In mitosis, a dramatic decrease in endocytosis rate has been reported (Berlin et al., 1978). We repeated those observations (Fig. 1) using either fluorescent dextran or FM1-43 uptake as a measure of endocytosis rate by measuring the number of vesicles per cell and the total fluorescence intensity. With fluorescent dextran, the endocytosis rate normally decreased to ∼10% of interphase level in metaphase. Optical methods for recording electrical activity in the goldfish optic tectum were evaluated. Tectal slices, with a short section of the optic nerve attached, were stained with a fluorescent styryl dye. Potential- dependent fluorescence changes following optic nerve stimulation were monitored with a photodiode. We found that large optical signals could be obtained. Experimental manipulations of the slice bathing solution permitted us to identify several events that contributed to the optical response, including activity in afferent fibers, excitatory and inhibitory postsynaptic potentials, and presumptive glial depolarizations. Here we explore drugs that cause loss of MCL1, particularly the potent new cyclin-dependent kinase inhibitor dinaciclib, which knocks down MCL1 by inhibiting CDK9. In this thesis, the syntheses of oligosaccharides for interaction studies with various lectins are described. The first section reports the syntheses of tetra, tri- and disaccharides corresponding to truncated versions of the glucosylated arm of Glc1Man9(GlcNAc)2, found in the biosynthesis of N-glycans. The thermodynamic parameters of their interaction with calreticulin, a lectin assisting and promoting the correct folding of newly synthesised glycoproteins, were established by isothermal titration calorimetry. In the second section, a new synthetic pathway leading to the same tetra- and trisaccharides is discussed. Adoption of a convergent strategy and of a different protecting group pattern resulted in significantly increased yields of the target structures. To monitor C3 activation in the serum, we measured iC3b by enzyme immunoassay. The cleavage fragment, iC3b, is a product of C3 activation that forms in the alternative pathway when C3b is cleaved by factor I. Typically this is something on the order of 0.5-2.5 µg of a 1 kb fragment on a 30 ml 1% mini gel. These numbers are guess-timates so your milage may vary. Run the gel normally and then place in a 0.002% methylene blue (w/v, Sigma M-4159) solution in 0.1X TAE (0.004M Tris 0.0001 M EDTA) for 1-4 h at room temp (22°C) or overnight at 4°C. Diffusion of the DNA does not seem to be a problem for fragments as small as 100 bp (3% Nusieve:1%agarose gel). This avoids background issues associated with staining with 0.02% methylene blue for 30-60 min and then destaining for what seems to be forever If destaining is needed to increase the visibility of the bands place the gel in 0.1X TAE with gentle agitation changing the buffer every 30 - 60 min until you are satisfied with the degree of destaining. Two experiments were conducted to determine the effect of CPC concentration and dwell time after treatment on microbial populations (tables 1 and 2). All dwell times after 1% CPC application to cattle hides resulted in lower APC and EB counts than controls (Table 1). However, for APC, 30 sec dwell time had the lowest counts and 30 min, 2 h, and 4 h were not different from one another. For EB, 30 min dwell time had the highest counts and 30 sec, 2 h, and 4 h were not different from one another. For CPC concentration (Table 2), APCs were not different among 0.5, 1, and 3% CPC after either 30 sec or 4 h dwell time, but 4 h counts were lower than 30 sec counts.
