Pharmacologically elevating brain endocannabinoids (eCBs) share anxiolytic and fear extinction-facilitating properties with classical therapeutics, including the selective serotonin reuptake inhibitor, fluoxetine. There are also known functional interactions between the eCB and serotonin systems and preliminary evidence that antidepressants cause alterations in brain eCBs. However, the potential role of eCBs in mediating the facilitatory effects of fluoxetine on fear extinction has not been established. Here, to test for a possible mechanistic contribution of eCBs to fluoxetines proextinction effects, we integrated biochemical, electrophysiological, pharmacological, and behavioral techniques, using the extinction-impaired 129S1/Sv1mJ mouse strain. Chronic fluoxetine treatment produced a significant and selective increase in levels of anandamide in the BLA, and an associated decrease in activity of the anandamide-catabolizing enzyme, fatty acid amide hydrolase. It is commonly thought that symptoms of schizophrenia arise as a consequence of increased dopamine transmission and that antipsychotic drugs alleviate symptoms by blocking this activity. The study team used D-amphetamine (a drug that can induce psychotic symptoms in humans) to model psychotic symptoms seen in schizophrenia in an animal model. D-amphetamine increases dopamine release disrupting the mouses ability to ignore relevant stimuli in the environment. This effect is reversed by administering antipsychotic drugs.. The research team was surprised to see the same behavioural effect in mice with D2 receptors genetically deleted. As a clinical psychologist with 25 years practice experience in the USA and now working in the UK for the last couple of years, I wonder how specific these results may be to a UK population sample. While I am unaware of comparable research in the US, my experience tells me that Americans are significantly more optimistic about medications in general, and psychopharmacology in particular, than are residents of the UK. I would predict that Americans would have a greater tendency to see a biological explanation of psychiatric symptomatology as good news, with the implication that the chemical imbalance might potentially be rectified efficiently through psychopharmacological means. Further, my guess is that a random, stratified sampling of Americans would find greater pessimism in the face of a purely psychological explanation for psychiatric symptoms, with the concomitant implication of a need for psychotherapy (with the likely expectation that this might take years or not be all that effective). ...

Last year, Dr. Costa published a paper in Neuropsychopharmacology revealing that the N-methyl-D-aspartic acid (NMDA) receptor antagonist memantine rescues performance deficits of the Ts65Dn mouse on a fear conditioning test. Dr. Costa is now leading a clinical trial of memantine based on his 2008 study in mice, and expects to publish the findings later this year. SAN JUAN, PUERTO RICO - The manufacturers of risperidone and olanzapine, two new drugs for the treatment of schizophrenia, are waging a spirited battle over which of these atypical antipsychotics is the best for the treatment of the disorder. About 1% of the population -- 2.5 million people in the United States -- suffer from schizophrenia. Symptoms of the disorder include positive symptoms: hallucinations, delusions and hearing voices; and negative symptoms: social withdrawal and little or inappropriate emotional responses.
