Five potential reference genes for RT-qPCR application, namely histone H3, beta-actin, GAPDH, ubiquitin and 18S rRNA, were evaluated for normalization of gene expression in four selected tissues (liver, kidney, thyroid and abdominal fat). Tissues were derived from fattening pigs exposed to different amounts and type of dietary iodine. Two software applications (geNorm and NormFinder) were used to evaluate the stability of the potential reference genes. All studied genes displayed high expression stability but different stability patterns between the investigated tissues. The results suggest GAPDH and 18S rRNA as reference genes applicable in all tissues investigated. Beta-actin and histone H3 are suitable reference genes for all tissues investigated except fat. In contrast, ubiquitin should be excluded from use as a reference gene in the porcine tissues analyzed due to variations in expression levels, despite the good expression stability.
A suitable reference gene is an important prerequisite for guarantying accurate and reliable results in quantitative real-time PCR (qRT-PCR) analyses. However, there is no absolute universality in reference genes among different species. Its hard to find an ideal reference gene to fit for different tissues and growth periods. Pitaya (Hylocereus) is commercially produced as a new fruit crop at a large scale in tropical and subtropical regions. To date, there is no report on the identification of the most reliable reference genes for qRT-PCR normalization in pitaya. In this study, six candidate reference genes i.e. Actin(1), GAPDH, UBC(1), UBC(2) EF1-α(1) and histone(1) were selected from thirty-nine typical candidate reference genes to determine the most stable reference genes for qRT-PCR normalization in different tissues, temperature stresses and fruit developmental stages of pitaya. Among the six candidate reference genes, Actin(1) and EF1-α(1) were the most stable gene according to calculations of

BACKGROUND: Normalizing through reference genes, or housekeeping genes, can make more accurate and reliable results from reverse transcription real-time quantitative polymerase chain reaction (qPCR). Recent studies have shown that no single housekeeping gene is universal for all experiments. Thus, suitable reference genes should be the first step of any qPCR analysis. Only a few studies on the identification of housekeeping gene have been carried on plants. Therefore qPCR studies on important crops such as cotton has been hampered by the lack of suitable reference genes.. RESULTS: By the use of two distinct algorithms, implemented by geNorm and NormFinder, we have assessed the gene expression of nine candidate reference genes in cotton: GhACT4, GhEF1alpha5, GhFBX6, GhPP2A1, GhMZA, GhPTB, GhGAPC2, GhbetaTUB3 and GhUBQ14. The candidate reference genes were evaluated in 23 experimental samples consisting of six distinct plant organs, eight stages of flower development, four stages of fruit ...
Quantitative real-time PCR (RT-qPCR) has become an accurate and widely used technique to analyze expression levels of selected genes. It is very necessary to select appropriate reference genes for gene expression normalization. In the present study, we assessed the expression stability of 11 reference genes including eight traditional housekeeping genes and three novel genes in different tissues/organs and developing seeds from four cultivars of tung tree. All 11 reference genes showed a wide range of Ct values in all samples, indicating that they differently expressed. Three softwares--geNorm, NormFinder and BestKeeper--were used to determine the stability of these references except for ALB (2S albumin), which presented a little divergence. The results from the three softwares showed that ACT7 (Actin7a), UBQ (Ubiquitin), GAPDH (glyceraldehyde-3-phosphate dehydrogenase) and EF1α (elongation factor 1-α) were the most stable reference genes across all of the tested tung samples and tung ...
The reliability of RT-qPCR data will be greatly improved by inclusion of a reference gene whose transcription level should be invariable in the different experimental conditions [4]. The present study is the first detailed survey on the stability of a large number of genes used as internal controls for RT-qPCR studies of differential expression of genes in peach.. Several approaches have been proposed to identify stability of gene expression and select the best reference genes in the context of the relevant experimental conditions [33-35, 40-45], but to date, there is no consensus on which method should be used to examine reference gene expression stability. A comparison of different algorithms of reference gene selection allows a better evaluation of the most reliable controls and reduces the risk of artificial selection of co-regulated transcripts [46]. In order to select suitable reference gene(s) for accurate normalization, we compared three different statistical approaches, geNorm, ...

Quantitative reverse transcriptase polymerase chain reaction (qRT-PCR) is a rapid and sensitive approach to identify miRNA and protein-coding gene expression in plants. However, because of the specially designated reverse transcription and shorter PCR products, very few reference genes have been identified for the quantitative analysis of miRNA expression in plants, and different internal reference genes are needed to normalize the expression of miRNAs and mRNA genes respectively. Therefore, it is particularly important to select the suitable common reference genes for normalization of quantitative PCR of miRNA and mRNA. In this study, a modified reverse transcription PCR protocol was adopted for selecting and validating universal internal reference genes of mRNAs and miRNAs. Eight commonly used reference genes, four stably expressed novel genes in Populus tremula, three small noncoding RNAs and three conserved miRNAs were selected as candidate genes, and the stability of their expression was examined

Reference genes are essential for studying mRNA expression with quantitative PCR (qPCR). We investigated 11 potential neutrophil reference genes (RPL19, GAPDH, ACTB, B2M, HPRT, G6PD, TFRC, PGK1, YWHAZ, SDHA and GYPC) for sheep under disease conditions of foot rot (FR) and with or without Se supplementation. Initial screening was based on gene expression level (,28 Cq cycles) and variability (SD , 1.5 Cq cycles) and excluded TFRC, GYPC and HPRT from further analysis. Expression stability of the remaining genes was evaluated using four software programs: geNorm, NormFinder, BestKeeper and the comparative delta Cq method. The neutrophil reference genes, G6PD, YWHAZ, GAPDH, RPL19 and SDHA, consistently ranked among the top five most stable genes under these experimental conditions. The SDHA gene expression was not stable in FR-diseased sheep receiving Se treatment and, thus, cannot be recommended as a reference gene. The commonly used genes, PGK1, ACTB and B2M, were not reliable reference genes, ...

Seashore paspalum (Paspalum vaginatum) is among the most salt- and cadmium-tolerant warm-season perennial grass species widely used as turf or forage. The objective of this study was to select stable reference genes for quantitative real-time polymerase chain reaction (qRT-PCR) analysis of seashore paspalum in response to four abiotic stresses. The stability of 12 potential reference genes was evaluated by four programs (geNorm, NormFinder, BestKeeper, and RefFinder). U2AF combined with glyceraldehyde-3-phosphate dehydrogenase (GAPDH) showed stable expression in Cd-treated leaves and cold-treated roots. U2AF and FBOX were the most stable reference genes in Cd-treated roots and cold-treated leaves. In Polyethylene Glycol (PEG)- or salt-treated roots, the reference gene U2AF paired with either ACT or CYP were stable. SAND and CACS exhibited the most stability in salt-treated leaves, and combining UPL, PP2A, and EF1a was most suitable for PEG-treated leaves. The stability of U2AF and instability of UPL and
0053] It follows that if the internal reference is not present (such as in the blood samples), iint ref should be zero and the ihigh volt will be substantially the same as ilow volt. Thus, the DI value will approach 1. When the internal reference is present, the value of DI will be greater than 1, depending on the amount of the reference compound relative to the amount of glucose. If the amount of internal reference added to the control solution provides a current similar to that from oxidizing the glucose-related mediator, the DI value can be about 2. The internal reference may be included in an amount suitable for control solutions corresponding to a high glucose concentration. It is typical to use several control solutions corresponding to low, normal, and high glucose concentration to test a glucose meter. If, for example the amount of the internal reference is chosen so that the DI value is 1.75 or greater for the highest glucose concentration in the control solution, the current from the ...

Pinus massoniaia Lamb has gained more and more attention as the most important tree species for timber and forestation in South China. Gene expression studies are of great importance to identify new and elite cultivars. Real-time quantitative PCR, a highly sensitive and specific method, is commonly used in the analysis of gene expression. The appropriate reference genes must be employed to normalize the calculation program for ascertaining repeatable and significant results. Herein, eleven housekeeping genes were evaluated during different stages of P. massoniana post nematode inoculation in this study. Three statistical approaches such as geNorm, NormFinder and BestKeeper were selected to analyze the stability of candidate genes. The results indicated that U2af and β-TUB were the most stable reference genes. These two genes could be used for the normalization in most of the experiments of P. massoniana, while Histone and AK were the least stable ones. In addition, EF expressed at the lowest average Ct

The selection of reference genes in sugarcane under Sugarcane mosaic virus (SCMV) infection has not been reported and is indispensable to get reliable reverse transcription quantitative PCR (RT-qPCR) results for validation of transcriptome analysis. In this regard, seven potential reference genes were tested by RT-qPCR and ranked according to their stability using BestKeeper, NormFinder and GeNorm algorithms, and RefFinder WEB-based software in an experiment performed with samples from two sugarcane cultivars contrasting for SCMV resistance, when mechanically inoculated with a severe SCMV strain and using mock inoculated plant controls. The genes Uridylate kinase (UK) and Ubiquitin-conjugating enzyme 18 (UBC18) were the most stable according to GeNorm algorithm and the Pearson correlation coefficients with the BestKeeper index. On the other hand, ribosomal protein L35-4 (RPL1), Actin (ACT) and Ubiquitin1 (UBQ1) were the least stable genes for all algorithms tested.
A correction for the undesirable effects of direct and indirect cross-interference from water vapour on ammonia (NH3) measurements was developed using an optical laser sensor based on cavity ring-down spectroscopy. This correction relied on new measurements of the collisional broadening due to water vapour of two NH3 spectral lines in the near infra-red (6548.6 and 6548.8cm−1), and on the development of novel stable primary standard gas mixtures (PSMs) of ammonia prepared by gravimetry in passivated gas cylinders at 100μmol mol−1. The PSMs were diluted dynamically to provide calibration mixtures of dry and humidified ammonia atmospheres of known composition in the nmol mol−1range and were employed as part of establishing a metrological traceability chain to improve the reliability and accuracy of ambient ammonia measurements. The successful implementation of this correction will allow the extension of this rapid online spectroscopic technique to exposure chamber validation tests under ...

Thousands of experiments and studies use the human reference genome as a resource each year. This single reference genome, GRCh38, is a mosaic created from a small number of individuals, representing a very small sample of the human population. There is a need for reference genomes from multiple human populations to avoid potential biases. Here, we describe the assembly and annotation of the genome of an Ashkenazi individual and the creation of a new, population-specific human reference genome. This genome is more contiguous and more complete than GRCh38, the latest version of the human reference genome, and is annotated with highly similar gene content. The Ashkenazi reference genome, Ash1, contains 2,973,118,650 nucleotides as compared to 2,937,639,212 in GRCh38. Annotation identified 20,157 protein-coding genes, of which 19,563 are | 99% identical to their counterparts on GRCh38. Most of the remaining genes have small differences. Forty of the protein-coding genes in GRCh38 are missing from Ash1;
A framework of establishing standard reference scale (texture) is proposed by multivariate statistical analysis according to instrumental measurement and sensory evaluation. Multivariate statistical analysis is conducted to rapidly select typical reference samples with characteristics of universality, representativeness, stability, substitutability, and traceability. The reasonableness of the framework method is verified by establishing standard reference scale of texture attribute (hardness) with Chinese well-known food. More than 100 food products in 16 categories were tested using instrumental measurement (TPA test), and the result was analyzed with clustering analysis, principal component analysis, relative standard deviation, and analysis of variance. As a result, nine kinds of foods were determined to construct the hardness standard reference scale. The results indicate that the regression coefficient between the estimated sensory value and the instrumentally measured value is significant ...

Pressure reference system (PRS) is an enhancement of the inertial reference system and attitude and heading reference system designed to provide position angles measurements which are stable in time and does not suffer by long term drift caused by the sensors imperfections. The measurement system uses behavior of the International Standard Atmosphere where atmospheric pressure descends with increasing altitude and two pairs of measurement units. Each pair measures pressure at two different positions that are mechanically connected with known distance between units, e.g. the units are mounted at the tips of the wing. In the horizontal flight, there is no pressure difference measured by the measurement system which means the position angle is zero. In case the airplane turns the tips of the wing mutually changes their position, one is going up and the second one is going down, and the pressure sensors in every unit measure different values which are translated into a position angle. The strapdown ...
In temperate climate, people spend on average 85% of their time in closed environments and a majority of that time in housing. Different sources (furniture, floors, walls, ceilings) may be responsible for the presence of pollutants in these closed environments. To address the health challenge raised by the quality of indoor air and provide useful elements for the management of this risk to public authorities, the French agency for food, environmental and occupational health and safety (Anses) works since 2004 in the development of indoor air guideline values (IAGVs), based exclusively on health criteria. Since the beginning of this work, Anses identified eleven indoor air pollutants of interest including formaldehyde known for its irritant effects. Since 2004, formaldehyde is classified by the International Agency for Research on Cancer (IARC) as proven carcinogen to humans (Group 1) and since 2007 is subject to IAGVs (short term IAGVs 50 μg·m−3 for an exposure of two hours and long term ...

Dermatophagoides farinae is an important source of indoor allergens that shows strong tolerance to external temperatures. However, the regularity and mechanism of tolerance are still unclear. Based on our previous RNA-seq and annotation of D. farinae under temperature stress, it is planned to identify differentially expressed genes (DEGs) involved in the temperature stress response by quantitative real-time PCR (qRT-PCR). However, the lack of reference genes directly limited the detection and confirmation of DEGs. Accordingly, in this study, we have selected six candidates as reference genes in D. farinae: 60S RP L11, 60S RP L21, α tubulin, GAPDH, Der f Mal f 6, and calreticulin, and evaluated their expression stabilities as affected by heat and cold stresses, using geNorm, NormFinder, BestKeeper, comparative ΔCt and RefFinder methods. Then the expression level of 15 DEGs were detected and verified. geNorm analysis showed that α tubulin and calreticulin were the most stable reference genes ...
There was an interview with the senior metrologist in Canada on As It Happens last night about this (itll be on the website tomorrow). Apparently, there are thirty or so national standards that were created using the Paris standard as a reference. These are the masses that appear to be getting heavier relative to the primary reference mass. Apparently, just as your stove gets greasy from cooking oils, the platinum-iridium national standards are getting dirtied by hydrocarbons in the atmosphere. The secondary standards get used far more often than the primary, and come in contact with the atmosphere much more often. So, in the experts view, the primary standard isnt getting lighter, the secondary national standards are getting heavier ...

Background: Reference genes serve an important role as an endogenous control/standard for data normalisation in gene expression studies. Although reference genes have recently been suggested for marsupials, independent analysis of reference genes on different immune tissues is yet to be tested. Therefore, an assessment of reference genes is needed for the selection of stable, expressed genes across different marsupial tissues. Methods: The study was conducted on red-tailed phascogales (Phascogale calura) using five juvenile and five adult males. The stability of five reference genes (glyceraldehyde-3-phosphate dehydrogenase, GAPDH; β-Actin, ACTB; 18S rRNA, 18S; 28S rRNA, 28S; and ribosomal protein L13A, RPL13A) was investigated using SYBR Green and analysed with the geNorm application available in qBasePLUS software. Results: Gene stability for juvenile and adult tissue samples combined show that GAPDH was most stable in liver and lung tissue, and 18S in small intestine and spleen. While all reference
The results of a questionnaire on the use of International Chemical Reference Substances and Infrared Reference Spectra were reported to the Committee. The majority of the answers came from national quality control laboratories (40.4%), pharmaceutical manufacturers (22.6%) and teaching and research institutions (19.8%). It was noted that the substances are mainly used as primary standards to calibrate working standards, but also to some extent directly as working standards. This probably reflects a single or infrequent use where it is not sensible to prepare a secondary standard. The Committee recommended that the WHO Collaborating Centre for Chemical Reference Substances should conduct a follow-up exercise and report on its results at the next meeting of the Expert Committee. ...
