in the seed, allowing the identification of the main protein bands by MS/MS and in some cases, even by peptide mass fingerprinting. Maize kernels do not contain enough of the essential sulphur-amino acid methionine (Met) to serve as a complete diet for animals, even though maize has the genetic capacity to store Met in kernels. Prior studies indicated that the availability of the sulphur (S)-amino acids may limit their incorporation into seed storage proteins. Serine acetyltransferase (SAT) is a key control point for S-assimilation leading to Cys and Met biosynthesis, and SAT overexpression is known to enhance S-assimilation without negative impact on plant growth. Therefore, we overexpressed Arabidopsis thaliana AtSAT1 in maize under control of the leaf bundle sheath cell-specific rbcS1 promoter to determine the impact on seed storage protein expression. The transgenic events exhibited up to 12-fold higher SAT activity without negative impact on growth. Storage parenchyma cells of developing legume cotyledons actively transport large amounts of storage proteins to protein storage vacuoles (PSV). These proteins are synthesized on the endoplasmic reticulum and pass through the Golgi apparatus. Clathrin coated vesicles (CCV) and small electron dense vesicles found near the trans-Golgi network (TGN) have both been implicated in the Golgi-to-vacuole transport step. Recent findings that protein storage cells contain more than one type of vacuole have necessitated a re-examination of the role of both types of vesicles in vacuolar protein transport. Immunoblots of highly purified CCV preparations and immunogold labelling with antibodies to the storage proteins vicilin and legumin, indicate that the dense vesicles, but not the CCV, are involved in storage protein transport in pea cotyledons. During seed development, the lytic vacuole (LV) is replaced by a protein storage vacuole (PSV) which specializes in accumulating seed storage proteins (SSPs). As seed protein reserves are mobilized upon germination, the PSV is once again replaced by the LV which takes on different roles in vegetative tissues. Cellular events occurring during these developmental transitions are not well understood, particularly, the transition between vacuole types. This research investigates whether PSVs can exist in leaves. To study vacuole transitions in leaves, an Arabidopsis thaliana line over-expressing the LEAFY COTYLEDON2 (LEC2) transcription factor was used. LEC2 is a master regulator of embryogenesis responsible for creating a cellular environment that promotes embryogenic development. Over-expression of LEC2 causes vegetative tissues to change their developmental fate to an embryonic state. A partial cDNA clone encoding Lupinus angustifolius Nodulin-45 was isolated by differential hybridisation. A genomic clone was also isolated, from which the DNA sequence was obtained for the 5′ end of the gene (including 1.2 kb of 5′ upstream region). The upstream region includes putative cis-elements, found upstream of other nodulin genes. Southern analysis indicates the presence of several Nodulin-45-like sequences in the lupin genome. The Nodulin-45 protein has a putative N-terminal endoplasmic reticulum-type signal sequence and also contains a large glycine-rich repeat sequence. The cDNA sequence is highly homologous to a Nodulin-45 cDNA sequence from Lupinus luteus (Szczyglowski et al., Plant Sci., 65 (1989) 87-95), although major sequence rearrangements are apparent between the L. luteus and L. angustifolius cDNAs.. ...

Duplication-Dependent CG Suppression of the Seed Storage Protein Genes of Maize | Genetics
Our results show that during a short evolutionary time span, individual α-zein genes have accumulated large variations in CG content. In particular, within the 19- and 22-kD zein gene families, extensively duplicated genes are more CG depleted compared to less duplicated genes. In addition, the 19-kD zein genes are more suppressed than the 22-kD zein genes, indicating that the former have undergone a greater number of gene duplications. This is supported by the fact that the 19-kD genes are more abundant compared to the 22-kD genes (Hagen and Rubenstein 1981; Wilson and Larkins 1984; Heindecker and Messing 1986).. A series of genetically related lines of common bean (Phaseolus vulgaris L.) integrate a progressive deficiency in major storage proteins, the 7S globulin phaseolin and lectins. SARC1 integrates a lectin-like protein, arcelin-1 from a wild common bean accession. Two pathways for the transport of integral membrane proteins appear to exist in protoplasts of suspension-cultured tobacco cells, as Jiang and Rogers (1998) have recently demonstrated by using chimeric integral membrane reporter proteins. Whereas γ-TIP and BP-80 seemed to share the same vesicular pathway to the prevacuolar compartment of these cells, α-TIP did not colocalize in the same transport compartments. Instead, after leaving the ER, α-TIP apparently reached an undefined compartment, bypassing the Golgi apparatus. In contrast to these results, α-TIP does indeed pass through the Golgi apparatus of developing pea cotyledons. Membrane anchorage was tested as a strategy to accumulate recombinant proteins in transgenic plants. Transmembrane domains of different lengths and topology were fused to the cytosolic HIV antigen p24, to promote endoplasmic reticulum (ER) residence or traffic to distal compartments of the secretory pathway in transgenic tobacco. Fusions to a domain of the maize seed storage protein γ-zein were also expressed, as a reference strategy that leads to very high stability via the formation of large polymers in the ER lumen. Although all the membrane anchored constructs were less stable compared to the zein fusions, residence at the ER membrane either as a type I fusion (where the p24 sequence is luminal) or a tail-anchored fusion (where the p24 sequence is cytosolic) resulted in much higher stability than delivery to the plasma membrane or intermediate traffic compartments. The E2F transcription factors and the RETINOBLASTOMA RELATED (RBR) repressor protein are principal regulators coordinating cell proliferation with differentiation, but their role during seed development is little understood. We show that in the fully developed embryos, cell number was not affected either in single or double mutants for the activator-type E2FA and E2FB Accordingly, these E2Fs are only partially required for the expression of cell cycle genes. In contrast, the expression of key seed maturation genes; LEAFY COTYLEDON 1-2 (LEC1-2), ABSCISIC ACID INSENSITIVE 3 (ABI3), FUSCA 3 (FUS3) and WRINKLED 1 (WRI1) are upregulated in the e2fab double mutant embryo. Phenotypes of five transgenic lines of narrow-leafed lupin (Lupinus angustifolius [L] cv Merrit) stably transformed with the isopentenyl pyrophosphate transferase (ipt) gene from Agrobacterium tumefaciens coupled to a flower-specific promoter (TP12) from Nicotiana tabacum [L.] are described. Expression of the transgene was detected in floral tissues and in shoot apical meristems on all orders of inflorescence. In each transgenic line there was significant axillary bud outgrowth at all nodes on the main stem with pronounced branch development from the more basal nodes in three of the lines. The lowest basal branches developed in a manner similar to the upper stem axillary branches on cv Merrit and bore fruits, which, in two lines, contained a significant yield of filled seeds at maturity. Abstract: In this study total 9 taxa belong to four different sections (Pratensis, Orobon, Orobus and Platystylis) collected from different regions of Turkey have been studied for the analysis of seed storage protein profiles to examine their relationship by sodium dodecyl sulphate polyacrilamide gel electrophoresis (SDS-PAGE) technique. Data obtained from electrophoresis of seed proteins were analysed using hierarchical clustering analysis and Euclidean distance was used for calculating the genetic resemblance. Dendogram was formed using average linkage. Electrophoretic protein profiles of seed cotyledons were showed that all taxa except for L. brachypterus var. brachypterus formed two clusters. 87 were evaluated for analysis of variability in seed storage proteins by using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Electrophorogram for each variety were scored and presence or absence of each band noted and was entered in a binary data matrix. Based on the data of SDS-PAGE gels cluster analysis was performed to check the variations among varieties. The overall result shows low degree of heterogeneity however different varieties reveal differential protein banding pattern. In those seeds there are two big protein fractions - globulins and albumins. The two globulin fractions are called Legumin - the major fraction and Vicilin - the minor one. There are not big differences between globulins and vicilins present in several leguminous plants. They are respectfully, very similar.
