Peripheral inflammatory hyperalgesia depends on the sensitization of primary nociceptive neurons. Inflammation drives molecular alterations not only locally but also in the dorsal root ganglion (DRG) where interleukin-1 beta (IL-1β) and purinoceptors are upregulated. Activation of the P2X7 purinoceptors by ATP is essential for IL-1β maturation and release. At the DRG, P2X7R are expressed by satellite glial cells (SGCs) surrounding sensory neurons soma. Although SGCs have no projections outside the sensory ganglia these cells affect pain signaling through intercellular communication. Therefore, here we investigated whether activation of P2X7R by ATP and the subsequent release of IL-1β in DRG participate in peripheral inflammatory hyperalgesia. Immunofluorescent images confirmed the expression of P2X7R and IL-1β in SGCs of the DRG. The function of P2X7R was then verified using a selective antagonist, A-740003, or antisense for P2X7R administered in the L5-DRG. Inflammation was induced by CFA,

Why the interest in such an obscure cell? There is not a lot of information in the literature.. I am a clinician with a special interest in melanoma and, as such, you are continually struck by the potential aggressive and lethal nature of invasive melanoma and its resistance to therapeutics. There are some striking differences in behaviour between melanoma and its non-melanoma skin cancer relatives. Obviously, the melanocyte as the cell of origin, rather than the keratinocyte, and inevitably the neural crest origin of the melanocyte.. The Satellite cell, its close relative the Schwann cell, the melanocyte and its malignant offspring the melanoma cell, all share this Neural crest cell origin.. During evolutionary development, aspects of the Neural crest began to appear in early chordate species and eventually reached full expression in vertebrates. Protochordates were thin transparent sessile filter-feeders sitting in burrows on the sea floor but with evolutionary development grew in size, became ...

Satellite cell activity is necessary for postnatal skeletal muscle growth. Severe phosphate (PO4) deficiency can alter satellite cell activity, however the role of neonatal PO4 nutrition on satellite cell biology remains obscure. Twenty-one piglets (1 day of age, 1.8 ± 0.2 kg BW) were pair-fed liquid diets that were either PO4 adequate (0.9% total P), supra-adequate (1.2% total P) in PO4 requirement or deficient (0.7% total P) in PO4 content for 12 days. Body weight was recorded daily and blood samples collected every 6 days. At day 12, pigs were orally dosed with BrdU and 12 h later, satellite cells were isolated. Satellite cells were also cultured in vitro for 7 days to determine if PO4 nutrition alters their ability to proceed through their myogenic lineage. Dietary PO4 deficiency resulted in reduced (P | 0.05) sera PO4 and parathyroid hormone (PTH) concentrations, while supra-adequate dietary PO4 improved (P | 0.05) feed conversion efficiency as compared to the PO4 adequate group. INTRODUCTION: The trigeminal ganglion consists of neurons which are of first order pseudounipolar carrying sensory pathway from the receptors. They receive sensation from receptors of face, mouth, scalp and duramater 1. These pseudounipolar neurons inside the ganglion are covered by satellite glial cells nothing but continuous with the Schwann cell sheath that surrounds the axon 2. These satellite glial cells expresses glial fibrillary acidic protein (GFAP) that is involved in inflammation and pain associated with migraine and neuralgia 3.. Migraine and trigeminal neuralgia, a major neurovascular disorder occurring due to the pathological changes in the cutaneous innervations of the trigeminal ganglion like the meningeal tissues, head and neck and scalp 4. Following any changes in the cutaneous sensation the neurons receiving the sensation gets excited and alters the normal functioning of surrounding neurons leading to pathological neuropathic pain 5.. The mechanism by which the pathologically ...
Peripheral sensory ganglia contain the somata of neurons mediating mechanical, thermal, and painful sensations from somatic, visceral, and oro-facial organs. Each neuronal cell body is closely surrounded by satellite glial cells (SGCs) that have properties and functions similar to those of central astrocytes, including expression of gap junction proteins and functional dye coupling. As shown in other pain models, after systemic pain induction by intra-peritoneal injection of lipopolysaccharide, dye coupling among SGCs in intact trigeminal ganglion was enhanced. Moreover, neuron-neuron and neuron-SGC coupling was also detected. To verify the presence of gap junction-mediated coupling between SGCs and sensory neurons, we performed dual whole cell patch clamp recordings from both freshly isolated and short term cultured cell pairs dissociated from mouse trigeminal ganglia. Bidirectional gap junction mediated electrical responses were frequently recorded between SGCs, between neurons and between ...
Peripheral neurotoxicity is a debilitating condition that afflicts up to 90% of patients with colorectal cancer receiving oxaliplatin-containing therapy. Although emerging evidence has highlighted the importance of various solute carriers to the toxicity of anticancer drugs, the contribution of these proteins to oxaliplatin-induced peripheral neurotoxicity remains controversial. Among candidate transporters investigated in genetically engineered mouse models, we provide evidence for a critical role of the organic cation transporter 2 (OCT2) in satellite glial cells in oxaliplatin-induced neurotoxicity, and demonstrate that targeting OCT2 using genetic and pharmacological approaches ameliorates acute and chronic forms of neurotoxicity. The relevance of this transport system was verified in transporter-deficient rats as a secondary model organism, and translational significance of preventive strategies was demonstrated in preclinical models of colorectal cancer. These studies suggest that ...
The effect of photodynamic therapy (PDT) on neurons is of critical importance when treating cancers within or adjacent to the nervous system. Neurons show reduced sensitivity to meta-tetrahydroxyphenyl chlorin (mTHPC) mediated PDT, so the aim of this study was to investigate whether neuron sparing is due to endogenous cellular antioxidant activity. Dorsal root ganglion (DRG) neurons and their associated satellite glia were subjected to mTHPC-PDT in a 3D co-culture system following incubation with antioxidant inhibitors: diethyl dithiocarbamate (DDC, SOD-1 inhibitor), 2-methoxyestradiol (2-MeOH2, SOD-2 inhibitor) and l-buthionine sulfoximine (l-BSO, glutathione synthase inhibitor). Sensitivity of each cell type was assessed using a combination of live/dead staining and immunofluorescence. Pretreatment with DDC and with l-BSO significantly increased the sensitivity of neurons to mTHPC-PDT and also affected satellite glial cell viability, whereas 2-MeOE2 caused only a small increase in neuron ...
Dorsal root ganglia (DRG) are highly vulnerable to frataxin deficiency in Friedreich ataxia (FA). An immunohistochemical and immunofluorescence study of DRG in 15 FA cases and 12 controls revealed that FA causes major primary changes in satellite cells and inflammatory destruction of neurons. Reaction product of the cytoplasmic markers and laminin confirmed proliferation of satellite cells and processes into multiple perineuronal layers and residual nodules. The formation of connexin 43-reactive gap junctions between satellite cells was strongly upregulated. Proliferating satellite cells in FA displayed many more frataxin- and ATP5B-reactive mitochondria than normal. Monocytes entered into the satellite cell layer, appeared to penetrate neuronal plasma membranes, and infiltrated residual nodules. Satellite cells and IBA1-reactive monocytes displayed upregulated ferritin biosynthesis, which was most likely due to leakage of iron from dying neurons.. Read the entire article HERE. ...
IBS is a prevalent, chronic GI disorder that negatively impacts the quality of life in up to 14% of the population.1 ,2 It is characterised by abdominal pain and discomfort associated with altered bowel habits.3-5 Although the pathophysiology of IBS is not completely understood, it is becoming clear that changes to peripheral cellular and sensory mechanisms play key roles in the associated pain.6 ,7 In particular, chronic visceral hypersensitivity (CVH) of colonic afferents is implicated in the development and maintenance of chronic visceral pain (CVP) in IBS.4 ,5 Characteristic features of CVH include nociceptor hypersensitivity8 and increased signalling of noxious colorectal distension (CRD) within the spinal cord.9-11 Recent evidence suggests sensory afferents display upregulation of numerous ion channels and receptors in animal models of CVH,7 ,10 ,12 making them targets for analgesic treatment.. Skeletal muscle undergoes a progressive age-related loss in mass and function. Preservation of muscle mass depends in part on satellite cells, the resident stem cells of skeletal muscle. Reduced satellite cell function may contribute to the age-associated decrease in muscle mass. Here, we focused on characterizing the effect of age on satellite cell migration. We report that aged satellite cells migrate at less than half the speed of young cells. In addition, aged cells show abnormal membrane extension and retraction characteristics required for amoeboid-based cell migration. Aged satellite cells displayed low levels of integrin expression. By deploying a mathematical model approach to investigate mechanism of migration, we have found that young satellite cells move in a random memoryless manner, whereas old cells demonstrate superdiffusive tendencies. Most importantly, we show that nitric oxide, a key regulator of cell migration, reversed the loss in migration speed and reinstated the ...
Translational control of gene expression is an important regulator of adult stem cell quiescence, activation and self-renewal. In skeletal muscle, quiescent satellite cells maintain low levels of protein synthesis, mediated in part through the phosphorylation of eIF2α (P-eIF2α). Pharmacological inhibition of the eIF2α phosphatase with the small molecule sal003 maintains P-eIF2α and permits the expansion of satellite cells ex vivo. Paradoxically, P-eIF2α also increases the translation of specific mRNAs, which is mediated by P-eIF2α-dependent read-through of inhibitory upstream open reading frames (uORFs). Here, we ask whether P-eIF2α-dependent mRNA translation enables expansion of satellite cells. Using transcriptomic and proteomic analyses, we show a number of genes associated with the assembly of the spindle pole to be upregulated at the level of protein, without corresponding change in mRNA levels, in satellite cells expanded in the presence of sal003. We show that uORFs in the 5′ UTR ...
The Earth is not a perfect sphere as its rotation causes it to be slightly flattened at the poles This oblateness can be modeled as a extra band of material encircling the equator The extra material exerts torque on an orbiting satellite causing the orbital planes to slowly precess in space rather than being fixed in an inertial frame The oblateness torque will also cause the line of the apsides t

ENGLISH ABSTRACT: Although muscle tissue demonstrates a remarkable capacity for regeneration following injury, this process is slow and often accompanied by the formation of scar tissue and a subsequent decrease in contractile capacity following regeneration. Treatment options are few and mostly supportive in nature. This regeneration process involves muscle stem cells (satellite cells) which ultimately give rise to the regenerated muscle. The contentious field of low level laser therapy (LLLT) has made remarkable claims in facilitating wound healing in soft tissue injuries of various types. Yet, the mechanism(s) invoked in these beneficial effects are poorly understood. We have investigated the effects of LLLT using a 638 nm laser on satellite cells in culture and in-vivo. Using an array of techniques we have measured, amongst other things, metabolic responses to laser irradiation, signaling pathways activated/altered and antioxidant status. In response to laser irradiation satellite cells in ...
Skeletal muscle satellite cells located between the plasma membrane and the basal lamina of muscle fibres, could for many years, only be studied in situ by electron microscopy. The introduction of immunohistochemistry and the discovery of molecular markers of satellite cells then made them accessible for light microscopic studies and a wealth of information is today available. Satellite cells are myogenic stem cells that can be activated from a quiescent state to proliferate for self-renewal or differentiate into myogenic cells. The satellite cells are involved in muscle growth during fetal and postnatal development and play a key role in repair and regeneration of damaged muscle fibres. The satellite cells are also essential for muscle fibre hypertrophy and maintenance of muscle mass in the adult. When the present thesis was initiated, studies on satellite cells in human skeletal muscle relied on the neuronal cell adhesion molecule (NCAM) as a marker for satellite cell identification. The ...
Myostatin, a member of the TGF- family, has been identified as a powerful inhibitor of muscle growth. the absence of myostatin involves little or no input from satellite cells. Hypertrophic fibers contain no more myonuclei or satellite cells and myostatin had no significant effect on satellite cell proliferation in vitro, while expression of myostatin receptors dropped to Amonafide (AS1413) IC50 the limits of detectability in postnatal satellite cells. Moreover, hypertrophy of dystrophic muscle arising from myostatin blockade was achieved without any apparent enhancement of contribution of myonuclei from satellite cells. These findings contradict the accepted model of myostatin-based Amonafide (AS1413) IC50 control of size of postnatal muscle and reorient fundamental investigations away from the mechanisms that control satellite cell proliferation and toward those that increase myonuclear domain, by modulating synthesis and turnover of structural muscle fiber proteins. It predicts too that any ...

A recent study by Rudnicki and colleagues has sought to address the question of satellite-cell heterogeneity directly (Kuang et al., 2007). The authors have shown that ∼90% of satellite cells on myofibres of adult Myf5cre/+ mice had had a `myogenic experience and expressed Myf5 at some point (as shown by the presence of YFP from the recombined targeted ROSA locus). The remaining ∼10% of satellite cells were YFP-negative, and were able to produce further YFP-negative and YFP-positive cells both in vitro and in vivo. When grafted into Pax7-null mice, these YFP-negative cells gave rise to approximately three times more Pax7-positive satellite cells than the YFP-positive cells, and a quarter of these remained YFP-negative. It was proposed by the authors that these YFP-negative cells correspond to a dedicated subset of satellite cells that have more stem-cell-like characteristics (satellite `stem cells), and that the YFP-positive cells are their transit-amplifying progeny that can undergo ...
This study demonstrated a number of novel findings regarding satellite cell proliferation in the skeletal muscles of the OZR model of metabolic syndrome. First, there was a decrease in the number of myonuclei in muscle fiber cross sections of the OZR compared with the LZR. Second, this study is the first to show that hypertrophy is not impaired in muscles of the OZR model of metabolic syndrome, if the stimulus is adequate. In addition, to the best of our knowledge, this is the first study to observe any form of loading-induced hypertrophy in any rodent model of metabolic syndrome. The third novel finding is that, although there is no change in the proportion of quiescent satellite cells in control muscles, there was a significant attenuation of satellite cell proliferation of adult muscle under normal control loading conditions in the OZR compared with the LZR. The attenuation of satellite cell proliferation in control soleus and plantaris muscles that we observed in this study may result in a ...

While the concept of deepfakes, or AI-generated synthetic imagery, has been decried primarily in connection with involuntary depictions of people, the technology is dangerous (and interesting) in other ways as well. For instance, researchers have shown that it can be used to manipulate satellite imagery to produce real-looking - but totally fake - overhead maps of cities. The study, led by Bo Zhao from the University of Washington, is not intended to alarm anyone but rather to show the risks and opportunities involved in applying this rather infamous technology to cartography.

The primary aim of our study was to validate the assessment of myonuclear and satellite cell number in biopsies from human skeletal muscle. We found that 25 type I and 25 type II fibers are sufficient to estimate the mean number of myonuclei per fiber. In contrast, the assessment of satellite cells improved when more fibers were included. Second, we report that small differences in counting satellite cells using CD56 and Pax7 antibodies can be attributed to the different staining profiles. Third, we provide support for the use of Ki67 in evaluating the proportion of active satellite cells. We observed very few (up to 1.3%) active satellite cells in healthy adult skeletal muscle at rest, but they increased significantly (up to 7-fold) following muscle activity. This study provides valuable tools to assess the behavior of satellite cells, both in pathological conditions and in response to physiological stimuli.. ...

Hi Peter, Congrats on working all the satellites - a very dedicated effort! Weather shouldnt be much of a factor for VHF and UHF communication; its not until you get into the microwaves that I have had trouble, and even with the AO-40 (RIP) 2.4 ghz downlink it wasnt too noticeable. Foliage is also a factor there, and somewhat for UHF; were you aiming through any big trees? Most of the variation from one pass to another comes from the somewhat random orientation of the satellites antennas compared to where the planet is, and the polarization of both stations antennas. Do you have linear or circularly polarized antennas? With the significant increase in small satellite activities and projects in Europe, the US, Asia, and other parts of the world, the availability of regular, affordable access to space for small satellites becomes even more critical than before. Obviously, there are many complicating factors in arranging a ride for your small satellite. These can be technical and programmatic mismatches between the available launch opportunity and the small satellite mission objectives and planning. But (more) often these complicating factors also relate to export restrictions, political considerations, lack of standardization or a mismatch in requirements and expectations between the small satellite developer and the launch provider or primary customer. Considering the number of rocket launches per year, one might expect sufficient excess capacity on board these launch vehicles to easily accommodate a large number of small satellites as auxiliary or piggyback payloads, In practice it is actually quite difficult to
