We routinely freeze cells in 10%DMSO - 90%FCS, and have NOT been successful in freezing hybridoma cells that have been adapted to serum free conditions. We have recently embarked on further efforts to do this by investigating alternative freezing media. I have seen some articles (I forget where) suggesting use of glycerol or glycerol/DMSO combinations as cryoprotectants for cell lines that are otherwise difficult to freeze. We have tried to freeze 3 or 4 different serum-free adapted hybrids, all with the same crummy results. Good luck, hope your luck is better than ours. Cytotoxic T lymphocyte (CTL)-mediated immunity plays a critical role in clearance of influenza virus-infected cells through contact-dependent interactions. Recent studies have described stationary tissue-resident memory (Trm) T cells which maintain CD69 and CD103 expression in peripheral tissues after local infections. These Trm cells are believed to contribute to protection after local reinfection, because they stay near the site of inoculation. Parabiosis studies previously showed that some non-migratory virus-specific CD8 T cells expressed both CD69 and CD103 in the airways after influenza virus infection. Since the TGFβ and TcR signaling pathways induce CD103 and CD69 expression on CD8 T cells in other models, I hypothesized that environmental factors and residual viral products support sustained surveillance of the lungs by pathogen-specific CTLs. Chicken reticuloendotheliosis virus (REV) causes the atrophy of immune organs and immuno-suppression. The pathogenic mechanisms of REV are poorly understood. The aim of this study was to use RNA sequencing to analyse the effect of REV on immunity and cell proliferation in chicken lymphocytes from peripheral blood in vitro. Overall, 2977 differentially expressed genes (DEGs) were examined from cells between infected with REV or no; 56 DEGs related to cell proliferation and 130 DEGs related to immunity were identified. MTT, Q-PCR and FCM indicated that REV reduced the number of lymphocytes by inhibiting the transition of S/G1 phase through FOXO and p53 pathways. Similarly, REV infection would destroy the immune defense of lymphocytes through MAPK-AP1 via Toll-like receptor-, NOD-like receptor- and salmonella infection pathways to reduce the secretion of IL8 and IL18. ABSTRACT. Antiviral effects of a synthetic Aluminum-Magnesium Silicate (AMS) were tested on Fowl Pox Virus (FPV). Five batches of the Nigerian brand of FPV vaccine were used as sources of the virus. The reconstituted vaccines were mixed with The Synthetic AMS on equal volume to weight basis and incubated at room temperature for one hour. They were centrifuged for 10 minutes at 2000 revolutions per minute. The incubation and centrifugation were repeated on a portion of each vaccine supernatant. The two sets of supernatants were tested by the Modified Passive Haemagglutination test, for FPV titres. Portions of the vaccines, not incubated with the AMS, were served as controls. Fowl Pox Virus titres of the vaccines increased from a mean of 2.8 ± 1.10 to 11.2 ± 4.38 when incubated with the AMS once.
