Since autoimmune diseases are listed as a possible risk factor for salivary duct stone formation, one might think this little bit of trivia would be better known. Just as stones can form in the kidneys and gallbladder, they can also form in the salivary ducts. The stones themselves dont hurt. Its when theyre big enough…

Oral mucocele (also termed mucous retention cyst, mucous extravasation cyst, mucous cyst of the oral mucosa, and mucous retention and extravasation phenomena) is a clinical term that refers to two related phenomena: Mucus extravasation phenomenon Mucus retention cyst The former is a swelling of connective tissue consisting of a collection of fluid called mucus. This occurs because of a ruptured salivary gland duct usually caused by local trauma (damage), in the case of mucus extravasation phenomenon, and an obstructed or ruptured salivary duct (parotid duct) in the case of a mucus retention cyst. The mucocele has a bluish translucent color, and is more commonly found in children and young adults. Although the term cyst is often used to refer to these lesions, mucoceles are not strictly speaking true cysts because there is no epithelial lining. Rather, it would be more accurate to classify mucoceles as polyps (i.e. a lump). The size of oral mucoceles vary from 1 mm to several centimeters and they ...
Tuft cells, sometimes referred to as brush cells, are chemosensory cells in the epithelial lining of the intestines and respiratory tract. The names tuft and brush refer to the microvilli projecting from the cells. Ordinarily there are very few tuft cells present but they have been shown to greatly increase at times of a parasitic infection. Several studies have proposed a role for tuft cells in defense against parasitic infection. In the intestine, tuft cells are the sole source of secreted interleukin 25 (IL-25). Tuft cells are differentiated from stem cells in the bases of intestinal glands and their increase is seen as a type-2 immune response via ILC2s, which secrete IL-13, causing an increase in the number of tuft cells. Type 2 immunity is involved in parasitic infections and allergic inflammation. The presence of α-gustducin in intestinal tuft cells raises the idea that tuft cells may be involved in the taste transduction pathway. Cells also express TRPM5, a signaling molecule in ...
Lethality: Ras signaling promotes the excretory duct cell fate, and mosaic analysis suggested that loss of this one specific cell can account for the zygotic lethality of let-60 null mutants (Yochem et al., 1997). The excretory duct cell is required for osmoregulation (Nelson and Riddle, 1984). let-60 loss-of-function mutants, which lack the excretory duct cell, die as rod-like larvae with a fluid-filled appearance (Figure 2). let-60 gain-of-function mutants sometimes have two excretory duct cells (Yochem et al., 1997). Ras signaling may have other (maternally-rescued) essential roles in addition to excretory duct formation, since mutations in let-23/EGFR, egl-15/FGFR and ptp-2/SHP-2 cause distinct scrawny and lethal defects that are rescued by constitutive forms of LET-60 Ras (DeVore et al., 1995; Gutch et al., 1998; Koga and Ohshima, 1995). Hyperactivation of egl-15/FGFR or let-60/Ras can also lead to lethality with a Clear, fluid-filled appearance (Kokel et al., 1998; Schutzman et al., ...
To elucidate the role of extracellular matrix and basement membrane associated proteins at the ectodermal-ectomesenchymal interface of developing salivary glands, human fetal submandibular glands (n=100), gestational age varying from 10 to 40 weeks, were evaluated for expression of tenascin, type IV collagen and laminin by using immunohistochemical methods and the results were compared with their expression in adult glands (n=20). Immunoreactive tenascin was present in the epithelial-mesenchymal interface of developing salivary ducts beginning from 10 weeks of gestational age during the early developing stages. In the late developmental stages, the intensity of immunoreaction was reduced around the proximal striated ducts and no immunoreaction was seen around the intercalated duct and acinar cells. Adult submandibular glands had immunoreactive tenascin confined to the periductal connective tissue of distal segment of striated and excretory ducts. Type IV collagen and laminin were observed in the ...
The salivary glands are essential for lubrication, defense and beginning digestion in the mouth. The largest of the salivary glands - the parotid - secretes important proteins into the saliva. As with all salivary glands, it has multiple secretion pathways, therefore it must sort proteins destined for saliva into the correct pathway for secretion. This can be tricky as there are seven possible pathways. One pathway takes proteins to the salivary duct, other pathways carry different proteins to the back side of the cell to be secreted into the blood or to form a supportive matrix for the cells. Transport along these pathways occurs by sorting the proteins into vesicles (hollow membrane sacs) that carry their cargo to the correct destination. Conventional thought was that cargo proteins are moved into the forming vesicles by attaching to sorting receptor proteins. Darling and his team have discovered a completely new approach, suggesting the reason no salivary sorting receptor protein has been ...

We previously published a detailed survey of the spatio‐temporal expression of Runx3 during embryonic development (Levanon et al, 2001). Runx3 expression was examined at embryonic day (E) 10.5 and between E14.5 and E16.5, and compared to the expression pattern of Runx1. Immunohistochemistry (IHC) and knock‐in (KI) β‐galactosidase activity (LacZ staining) were used in parallel throughout this analysis to rigorously determine the expression patterns of the two TFs. Runx3 and Runx1 were readily detected in different compartments of the haematopoietic system and also in the dorsal root ganglia (DRG), epidermal appendages and developing skeletal elements (Levanon et al, 2001). However, regarding epithelia an interesting distinction was noted in the expression pattern of Runx1 and Runx3. While Runx1 was expressed in various epithelia including mucosa of the oesophagus and stomach, the salivary glands ducts and the olfactory and respiratory mucosa, Runx3 expression was undetectable in these ...
We defined here Elp3 as a target of Wnt signaling dispensable for intestinal homeostasis but essential for Tuft cell differentiation, Wnt-driven tumor initiation, and radiation-induced regeneration in the intestine. Although nuclear levels of β-catenin and constitutive Wnt-dependent gene transcription do not require Elp3 in the intestine, our data demonstrate that the translation of Sox9, a protein induced by Wnt, is controlled by Elp3 to maintain a pool of Lgr5+/Dclk1+/Sox9+ cancer stem cells.. Tuft cells derive from Lgr5+ crypt columnar stem cells but their differentiation and biological roles remain poorly understood (Gerbe et al., 2011; Bjerknes et al., 2012). We found here that Elp3 is critical for Tuft cell differentiation, as its absence leads to dramatic reduction of Dclk1+ Tuft cells in the intestinal epithelium. The reduction of differentiated Tuft cells in Elp3-deficient mice did not lead to any detectable functional defect or to significant change in intestinal architecture, ...

Salivary Gland (SG) Cancers are a rare and heterogeneous group of tumors, usually approached by multidisciplinary teams in high specialized centers. Until today no standard of care exists to treat these cancers. The identification of a target, the androgen receptor, in SG tumors has allowed for new treatment strategies options for this rare group of diseases. As a matter of fact, strong positivity for androgen expression has been found in salivary duct carcinoma and adenocarcinomas. The purpose of this study is therefore to evaluate the efficacy and safety of chemotherapy versus androgen deprivation therapy (ADT) in patients with recurrent and/or metastatic AR expressing SGCs.. The study will include two cohorts of patients: Cohort A, which comprises chemo-naïve patients, and Cohort B, which comprises pretreated patients. ...

I have made several trips to the dentist. He said it was a blocked minor salivary duct, and he could remove it surgically. He said there was no stone present, that it was just inflamed. I went to my doctor and he said it could be a sinus infection and gave me Penicillin which did nothing. Today I went back to the dentist, and he wants me to go to an oral surgeon for a diagnoses. I have used numerous mouthwashes and am drinking plenty of water. What could this pain be? I am so fed up with it and now am very concerned that it may be oral cancer ...
Antibodies to the epithelial cells of the salivary duct occur in 60 % of patients with Sjögrens syndrome. Sjögrens syndrome is a systemic autoimmune disease in which immune cells attack and destroy the exocrine glands that produce tears and saliva. Sjögrens syndrome can exist as a disorder in its own right (primary Sjögrens syndrome) or it may develop years after the onset of an associated rheumatic disorder, such as rheumatoid arthritis, SLE, scleroderma, primary biliary cirrhosis, etc. (secondary Sjögrens syndrome). ...
Tubular epithelial cells are one of the most abundant cell types in multicellular organisms. Tubular cells transport gases and liquids, and funnel harmful excretory waste from our bodies. It is clear that Receptor Tyrosine Kinase (RTK) signaling is essential for the formation of many tubular organs such as our kidneys and blood vessels. However, which steps in tube development require RTK signaling is less well understood. The C.elegans excretory system is a primitive renal system with just three essential cells (duct, pore, and canal cells), providing a simple yet dynamic system to study tube specification and morphogenesis. In the C.elegans excretory system, we demonstrated that the EGF-Ras-Erk signaling pathway specified the excretory duct tube versus the pore tube fate. In addition, EGF-Ras-Erk signaling influenced the positions that the duct and pore cells adopted within the tubular network. And finally, after position and fate determination, EGF-Ras-Erk signaling had a continued role in

Back to the future of literature: Niffeneggers Wife Blake Hinton / Reviewer. Out of all genres to write about, time travel is easily the hardest. Its the hardest, not in the writing aspect, but because of the fact the story has been done hundreds of times before. In this day and age, a story simply about time travel is boring. To have true success is to bring a new element to the table. Jack Finney did it with his classic time travel novel Time And Again. Now, decades later, author Audrey Niffenegger does it again with her new novel, The Time Travelers Wife. What makes this book so innovative is the creative plot and unique style. In fact, the story is very reminiscent of Kurt Vonneguts Slaughter-House Five. In Wife, the two main characters are a married couple, Henry and Clare De Tamble. Henry is a Chicago librarian who is the victim of a new disease named chrono displacement disorder. Because of this disease, Henry is unstuck in time. Without warning, Henry pops backward and ...

The shedding of severe acute respiratory syndrome coronavirus (SARS-CoV) into saliva droplets plays a critical role in viral transmission. The source of high viral loads in saliva, however, remains elusive. Here we investigate the early target cells of infection in the entire array of respiratory tissues in Chinese macaques after intranasal inoculations with a single-cycle pseudotyped virus and a pathogenic SARS-CoV. We found that angiotensin-converting enzyme 2-positive (ACE2 +) cells were widely distributed in the upper respiratory tract, and ACE2 + epithelial cells lining salivary gland ducts were the early target cells productively infected. Our findings also have implications for SARS-CoV early diagnosis and prevention. Copyright © 2011, American Society for Microbiology. All Rights Reserved ...
Purpose: We sought to identify genomic alterations (GAs) in salivary gland adenocarcinomas, not otherwise specified (NOS), salivary duct carcinomas (SDC), carcinoma ex pleomorphic adenoma (ca ex PA), and salivary carcinoma, NOS. Experimental design: DNA was extracted from 149 tumors. Comprehensive genomic profiling (CGP) was performed on hybridization-captured adaptor ligation-based libraries of 182 or 315 cancer-related genes plus introns from 14 or 28 genes frequently rearranged for cancer and evaluated for all classes of GAs. Results: 590 GAs were found in 157 unique genes (mean 3.9/tumor). GAs in the PI3K/AKT/mTOR pathway were more common in SDC (53.6%) than other histologies (p=0.019) Cyclin dependent kinase GAs varied amongst all histotypes: adenocarcinoma, NOS (34.6%); SDC (12.2%); ca ex PA (16.7%); carcinoma, NOS (31.2%) (p=0.043). RAS GAs were observed: adenocarcinoma, NOS (17.3%); SDC (26.8%); ca ex PA (4.2%); and carcinoma, NOS (9.4%) (p=0.054). ERBB2 GAs, including amplifications and ...

Background: The purpose of the present study was to elucidate whether myoepithelial cells proliferate mitotically during regeneration of rat submandibular glands after atrophy.. Methods: The excretory duct of the right submandibular gland of rats was doubly ligated near the hilum with metal clips, which were removed after 7 days of ligation (day 0). The regenerating right submandibular glands were removed from 0 to 14 days after removal of the clips. The removed tissue was examined with immunohistochemical double staining for proliferating cell nuclear antigen (PCNA) as a marker of proliferating cells and actin as a marker of myoepithelial cells, as well as with transmission electron microscopy (TEM).. Results: The PCNA-positive myoepithelial cells were observed at the periphery of transitional duct-acinar structures, ducts and acini in the regenerating glands at every time-point, and the PCNA-labeling index of myoepithelial cells increased greatly especially between day 2 and 4. The mitosis of ...
Objectives: The objectives of this study were to investigate the feasibility of clinical application of magnetic resonance (MR) virtual endoscopy as a presurgical procedure before sialoendoscopy and to evaluate its value in the diagnosis of obstructive salivary gland diseases and preoperative visualization of endoluminal views.. Study Design: This study presents our initial experience to use MR virtual endoscopy for the presurgical visualization of salivary duct lumen and ductal pathologies in comparison to the sialoendoscopy findings in a feasibility study.. Methods: Six consecutive patients with suspected obstructive salivary gland diseases underwent MR sialography with a three-dimensional fast imaging using steady-state acquisition. The three-dimensional MR data were transferred to an independent workstation and were postprocessed with navigator software to generate three-dimensional reconstruction and virtual endoscopic images. The fly-through mode was used to imitate the sialoendoscopic ...
Metaplasia is a change in which one terminally differentiated cell type is replaced by another cell type of the same germ line. Usually, a specialized type of epithelium is replaced by less specialized type of epithelium. Metaplasia is often, but not always, an adaptive change that occurs in response to repeated epithelial damage and is therefore often accompanied by other lesions such as inflammation or necrosis. Squamous metaplasia may be reversible if the cause is removed. Squamous metaplasia is a common form of metaplasia, presumably because squamous epithelium is more resistant to damage than other forms of epithelia. Squamous metaplasia is usually the result of chronic irritation, but it can have other causes (e.g., hypovitamnosis A). In the salivary ducts, metaplasia of the normally cuboidal ductal epithelium to stratified squamous epithelium has been seen in response to chemicals, ionizing radiation, viral infections, vitamin A deficiency, and blockage of ducts by salivary calculi. ...

Results of this study indicate that arctic animals possess the intralobular heterogeneity of vitamin A-storage capacity in the hepatic stellate cells.. In earlier studies, a specific zonality for intralobular vitamin A-storage was reported. Although our results agree with other reports as to the presence of the heterogeneity of vitamin A-storing lipid droplets in the hepatic stellate cells in the liver lobule [4, 5], the location of the highest-storage zone disagree with these reports. This discrepancy might be explained by the observation methods used in the earlier studies. Whereas those authors used only the light microscopical method [4, 5], in the present study we examined all tissues by electron microscopy. Moreover, we certified that the tendency of vitamin A storage observed in liver lobules by light microscopy strongly supported that of intralobular heterogeneity revealed by electron microscopical morphometry.. The existence of the intralobular heterogeneity of vitamin A storage would ...

having been able to make milk out of grass. we have, secondly, the organ for its reception and retention; we have, thirdly, the excretory duct, annexed to it; and we have, lastly, the determination of the milk to the breast, at the particular juncture when it is about to be wanted. We have all these properties in the subject before us; and they are all indications of design. The last circumstance is the strongest of any. If I had been to guess beforehand, I should have conjectured, that at the time when there was an extraordinary demand for nourishment in one part of the system, there would be the least likelihood of a redundancy to supply another part. The advanced pregnancy of the female has no intelligible tendency to fill the breast with milk. Head and neck cancers are common in several regions of the world and the treatment usually includes radiotherapy. This treatment can generate adverse effects to the salivary flow, with a relationship between the dose and the damage caused. Salivary gland cells are highly permeable to water and therefore, they express aquaporins (AQPs). This study analyzed changes in the expression and location of these proteins and identified morphological changes induced by low radiation in rat submandibular gland. Female rats were divided into control and irradiated groups. Immunohistochemistry analysis allowed confirming the presence of AQP1 in the blood vessel endothelium. Intense and steady labelling granules were also observed in the cytoplasm of submandibular gland ductal cells. In addition, there was AQP5 positive labelling in ductal cells delimiting the lumen of intercalated duct, in the cytoplasm and membrane of acinar cells. Finally, the decrease of AQP labelling in irradiated animal glands validated ...
A stricture in a salivary gland duct is essentially a narrowing of the duct. When salivary flow is stimulated the duct may be too narrow to allow it to pass through quickly and act as a bottle neck. The saliva builds up and causes a swelling of the salivary gland in a similar way to a stone causing a local blockage. The swelling usually settles within an hour or so.. Strictures often affect the parotid gland and can be treated by stretching the duct with an endoscope or a balloon. This is usually carried out under a local anaesthetic.. Both treatment of stones and strictures can often be managed by the use of sialdensocopy which is a specialist procedure undertaken regularly by very few surgeons in the UK. ...

The esophagus is schematically characterized by the same wall structure as all other sections of the gastrointestinal tract. There is a submucosal layer of connective tissue, and a muscular layer (muscularis) provides for the targeted transport of the food to the stomach via contractions of the smooth muscles (peristalsis). The mucosa is composed of a multi-layered, non-keratinized [[squamous epithelium], where the excretory ducts of the esophageal glands are spread in. While the muscularis consists of striated muscles in the upper third, the lower third only consists of smooth muscles. In the middle third, you can find striated muscles as well as smooth muscles, while the proportion of smooth muscles grows toward aboral direction. ...

Results of three-dimensional steady flow calculations are compared with existing pressure and velocity-measurements of two manifold-type junctions. The junctions consist of a main duct and a side branch, both with the same rectangular cross section, with the side branch joining the main duct at an angle of either 90 or 45 degrees. Both combining and dividing flow configurations are considered for different total mass flow rates and different side-branch-to-main-duct mass flow ratios. One objective of this investigation was to assess the effects of numerical differencing scheme and mesh refinement on solution accuracy, and both parameters showed strong influences on the computed results. It is shown that calculations should be made with the highest possible level of numerical accuracy and grid resolution in regions of flow recirculation.Comparisons of computed and measured velocities, static pressures, and flow loss coefficients are presented in this paper. For most cases considered, the model ...
You might be seeing members in your family falling sick but not able to recognise the cause of their sickness.
