Protein instability and immunogenicity are two main roadblocks to the clinical success of novel protein drug delivery systems. In this commentary, we discuss the need for more extensive analytical characterization in relation to concerns about protein instability in injectable drug delivery systems for sustained release. We then will briefly address immunogenicity concerns and outline current best practices for using state-of-the-art analytical assays to monitor protein stability for both conventional and novel therapeutic protein dosage forms. Next, we provide a summary of the stresses on proteins arising during preparation of drug delivery systems and subsequent in vivo release. The stability of the native state of proteins is, therefore, of paramount importance both in vivo and for many biotechnological applications in vitro. Protein stability is determined by the difference between the free energies of the native and non-native states. It follows that protein stabilization can be achieved via two different strategies: (i) positive design by introducing favorable interactions between residues in contact in the native state; and (ii) negative design by introducing unfavorable interactions between residues in contact in the non-native states. We propose a model of competition of nn species in a chemostat, with constant input of some species. We mainly emphasize the case that can lead to coexistence in the chemostat in a non-trivial way, i.e., where the n−1n−1 less competitive species are in the input. We prove that if the inputs satisfy a constraint, the coexistence between the species is obtained in the form of a globally asymptotically stable (GAS) positive equilibrium, while a GAS equilibrium without the dominant species is achieved if the constraint is not satisfied. While forces driving stability are now well understood, much less is known about designing specificity. We use a Protein-fragment Complementation Assay (PCA), which utilises semi-rational design to select antagonists of Jun and Fos peptides that interact in the oncogenic Activator Protein-1 transcriptional regulator. We have devised a Competitive and Negative Design Initiative (CANDI) technique to increase target-specificity in the PCA system by expressing potential off-targets during selection. Peptides generated have allowed us to predict coiled coil stability and specificity based only the primary sequence, and to consequently design peptides. I usually would bring one of these when doing casual work in the field so I do not have to worry about losing it or damaging it.. But since these meters are so inaccurate, even if brand new ones might be spot on their long term stability can not be guaranteed. My CEN-TECH multimeter for instance, had an error of more than 10% when it was just bought. So it is a good idea to calibrate these cheap meters at least once a while so that the measured values do not drift too much.. A delayed predator-prey system with Holling type II functional response and stage structure for both the predator and the prey is investigated. By analyzing the corresponding characteristic equations, the local stability of each of the feasible equilibria of the system is addressed and the existence of a Hopf bifurcation at the coexistence equilibrium is established. By means of persistence theory on infinite dimensional systems, it is proved that the system is permanent. By using Lyapunov functions and the LaSalle invariant principle, the global stability of each of the feasible equilibria of the model is discussed. Numerical simulations are carried out to illustrate the main theoretical results.
