N2 - Riboflavin (vitamin B2) is the precursor of flavin mononucleotide (FMN) and flavin adenine dinucleotide, which are essential coenzymes in all free-living organisms. Riboflavin biosynthesis in many Bacteria but not in Archaea is controlled by FMN-responsive riboswitches. We identified a novel bifunctional riboflavin kinase/regulator (RbkR), which controls riboflavin biosynthesis and transport genes in major lineages of Crenarchaeota, Euryarchaeota and Thaumarchaeota. RbkR proteins are composed of the riboflavin kinase domain and a DNA-binding winged helix-turn-helix-like domain. Using comparative genomics, we ...

Purpose: Infectious keratitis is a potentially sight-threatening condition which may cause severe visual loss. Antimicrobial treatment does not always guarantee rapid and complete healing, since the development of resistance to antimicrobial agents and misdiagnosis of fungal keratitis contribute to an increased rate of refractory keratitis. Recently, the antimicrobial properties of CXL against common pathogens in vitro, and CXL has been successfully used in the treatment of refractory infectious keratitis with corneal melting. In the present report we provide evidence that fluorescein competes riboflavin in UV-A absorption during CXL.. Methods: We analyzed the UVA-absorption of riboflavin and fluorescein by spectrophotometry (Lightwave S2000, Biochrom WPA, Cambridge, UK). The following concentrations were tested: 0.1%, 0.01%, 0.001% for riboflavin and 10%, 1%, 0.1%, 0.01% for fluorescein. NRS1 strains Network on Antimicrobial Resistance in Staphylococcus aureus were used in all experiments. ...
Corneal collagen cross-linking with riboflavin (vitamin B2) and UV-A light is a surgical treatment for corneal ectasia such as keratoconus, PMD, and post-LASIK ectasia. It is used in an attempt to make the cornea stronger. According to a 2015 Cochrane review, there is insufficient evidence to determine if it is useful in keratoconus. In 2016, the US Food and Drug Administration approved riboflavin ophthalmic solution crosslinking based on three 12-month clinical trials. A 2015 Cochrane review that looked at all high quality evidence on corneal collagen cross-linking found that it was insufficient to determine if it is useful in keratoconus. Among those with keratoconus who worsen CXL may be used. In this group the most common side effects are haziness of the cornea, punctate keratitis, corneal striae, corneal epithelium defect, and eye pain. In those who use it after post-LASIK ectasia, the most common side effects are haziness of the cornea, corneal epithelium defect, corneal striae, dry eye, ...

1.3 mg/day. Therefore, if the PD patients had a normal absorptive capacity for vitamin B2, their large ingestion of red meat (up to 700 g/day), associated with milk, rice and beans, fruits and vegetables, should have provided a normal riboflavin status. In contrast, 31 consecutive PD patients had laboratory evidence for riboflavin deficiency (Table 2) suggesting that patients with sporadic PD belong to the subset of the general population (10-15%) (3) that may express a flavokinase with low affinity for vitamin B2, leading to a decreased absorption.. However, the digestion of red meat releases hemin, a highly diffusible toxin that, when not properly inactivated, increases intracellular iron concentrations and enhances hydroxyl radical production (Fenton reaction). Most of the absorbed hemin is destroyed by the enzyme heme oxygenase (HO) in the digestive tract and liver (26). Because HO is oxidized during the catabolization of hemin to biliverdin, the HO molecules must be reduced through the ...
Riboflavin deficiency is usually associated with a low intake of milk and animal proteins. Pasteurization of milk results in an appreciable loss of this vitamin. Ectodermal tissues such as skin, nervous system and eye are the most vulnerable to its lack. Clinically riboflavin deficiency is seen usually along with the deficiency of multiple factors such as in poorer population and in alcoholics. It is also seen in newborn infants subjected to phototherapy for hyperbilirubinemia. Breast fed infants are more liable to show deficiency because of lower riboflavin content of human milk. Its deficiency is also seen in patients on chronic peritoneal dialysis and hemodialysis. Following are the features seen with its deficiency.. ...

NICE encourages further research into CXL using riboflavin and UVA for keratoconus and keratectasia, especially epithelium-on (transepithelial) CXL and the combination (CXL-plus) procedures. Details of the techniques used should be clearly described. Reported outcomes should include visual acuity, corneal topography and quality of life. Data on long-term outcomes for all types of CXL using riboflavin and UVA for keratoconus and keratectasia would be useful - specifically data about prevention of progression to corneal transplantation and about repeat procedures and their efficacy ...

As co-enzymes, the B vitamins are essential components in most major metabolic reactions. They play an important role in energy production, including the metabolism of lipids, carbohydrates, and proteins. B vitamins are also important for blood cells, hormones, and nervous system function. As water-soluble substances, B vitamins are not generally stored in the body in any appreciable amounts. Thiamin and riboflavin are all essential coenzymes in energy production. Thiamin is converted quickly into thiamin pyrophosphate, which is required for glycolytic and Krebs cycle reactions. Thiamin also appears to be related to nerve impulse transmission. Riboflavin is a component of the coenzymes FAD and FMN, which are intermediates in many redox reactions, including energy production and cellular respiration reactions. Vitamin B-6 is a coenzyme in amino acid metabolism. It is necessary for the metabolism of homocysteine and the conversion of tryptophan into niacin. Vitamin B-6 dependent enzymes are also ...

Riboflavin synthase (RS-α) catalyzes the biosynthesis of riboflavin (vitamin B2) by dismutation of 6,7-dimethyl-8-(1-D-ribityl)lumazine (Lum) (EC 2.5.1.9). Riboflavin synthases of bacteria and fungi are structurally and evolutionary related to antenna proteins involved in bioluminescence of marine bacteria [1,2]. These proteins seem to have evolved from the duplication of a domain of about 100 residues, the lumazine-binding repeat. The 3D structure of RS-α, which is an asymmetric homotrimer, shows that both domains form a 6-stranded antiparallel β-barrel (see ,PDB:1PKV,) [3], while a C-terminal helix is involved in trimerization. The Lum-binding domain of RS-α forms two Greek-key folds with the topology BBHBBBHB, where most of the substrate binding sites are located in β-strands (B) 4 and 5 and in helix (H) 2 [3,4,5]. Some proteins known to contain a lumazine-binding repeat: ...

Corneal Collagen Crosslinking with Riboflavin (also known as C3R) is a procedure that has been shown to strengthen the weak corneal structure. The cornea is made up of many layers of collagen arranged in a very regular pattern with cross links for strength. C3R works by increasing collagen cross-linking thereby strengthening the cornea. Studies have shown the cornea is strengthened by over 300% following this procedure.. During the procedure, riboflavin eye drops are applied to the cornea. The surgeon then uses UV light to activate the riboflavin, inducing the strengthening cross-links in the cornea. This is a short procedure performed as a day-case and is offered if the keratoconus is progressive. The aim of treatment is to prevent any further deterioration. ...

Comparative map before and after CXL-plus (trans-PTK-PRK + accelerated CXL). SCVA improved from 20/50 to 20/25. Epi-off corneal crosslinking (CXL) is a proven treatment for corneal ectatic disease. It halts the ectatic process by strengthening the cornea by means of creating covalent bonds between collagen fibrils. In the last three years many efforts have been made by researchers and ophthalmic companies to improve the quality and the time of treatment through development of new riboflavin solutions and new irradiance protocols.. However, even though the main target of halting corneal ectasia is reached in most cases, few patients affected by moderate to severe keratoconus have a consistent visual acuity (VA) improvement. When performed on its own, the CXL procedure is not intended to improve vision, but rather to stabilise disease, and therefore the patient still remains to be rehabilitated visually, thereby affecting their quality of life. Many face the need to wear customised contact lenses ...
The chicken riboflavin carrier protein (RCP) is an estrogen induced egg yolk and white protein. Eggs from hens which have a splice mutation in RCP gene fail to hatch, indicating an absolute requirement of RCP for the transport of riboflavin to the oocyte. In order to understand the mechanism of regulation of this gene by estrogen, the chicken RCP gene including 1 kb of the 5′ flanking region has been isolated. Characterization of the gene structure shows that it contains six exons and five introns, including an intron in the 5′ untranslated region. Sequence analysis of the 5′ flanking region does not show the presence of any classical, palindromic estrogen response element (ERE). However, there are six half site ERE consensus elements. Four deletion constructs of the 5′ flanking region with varying number of ERE half sites were made in pGL3 basic vector upstream of the luciferase-coding region. Transient transfection of these RCP promoter deletion constructs into a chicken hepatoma cell ...

Clone REA654 recognizes the human variable β2 subunit of the α/β T cell receptor (TCR Vβ2). The TCR is a heterodimeric glycoprotein associated with the CD3 antigen. The α and β TCR chains are composed of constant and variable regions, each encoded by distinct gene segments. TCR Vβ2 is a variant of the TCR β chain. α/β T cells express a diverse α/β TCR repertoire that specifically co-recognizes self or foreign antigen bound to antigen-presenting molecules, which thereby leads to T cell-mediated immunity. For example, the TCR can directly bind to peptide fragments, riboflavin precursors, and lipid antigens that are presented by major histocompatibility complex (MHC) molecules, MR1 and CD1, respectively. In each case, the antigen sits within the antigen-binding cleft, whereupon the TCR recognizes a composite surface formed by the antigen-presenting molecule and surface-exposed regions of the antigen itself. This co-recognition paradigm is a central tenet of α/β T cell-mediated immunity and

Brown-Vialetto-Van Laere syndrome represents a phenotypic spectrum of motor, sensory, and cranial nerve neuropathy, often with ataxia, optic atrophy and respiratory problems leading to ventilator-dependence. Loss-of-function mutations in two riboflavin transporter genes, SLC52A2 and SLC52A3, have recently been linked to Brown-Vialetto-Van Laere syndrome. However, the genetic frequency, neuropathology and downstream consequences of riboflavin transporter mutations are unclear. By screening a large cohort of 132 patients with early-onset severe sensory, motor and cranial nerve neuropathy we confirmed the strong genetic link between riboflavin transporter mutations and Brown-Vialetto-Van Laere syndrome, identifying 22 pathogenic mutations in SLC52A2 and SLC52A3, 14 of which were novel. Brain and spinal cord neuropathological examination of two cases with SLC52A3 mutations showed classical symmetrical brainstem lesions resembling pathology seen in mitochondrial disease, including severe neuronal ...

6,7-Dimethyl-8-ribityllumazine is the biosynthetic precursor of riboflavin, which, as a coenzyme, plays a vital role in the electron transfer process for energy production in all cellular organisms. The enzymes involved in lumazine biosynthesis have been studied in considerable detail. However, the conclusive mechanism of the reaction catalyzed by lumazine synthase has remained unclear. Here, we report four crystal structures of the enzyme from the hyperthermophilic bacterium Aquifex aeolicus in complex with different inhibitor compounds. The structures were refined at resolutions of 1.72 Angstrom, 1.85 Angstrom, 2.05 Angstrom and 2.2 Angstrom, respectively. The inhibitors have been designed in order to mimic the substrate, the putative reaction intermediates and the final product. Structural comparisons of the native enzyme and the inhibitor complexes as well as the kinetic data of singlesite mutants of lumazine synthase from Bacillus subtilis showed that several highly conserved residues at ...
Investigators at Great Ormond Street Hospital, London, UK, and multiple centers internationally report the response to high-dose oral riboflavin therapy in 18 patients from 13 families with mutations in SLC5ZA2, encoding riboflavin transporter RTVT2, a new causative gene for Brown-Vialetto-Van Laere syndrome (BVVLS), a progressive neurodegenerative disorder leading to death in childhood. BVVLS is characterized by cranial neuropathies, pontobulbar palsy, sensorimotor neuropathy manifesting with sensory ataxia, weakness of upper limbs and axial muscles, with preserved strength of lower limbs, optic atrophy, sensorineural hearing loss, and respiratory insufficiency. Riboflavin therapy resulted in significant sustained clinical and biochemical improvement in 2 patients and preliminary response in 13 patients. [1]. COMMENTARY. BVVLS is a similar disorder to Fazio Londe syndrome caused by subtly different mutations of the same gene, and with the additional clinical feature of sensorineural deafness ...
This chapter reviews current information on the nature and regulation of the genes involved in the synthesis of riboflavin, biotin, folic acid, thiamine, lipoic acid, and pantothenic acid in Bacillus subtilis, and of cobalamin in Bacillus megaterium. More is known about riboflavin (rib) genes in B. subtilis than in any other microorganism. It is possible that flavin mononucleotide (FMN) itself directly regulates riboflavin gene expression, since various nucleotides, including flavin mononucleotides and riboflavin, are reported to bind specifically to RNA aptamers. Folic acid derivatives, such as tetrahydrofolate, are required as coenzymes in numerous one-carbon-atom transfer reactions in B. subtilis and other organisms. The major folic acid biosynthetic operon, located at 7°, encodes nine genes, six of which are required for folate synthesis. Cobalamin (vitamin B12) is the most structurally complex molecule of all the vitamins synthesized by bacteria. More than 30 biosynthetic genes are required for

Corneal collagen crosslinking (CXL) is a minimally invasive procedure that is currently supposed to arrest the progression of corneal ectatic conditions. CXL induces crosslinks in the stromal collagen of the cornea.
The mean Kmax value of the KeraVio-treated eyes decreased by 0.81 D at 6 months, although the value had increased by 6.03 D in the year before baseline. Thus, our study demonstrates that KeraVio has a beneficial effect on corneal topography in patients with corneal ectasia over the course of 6 months. Kmax, believed to be an important indicator of CXL success, decreased significantly after the procedure. Previous studies reported a reduction of 1-2 D in the mean Kmax of treated keratoconic eyes at 1 year.15 27 Ferdi et al 28 reported a systematic review and meta-analysis of keratoconus natural history data, including 11 529 eyes from 41 publications. Their meta-analysis of Kmax demonstrated a significant increase of 0.7 D at 12 months. Although no untreated control group was included in this pilot study, we believe that the efficacy of KeraVio was demonstrated by the stabilisation of progressive corneal disorder. In our study, the Kmax decreased by 2.0 D or more in four eyes (20%) and increased ...

Odds are we don t all have the same underlying issue. My dentist & I think my case was viral due to the timing of the onset and the dramatic response to the L-lysine. The research on herpes & lysine suggest that it s the balance of arginine & lysine which impacts viral replication. High arginine to lysine encourages replication. High lysine to arginine stops replication. Peeling lips can also be symptom of riboflavin deficiency.
