The transforming growth factor-beta (TGF-beta) receptor type III is a low abundance cell surface component that binds TGF-beta 1 and TGF-beta 2 with high affinity and specificity, and is present in many mammalian and avian cell types. Type III TGF-beta receptors affinity-labeled with 125I-TGF-beta migrate in sodium dodecyl sulfate-polyacrylamide electrophoresis gels as diffuse species of 250-350 kDa. Here we show that type III receptors deglycosylated by the action of trifluoromethanesulfonic acid yield affinity-labeled receptor cores of 110-130 kDa. This marked decrease in molecular weight is also achieved by combined treatment of type III receptors with heparitinase and chondroitinase ABC. Digestion of receptor-linked glycosaminoglycans by treatment of intact cell monolayers with heparitinase and chondroitinase does not prevent TGF-beta binding to the type III receptor core polypeptide and does not release the receptor polypeptide from the membrane. Transforming growth factor beta1 (TGFbeta1) is a potent growth inhibitor for most cells, including neoplastic cells. However, there are several types of malignant cells that are resistant to its growth-inhibitory effect. LMC19, a highly malignant rat urothelial cell line, lacks TGFbeta1 receptor (TbetaRI) and is insensitive to the growth-suppresive effect of TGFbeta1. We transfected an expression vector containing human TbetaRI into this cell line. In the present study, we demonstrate a novel mechanism that causes down-regulation of TβRII. This mechanism involves the binding of an Sp1-HDAC1 complex to a specific Sp1 site of the TβRII promoter in PDAC cells. Treatment of BxPC-3 and MIA PaCa-2 cells with a HDAC inhibitor, TSA, strongly activates TβRII promoter and induces TβRII expression. An increase in the association of TβRII promoter chromatin with acetylated histone H4 was observed after TSA treatment. These findings suggest that histone deacetylation plays a role in repression of the TβRII gene in PDAC. Luciferase reporter assays using serial deletion constructs revealed that a major TSA response region is located within the NRE-2 (−100 to −67) region of the TβRII promoter. Analysis using site-specific mutation constructs demonstrated that a specific Sp1 site (Sp1C) and an inverted CCAAT element located at −102 and −83 bp relative to the transcription start site of TβRII promoter are required for TSA-mediated reversal ...

We report on a 2-year-old Polish girl with typical manifestations of Loeys-Dietz syndrome (LDS), a rare genetic condition belonging to the group of Marfan-related disorders. The characteristic LDS symptoms observed in the girl included craniofacial dysmorphism (craniosynostosis, cleft palate, hypertelorism), arachnodactyly, camptodactyly, scoliosis, joint laxity, talipes equinovarus, translucent and hyperelastic skin, and umbilical hernia. Mild dilatation of the ascending aorta and tortuous course of the left internal carotid artery were recognized during her second year of life. Molecular genetic testing revealed a heterozygous missense mutation (c.1582C,T, p.R528C) in the transforming growth factor beta receptor II gene (TGFBR2). This mutation has been previously associated with LDS in 5 unrelated cases, and was never reported in patients with other Marfan-related disorders.
