One of the most extreme and fascinating examples of naturally occurring mutagenesis is represented by circular permutation. Circular permutations involve the linking of two chain ends and cleavage at another site. Here we report the first description of the folding mechanism of a naturally occurring circularly permuted protein, a PDZ domain from the green alga Scenedesmus obliquus. Data reveal that the folding of the permuted protein is characterized by the presence of a low energy off-pathway kinetic trap. This finding contrasts with what was previously observed for canonical PDZ domains that, although displaying a similar primary structure when structurally re-aligned, fold via an on-pathway productive intermediate. Although circular permutation of PDZ domains may be necessary for a correct orientation of their functional sites in multi-domain protein scaffolds, such structural rearrangement may compromise their folding pathway. This study aimed to identify and determine the carotenoids from green microalga, Scenedesmus protuberans using analytical techniques. Identification of carotenoids was realized by comparing their absorption and mass spectral data with those of reference standards available and reported values. Chromatographic data were then combined with the spectroscopic information. The separation of carotenoids was achieved by C30 column and high-performance liquid chromatography-diode array detection was used for their determination. In the present work, the carotenoid content of S. protuberans was found to be 1.45 mg/g of violaxanthin, 2.47 mg/g of all-trans-lutein, 0.15 mg/g of all-trans-α-carotene, 0.55 mg/g of all-trans-β-carotene, and 0.20 mg/g of 9 or 9′-cis-β-carotene. The ability to defend against grazers is a main driving force in phytoplankton evolutionary histories. This leads to trade-offs with other life-history traits, modifying eco-evolutionary dynamics. One well-known example is the chlorophyte Scenedesmus sp., which forms colonies when being grazed by the predator Daphnia. I see that this discussion is several months old, but Id be interested in knowing if you ever solved the mystery. Ive been using pre-reduced indigo, Thiox and Soda Ash. Ive also experienced the whitening/bleaching out at the fold lines. I tried halving my recipe while keeping the amount of water the same, thus diluting its strength. The vat became the nice yellowy-green its supposed to be. Not much of a flower, though. Ive made several vats this way, dyed yards of 100% Robert Kaufman pfd cotton, Dharma 50/50 cotton/linen blend, 100% cotton sateen and some 100% cotton denim. Sometimes I still get the bleaching at the fold line. also if a piece is folded the top side changes. The lighter areas have gotten even lighter or turned white. Supercritical carbon dioxide (scCO2) was used to extract components of interest from Scenedesmus dimorphus, a microalgae species, under varied algal harvesting and extraction conditions. Liquid chromatography-mass spectrometry (LC-MS) was used to quantify the concentration of fatty acid methyl esters (FAME) and the FAME profile of transesterified lipids, phospholipids and pigments extracted under varied supercritical temperatures and pressures. The scCO2 extraction results are compared with conventional solvent extraction to evaluate differences in the efficiency and nature of the extracted materials. Algae harvested by centrifugation (vs. lyophilization) demonstrated a similar extraction efficiency in scCO2, indicating potential energy benefits by avoiding conventional algal mass dehydration prior to extraction. Fatty acids (FA) were widely used as taxonomic indicators to distinguish algal groups by the presence, the abundance and the distribution of particular FA. This work aimed to compare between species FA composition of three green algae (Chlorophyceae: Chlorella vulgaris, Pseudokirchneriella subcapitata and Scenedesmus subspicatus) grown under comparable conditions, and to assess the suitability of using FA as indicators of taxonomic groups and environmental conditions for microalgae.
