Sarcocystis are intracellular protozoan parasites that are characterised by their ability to invade muscle tissue and form intramuscular sarcocysts. A muscular sarcocystosis outbreak was reported by travellers returning from Tioman Island in 2011 and 2012 where Sarcocystis nesbitti was identified as the main cause. The source of the S. nesbitti that was involved has remained elusive, although water is hypothesised to be the main cause of transmission. A surveillance study was therefore undertaken in the northern regions of Tioman Island to identify the source of S. nesbitti by screening rivers, water tanks, wells and seawater. Water samples were collected from rivers, water tanks, wells and seawater on Tioman Island over the course of April to October 2015. Water samples were indirectly screened for Sarcocystis species by obtaining sediment from respective water sources. PCR amplification of the 18S rRNA gene region was conducted to identify positive samples. Microscopy was

Procedures - Sequencing of the nuclear small subunit ribosomal RNA gene from a new S neurona isolate (UCD 1) was performed. The sequence was compared with that of other closely related Sarcocystidae parasites. From this sequence, conserved DNA sequence primers were selected and an oligonucleotide probe was designed to hybridize with a unique ...
Sarcocystis neurona is the most common cause of Equine Protozoal Myeloencephalitis (EPM), affecting 0.5-1% horses in the United States during their lifetimes. The objective of this study was to evaluate the equine immune responses in an experimentally induced Sarcocystis neurona infection model. Neurologic parameters were recorded prior to and throughout the 70-day study by blinded investigators. Recombinant SnSAG1 ELISA for serum and CSF were used to confirm and track disease progression. All experimentally infected horses displayed neurologic signs after infection. Neutrophils, monocytes, and lymphocytes from infected horses displayed significantly delayed apoptosis at some time points. Cell proliferation was significantly increased in S. neurona-infected horses when stimulated nonspecifically with PMA/I but significantly decreased when stimulated with S. neurona compared to controls. Collectively, our results suggest that horses experimentally infected with S. neurona manifest impaired ...
Sarcocystosis is caused by species of Sarcocystis, an intracellular protozoan parasite in the phylum Apicomplexa. These parasites have an indirect life cycle, cycling between a definitive and an intermediate host. Intestinal infections occur in the definitive host, and tissue invasion is seen in the intermediate host. More than a hundred species of Sarcocystis are parasites of domestic and wild animals. Many of these infections are asymptomatic, particularly in the definitive host. Equine protozoal myeloencephalitis is usually caused by Sarcocystis neurona. Neospora caninum and/or Neospora hughesi, not discussed in this outline, have also been implicated in some cases. Symptomatic infections caused by Sarcocystis species are reported occasionally in other domestic animals. Humans are a definitive host for S. suihominis, found in pork, and S. hominis, found in beef. These parasites infect the intestines. Humans can be intermediate hosts for a variety of other Sarcocystis species. The parasites are found
The aim of the study was to diagnose Sarcocystis sp. infections in cattle and to detect coinfections by Toxoplasma gondii and/or Neospora caninum. Blood, diaphragm, esophagus, and myocardium from 90 beef cattle from Argentina were collected. Histopathological, immunohistochemical, polymerase chain reaction assays, and direct microscopical examination were carried out. Sarcocysts from myocardium were measured and counted. Indirect fluorescent antibody test (IFAT) for the three protozoans was performed. Sarcocystis cruzi sarcocysts were found in 100% of myocardium samples. Sarcocysts per gram ranged from 8 to 380 with higher values found in adult cattle (p < 0.001). T. gondii and N. caninum were not detected by immunohistochemistry. T. gondii DNA was found in myocardium of 2/20 seropositive animals, while N. caninum DNA was not found. Antibodies against S. cruzi were detected in all samples, those against N. caninum in 73% and against T. gondii in 91% of the samples (IFAT titer >/=25). It is ...
The present study examined the seroprevalence of Toxoplasma gondii and Sarcocystis neurona in a population of beavers (Castor canadensis) from Massachusetts. Sixty-two blood samples were collected during the field seasons over 3 consecutive years from different animals. Blood was collected onto filter paper and shipped to the Department of Biomedical Sciences, Virginia Tech, Blacksburg, Virginia, for parasite testing. The samples were tested at dilutions of 1:25. 1:50, and]:100 against each parasite antigen by modified agglutination tests to determine whether antibodies to either parasite were present in the blood. Six of 62 samples (10%) were positive for T. gondii, with 2 samples having titers of 1:25 and 4 having titers of 1:50. Four of 62 samples (6%) were positive for S. neurona, with 2 samples having titers of 1:25 and 2 having titers of 1:50 ...

Citation: N/A Interpretive Summary: Sarcocystis neurona is a single-celled parasite of horses. It causes a fatal illness (EPM) in horses in Americans. Its life cycle is not fully known. Opossums are its definitive (reservoir) host. Horses become infected by ingesting the resistant stage of the parasite shed in opossum feces. Scientists at the Beltsville Agricultural Research Center have found S. neurona sarcocysts (muscle cysts) in muscles of naturally infecte skunks which could be a source of infection for the opossum. The also describe a new species of Sarcocystis, S. mephitisi in muscles of skunks. These results will be of interest to biologists, horse owners and parasitologists. Technical Abstract: Two structurally distinct types (A, B) of microscopic sarcocysts were found in muscles of 4 of 5 feral skunks. Type A sarcocysts had up to 6 m thick sarcocyst wall. The villar protrusions (Vp) on the sarcocyst wall were up to 5 m long. The Vp were constricted at the base, expanded in the middle, ...
Citation: N/A Interpretive Summary: Sarcocystis neurona and Toxoplasma gondii are single celled parasites of animals. The former is transmitted via infected feces of opossums and the latter by cats. Both parasites are widely distributed in the Americas. Scientists at the Beltsville Agricultural Research Center and Virginia Tech; Blacksburg report dual infection by these parasites causing encephalitis in a sea otter. Results indicate contamination of sea water by these organisms. The results will be of use to parasitologists, veterinarians and wildlife biologists. Technical Abstract: Dual Sarcocystis neurona and Toxoplasma gondii infection was observed in a Northern sea otter from Washington, USA. The animal was found stranded, convulsed, and died shortly thereafter. Encephalitis caused by both S. neurona and T. gondii was demonstrated in histological sections of brain stained with parasite specific antisera. The lesions induced by T. gondii suggested that the sea otter was suffering from ...

An adult female sea otter housed for 5 years in an outdoor habitat in an aquarium developed signs of neurologic disease. Bilateral caudal paresis was evident initially and other neurologic signs consistent with CNS disease developed rapidly. Diagnostic work-up included CBC, serum biochemical analyses, determination of serum antibody titers, radiography of the vertebral column, CSF analysis, muscle biopsy, computed tomography of the brain, and assays for mercury, lead, and thiamine. A tentative diagnosis of encephalitis caused by a Sarcocystis neurona-like organism was made on the basis of detection of CSF antibodies by use of Western blot analysis. Response to treatment was not satisfactory and the sea otter was euthanatized. Immunohistochemical staining revealed S neurona-like organisms within foci of inflammation in the brain and spinal cord. This report provides evidence that, for sea otters, there may be a mode of transmission of an S neurona-like organism that does not involve ...

The effect of temperature and relative humidity (RH) on the survival of sporocysts of S. cruzi were investigated in vitro. Under all experimental conditions (temperature of 4°C, 37°C or room temperature; RH of 18%, 75%, or 100%) some sporozoites retained their viability to excyst for at least 90 days. The best conditions for survival were 4°C at 100% RH (more than 240 days) and 37°C at 18% RH (more than 180 days). Sporocysts maintained at room temperature at all humidities had the lowest level of survival. It is concluded that sporocysts of S. cruzi are able to survive in most environments for several months and that the fluctuation of the daily ambient temperature is likely to influence the viability of the sporocysts.. ...

The widespread protozoan parasite Toxoplasma gondii interferes with host cell functions by exporting the contents of a unique apical organelle, the rhoptry. Among the mix of secreted proteins are an expanded, lineage-specific family of protein kinases termed rhoptry kinases (ROPKs), several of which have been shown to be key virulence factors, including the pseudokinase ROP5. The extent and details of the diversification of this protein family are poorly understood. In this study, we comprehensively catalogued the ROPK family in the genomes of Toxoplasma gondii, Neospora caninum and Eimeria tenella, as well as portions of the unfinished genome of Sarcocystis neurona, and classified the identified genes into 42 distinct subfamilies. We systematically compared the rhoptry kinase protein sequences and structures to each other and to the broader superfamily of eukaryotic protein kinases to study the patterns of diversification and neofunctionalization in the ROPK family and its subfamilies. We identified

DIAGNOSIS: TOXOPLASMOSIS. DISCUSSION:. The immunohistochemical study was positive for Toxoplasma gondii demonstrating positive staining for the bradyzoites within the cysts and pseudo-cysts as well as individual tachyzoites in the necrotic tissue. Histopathological diagnosis of toxoplasmosis could be difficult due to the fact that cysts are difficult to recognize. These cysts may resemble those seen in other infections such as Leishmania donovani, Trypanosoma cruzi, Sarcocystis, Besnoitia and so on (1) Immunohistochemical stains now reveal the true nature of the microorganism, although electron microscopy can be used. Toxoplasmosis is a common CNS infection in AIDS patients (2,3). The characteristic lesion is a necrotizing acute abscess (1). Toxoplasmosis is well known as an intrauterine infection with devastating consequences but it is far more rare in settings outside of these. Although reported in post-transplanted patients, there have been virtually no reports of CNS toxoplasmosis in ...

Summary The pathogenesis of toxoplasmosis is discussed under the headings of Host, Microorganism, Treatment and Time, in an attempt to integrate the many factors operating during infection. The obligate intracellular development of Toxoplasma, its ability to grow in many cells and the formation of a relatively inert cyst are the major microorganismal attributes determining pathogenesis. Whereas many natural hosts appear relatively resistant to many of the strains, the inability of some hosts to overcome infection or to develop an effective immunity and, furthermore, the presence of an infection-immunity (premunition), the development of hypersensitivity with chronic infection and the results of cyst rupture are the major host factors complicating pathogenesis. Our knowledge of the shifting relative importance of each factor while infection progresses is inadequate. The analysis of the pathogenesis must take into account the effects of superimposition of different rates of a variety of processes. It has

Through 2 international traveler-focused surveillance networks (GeoSentinel and TropNet), we identified and investigated a large outbreak of acute muscular sarcocystosis (AMS), a rarely reported zoonosis caused by a protozoan parasite of the genus Sarcocystis, associated with travel to Tioman Island, Malaysia, during 2011-2012.; Clinicians reporting patients with suspected AMS to GeoSentinel submitted demographic, clinical, itinerary, and exposure data. We defined a probable case as travel to Tioman Island after 1 March 2011, eosinophilia (,5%), clinical or laboratory-supported myositis, and negative trichinellosis serology. Case confirmation required histologic observation of sarcocysts or isolation of Sarcocystis species DNA from muscle biopsy.; Sixty-eight patients met the case definition (62 probable and 6 confirmed). All but 2 resided in Europe; all were tourists and traveled mostly during the summer months. The most frequent symptoms reported were myalgia (100%), fatigue (91%), fever ...
Equine protozoal myeloencephaltis is a serious health concern for horse owners. Although more than half of horses infected with this disease will respond to treatment, about 40% do not and eventually die from the deterioration of their central nervous system. Sadly, only one-tenth of infected horses completely recover from the infection. Learning the equine protozoal myeloencephalitis symptoms can help you get treatment for your horse quickly.. Recognizing Symptoms. The equine protozoal myeloencephalitis symptoms can be different in the infected horses because the infection causes lesions to appear in different areas of the brain. Depending on where these lesions show up, the symptoms change. Most of the more common symptoms involve lameness, loss of coordination, changes in the horses gait, and muscle atrophy. Other symptoms can include head tilting, leaning against the stall wall for balance, paralysis of the face, and loss of feeling in the head and neck area. When your horse begins showing ...
Sarcocystosis is characterized by the invasion of muscles and sometimes other tissues by protozoa of the genus Sarcocystis, of which S. hominis (intermediate host domestic cattle) and S. suihominis (domestic pig) are the most significant to humans, to whom they are transmitted by ingestion of uncooked beef or pork. Humans serve as either intermediate or final host: (1) intermediate host-presence of cysts in muscle is usually asymptomatic, but may cause myositis or myopathy; detected on clinical examination or muscle biopsy; (2) final host-may be asymptomatic or cause fever and gastrointestinal upset; oocysts or sporocysts can be detected in faeces. There is no specific treatment. Prevention is by not eating uncooked meat from any animal....

Until the availability of molecular testing, it was almost impossible to definitively identify the causative agent and infection status of an affected horse. If a horse showed signs of neurologic problems, the veterinarian began a process of elimination to determine what was NOT causing the symptoms. Traditional EPM tests were only effective at determining that the horse did not have EPM. If traditional EPM test results were positive, that only definitively revealed that the horse had been exposed in the past to the parasites that cause EPM. Testing could not show whether the horse had an active infection by those parasites. ...

Maureen Long is a PeerJ user. Bio: Our laboratory investigates primarily vector-borne pathogens that affect humans and animals. Research interests include several equine and large animal infectious diseases with emphasis currently on mosquito borne flaviviruses and alphaviruses. Other pathogens currently under investigation include the human pathogens Dengue virus and recently Zika virus. We are interested primarily in using an -omics approach to determine why certain animals/people actually develop diseases while others do not. We have studied WNV in horses and birds and performed surveillance in horses and people for several mosquito-borne viruses. We have just complete work using a transcriptomic approach to comparing viral infection of the brain (West Nile virus) to protozoal infection of the brain (Sarcocystis neurona) in the horse. A $1.86 million grant from the National Science Foundation - National Institutes of Health Ecology of Infectious Disease Grant Program will expand the investigations of a UC Davis-led research team tracking how a deadly parasite moves from land to sea to infect and kill sea otters (Enhydra lutris nereis) in California. Weve already identified the single-celled parasite, Toxoplasma gondii, that causes high rates of mortality in threatened southern sea otters, says Patricia Conrad, professor of parasitology at the UC Davis School of Veterinary Medicine. Conrad, the principal investigator, says, Toxoplasma infections are killing southern sea otters and are one of several factors reducing the potential for sea otter population recovery. Sea otters do not prey on known intermediate hosts for T. gondii, says Conrad, so the most likely source of infection is highly infectious, egg-like oocysts shed in the feces of cats and transported via freshwater runoff into the marine ecosystem. Toxoplasma ...

The U.S. Food and Drug Administration announced May 15 that four horses have died after receiving compounded drug products, containing pyrimethamine and toltrazuril, used to treat equine protozoal myeloencephalitis. The compounds originated at Wickliffe Pharmacy in Lexington, Ky.. Six additional horses have suffered adverse effects, including fever and seizure, from the medication but are now recovering.. One lot of the compound was a paste and one was an oral suspension. The FDA reports that all of the products in the lots are accounted for and are no longer in distribution.. [The] FDA is working with the pharmacy and state partners to investigate these adverse events, stated an FDA release. At this time, FDA testing indicates that one lot of product contained higher levels of pyrimethamine than the labeling indicates. Adverse events associated with high doses of pyrimethamine include seizures, fever and death. Two of the horses were located in Kentucky and eight in Florida.. These ...
Competing Friday in the 53rd annual Abdallah Shrine Rodeo at the Leavenworth County Fairgrounds had extra significance for Paige Boylan.. The 2011 Tonganoxie High School graduate not only competed in her hometown in the barrel-racing event of the Shrine Rodeo for the first time but also its is part of what she is calling a rebuilding year with her teammate: her quarterhorse Queenie.. A year ago, her horse was diagnosed with equine protozoal myeloencephalitis, a condition in which a parasite affects the central nervous system. Three months later, Queenie suffered a bone infection in one of her back legs and lost 200 pounds in the process.. Ive had a big support group throughout my horse being sick and people upping my confidence and my horse getting better, she said. I thought she was done after last year.. But a resilient Queenie has gained back the weight and now weighs nearly 1,100 pounds.. Looking at her today, you would have never known she was sick last year, Boylan said.. Though an ...
Southern Sea Otters (Enhydra lutris nereis) are a fixture in Monterey Bay, California. On our whale watch last year, we saw this little whiskered fellow hanging out by a kelp forest and munching on a clam. At first, he was floating on his back in the water with a clam on his chest and next he had cracked the clam and was eating. They are such delightful creatures. Although the Southern Sea Otter population is much larger than earlier in the 20th Century, they are still dying at alarming rates and struggling to survive. The Monterey Bay Aquarium is doing a lot of research on these otters and has a wonderful exhibit of otters who are unable to be released back into the wild ...

Sea otter (Enhydra lutris nereis) sighting data can be selected to dynamically generate a graph based on the criteria you select. You can either graph the data using monthly sightings over the course of a single year, or you can graph yearly sightings for two or more years.. When viewing monthly sightings, the number of sea otters sighted are plotted for each age class (i.e. adults, adults with pups, large pups, and/or small pups) you selected for every month, from January to December. At the top of each bar on the graph is the actual number of sightings. Bars are color-coded by sea otter age class. Below the graph are four summary statistics based on the set of data you selected for each age class over the whole year: 1) total sea otter sightings, 2) monthly mean number of sea otter sightings, 3) standard deviation for the monthly mean number of sea otter sightings, and 4) the standard error of the mean.. When viewing yearly sightings, the number of sea otters sighted are plotted for each age ...

Adriano Aguzzi, a professor of neuropathology and director of the Swiss National Creutzfeldt-Jakob Disease Surveillance Center in Zurich, Switzerland, said in an e-mail that IHC can be as sensitive as the Western blot method in certain specific instances. Aguzzis institution, however, routinely runs a Western blot test enhanced with another step on every suspect case, because of its unrivalled specificity and because in most cases we find it to be much more sensitive than immunohistochemistry.. In the months since the November announcement, the USDA came under intense pressure from organizations such as Consumers Union for failing to conduct a Western blot test on the suspect cow.. Activists at Consumers Union pointed to reports in veterinary literature which suggested IHC could miss some cases.. Michael Hansen, a senior research associate with Consumers Union, says he confronted USDA officials about the issue during the June 6 meeting of Institute of Medicine of the National ...

Initially the first doctor advise me for HIV 1&2 P24 ANTIBODIES screening Combo test (CMIA) along with VDRL (RPR) SERUM which came non reactive on the 23rd March 2016. Also on the 08 April I got a negative result for Western Blot test as well. After four weeks from the initial infection in my throat which invoved sowllen Cervical lymph nodes I have been diagnosed with Infectious Mono caused by EBV which came positive in my blood report. As per the articles I am reading on the net since then says that mono and HIV have almost same symptoms. So after when I got ...
Shodno ...
