Both beta(1)- and beta(2)-adrenergic receptors (beta(1)- and beta(2)-AR) have important roles in heart function mainly in response to catecholamines. Some specific polymorphisms in the beta(1)- and beta(2)-AR genes, named ADRB1 and ADRB2, respectively, have been implicated in several cardiovascular and noncardiovascular phenotypes. In this study, we aimed to investigate the possible relationship between Ser49Gly and Arg389Gly polymorphisms of the ADRB1 and Arg16Gly and Gln27Glu polymorphisms of the ADRB2 gene with ST elevation myocardial infarction (MI) in a Turkish population. One hundred patients with ST elevation MI and 100 healthy control subjects were genotyped using the PCR-RFLP method. Although the Arg389 allele of the ADRB1 gene was associated with an elevated risk of MI, the Glu27 allele of the ADRB2 gene was associated with a decreased risk of MI. Carriers of the ADRB1 Arg389 allele (heterozygotes + homozygotes) had an approximately 3.5-fold increased risk for MI than Gly389 ...

Macrophages play an integral role in eliminating infection during the inflammatory response. Catecholamines are known to alter macrophage function by binding to adrenergic receptors on the macrophage membrane. Stimulation through β-adrenergic receptors (β-AR) is known to decrease macrophage function while stimulation through the α-adrenergic receptor (α-AR) is thought to have a stimulatory effect on immune function. The RAW264.7 murine macrophage cell line is used as a model of macrophage function, but little is known about the expression of adrenergic receptors on these cells. In these studies, macrophages were treated with the catecholamine norepinephrine, the non-selective α-adrenergic receptor agonist clonidine, or the selective α2-adrenergic receptor antagonist RS79948 (RS), prior to lipopolysaccharide (LPS) activation, and then analyzed for the production of the proinflammatory cytokines TNF-α and IL-6. Low concentrations of clonidine increased cytokine production while RS reversed these
TY - JOUR. T1 - α-1 Adrenergic receptors stimulation induces the proliferation of neural progenitor cells in vitro. AU - Hiramoto, Takeshi. AU - Ihara, Yoshiaki. AU - Watanabe, Yasuhiro. PY - 2006/11/6. Y1 - 2006/11/6. N2 - The proliferation of neural progenitor cells (NPCs) is regulated by classical neurotransmitters such as dopamine, serotonin and acetylcholine, via its own receptors. Previous studies have reported that the depletion of l-norepinephrine decreases the proliferation of NPCs in the adult rat hippocampus and it has been suggested that l-norepinephrine regulates the proliferation of NPCs. However, it remains unknown whether or not adrenergic receptors are involved in the increased proliferation of NPCs. In the present study, an MTT cell proliferation assay was carried out in order to investigate the roles played by adrenergic receptors in the proliferation of NPCs. We demonstrated that l-epinephrine enhanced the proliferation of embryonic NPCs in vitro. In addition, the α-1 ...
The ß-adrenergic receptors are linked to G proteins. The ß-receptor has three known subtypes. Beta-1 receptors primarily regulate myocardial tissue and affect the rate of contraction via impulse conduction. Beta-2 receptors regulate smooth muscle tone and influence vascular and bronchiolar rela...
Adrenergic receptors are a class of G protein-coupled receptors. They are the targets for epinephrine and norepinephrine and thus represent a critical component in the sympathetic nervous system for maintenance of homeostasis and response to disease. Adrenergic receptors carry out signaling via coupling to guanine nucleotide binding proteins (G-proteins).

The mechanisms of desensitization and resensitization of the dopamine D1 receptor are not well understood. Although the high homology of the D1 receptor with the β2-adrenergic receptor might indicate that similar mechanisms regulate the responsiveness of the receptors, even subtypes of β-adrenergic receptors differ substantially in this respect (Shiina et al., 2000). For β-adrenergic receptors, a distinction has been made between desensitization that is homologous (resulting from stimulation of the same receptor) or heterologous (resulting from stimulation of a different receptor subtype, or nonreceptor-mediated activation of a second messenger) (Harden, 1983). A mechanism for heterologous desensitization is phosphorylation of the receptor by PKA (Clark et al., 1989; Hausdorff et al., 1989), whereas homologous desensitization of the β2-adrenergic receptor involves phosphorylation of the agonist-occupied receptor by both GRK (Benovic et al., 1986) and PKA (Post et al., 1996; Moffett et al., ...

Genomics has significantly improved our understanding of chronic obstructive pulmonary disease (COPD) over the past decade. Genome-wide association studies have identified a number of genetic variants associated with lung function and susceptibility to COPD,1 2 and have helped explain why only some smokers develop COPD and why never-smokers can also be affected by the disease. Once COPD is diagnosed, reducing the burden of respiratory exacerbations is a clinical priority because these events are associated with accelerated lung function decline3 and increased mortality.4 However, not all patients with COPD experience exacerbations.5 6 Therefore, identifying individuals at increased risk of exacerbations may help target management and improve outcomes. Beyond the susceptibility to COPD, genomics may also help inform the morbidity of COPD. A previously reported twin study suggests that more than 60% of the risk of severe COPD exacerbations can be attributed to genetic factors.7 Among those, ...
alpha(1B)-Adrenergic receptors mediate many of the actions of the natural catecholamines, adrenaline and noradrenaline. They belong to the seven transmembrane domains G protein-coupled receptor superfamily and exert their actions mainly through activation of Gq proteins and phosphoinositide turnover/calcium signaling. Many hormones and neurotransmitters are capable of inducing alpha(1B)-adrenergic receptor phosphorylation and desensitization; among them: adrenaline and noradrenaline, phorbol esters, endothelin-I, bradykinin, lysophosphatidic acid, insulin, EGF, PDGF, IGF-I, TGF-beta, and estrogens. Key protein kinases for these effects are G protein coupled receptor kinases and protein kinase C. The lipid/protein kinase, phosphoinositide-3 kinase also appears to play a key role, acting upstream of protein kinase C. In addition to the agents employed for cells stimulation, we observed that paracrine/autocrine mediators also participate; these processes include EGF transactivation and ...
We11,12 and others13-15 have shown that βAR stimulation in cardiac myocytes results in apoptosis that is mediated by ROS-dependent activation of JNK and the mitochondrial death pathway.16 This study demonstrates that βAR-stimulated JNK activation and apoptosis in ARVMs require Rac1. We further show for the first time that statins inhibit βAR-stimulated JNK activation and apoptosis and that these effects are due to inhibition of βAR-stimulated Rac1 activation.. A new finding of this study is that βAR stimulation causes Rac1 activation. Furthermore, adenoviral overexpression of the N17Rac1 dominant-negative mutant inhibited βAR-stimulated apoptosis and 3 known steps in the signaling pathway leading to βAR-stimulated apoptosis: JNK activation, cytochrome C release, and caspase-3 activation. Thus, in cardiac myocytes, βAR stimulation couples to Rac1 activation, which is upstream of JNK in the mitochondrial death pathway and necessary for βAR-stimulated apoptosis.. Little is known about the ...

Duchenne muscular dystrophy (DMD) is a progressive neuromuscular disease resulting in severe respiratory derangements. As such, DMD patients are at a high risk of nocturnal hypoventilation, thereby requiring nocturnal ventilation (NV). To this end, NV is an important clinical milestone in the management of DMD. Emerging evidence suggests that ß2 adrenergic receptors (ADRB2) may play a role in determining respiratory function, whereby more functional ADRB2 genotype variants (e.g., Gly16) are associated with improved pulmonary function and respiratory muscle strength. These findings suggest that the more functional ADRB2 genotype may help to preserve respiratory function in patients with DMD. The purpose of this study was to identify the influence of ADRB2 genotype on the risk of NV use in DMD. Data from the CINRG Duchenne Natural History Study including 175 DMD patients (3-25 yrs) were analyzed focusing on ADRB2 genotype variants. Time-to-event analyses were used to examine differences in the age at

The deubiquitinase ubiquitin-specific protease 20 is a positive modulator of myocardial β1-adrenergic receptor expression and signaling.
Similar to the scenario in neonatal rat cardiomyocytes, β1-AR stimulation with isoproterenol induces a robust increase in cAMP accumulation that is associated with an increase in the amplitude and an acceleration of the kinetics of both the calcium transient and the twitch in adult rat ventricular myocytes. In contrast, only rather high concentrations of the β2-AR agonist zinterol (10-5 mol/L) modulate contractile function in adult rat ventricular myocytes (Figure 2⇑). Parenthetically, 2 laboratories have presented data to support the theoretical argument that 10-5 mol/L zinterol (in the absence of a β1-AR blocker) acts as a mixed β1-/β2-AR agonist; β2-AR selectivity is achieved only if a β1-AR blocker (such as CGP 20712A) is included in the assay.11 73 In contrast, Xiao and Lakatta74 maintain that the effects of high concentrations of zinterol (even without a β1-AR blocker) are attributable entirely to the minor population of β2-ARs in the preparation, and they present results to ...
MN-221 is a novel, highly selective ß2-adrenergic receptor agonist being developed for the treatment of exacerbations of asthma/COPD. We licensed MN-221 from Kissei Pharmaceutical Co., Ltd. Preclinical studies conducted in vitro and in vivo showed MN-221 to be highly selective for the ß2-adrenergic receptor. In these studies, the ß1-adrenergic receptor stimulating activity of MN-221 was less than that of other ß2-adrenergic receptor agonists in isolated rat atrium and in vivo cardiac function tests in rats, dogs and sheep, thereby suggesting that MN-221 may stimulate the heart less than older, less selective ß2-adrenergic receptor agonists. In addition, in vitro studies also suggested that MN-221 may act as only a partial ß1-adrenergic receptor agonist in cardiac tissue, while acting as a full ß2-adrenergic receptor in lung tissue. We believe that this improved receptor binding and functional selectivity may result in fewer cardiovascular side effects than are commonly observed with other ...
