The group recently developed a novel three-component labelling reaction for formation of trisubstituted iodotriazoles directly from aqueous radioiodide, and the parent azide and alkynes. The method is technically straightforward, high yielding and robust, allows rapid access to highly functionalized tracers. We have used this to prepare dual optical and nuclear labelling reagents, and demonstrated imaging of antibody distribution from the cellular level to the whole body (multiscale imaging). The group has also made progress on new 18F chemistry, which is currently implemented for development of metabolic tracers for imaging of drug efflux pumps. Reactive oxygen species (ROS) play important roles in cell signaling and homeostasis. However, an abnormally high level of ROS is toxic, and is implicated in a number of diseases. Positron emission tomography (PET) imaging of ROS can assist in the detection of these diseases. For the purpose of clinical translation of [18F]6-(4-((1-(2-fluoroethyl)-1H-1,2,3-triazol-4-yl)methoxy)phenyl)-5-methyl-5,6-dihydrophenanthridine-3,8-diamine ([18F]DHMT), a promising ROS PET radiotracer, we first manually optimized the large-scale radiosynthesis conditions and then implemented them in an automated synthesis module. Our manual synthesis procedure afforded [18F]DHMT in 120 min with overall radiochemical yield (RCY) of 31.6% ± 9.3% (n = 2, decay-uncorrected) and specific activity of 426 ± 272 GBq/µmol (n = 2). Fully automated radiosynthesis of [18F]DHMT was achieved within 77 min with overall isolated RCY of 6.9% ± 2.8% (n = 7, decay-uncorrected) and specific activity of 155 ± 153 GBq/µmol (n = 7) at the end of

PURPOSE: The radiolytic products of eight beta-blockers were studied in order to understand the mechanisms of irradiation of drugs in the solid state. METHODS: The drugs were analyzed by high-performance liquid chromatography coupled to a diode array detector in order to observe the degradation of the main compound after irradiation and in order to study the nonvolatile final products on more concentrated solutions of irradiated drugs. RESULTS: The first test assessed that the main compound was not significantly degraded after gamma irradiation for any of the eight beta-blockers. A more complete study, which consisted on separating the nonvolatile products and on quantifying them, indicated first that the radiolytic products could reach the number of 14 and moreover that some could exceed the 0.1% threshold at 30 kGy. Somatostatin analogs labeled with different radionuclide are vastly investigated for diagnosis or treatment of somatostatin receptor positive tumors. Labeling somatostatin analogs with 99mTc as alternative of 111In needs doing different quality control method for determination of labeling yield and complex stability for clinic applications. Methods: For determining radiochemical purity, three different methods were compared and evaluated including HPLC, TLC and Sep-Pak. Results: All the three methods were suitable for determining of radiochemical purity in peptide kits, although using HPLC was more effective rather than the others. Conclusion: HPLC was the best method for determination of radiochemical purity of the peptide kits.
