Pyruvic acid, central to leaf carbon metabolism, is a precursor of many volatile organic compounds (VOCs) that impact air quality and climate. Although the pathways involved in the production of isoprenoids are well-known, those of several oxygenated VOCs remain uncertain. We present concentration and flux measurements of pyruvic acid and other VOCs within the tropical rainforest (TRF) biome at Biosphere 2. Pyruvic acid concentrations varied diurnally with midday maxima up to 15 ppbv, perhaps due to enhanced production rates and suppression of mitochondrial respiration in the light. Branch fluxes and ambient concentrations of pyruvic acid correlated with those of acetone, acetaldehyde, ethanol, acetic acid, isoprene, monoterpenes, and sesquiterpenes. The aim of this study was to examine effects of sodium pyruvate on viability as well as on synthesis of reactive oxygen species (ROS), lipid peroxidation and DNA integrity of cryopreserved bovine sperm. In each of 23 Simmental AI bulls three ejaculates were collected. In a split sample design ejaculates were diluted by using Triladyl(®) extender without and with the addition of 5mM sodium pyruvate. In acute ischaemia, glucose-insulin-potassium administration reduces mortality and β-adrenoceptor antagonists have favourable effects on the outcome of ischaemic heart disease. The present study was designed to investigate whether insulin (1.4×10−7 M) and the β-adrenoceptor antagonist, propranolol (10−5 M), increase hypoxic vasodilation in correspondence with changes in glycolysis. Porcine coronary arteries, precontracted with 10−5 M prostaglandin F2α, were mounted in a pressure myograph and a microdialysis catheter was inserted in the tunica media. Hypoxic vasodilation, interstitial lactate/pyruvate ratio and interstitial glucose were measured at low (2 mM) and high (20 mM) glucose concentrations. This study demonstrates robust survival of NSPCs under conditions of anoxia and mitochondrial inhibition, with increased importance of glycolysis for survival compared to primary cortical neurons. Using an inducible Cre/loxP approach, we also show that sustained deletion of exon 1 of the Hif1a gene within undifferentiated NSPCs moderately increases mitochondrial pyruvate utilization, but has no effect on other parameters of metabolism that were tested here, including the ability of NSPCs to withstand prolonged anoxia.
