Adenosine was coupled to human serum albumin by two different procedures that preserved the purine and ribose rings. The conjugates were evaluated for antigenicity in rabbits and guinea pigs. A conjugate containing 2′(3′)-O-succinoyladenosine failed to elicit antibodies, whereas one containing laevulinic acid (O2′, 3′-adenosine-acetal) elicited antibodies in all animals injected. The affinity and specificity of binding of adenosine to three selected antisera were evaluated. Dissociation constants of 31-187 nM were observed. Displacement of adenosine binding to all antisera by adenine 5′-nucleotides, adenine, inosine and hypoxanthine required more than 1000-fold higher concentrations than of adenosine itself. Current immunoassays for interleukin-1 beta (IL-1 beta) are effective for analyzing fluids derived from cultured cells. However, IL-1 beta determinations in human plasma or serum samples are technically complicated by higher protein and lipid concentrations, physicochemical differences which exist between samples from healthy subjects and those experiencing acute phase responses, and by the fact that IL-1 beta can be produced and degraded in the blood collection tube after the sample is drawn. A simple chloroform extraction process has been developed which eliminates several of the interfering factors from plasma samples and increases the amount of IL-1 beta detected by radioimmunoassay and lymphocyte activation assay. In the radioimmunoassay, rabbit sera was found to influence the accuracy and variability of plasma measurements. The aim of the study was to evaluate our in-house assay for precision, detection limit, interference of haemolysis, suitability of serum versus plasma and biological variation within one subject.. Method: All samples were obtained from one subject on two occasions following an over night fast and the consumption of 1000 and 3000 kcal meals on the respective days. The involvement of central catecholaminergic neurons in tonic gonadotrophin release was investigated. Male rats were given a single cerebral intraventricular injection of 6-hydroxydopamine hydrochloride (6-OHDA; 170 μg free base) dissolved in 0·001 n-HCL. Plasma LH and FSH were measured by double antibody radioimmunoassay. In the 6-OHDA-treated animals, LH was significantly reduced (P,0·001 after 1 hr and remained consistently lower for 8 hr. After a transient elevation, the controls showed no significant change (P,0·01). No difference in LH concentrations was found between experimental groups sampled at 2 days or later (P,0·01). Levels of FSH were less consistently different. Administration of 6-OHDA dissolved in different vehicles, with varied pH and osmolarity, gave similar results which suggests that the quinone derivative of 6-OHDA is effective in producing the catecholaminergic impairment. The pneumococci possess, in addition to type-specific capsular polysaccharides, a number of antigens common to the species. Antibodies to phosphocholine (PC), a major determinant of C-carbohydrate, have been shown to protect mice from experimental pneumococcal infection, but little is known of the role of anti-PC antibodies in humans or the extent to which anti-PC levels are affected by carriage or infection. We examined 115 sera from 30 infants, who were followed propsectively from birth through 4 years of age, for the presence of immunoglobulin M antibody to PC, using a solid-phase radioimmunoassay method. Infants were found to develop antibody to PC in response to pneumococcal carriage and infection, and nearly all infants developed some antibody. Antibody levels increased with age. Summary. Baboon CG from the urine and placenta (Days 33-45 of gestation) was purified by ammonium acetate/alcohol extraction followed by ionic exchange, gel filtration and affinity chromatography. The CG activity was monitored using a double-antibody radioimmunoassay utilizing anti-baboon CG and hCG both as the labelled ligand and standard. Biological activity was measured by the rat luteal cell radio-receptor assay. The purified preparation exhibited heterogeneity in terms of its behaviour during ionic exchange chromatography and isoelectric focussing. Two new monoclonal antibodies (Lym-1 and Lym-2), reactive with the cell surface of B-lymphocytes and derived tumors, have been produced using tumor cell nuclei preparations as immunogens. Specificity screens using live cell radioimmunoassay techniques with 52 well-characterized human lymphoma and leukemia cell lines showed that both Lym-1 and Lym-2 bound to cell lines of B-cell lineage but were unreactive with those of T-cell, myeloid, or erythroid derivation. The B-cell specificity of these reagents was confirmed on 36 lymphoma and 15 leukemia biopsy specimens by using immunoperoxidase or immunofluorescence techniques. Additionally, flow cytometric analysis of 22 lymphoma biopsies showed that the majority of B-cell tumors were Lym-1 and/or Lym-2 positive and that within a given biopsy, a high percentage of the malignant cell population was stained. p,Serum specimens from 12 patients with type A hepatitis were analyzed for immunoglobulin M-type antibody to hepatitis A virus (IgM anti-HA). A recently developed solid-phase radioimmunoassay kit for IgM anti-HA (HAVAB-M, Abbott Laboratories) and a competitive binding radioimmunoassay kit (HAVAB, Abbott Laboratories) with or without 2-mercaptoethanol treatment, as modified by Yano et al. (Acta Hepatol. Jpn. 21, 704-712, 1980) were used to obtain an M-index. All specimens obtained within 60 days of the onset of illness and specimens from 2 of 4 patients later than 60 days after the onset were positive with the HAVAB-M test. According to recent investigations neopterin (a pyrazinopyrimidine derivative) is a biochemical marker that reflects the activity of the proinflammatory immunocellular system of the synovial tissue in rheumatoid arthritis (RA). Interferon gamma, derived from antigen activated T lymphocytes, stimulates macrophages to synthesise and release neopterin into the culture supernatant in vitro. To extend this in vitro model to a clinical level a sensitive new radioimmunoassay technique was used to measure neopterin concentrations in the synovial fluid (SF) of 17 patients with active RA, nine with osteoarthritis, and six with acute gout, and in that of 12 controls undergoing meniscectomy. The SF neopterin concentrations were significantly higher in patients with RA than in the other groups of patients, particularly the controls. Oestrus was synchronised with PGF2-alpha in four pluriparous West African Dwarf (WAD) goats and blood collected through jugular venupuncture. The length of the oestrous cycle and oestrus duration were observed in three successive cycles while peripheral plasma progesterone and oestradiol-17B levels were determined by the radioimmunoassay technique. The mean oestrous cycle lengths and oestrus duration were 19.4 ± 1.7 d and 33.3 ± 6.6 h, respectively. Progesterone levels were very low during the first four days of the cycle (day 1 = day of oestrous) with the lowest level of 0.3 ± 0.02 ng/ml being recorded on day 2 of the cycle. There was a steady rise in the levels during the cycle until day 15 when the highest level of 2.2 ± 0.05 ng/ml was recorded which was followed by a sharp decline on day 20 (P ,0.05). 490 blood samples have been examined for the comparison of the various methods concerned with the detection of the Australia Antigen (Au). The following methods have been comparatively examined: immunodiffusion (ID) counterelectrophoresis (CEP), complement fixation (CF) passive hemoagglutination (HA) and solid phase radioimmunoassay RIA. On the basis of the results so far obtained the RIA appears the most sensitive and specific method, followed by the HA. In order to obtain reproducible results for diagnostic purposes we suggest to carry out routinely two different techniques together (possibly RIA and HA), while for screening purposes just one technique, choosed between the two most indicative ones, shall be employed.

The polymer in turn has bonded to it, by covalent bonds, antibodies capable of binding a specific antigen thereto and stoichiometrically releasing the bound antigen by the use of a rinsing solution which does not adversely affect the binding of the antibody to the polymer. Preferably, each of the porous particles includes an impervious core which has joined thereto sufficient layers of microparticles to form an outer porous coating on the core. Methods of forming such an immunoadsorbent and for using the same and structure for supporting the same are also disclosed.

The in vivo release of cholecystokinin (CCK)-like material (CCKLM) was measured in the frontal cortex of freely moving rats using the microdialysis technique combined with a sensitive radioimmunoassay. Local perfusion of K+ (100 mM)-enriched artificial CSF resulted in a 10-fold increase in CCKLM outflow, as compared with that occurring under basal resting (K+ = 3.0 mM) conditions, and this effect could be completely prevented by removal of Ca2+ in the perfusing fluid. Chromatographic analyses demonstrated that CCK-8S contributed to 70% of CCKLM. Stressful stimuli such as a 2-min exposure to diethyl ether and a 30-min restraint produced a marked but transient increase in cortical CCKLM release. In addition, anxiety-like behavior induced by the systemic administration of yohimbine (5 mg/kg i.p.) was associated with a long-lasting enhancement in the peptide outflow. Anti-warfarin antiserum was prepared in rabbits by immunization with a synthesized warfarin antigen, 4-azo-warfarin human serum albumin, which possesses two enantiomorphic haptenic sites of warfarin on the molecule. The antiserum recognized both R- and S-warfarin to the same degree, 50% cross reactivities of racemic warfarin, respectively. One of the warfarin metabolites, racemic warfarin alcohol, showed 1% cross reactivity, and R- or S-warfarin alcohol have half the reactivity of the racemic alcohol. Rabbit plasma warfarin levels were determined by radioimmunoassay using this antiserum and racemic [14C]warfarin and by fluorometric assay after isolation by thin layer chromatography. The U937 cell line has been shown to generate tumour fibrosis factor α (TNF-α) in response to soluble stimuli such as PMA and LPS, but only after treatment with GM-CSF. We report here the generation of TNF-α from U937 cells following phagocytosis of opsonised zymozan particles without the need for pre-treatment with GM CSF. The release of TNF-α from U937 cells was demonstrated by a specific radioimmunoassay, L929 cell killing and neutrophil priming. The biological activities in the cell supernatant were inhibited by TNF-α affarserum. Phagocytosis was required for TNF-α production. Non-opsonised zymozan or latex particles which were not phagocytosed or pretreatment with cytochalasin B, which inhibited phagocytosis of opsonised zymozan particles, all failed to trigger TNF-α production. Phagocytosis failed to trigger detectable IL-1 generation, and production of IL-6 was insufficient to produce biological effects on neutrophils.
