Single particle diffractive imaging data from Rice Dwarf Virus (RDV) were recorded using the Coherent X-ray Imaging (CXI) instrument at the Linac Coherent Light Source (LCLS). RDV was chosen as it is a well-characterized model system, useful for proof-of-principle experiments, system optimization and algorithm development. RDV, an icosahedral virus of about 70 nm in diameter, was aerosolized and injected into the approximately 0.1 μm diameter focused hard X-ray beam at the CXI instrument of LCLS. Diffraction patterns from RDV with signal to 5.9 Ångström were recorded. A viroplasm is an inclusion body in a cell where viral replication and assembly occurs. They may be thought of as viral factories in the cell. There are many viroplasms in one infected cell, where they appear dense to electron microscopy. Very little is understood about the mechanism of viroplasm formation. A viroplasm is a perinuclear or a cytoplasmic large compartment where viral replication and assembly occurs. The viroplasm formation is caused by the interactions between the virus and the infected cell, where viral products and cell elements are confined. Virosomes are also called virus factories, and virus inclusions. Viroplasms have been reported in many unrelated groups of Eukaryotic viruses that replicate in cytoplasm, however, viroplasms from plant viruses have not been as studied as viroplasms from animal viruses. It is thought that the high fidelity segmented genome assembly involves multiple sequence-specific RNA-RNA interactions between single-stranded RNA segment precursors. These are mediated by virus-encoded non-structural proteins with RNA chaperone-like activities, such as rotavirus (RV) NSP2 and avian reovirus sigma NS. Here, we compared the abilities of NSP2 and sigma NS to mediate sequence-specific interactions between RV genomic segment precursors. Despite their similar activities, NSP2 successfully promotes inter-segment association, while sigma NS fails to do so. Antheraea mylitta cytoplasmic polyhedrosis virus (AmCPV), a cypovirus of Reoviridae family, infects Indian non-mulberry silkworm, Antheraea mylitta, and contains 11 segmented double stranded RNA (S1-S11) in its genome. Some of its genome segments (S2 and S6-S11) have been previously characterized but genome segments encoding viral capsid have not been characterized. In this study genome segments 1 (S1) and 3 (S3) of AmCPV were converted to cDNA, cloned and sequenced. S1 consisted of 3852 nucleotides, with one long ORF of 3735 nucleotides and could encode a protein of 1245 amino acids with molecular mass of ~141 kDa. Similarly, S3 consisted of 3784 nucleotides having a long ORF of 3630 nucleotides and could encode a protein of 1210 amino acids with molecular mass of ~137 kDa. Method and apparatus for forming and unloading a completed stack of articles. An assembly station includes a rotatable turret from which at least one stacker arm projects. The turret rotates to each of a number of article loading positions to sequentially load articles onto the arm in a desired order. The turret is then advanced to allow the articles to be concurrently unloaded from the stacker arm at an unloading position. In an automated disc drive manufacturing environment, the articles comprise respective numbers of magnetic recording discs and intervening disc spacers which are arranged into a disc/spacer stack as the stacker arm is advanced to a number of disc feeder and spacer feeder stations. The completed disc/spacer stack is then unloaded onto a spindle motor hub. Additional arms preferably project from the turret so that, as the turret rotates, multiple stacks are simultaneously formed and sequentially unloaded.

Sedoreovirinae (sedo = smooth) is a subfamily of the Reoviridae family of viruses. Viruses in this subfamily are distinguished by the absence of a turreted protein on the inner capsid to produce a smooth surface. The S1 gene nucleotide sequences of 10 type 3 (T3) reovirus strains were determined and compared with the T3 prototype Dearing strain in order to study sequence diversity in strains of a single reovirus serotype and to learn more about structure-function relationships of the two S1 translation products, sigma 1 and sigma 1s. Analysis of phylogenetic trees constructed from variation in the sigma 1-encoding S1 nucleotide sequences indicated that there is no pattern of S1 gene relatedness in these strains based on host species, geographic site, or date of isolation. This suggests that reovirus strains are transmitted rapidly between host species and that T3 strains with markedly different S1 sequences circulate simultaneously. The genome of orbiviruses (Reoviridae family) comprises 10 segments of double-stranded RNA. The fourth largest segment of the tick-borne Kemerovo (KEM) group orbiviruses is the genetic determinant of neurovirulence in experimentally infected mice, and segment 6 determines serotype. Reassortant viruses derived from a cross between two KEM-related viruses, Great Island (GI) and Wexford (WEX), that had the heterotypic gene combination W4G6 (segment 4 of WEX virus and segment 6 from GI virus) were nonpathogenic in mice. This apparent genetic modulation of neurovirulence may have resulted from steric interaction between the two outer capsid proteins of nonpathogenic reassortants. Further data are consistent with this hypothesis. Reassortants generated from additional KEM group viruses showed various degrees of enhanced neurovirulence in terms of their PFU/LD50 (ratio of infectivity in cell culture and in mice) and ASTmax (the average survival time at the highest virus dilution resulting in 100% mortality).

In this study, we evaluated the efficacy of human adjuvant vector cells for use as cancer vaccines in dogs and immunodeficient mice using human materials. We used allogeneic human cells, the CD1d-HEK293 cell line loaded α-GalCer and transfected with mRNA-encoding tumor antigen as artificial adjuvant vector cells. Our approach uses antigen-expressing vector cells expressing 2 antigens, which allows for host DC cross-presentation of tumor antigens to naïve T cells while simultaneously presenting iNKT cell ligand to iNKT cells (34, 35). This strategy induces activation of tumor antigen-specific CTLs as well as iNKT cells. An additional benefit of this strategy is the flexibility of mRNA transduction, which uses mRNA derived from tumor cell lines or tumors from third party patients and encodes for tumor antigens without the need for HLA matching. In HepG2 cells, we constructed HepG2-PXR cell line that stably overexpresses PXR in order to better study the effect of PXR on lipogenesis. Human PXR expression plasmid, pCMV-3Xflag-PXR, and control vector plasmid, pCMV-3Xflag, were transfected into HepG2 cells, which were then selected by G418 for 14 days. The cell colonies were selected and expanded. The PXR and vector cell lines were named HepG2-PXR and HepG2-Vector, respectively. The expression of PXR at both mRNA and protein levels was verified. RT-PCR analysis showed that the mRNA level of PXR in HepG2-PXR cells was much Esistant and dyslipidemic. Attending the meeting to answer any questions concerning the roofing portions of the project was Doug Copley of Tremco. Copley has consulted with Plymouth Schools Maintenance Director Dave Schoof over the last several months to help identify needed specific roofing replacement sites. Bluetongue virus (BTV) is the type species of the genus Orbivirus, family Reoviridae (1, 2), that exists as at least 27 distinct serotypes (3, 4). The BTV can infect most ruminants, causing severe "bluetongue" disease (BT). The BTV particle is icosahedral and nonenveloped and is composed of a three-layered protein capsid surrounding 10 double-stranded RNA (dsRNA) genome segments ranging in size from 3,944 to 822 bp. N2 - Reassortants of bluetongue virus Serotype 11 (BTV-11) were isolated from a yearling heifer experimentally infected with two electrophoretically different strains (UC-2 and UC-8) by subcutaneous inoculation. Viruses were recovered by direct titration of sonicated blood samples onto Vero cell monolayers, which were overlaid with agarose and later plaque purified. The parental electropherotype of UC-8 was identified as the predominant virus strain during the infection; UC-2 was not isolated. UC-2 inffectivity was shown by reassortants which contained genome segments that were identical in migration pattern to the parental UC-2 electropherotype.
