The gene encoding a thermostable cellulase of family 12 was previously isolated from a Rhodothermus marinus through functional screening. CelA is a protein of 260 aminoacyl residues with a 28-residue amino-terminal signal peptide. Mature CelA was poorly synthesized in some Escherichia coli strains and not at all in others. Here we present an alternative approach for its heterologous production as a secreted polypeptide in Streptomyces. CelA was successfully over-expressed as a secreted polypeptide in Streptomyces lividans TK24. To this end, CelA was fused C-terminally to the secretory signal peptide of the subtilisin inhibitor protein (Sianidis et al. in J Biotechnol. 121: 498-507, 2006) from Streptomyces venezuelae and a new cloning strategy developed. In the present study, TreZ from Zunongwangia sp. was isolated and expressed in E. coli BL21 (DE3). The optimum temperature for TreZ is 50 °C, which is higher than that of many previously reported trehalases, such as those in Apis mellifera L 7 °C [22], Rhodotorula rubra 30 °C [23], Saccharomyces cerevisiae 40 °C [24], and Rhizopus microsporus var. rhizopodiformis 45 °C [25]. This paper deals with mechanical fault monitoring in induction machines from stator current measurements. The considered faults lead to amplitude and/or phase modulations of the measured stator current. The different faults can be characterized by their time-frequency signatures via the Wigner distribution. The time-frequency representations apply to complex signals that may be obtained through the Hilbert transform of the real measured signal. In case of high frequency modulations, it can not be taken advantage of the time frequency signatures. This study proposes an alternative complex signal representation for modulated stator currents. The so-called space vector is obtained through the Concordia transform. From three stator current measurements, the Concordia transform builds a complex vector which conveniently carries the information about phase and amplitude modulations. On Mars, significant amounts (0.4 - 0.6 %) of perchlorate ions were detected in dry soil by the Phoenix Wet Chemistry Laboratory and later confirmed with the Mars Science Laboratory. Therefore, the ability of Hydrogenothermus marinus, a desiccation tolerant bacterium, to survive and grow in the presence of perchlorates was determined. Results indicated that H. marinus was able to tolerate concentrations of sodium perchlorate up to 200 mM (≙ 1.6 %) during cultivation without any changes in its growth pattern. After the addition of up to 440 mM (≙ 3.7 %) sodium perchlorate, H. marinus showed significant changes in cell morphology; from single motile short rods to long cell chains up to 80 cells. Furthermore, it was shown that the known desiccation tolerance of H. marinus is highly influenced by a pre-treatment with different perchlorates; additive effects of desiccation and perchlorate treatments are visible in a reduced survival rate.
