Tumor radioresistance leading to local therapy failure remains a major obstacle for successful treatment of high-grade glioma. We hypothesized that distinct radiobiological features of particle therapy with carbon ions may circumvent glioma radioresistance. We demonstrate that carbon irradiation (CIR) efficiently eradicates radioresistant patient-derived glioma stem cells (GSCs), leading to growth inhibition and prolonged survival. The impact of CIR at the tumor-stroma interface was further investigated in 2 syngeneic mouse and 2 orthotopic GSC xenograft models. Intriguingly, tumor regressions and long-term local controls were observed at doses greater than or equal to 15-Gy CIR. Fractionated CIR further prolonged survival. BACKGROUND: The aim of this study was to investigate the biological effect of the alpha-particle-emitting isotope astatine-211 on the human cell line Colo-205 and to compare it with that of low-dose-rate gamma-radiation. MATERIALS AND METHODS: Plastic (PMMA) rotating phantoms were constructed, allowing precise dosimetry on a cellular level for both types of radiation. Growth assays using 96-well plates were used to estimate apparent cell survival for the two types of radiation. From this, the relative biological effect (RBE) could be estimated. RESULTS: Irradiation of the cells with 211At resulted in an RBE of 25.1 +/- 6.7 at 37% survival, and 17.3 +/- 2.5 at 10% survival, when compared with low-dose-rate gamma-irradiation. Purpose: Estimation of the actual delivered 4-dimensional (4D) dose in treatments of patients with mobile hepatocellular cancer with scanned carbon ion beam therapy. Methods and Materials: Six patients were treated with 4 fractions to a total relative biological effectiveness (RBE)-weighted dose of 40 Gy (RBE) using a single field. Respiratory motion was addressed by dedicated margins and abdominal compression (5 patients) or gating (1 patient). 4D treatment dose reconstructions based on the treatment records and the measured motion monitoring data were performed for the single-fraction dose and a total of 17 fractions. Background and purpose: To determine the influence of the number of fractions (or the dose per fraction) on the proton relative biological effectiveness (RBE). Materials and methods: Intestinal crypt regeneration in mice was used as the biological endpoint. RBE was determined relative to cobalt-60 gamma rays for irradiations in one, three and ten fractions separated by a time interval of 3.5 h. Proton irradiations were performed at the middle of a 7-cm Spread Out Bragg Peak (SOBP). The spatial pattern of energy depositions is crucial for understanding the mechanisms that modify the relative biological effectiveness of different radiation qualities. In this paper, we present data on energy-deposition properties of mono-energetic protons (1-20 MeV) and their secondary electrons in liquid water. Proton-impact ionization was described by means of the Hansen-Kocbach-Stolterfoht doubly differential cross section (DDCS), thus modelling both the initial energy and angle of the emitted electron. Excitation by proton impact was included to account for the contribution of this interaction channel to the electronic stopping power of the projectile. The Proton Irradiation Facility (PIF) was designed primarily for single-event effect (SEE) studies and characterization of electronic components and detectors for the space environment. The energy spectrum of protons trapped in earth orbits peaks in the 10-100 MeV region (depending on the amount of intervening absorber) and drops by about an order of magnitude at 500 MeV. This makes the TRIUMF cyclotrons variable-energy capability of up to 500 MeV ideal for studies of space effects. The scientists divided the GIST patients into four groups--extremely low risk, low risk, moderate risk, and high risk--on the basis of the size of the tumor and an index that calculates how many cells are dividing. Of the 170 patients with GIST tumors classified as extremely low risk, low risk, and moderate risk, only one patient had a tumor that could not be operated on, and three patients had recurrent tumors. This can be compared with the 89 patients with high-risk tumors, among whom 36 had inoperable tumors and 35 developed new tumors after surgery. The aim of this study is to quantify patterns of age-related shape change in the human thorax using Procrustes superimposition. Landmarks (n=106) selected from anonymized computed tomography (CT) scans of 63 adult males free of skeletal pathology were used to describe the form of the rib cage. Multivariate linear regression was used to determine a relationship between landmark location and age. Linear and quadratic models were also investigated. A permutation test employing 1×105 random trials was used to assess the model significance for both model formulations. In this paper, we present a computational, multiscale system of models to better assess the relative biological effectiveness (RBE) and compound biological effectiveness (CBE) of several neutron sources as applied to BNCT using boronophenylalanine (BPA) and a potential monoclonal antibody (mAb) that targets HER-2-positive cells with Trastuzumab. The multiscale model is tested against published in vitro and in vivo measurements of cell survival with and without boron. Electron paramagnetic resonance (EPR) dosimetry using the well established dosimeter material alanine is a generally accepted dosimetric method for measurements of high absorbed doses. Alanine EPR dosimetry is however not sensitive enough for high precision measurements of low (, 5 Gy) absorbed doses using reasonably measurement times and small dosimeters. Although cancer is the leading cause of disease-related mortality in children, the relative rarity of pediatric cancers poses a significant challenge for developing novel therapeutics to further improve prognosis. Patient-derived xenograft (PDX) models, which are usually developed from high-risk tumors, are a useful platform to study molecular driver events, identify biomarkers and prioritize therapeutic agents. Here, we develop PDX for Childhood Cancer Therapeutics (PCAT), a new integrated portal for pediatric cancer PDX models.
