Small, non-coding Y RNAs are abundant vertebrate RNAs. All four Y RNAs have an essential role in DNA replication and display functional redundancy. As Y RNAs are enriched in human solid tumours, understanding their molecular mechanism during replication and cell proliferation is of high importance. In this issue (p. 1239) Torsten Krude and Eyemen Kheir investigate the expression levels and dynamic association between Y RNAs and chromatin during the cell cycle of cancer and non-cancer cell lines. Background To our knowledge there are no studies assessing anti-Ro/SSA and anti-La/SSB autoantibodies in a large population of childhood-systemic lupus erythematosus (cSLE) patients. Methods This was a retrospective multicenter cohort study performed in 10 Pediatric Rheumatology services, São Paulo state, Brazil. Anti-Ro/SSA and anti-La/SSB antibodies were measured by enzyme linked immunosorbent assay (ELISA) in 645 cSLE patients. Results Anti-Ro/SSA and anti-La/SSB antibodies were evidenced in 209/645 (32%) and 102/645 (16%) of cSLE patients, respectively. Analysis of cSLE patients with and without anti-Ro/SSA antibodies revealed higher frequencies of malar rash (79% vs. 71%, p = 0.032), photosensitivity (73% vs. 65%, p = 0.035), cutaneous vasculitis (43% vs. 35%, p = 0.046) and musculoskeletal involvement (82% vs. 75%, p = 0.046) in spite of long and comparable disease duration in both groups (4.25 vs. 4.58 years, p = 0.973). Each of the two protein heterodimers purified from SRP elicited a substantial change in fluorescein emission upon association with the modified RNA. The binding of SRP9/14 to singly-labeled SRP RNA-Fl increased fluorescein emission intensity by 41% at pH 7.5 and decreased its anisotropy from 0.18 to 0.16. There may be some significant variations in HEp-2 immunofluorescence assay (IFA) staining depending on the manufacturer of HEp-2 cell slides employed. Some autoantigens and epitopes may not be available in some cell preparations. To ensure that the HEp-2 slide in use can detect anti-Ro60/SS-A and anti-La/SS-B staining, appropriate reference materials, such as those from www.AutoAb.org, should be used to verify the HEp-2 slide and other reagents employed in the assay. Experts in the field also advise to have low titer positive controls to test each new batch of HEp-2 slides for appropriate sensitivity.
