Exosomes are enriched for a specific subset of host-derived proteins, nucleic acids, lipids and carbohydrates, though they also incorporate most host cell molecules at baseline levels. Various models of exosome biogenesis have been proposed, but the field lacks the robust mechanistic studies that are needed to obtain a molecular understanding of vesicle secretion. To shed light on this process, our laboratory has taken a cargo-based approach in which we focus on the cis-acting signals that are necessary and sufficient for the budding of specific proteins. We have demonstrated, using primary biopsies and cell lines, that MCT1 activity is highly associated with MYCN amplification in human neuroblastoma. Thus, as is the case with MYCN, MCT1 expression seems to correlate with high-risk disease features such as the presence of metastatic disease at diagnosis. However, MYCN is unlikely to be a good therapeutic target because it is a transcription factor affecting the activity of numerous proteins (Dang et al., 1999). This work supports the hypothesis that MCT1 may be a more useful therapeutic target. The functional assays of MCT activity show that inducibility of the gene at low pH or amount of protein by Western blot are not predictive of how much an MCT inhibitor can affect intracellular pH. Excessive workloads, a lack of support from line managers and unrealistic targets have contributed to a stressed-out workforce, a survey has found.. The purpose of this study was to examine leadership in the public sector based on leadership models, organizational factors, conditions and health of employees. The study was divided into sub-study one and two. In sub-study one the design was an observational study using cross sectional design in which 67 first line managers in the public sector answered a survey about conditions and leadership models. Sub-study two was an observational study using case-control design focusing on a health promoting leadership intervention in the municipal sector. Two semi-structured interviews and 41 surveys were collected. The first-line managers in each group were interviewed and their employees had to answer a questionnaire.
