The structurally related peptides neuropeptide Y (NPY), peptide YY (PYY) and pancreatic polypeptide (PP) are endogenous agonists of the NPY receptor (YR) family, which in humans comprises four functionally expressed subtypes, designated Y1R, Y2R, Y4R and Y5R. Non-peptide antagonists with high affinity and selectivity have been described for the Y1R, Y2R and Y5R, but such compounds are still lacking for the Y4R. In this work, the structures of the high affinity selective (R)-argininamide-type Y1R antagonists BIBP3226 and BIBO3304 were linked via the guanidine or urea moieties to give homo-dimeric argininamides with linker lengths ranging from 31 to 41 atoms. In different behavioral paradigms including the elevated plus maze (EPM), it was observed previously that deletion of the neuropeptide Y Y2 receptor subtype results in potent suppression of anxiety-related and stress-related behaviors. To identify neurobiological correlates underlying this behavioral reactivtiy, expression of c-Fos, an established early marker of neuronal activation, was examined in Y2 receptor knockout (Y2(-/-)) vs. wildtype (WT) mice. Mice were placed on the open arm (OA) or closed arm (CA) of the EPM for 10 min and the effect on regional c-Fos expression in the brain was investigated. The number of c-Fos positive neurons was significantly increased in both WT and Y2(-/-) lines after OA and CA exposure in 51 of 54 regions quantified. These regions included various cortical, limbic, thalamic, hypothalamic, and hindbrain regions. DESCRIPTION (provided by applicant): The purpose of the proposed studies is to assess the feasibility of developing a novel Neuropeptide Y (NPY) obesity treatment. Extensive preclinical and clinical studies indicate that NPY receptors are important mediators of appetite and obesity. The proposed Specific Aims will determine if chronic treatment with selective NPY Y2 or Y4 receptor agonists demonstrate significant anti-obesity activity in a well established rodent model of obesity. Preliminary studies indicate that proprietary Y2 (PD4048) and Y4 (PD4074) receptor agonists administered acutely demonstrated significant anti-obesity effects as judged by decreased food intake and enhanced metabolic rate. Additional Preliminary Studies demonstrate that acute co-administration of our selective Y2 and Y4 agonists resulted in a synergistic effect on food intake that was significantly greater than either drug administered alone. Neuropeptide Y (NPY) is a downstream modulator of leptin action, possibly at the level of the arcuate nucleus where NPY neurons are known to express both leptin receptors and Y2 receptors. In addition to the well-described role of NPY and leptin in energy balance and obesity, intracerebroventricular administration of NPY or leptin also causes bone loss. Here we show that Y2 receptor-deficient mice have a twofold increase in trabecular bone volume as well as greater trabecular number and thickness compared with control mice. We also demonstrate that central Y2 receptors are crucial for this process, since selective deletion of hypothalamic Y2 receptors in mature conditional Y2 knockout mice results in an identical increase in trabecular bone volume within 5 weeks. In looking at the K-6 algebra-related standards in the draft document released recently by the Common Core State Standards Initiative (Proposed Standards Go Public, March 17, 2010), one cant help but wonder if these standards arent overly formalistic. Although such an approach may have some value for future mathematicians, the standards could discourage the average and below-average student.. For example, in the 6th grade standards, students are expected to see the need for and to understand that through the use of the multiplicative identity, the distributive law, and the commutative law they can get from y + y + y to 3y as follows: y + y + y = y(1 + 1 + 1) = y(3) = 3y. Starting from the working hypothesis that specific chemical labelling may be an attractive approach to detect and study G protein-coupled receptors (GPCRs) we synthesized catalytically active antagonists of the neuropeptide Y1 receptor (Y1R). An argininamide-type Y1R antagonist scaffold was combined with a DMAP moiety via spacers of different length and chemical nature. These hybrid compounds have Y1R affinities in the two-digit nanomolar range and are capable of catalysing acyl-transfer reaction to surrogates of bionucleophiles, as demonstrated in the absence of cells by using esters of fluorescent dyes as substrates in buffer. The present study demonstrates for the first time that orexin neurons in the LHA coexpress the NPY Y4 receptor subtype and are activated by a Y4-specific ligand, rPP. Using a newly characterized NPY Y4 antibody, a low level of Y4-IR was observed in small, round cells throughout the hypothalamus. The effects of various neuropeptide Y (NPY) and peptide YY (PYY) fragments on electrically-evoked twitches in the rat isolated vas deferens were studied and compared with the effects of full length NPY and PYY. The aim was to identify the shortest NPY/PYY fragments that are capable of suppressing the contractions. 2. NPY (1-36) and C-terminal fragments of NPY (from 11-36 to 22-36) suppressed the electrically-evoked twitches in a concentration-dependent manner. In this study, the correlation between noradrenaline, and neuropeptide Y was studied in the cerebrospinal fluid from patients with ischemic brain stroke and healthy controls. The results obtained showed linear relationships of neuropeptide Y with the levels of noradrenaline, in healthy controls as well as in the patient group. The results can help in the understanding of cotransmission between amines and neuropeptide, and the results could be taken into consideration prior to the designing of a therapeutic approach.

Mobilization of hematopoietic stem and progenitor cells (HSPCs) from the bone marrow (BM) into the peripheral blood is a complex process that is enhanced dramatically under stress-induced conditions. A better understanding of how the mobilization process is regulated will likely facilitate the development of improved clinical protocols for stem cell harvesting and transplantation. (1) showed that the truncated cleaved form of neurotransmitter neuropeptide Y (NPY) actively promotes a breach of BM vascular sinusoidal portals, thereby augmenting HSPC trafficking to the circulation. Severe fever with thrombocytopenia syndrome (SFTS) is an emerging disease in China, South Korea, and Japan caused by the tick-borne SFTS virus (SFTSV). Severe and fatal SFTS presents as a hemorrhagic fever characterized by high viral load, uncontrolled inflammatory response, dysregulated adaptive immunity, coagulation abnormalities, hemorrhage, and multiorgan failure with up to 33% case fatality rates (CFRs). Despite its public health significance in Asia, vaccines and specific therapeutics against SFTS are still unavailable. A better understanding of the pathogenesis of SFTS is crucial to improving medical countermeasures against this devastating disease.
