Oestrus was synchronised with PGF2-alpha in four pluriparous West African Dwarf (WAD) goats and blood collected through jugular venupuncture. The length of the oestrous cycle and oestrus duration were observed in three successive cycles while peripheral plasma progesterone and oestradiol-17B levels were determined by the radioimmunoassay technique. The mean oestrous cycle lengths and oestrus duration were 19.4 ± 1.7 d and 33.3 ± 6.6 h, respectively. Progesterone levels were very low during the first four days of the cycle (day 1 = day of oestrous) with the lowest level of 0.3 ± 0.02 ng/ml being recorded on day 2 of the cycle. The following methods have been comparatively examined: immunodiffusion (ID) counterelectrophoresis (CEP), complement fixation (CF) passive hemoagglutination (HA) and solid phase radioimmunoassay RIA. On the basis of the results so far obtained the RIA appears the most sensitive and specific method, followed by the HA. In order to obtain reproducible results for diagnostic purposes we suggest to carry out routinely two different techniques together (possibly RIA and HA), while for screening purposes just one technique, choosed between the two most indicative ones, shall be employed.

The in vivo release of cholecystokinin (CCK)-like material (CCKLM) was measured in the frontal cortex of freely moving rats using the microdialysis technique combined with a sensitive radioimmunoassay. Local perfusion of K+ (100 mM)-enriched artificial CSF resulted in a 10-fold increase in CCKLM outflow, as compared with that occurring under basal resting (K+ = 3.0 mM) conditions, and this effect could be completely prevented by removal of Ca2+ in the perfusing fluid. Chromatographic analyses demonstrated that CCK-8S contributed to 70% of CCKLM. Stressful stimuli such as a 2-min exposure to diethyl ether and a 30-min restraint produced a marked but transient increase in cortical CCKLM release. The U937 cell line has been shown to generate tumour fibrosis factor α (TNF-α) in response to soluble stimuli such as PMA and LPS, but only after treatment with GM-CSF. We report here the generation of TNF-α from U937 cells following phagocytosis of opsonised zymozan particles without the need for pre-treatment with GM CSF. The release of TNF-α from U937 cells was demonstrated by a specific radioimmunoassay, L929 cell killing and neutrophil priming. The biological activities in the cell supernatant were inhibited by TNF-α affarserum. Phagocytosis was required for TNF-α production. Non-opsonised zymozan or latex particles which were not phagocytosed or pretreatment with cytochalasin B, which inhibited phagocytosis of opsonised zymozan particles, all failed to trigger TNF-α production. Phagocytosis failed to trigger detectable IL-1 generation, and production of IL-6 was insufficient to produce biological effects on neutrophils. The choriogonadotropin beta-subunit radioimminoassay has been used extensively to measure human choriogonadotropin in the sera of pregnant women and individuals with trophoblastic and nontrophoblastic tumors. We circumvented the non-parallelism between the standards and serial dilutions of urine containing choriogonadotropin by adding pooled urine from men to the standard tubes and limiting the volume of urine to 100 microliter. This modified assay has a sensitivity of 3 int. units/liter of urine and is specific for choriogonadotropin concentrations of 40 int. units/liter of urine.
