Effects of supplementation with ruminal epithelial cells on fiber-degrading activity and cell growth of Ruminococcus albus (R. albus, strain 7) was tested using a basal substrate of rice straw and formulated concentrate. Cultures of R. albus alone and R. albus with rumen protozoa were grown at 39째C for 48 h with an 8.4% crude protein (CP) substrate, 33% of the CP supplemented with either ruminal epithelial cells or defatted soybean meal. The ruminal epithelial cells had lower amounts of rumen soluble and degradable protein fractions as compared to defatted soybean meal, as determined by an enzymatic method, and the same was found with amino acid composition of protein hydrolysates. Ruminal epithelial cells were directly utilized by the R. albus, and resulted in greater growth of cell-wall free bacteria compared to defatted soybean meal. The effect of epithelial cells on bacterial growth was enhanced by the presence of rumen protozoa. In consistency with cultures of R. albus and R. albus with ...
In this study we hypothesised that dysbiosis is important not only in CD but also in UC pathophysiology and that this dysbiosis may partly overlap with the dysbiosis observed in CD. We therefore evaluated if the predominant microbial signature as we previously described in CD is also present in UC.24 The CD-associated signature was characterised by five bacterial species, namely a decrease in F prausnitzii, B adolescentis, D invisus and an uncharacterised species of Clostridium cluster XIVa, and an increase in R gnavus. In contrast, when we evaluated the culturable species F prausnitzii, B adolescentis and R gnavus in the present UC cohort, we did not find the same signature in UC patients based on DGGE analysis. Our study has shortcomings including the mixed patient population, a potential impact of differences in duration of therapy on the microbiota and the lack of information on dietary aspects. It might be possible that disease-relevant species could be missed using DGGE instead of ...
Faecalibacterium prausnitzii is an anti-inflammatory commensal bacterium identified on the basis of human clinical data. The mechanisms underlying its beneficial effects are still unknown. Gnotobiotic mice harboring F. prausnitzii (A2-165) and Escherichia coli (K-12 JM105) were subjected to 2,4,6-trinitrobenzenesulfonic acid (TNBS)-induced acute colitis. The inflammatory colitis scores and a gas chromatography-time of flight (GC/TOF) mass spectrometry-based metabolomic profile were monitored in blood, ileum, cecum, colon, and feces in gnotobiotic mice. The potential anti-inflammatory metabolites were tested in vitro. We obtained stable E. coli and F. prausnitzii-diassociated mice in which E. coli primed the gastrointestinal tract (GIT), allowing a durable and stable establishment of F. prausnitzii. The disease activity index, histological scores, myeloperoxidase (MPO) activity, and serum cytokine levels were significantly lower in the presence of F. prausnitzii after TNBS challenge. The ...

A set of seven new oligonucleotide probes is presented to extend the set of probes for the predominant microbiota of the human gastrointestinal tract and in particular for fecal bacteria. Five of the seven probes each detected an average of more than 1% of the total bacterial microbiota. Especially the probes for the ruminococci and related Clostridium group IV bacteria are valuable, since these probes detected 10% of the fecal bacteria, and this group of bacteria is likely to have interesting metabolic features, such as degradation of complex carbohydrates. The numbers of the ruminococcus group are within the range earlier estimated in human feces by culture techniques (7), in which a mean of 1.6 × 1010 g−1 (dry weight) was found. Also the numbers of the Eubacterium cylindroides group are within the reported ranges. The numbers of C. innocuum, E. cylindroides, and Eubacterium dolichum enumerated previously showed a large variation between the subjects with mean numbers of 4 × 108, 4 × 109, ...
21st March 2015 Clostridia: who are they ? The clostridia or Clostridiales, with Clostridium and other related genera, are Gram-positive sporulating bacteria. They are obligate anaerobes, and belong to the taxonomic phylum Firmicutes. This phylum includes clostridia, the aerobic sporulating Bacillales (Bacillus, Listeria, Staphylococcus and others) and also the anaerobic aero-tolerant Lactobacillales (id est, lactic…
This is one of the shells of a Productus, a common type of brachiopod. This bed of the Petershill limestone is stuffed full of these brachiopods, among other fossils. See [[[3670310]]].
The composition of the fecal microbiota of UC patients differs from that of healthy individuals: we found a reduction in R hominis and F prausnitzii, both well-known butyrate-producing bacteria of the Firmicutes phylum. These results underscore the importance of dysbiosis in IBD but suggest that dif …
It has been suggested that increased risk for chronic inflammatory diseases associated with C-section is a result of the long-term dysbiosis observed in children delivered by C-section (reviewed in Ref. 39). In this study, we demonstrated in a murine model that mode of delivery had a major impact on the early composition of the gut microbiota in mice, as analysis by 16S rRNA gene sequencing confirmed a separate clustering of the vaginally delivered and C-section pups. A distinct bacterial profile enriched in especially Bacteroides and Lachnospiraceae and with lower abundance of Rikenellaceae and Ruminococcus was characteristic for the C-section pups. On the contrary, C-section has been shown to cause decreased colonization rate of Bacteroides and increased prevalence of Clostridia in humans (40). These differences may, besides genetic differences, be because of the initial colonization pattern being very susceptible to environmental changes. When human babies are not exposed to maternal vaginal ...

DNA from the batch culture fluid and rumen contents in the feeding study was extracted and purified using the repeated bead beating plus column method [19] with a DNA stool mini kit (QIAamp DNA Stool Mini Kit; Qiagen, Hilden, Germany). The purified DNA was quantified using a NanoDrop 2000 (Thermo Fisher Scientific, Waltham, MA, USA) and diluted to 10 ng/μL before being subjected to polymerase chain reaction (PCR) amplification. The diluted DNA was used for quantitative real-time PCR (qPCR) to monitor the abundance of rumen representatives including total bacteria, total archaea, fungi, protozoa, Fibrobacter succinogenes (F. succinogenes), Ruminococcus albus (R. albus), Ruminococcus flavefaciens (R. flavefaciens), genus Prevotella, Prevotella bryantii, Prevotella ruminicola (P. ruminicola), Anaerovibrio lipolytica (A. lipolytica), and Butyrivibrio group. Details of qPCR such as primers, standards, PCR conditions, and calculations were as described by Koike et al [20] and Ohene-Adjei et al [21]. ...
The nucleotide positions of the target site for the forward primer on T. bryantii 16S rRNA gene sequences were 380-400 while those of the reverse primer were 934-953, yielding a 575-bp PCR product. The primer set was designed to cover all rumen Treponema and named g-TrepoF. The online basic local alignment search tool (blast) program (http://blast.ncbi.nlm.nih.gov/Blast.cgi) was used to determine the specificity of the forward primer.. The specificity of the primers was further tested by PCR amplification using genomic DNA from pure cultures of 16 representative rumen bacterial strains including T. bryantii ATCC33254, F. succinogenes ATCC19169, Ruminococcus albus 8, Ruminococcus flavefaciens C94, Prevotella ruminicola 23, Prevotella bryantii B14, Prevotella brevis GA33, Butyrivibrio fibrisolvens. The physiological responses of unfed and postprandial red rock crabs (Cancer productus J.W. Randal, 1840) were investigated during periods of emersion. During aerial exposure, oxygen uptake quickly fell to very low levels and was no longer detectable in the haemolymph after 12 h. The resulting anaerobic respiration led to a build up in lactic acid and the resulting acidosis was more pronounced in the postprandial crabs. There was also a concomitant rise in PCO2 and CCO2, and in both cases these were higher in postprandial animals. Higher ammonia levels in postprandial crabs showed that cellular activities were still proceeding anaerobically, suggesting that although crabs can delay mechanical digestion during emersion, once intracellular digestion occurs they may be committed to these processes. Increased mortality rates of postprandial animals were probably due to a combination of the high lactate and CO2 levels coupled with an increased ammonia concentration. For C. productus stranded in the ...
Objective Bacteria play an important role in the onset and perpetuation of intestinal inflammation in inflammatory bowel disease (IBD). Unlike in Crohns disease (CD), in which dysbiosis has been better characterised, in ulcerative colitis (UC), only small cohorts have been studied and showed conflicting data. Therefore, we evaluated in a large cohort if the microbial signature described in CD is also present in UC, and if we could characterise predominant dysbiosis in UC. To assess the functional impact of dysbiosis, we quantified the bacterial metabolites.. ...

A few weeks ago (June 2012), a paper in Nature by a group of researchers suggested that despite the vast geographical and nutritional differences in the human population, that just three predominant bacterial clusters (referred to as enterotypes hereafter) could explain all of our gastric microbial mixes.[1] This they suggest indicates the existence of a limited number of well-balanced host-microbial symbiotic states that might respond differently to diet and drug intake.. Each of these three enterotypes are identifiable by the variation in the levels of one of three genera: Bacteroides (enterotype 1), Prevotella (enterotype 2) and Ruminococcus (enterotype 3). These enterotypes are not as sharply delimited as, for example, human blood groups; they are, in contrast, densely populated areas in a multidimensional space of community composition. They are nevertheless likely to characterise individuals, in line with previous reports that gut microbiota are quite stable in individuals and can even be ...

consuming timothy hay and a commercial concentrate (60:40, w/w) twice daily (09:00 and 17:00) at 2% of body weight with free access to water and mineral block were used as rumen fluid donors. In vitro fermentation experiment, with timothy hay as substrate, was conducted for up to 72 h, with E. stolonifera extract added to achieve final concentration 1%, 3%, and 5% on timothy hay basis. Results: Administration of E. stolonifera extract to a ruminant fluid-artificial saliva mixture in vitro increased the total gas production. Unexpectedly, E. stolonifera extracts appeared to increase both methane emissions and hydrogen production, which is contrasts with previous observations with brown algae extracts used under in vitro fermentation conditions. Interestingly, real-time polymerase chain reaction indicated that as compared with the untreated control the ciliate-associated methanogen and Fibrobacter succinogenes populations decreased, whereas the Ruminococcus flavefaciens population increased as a ...
The authors state in the results: Ten human subjects consumed crackers for three weeks each containing either RS2, RS4, or native starch in a double-blind, crossover design. Multiplex sequencing of 16S rRNA tags revealed that both types of RS induced several significant compositional alterations in the fecal microbial populations, with differential effects on community structure. RS4 but not RS2 induced phylum-level changes, significantly increasing Actinobacteria and Bacteroidetes (+++)while decreasing Firmicutes(-). At the species level, the changes evoked by RS4 were increases in Bifidobacterium adolescentis and Parabacteroides distasonis, while RS2 significantly raised the proportions of Ruminococcus bromii and Eubacterium rectale when compared to RS4. The population shifts caused by RS4 were numerically substantial for several taxa, leading for example, to a ten-fold increase in bifidobacteria in three of the subjects, enriching them to 18-30% of the fecal microbial community. The ...
Coprococcus (ATCC 27759) is a genus of anaerobic cocci which are all part of the human faecal flora, but rarely seen in human clinical specimens. Coprococcus includes those gram-positive, anaerobic cocci that actively ferment carbohydrates, producing butyric and acetic acids with formic or propionic and/or lactic acids. Fermentable carbohydrates are either required or are highly stimulatory for growth and continued subculture. - Lillian V. Holdeman & W. E. C. Moore. The genus is bio-chemically closely related to Ruminococcus, and phylogenetically to the genus Lachnospira. Coprococcus eutactus is an obligately anaerobic, nonmotile, gram-positive coccus occurring in pairs or chains of pairs. Cells may lose colour readily and acquire a slightly elongate shape in a medium containing a fermentable carbohydrate, but are normally round, and 0.7 to 1.3 µm in diameter. Coprococcus may be used as a microbial biomarker to assess the health of the human gastro-intestinal tract. Gut microorganisms ...

The European Union is facing new challenges. Its population is ageing fast and recent enlargements widened the health gaps between Member States, stated European Union Health Commissioner Markos Kyprianou. Migration, the mobility of health professionals and patients, the risks posed by pandemics and bioterrorism, all represent additional challenges. So do globalisation and climate change. The time has come for the European Union to develop a strong and comprehensive strategy on health.. The European Commission underlines the importance of research to the strategy, noting that health policy must be based on the best scientific evidence derived form sound data and information, and relevant research.. Research has an important role to play in achieving the three key objectives of the strategy. For example, our ageing population means that the development of geriatric medicine needs to be actively promoted, and we need to improve our understanding of neurodegenerative diseases such as ...
Fermentation of legume fodder tree leaves by rumen microorganisms was evaluated. The substrates were sun-dried, ground leaves. Gas and volatile fatty acid (VFAs) production were estimated. Using gas production as an index of fermentation at 12 h, the leaves tested ranked as follows; Chamaecytisus palmensis,Gliricidia sepium,Sesbania sesban,Tephrosia bracteolata,Leucaena pallida,Vernonia amygdalina,Acacia sieberiana,Sesbania goetzei,Acacia angustissima. Using VFA production, the ranking was as follows; G. sepium,S. sesban,S. goetzei,L. pallida,C. palmensis/amygdalina,T. bracteolata, A. sieberiana,A. angustissima. Absolute gas or VFA production rates, were also used to rank the leaves. Extracts (70 percent acetone) of A. angustissima inhibited the growth of Ruminococcus albus 8, R. flavefaciens FD-1, Prevotell aruminicola D3ID and Streptococcus bovis JBI while the growth of Selenomonas ruminantium D was depressed when 0.6 ml extracts were added. Tryptamine, also known as TrpN, is a catabolite of tryptophan converted by the gut microbiota. After absorption through the intestinal epithelium, tryptophan catabolites enter the bloodstream and are later excreted in the urine. Both Clostridium sp. and Ruminococcus sp. have been found to convert tryptophan into tryptamine (PMID: 30120222 ). Tryptamine is a monoamine compound that is a common precursor molecule to many hormones and neurotransmitters. Biosynthesis generally proceeds from the amino acid tryptophan, with tryptamine acting as a precursor for other compounds. Substitutions to the tryptamine molecule give rise to a group of compounds collectively known as tryptamines. The most well-known tryptamines are serotonin, an important neurotransmitter, and melatonin, a hormone involved in regulating the sleep-wake cycle. Tryptamine has been detected, but not quantified in, several different foods, such as onion-family vegetables, acerola, Japanese walnuts, custard apples, and green zucchinis. ...
Ayurveda has a rich history and its significance woven deeply in the Indian culture. The concept of prakriti (a persons nature or constitutional type determined by the proportion of three doshas, namely - vata, pitta and kapha) in Ayurveda is deeply rooted in personalized health management. While the attributes of prakriti has been established to have a genomic basis, there is dearth of elaborate evidences linking prakriti with manifestation of diseases. Next generation sequencing studies have provided a causal link between variation in the gut microbiome and its effect on an individuals fitness. Separately, reports have identified gut microbial patterns associated with several host variables such as geography, age, diet and extreme prakriti phenotypes. Recently, few reports have identified a core gut microbiome consisting of Bacteroides, Faecalibacterium, Prevotella and Ruminococcus prevalent across the Indian population; however, a few bacterial genera were specifically enriched in ...

Capable tools are desired for urban spatial policies planning in China to safeguard its sustainable development strategy. This study develops an activity-based Land Use/Transport Interaction (LUTI) model to forecast the urban activity impacts of the land-use policies. Essentially, its endogenized and interactive features in residential and employment distribution modeling mark it out from the traditional Lowry models. The LUTI model proposed consists of four models, i.e., a transport sub-model, a residential location model, an employment location model and a real estate rent model. It is then applied to the Beijing metropolitan area to characterize the urban activity evolution trend under the land use policies of recent years. The results show that with the increasing number of floorspace developed on the outskirts, more residents and employers are relocating there and sub-centers are formed to divide the service of central Beijing. This trend is consistent with the objective of government ...
