Spleen cells derived from BN rats receiving HgCl2 were fused with the nonsecreting rat myeloma cell line IR983F. We screened 59 supernatants from immunoglobulin-secreting hybrids for antibody activity against actin, tubulin, autologous and heterologous myosin, myoglobin, dsDNA, peroxidase, and the haptens TNP, NIP, NNP, and NBrP. Six monoclonal antibodies (mAb) were found to react with antigen(s) of the panel. At least three groups of antibody specificities were identified: clones reacting with TNP (1 IgM, 1 IgE); clones reacting with horseradish peroxidase (1 IgM); and clones possessing widespread reactivity for several antigens as found for mouse natural autoantibodies (2 IgM, 1 IgE). We also analyzed the idiotypic (Id) determinants of the 59 mAb by using anti-Id antibodies described elsewhere prepared in rabbits against the BALB/c D23 natural monoclonal autoantibody and recognizing a BALB/c recurrent Id (Id D23) of natural polyspecific autoantibodies. We found that all rat mAb that possessed ...
The striatum integrates sensory and motor information to enable action selection and behavioral reinforcement. There is a large topographical projection from the rat barrel cortex to widely distributed areas of the striatum. The response of the striatum to sensory stimuli at a network level is, however, not comprehensively understood. We used a 10Hz/100ms air puff, allowing un-damped movement of multiple whiskers, to look at functional connectivity in contralateral cortex and striatum in response to sensory stimulation. Simultaneous recordings of cortical and striatal local field potentials (LFPs) were made under isoflurane anaesthesia in 15 male Brown Norway rats. Four electrodes were placed in barrel cortex while the dorsolateral striatum was mapped with 500µm-spaced micro-electrodes spanning the left striatum medio-laterally and moved both dorso-ventrally and rostro-caudally with 500µm steps. This resulted in a maximum of 315 striatal positions per animal. Significant event-related ...
Dose-dependent specific antibody production, antigen-dependent pulmonary inflammation, and thiol changes in the lung and associated lymph nodes were examined in a Brown Norway rat model of pulmonary sensitization. Cysteine (CYSH), glutathione (GSH), and markers of inflammation in bronchoalveolar lavage fluid (BALF) were measured following ovalbumin (OVA) inhalation challenge. Alveolar macrophages
This study shows that the colony of inbred BN rats used here presents, in addition to several arterial phenotypes of potential pathophysiological and fundamental interest recently reported (20), an apparently nonsymptomatic form of congenital HN. The relevance of such a form of HN to human pathology is questionable (31), but since a significant proportion of cases of HN detected antenatally either disappear or remain nonsymptomatic, understanding mechanisms involved in this mild form might enable its distinction from the other, more severe forms that, if unchecked, lead to destruction of renal parenchyma and ultimate renal failure (4). Thus the BN rat represents an interesting animal model for the identification of genes that may play a role in this condition. This study is the first to attempt to identify QTLs for HN by genetic linkage.. The first major observation in this study is that in the BC population HN is not linked to any of the other principal arterial phenotypes exhibited by the BN ...
We investigated the potential role of intercellular-adhesion molecule-1 (ICAM-1) in allergen-induced bronchial hyperresponsiveness (BHR) and inflammation in sensitised Brown-Norway rats. Rats were sensitised with ovalbumin (OA) intraperitoneally and 21 days later they were either exposed to 0.9% NaCl or 1% OA aerosol for 15 min. Rats exposed to OA aerosol were pretreated either with ICAM-1 antibody (3 mg/kg i.p. and i.v., 45 min prior to OA exposure) or with the diluent for the antibody. Eighteen to twenty-four hours after OA or 0.9% NaCl exposure, rats were anaesthetised, tracheostomised and mechanically ventilated, and airway responsiveness to acetylcholine (ACh) aerosol was measured as the provocative concentration of ACh needed to increase pulmorary resistance by 100% (PC100). Mean -log PC100 was increased in rats exposed to OA but pretreated with diluent (2.75 +/- 0.06) compared to rats treated with ICAM-1 antibody (2.51 +/- 0.08 ...

BACKGROUND: The aim of the current study was to investigate the possibility of increasing the maximal tolerated dose (MTD) of a tumor-selective radiolabeled antibody when radioimmunotherapy (RIT) is combined with extracorporeal depletion of radioimmunoconjugates from the circulation. Furthermore, the authors evaluated whether this increase in dose improved the therapeutic effect on solid manifest tumors in an immunocompetent animal model. METHODS: Rats were injected with high activities/body weight of lutetium (Lu-177)- or yttrium (Y-90)-labeled antibody conjugates (monoclonal antibody tetraazacyclododecanetetraacetic acid-biotin) and subjected to removal of the conjugate from the circulation by extracorporeal affinity adsorption treatment 24 hours postinjection. Myelotoxicity was assessed by analysis of blood parameters for 12 weeks. The effect of increased doses in combination with extracorporeal affinity adsorption treatment was evaluated with respect to myelotoxicity and therapeutic effect ...
In this study, immunologic interactive effects between timothy grass pollen grains and their PCGs were performed by an evaluation of allergy (humoral and cellular responses) and inflammatory parameters on Brown Norway rats, a good animal model of pollen allergy [17]. In parallel, allergic and inflammatory responses of these interactive effects were compared with those obtained on rats sensitized and challenged with only one aeroallergen, pollen grains or PCGs.. Our findings showed that combined effects between pollen and PCGs induce humoral responses. Previous studies showed that PCGs and pollen grains induced humoral responses on Brown Norway rats [9, 10, 16] when each aeroallergenic source was used for both sensitization and challenge. PCGs can also trigger IgE-mediated reactions in grass pollen-sensitive patients [4].. Humoral responses on allergic PG and GP groups were influenced mainly by PCGs. On these groups (PG and GP groups), the IgE and IgG1 levels were not different from those ...
Purpose: Circumpapillary SD-OCT scans of the neural retina are routinely used to quantify structural changes in glaucoma. The purpose of this study was to evaluate structural changes in a rat model of glaucoma using computationally generated and scaled circumpapillary scans.. Methods: SD-OCT images were captured using the Spectralis system in 18 normal aged male Brown Norway rats (36 eyes; weight = 422 ± 32 g). Intraocular pressure was surgically elevated in six of these animals (6 eyes) by injecting hypertonic saline into an episcleral vein. Pre- and post-operative images were corrected for lateral magnification using individual ultrasonic axial biometric measurements. Circumpapillary scans (1 mm diameter) were generated from 30° x 25° horizontal volumetric image data sets centered on the optic nerve head. Global and regional (superior, inferior, nasal, and temporal) mean NFL/RGLC, NFL/RGCL+IPL, and total retinal thicknesses were calculated from the circumpapillary scans. Statistical ...
Purpose : Recent studies suggest that menopause or estrogen receptor mutations modulate the risk for glaucoma. Ocular biomechanical properties influence mechanical loads at the optic nerve head and may contribute to loss of retinal ganglion cells (RGC). Estrogen is known to alter biomechanical properties in other tissues; therefore, we hypothesize that an estrogen deficiency contributes to the etiology of glaucoma by altering ocular tissue biomechanics. Here, we examined the impact of menopause on ocular compliance in rats, a measure of corneoscleral biomechanical properties. We then assessed the effect of menopause on visual function in the established Morrison ocular hypertension (OHT) model. Methods : Female Brown-Norway rats (3-4 months) were randomly assigned to pre- or post-menopausal groups. Menopause was induced surgically by ovariectomy (OVX). We measured ocular compliance in non-OVX (n=8) and OVX (n=11) rats using the iPerfusion system. In separate cohorts of non-OVX (n=6-8) and OVX ...
... in the field of allergy. even when they are not given until after starting HgCl2 administration. IFN- is definitely a pivotal cytokine in ameliorating the Th2 response and actions aimed at selective up-regulation of this cytokine may be of restorative value in suppression of undesirable IgE reactions. < 005 was taken to indicate statistical significance. RESULTS Exogenous type-1 cytokines suppress IgE production in HgCl2-treated BN rats HgCl2 treatment of BN rats resulted in designated elevation of serum IgE concentrations, as previously reported [7]. IgE levels were barely above normal at day time 7, then rose rapidly to maximum levels by day time 14. Administration of exogenous recombinant rat IFN- at a dose of 6 104 U/day time Motesanib had little effect (= 069, two-tailed MannCWhitney = 0009 HgCl2 only, = 001 group treated with 6 104 U/day time; two-tailed MannCWhitney = 0026). IgE levels at day time 14 were ...
Norway rats are primarily nocturnal. Outdoors, they burrow in soil near stream/river banks, piles of garbage or under concrete slabs. Norway rats are social and often build burrows close to one another. Indoors, they often nest in basements, piles of debris or undisturbed materials. They are known to gnaw through almost anything - including plastic or lead pipes - to obtain food or water.. ...
The nasal vascular permeability of ovablumin (OVA)-sensitized Brown Norway rats was evaluated by analyzing a brilliant blue concentration in perfusate from the nose after exposure of the nasal mucus to OVA. Oral administration of ,I,Lactobacillus,/I, GG and ,I,L. gasseri,/I, TMC0356 significantly inhibited the increase in nasal vascular permeability (,I,P,/I,,0.01). The serum IgE of the tested rats also decreased, although the change was not statistically significant. These results indicate that ,I,Lactobacillus,/I, GG and ,I,L. gasseri,/I, TMC0356 might alleviate nasal allergic symptoms by suppressing the increase in nasal vascular permeability caused by local inflammation associated with allergic rhnititis.. ...

TY - JOUR. T1 - An Animal Model for Wheat Allergy Skin Sensitisation: A Comparative Study in Naive versus Tolerant Brown Norway Rats. AU - Ballegaard, Anne-Sofie Ravn. AU - Madsen, Charlotte Bernhard. AU - Bøgh, Katrine Lindholm. N1 - © 2018 The Author(s) Published by S. Karger AG, Basel.. PY - 2019. Y1 - 2019. N2 - BACKGROUND: Allergic sensitisation to foods may occur in infancy without prior oral exposure to the offending food, leading to the assumption that food allergy sensitisation may occur through the skin. Concerns have been raised regarding the safety of use of personal care products containing hydrolysed wheat proteins, since these products have been shown to induce allergy through the skin, and even cause an abrogation of an already established oral tolerance.OBJECTIVE: To establish an animal model for food allergy skin sensitisation and compare the sensitising capacity of an unmodified and an acid-hydrolysed gluten product via slightly damaged skin in naïve versus tolerant ...
The effects of cypermethrin, copper and glyphosate on the endocrine system and subsequent response to female pheromones were investigated in mature male brown trout (Salmo trutta) parr. Responses measured were the amount of strippable milt, blood plasma levels of both an androgen (11-ketotestosterone (11-KT)) and a progestin (17α,20β-dihydroxy-4-pregnen-3-one (17,20b-P)), and behavioural changes. This was done in a two phased investigation where parr were exposed to one of the following via ambient water: 1) 0.1 or 1.0 μg L-1 cypermethrin, 2) 10 or 100 μg L-1 copper (Cu2+), or 3) 150 μg L-1 glyphosate for a 96 hour period. Phase one was a priming experiment exposing parr to a treatment followed by priming with PGF2α or ovarian fluid (OVF). Atlantic salmon (Salmo salar) parr were, also exposed to glyphosate during phase I. The second phase was centered on behavioural observations. Exposed parr were placed in a 35,000 L stream aquarium together with two ovulated females and four anadromous ...
Complexes of 125I p30, a viral core polypeptide, and rat anti-p30 antibody, preformed in vitro, were injected into the heart of BN rats bearing Moloney sarcomas (MST) and of BN rats bearing an unrelated tumor or unexposed to tumor. Complexes were cleared from the circulation of MST-bearing rats more rapidly than from sera of controls and were almost completely eliminated after 30 hr. There was no relationship between rate of disappearance and size of tumor or levels of circulating complexes. Disappearance rates in rats with progressing and regressing tumors were similar. Uncomplexed labeled p30 was cleared from the circulation of tumor-bearing and control rats with kinetics similar to those of labeled complexes. Complexes were localized in the spleen of tumor-bearing and control rats, but much more in spleens of MST-bearing rats. No other tissues, including tumor, concentrated complexes, nor was there binding to peripheral blood and spleen cells. The data suggest that augmented clearance and ...

The Brown Norway (BN) rat model for respiratory sensitisation has used both dermal and inhalation routes for induction. The elicitation phase was typically four inhalation exposures, spaced at approximately 15-day intervals. After the last challenge the measurements made included: respiratory responses using whole body plethysmography, analysis of inflammatory endpoints using lung lavage fluid and IgE levels in lung lavage fluid or serum. Lung and lymph node weights were measured post mortem and the respiratory tract was used for histopathological investigations. Controls did not receive the induction treatment but were challenged by inhalation as for the test groups, to control for short term irritation effects. On occasion respiratory methacholine challenge was performed the day following MDI challenge to test for non-specific respiratory hyperreactivity. The test material in these studies was polymeric MDI. Both inhalation and dermal routes of induction were investigated in a study in BN ...
From our stock of SDRs (Sprague-Dawley rats), we established a mutant strain having small opaque eyes and named it HiSER (Hirosaki small-eye rat). The HiSER phenotype is progressive and autosomal recessive. In HiSER eyes, disruption and involution of the lens, thickening of the inner nuclear layer, detachment and aggregation of the retina, rudimentary muscle in the ciliary body and cell infiltration in the vitreous humour were observed. Genetic linkage analysis using crossing with Brown Norway rat suggested that the causative gene(s) is located on chromosome 10. Microarray analysis showed that the expression level of the Cryba1 gene encoding βA3/A1-crystallin on chromosome 10 was markedly decreased in HiSER eyes. Genomic PCR revealed deletion of a 3.6-kb DNA region encompassing exons 4-6 of the gene in HiSERs. In HiSER eyes, a chimaeric transcript of the gene containing exons 1-3 and an approximately 250-bp sequence originating from the 3′-UTR of the Nufip2 gene, located downstream of the ...

Rats are long-tailed rodents that are present in the world in various sizes, colors and shapes. They are larger in size when compared to mice. The two most common types are brown rats and black rats. The brown rats are also known as Norway rats. The black rats are believed to have originated in the Asian regions before slowly making their way to other parts of the world. An adult black rat can be around 30 to 46 cms in length. They are omnivorous in nature and feed on both plants and small insects or animals. They are also very generic in their food and eat about everything they can find. Not only do they feed on fruits, seeds, leaves and stems but also the food stuffs meant for cows, dogs, cats and chickens. They also are a big threat to farmers and destroy agricultural based crops. A Norway rat is around 25 cm in length with a tail that is usually as long as they are. They weigh around 250 to 350 grams. They are known to have originated from China and are an important model of scientific ...
