Background Dibutyl phthalate (DBP) is predominantly used as a plasticizer inplastics to make them flexible. Extensive use of phthalates in both industrial processes and other consumer products has resulted in the ubiquitous presence of phthalates in the environment. In order to better determine the level of pollution in the environment and evaluate the potential adverse effects of exposure to DBP, immunoassay for DBP was developed. Methodology/Principal Findings A monoclonal antibody specific to DBP was produced from a stable hybridoma cell line generated by lymphocyte hybridoma technique. An indirect competitive enzyme-linked immunosorbent assay (icELISA) employing direct coating of hapten on polystyrene microtiter plates was established for the detection of DBP. Polystyrene surface was first oxidized by permanganate in dilute sulfuric acid to generate carboxyl groups. Then dibutyl 4-aminophthalate, which is an analogue of DBP, was covalently linked to the carboxyl groups of polystyrene surface with

Nanosphere lithography, an inexpensive and high throughput technique capable of producing nanostructure (below 100 nm feature size) arrays, relies on the formation of a monolayer of self-assembled nanospheres, followed by custom-etching to produce nanometre size features on large-area substrates. A theoretical model underpinning the self-ordering process by centrifugation is proposed to describe the interplay between the spin speed and solution concentration. The model describes the deposition of a dense and uniform monolayer by the implicit contribution of gravity, centrifugal force and surface tension, which can be accounted for using only the spin speed and the solid/liquid volume ratio. We demonstrate that the spin recipe for the monolayer formation can be represented as a pathway on a 2D phase plane. The model accounts for the ratio of polystyrene nanospheres (300 nm), water, methanol and surfactant in the solution, crucial for large area uniform and periodic monolayer deposition. The ...
Bio VI Surface functionalization of biological inert polymers, for example polypropylene and polystyrene, with material binding peptides facilitates an efficient immobilization of enzymes, bioactive peptides or antigens at ambient temperature in water. The developed Peptide Polymer evolution protocol (PePevo) is robust directed evolution protocol that enables to tailor polymer binding anchor peptides for efficient binding under application conditions. Key for a successful directed evolution campaign was to develop an epPCR protocol with a very high mutation frequency, 60 mutations per kb, to ensure sufficient diversity in the peptides LCI and Tachystatin A2. LCI and Tachystatin A2 were genetically fused to the reporter eGFP to quantify peptide binding on polypropylene and polystyrene surfaces by fluorescence analysis. PePevo was validated in two directed evolution campaigns for both peptides and polymers, LCI and polypropylene as well as Tachystatin A2 and Polystyrene. The nonionic surfactant ...

The local surface properties of polystyrene (PS) dishes incubated for 14 h with gram‐negative cells (Shewanella putrefaciens) were investigated by atomic force microscopy (AFM) in aqueous solutions at two pH values (4 and 10). The AFM images and force curves revealed the presence of a polymeric layer adsorbed onto the PS surfaces in acid media. In contrast, no evolution was observed in the case of basic media
The fabrication of cell sheets is a major requirement for bottom-up tissue engineering purposes (e.g. cell sheet engineering) and regenerative medicine. Employing thermoresponsive polymer coatings as tissue culture substrates allows for the mild, temperature-triggered detachment of intact cell sheets along with their extracellular matrix (ECM). It has been shown before that biocompatible, thermoresponsive poly(glycidyl ether) monolayers on gold substrates can be utilized to harvest confluent cell sheets by simply reducing the temperature to 20 °C. Herein, we report on the first poly(glycidyl ether)-based coating on an application-relevant tissue culture plastic substrate. We devised a simple, substrate geometry-independent method to functionalize polystyrene (PS) surfaces from dilute ethanolic solution via the physical adsorption process of a thermoresponsive poly(glycidyl ether) block-copolymer (PGE) bearing a short, hydrophobic, and photoreactive benzophenone (BP) anchor block. Subsequently, ...
Mueller matrices of dense aqueous suspensions of different concentrations are measured with a phase-modulated Mueller ellipsometer as a function of the scattering angle. Different concentrations of a solution containing 404-nm-diameter polystyrene latex spheres dispersed in water were prepared. Experimental results are compared with a three-dimensional Monte Carlo simulation of the propagation of photons with the cell geometry accounted for. The Fresnel laws and the Mie theory determine the changes in direction and polarization during the propagation of the photon. Excellent agreement over the whole angular range is found between experimental and simulated Mueller matrices.. © 2001 Optical Society of America. Full Article , PDF Article ...

The Polystyrene Packaging Council (PSPC) provides facilities to recycle all of you polystyrene waste, such as food packaging, protective and display packaging. Polystyrene is produced from naphtha, a product remaining after the transformation process of petroleum in refineries. A key benefit of polystyrene is that it can be recycled and the PSPC is initiating and setting up recycling programmes all over South Africa.. Polystyrene is generally not a high profile target in recycling terms but many companies, local authorities and individuals may not have considered the implications of just how much polystyrene they are dumping. The recycling of polystyrene is within everyones reach and can be done easily with the help of the PSPC.. Find out if there is a local drop off site for polystyrene or a kerbside pick-up service in your area and use it. You can also help to start a school, church or community collection programme.. The PSPC is also embarking on training session for schools to help learners ...

Rapid Electrokinetic Patterning (REP) is an optoelectric technique for trapping and translating micro- and nanoparticles non-invasively. It uses a combination of laser-induced AC electrothermal flow and particle-electrode interactions in the presence of a uniform AC electric field. The trapping is governed by laser power, electric field strength, AC frequency and dielectric properties of the particle and the medium. A REP trap has an AC frequency, termed critical frequency, above which particles cannot be trapped. It is expected to be dependent on dielectric properties of the particle and the medium. However, we propose that the particle concentration and AC field strength also influence the critical frequency. In our experiments, we test 1 µm polystyrene microspheres in REP under the condition of three particle concentrations and two electric field strengths while keeping the laser power fixed at 25mW. We find that as the particle concentration increases, the critical frequency increases. Besides, by
RNP : Recombinant RNP-68 and RNP-A antigens are bound to polystyrene microspheres, which are impregnated with fluorescent dyes to create a unique fluorescent signature. RNP antibodies, if present in diluted serum, bind to the RNP antigens on the microspheres. The microspheres are washed to remove extraneous serum proteins. Phycoerythrin (PE)-conjugated antihuman IgG antibody is then added to detect IgG anti-RNP bound to the microspheres. The microspheres are washed to remove unbound conjugate, and bound conjugate is detected by laser photometry. A primary laser reveals the fluorescent signature of each microsphere to distinguish it from microspheres that are labeled with other antigens, and a secondary laser reveals the level of PE fluorescence associated with each microsphere. Results are calculated by comparing the median fluorescence response for RNP microspheres to a 4-point calibration curve.(Package insert: Bioplex 2200 ANA Screen. Styrofoam is a trademarked brand of closed-cell extruded polystyrene foam (XPS), commonly called Blue Board manufactured as foam continuous building insulation board used in walls, roofs, and foundations as thermal insulation and water barrier. This material is light blue in color and is owned and manufactured by The Dow Chemical Company.[1] In the United States and Canada, the colloquial use of the word styrofoam refers to another material that is usually white in color and made of expanded (not extruded) polystyrene foam (EPS). It is often used in disposable coffee cups and coolers, and as cushioning material in packaging.[1] The trademarked term is used generically although it is a different material from the extruded polystyrene used for Styrofoam insulation. The Styrofoam brand polystyrene foam, which is used for craft applications, can be identified by its roughness and the crunch it makes when cut.[citation needed] Additionally, it is moderately soluble in many organic solvents, ...

Alternative, non-electrochemistry-based technologies for continuous glucose monitoring are needed for eventual use in diabetes mellitus. As part of a programme investigating fluorescent glucose sensors, we have developed fibre-optic biosensors using glucose/galactose binding protein (GBP) labelled with the environmentally sensitive fluorophore, Badan. GBP-Badan was attached via an oligohistidine-tag to the surface of Ni-nitrilotriacetic acid (NTA)-functionalized agarose or polystyrene beads. Fluorescence lifetime increased in response to glucose, observed by fluorescence lifetime imaging microscopy of the GBP-Badan-beads. Either GBP-Badan agarose or polystyrene beads were loaded into a porous chamber at the end of a multimode optical fibre. Fluorescence lifetime responses were recorded using pulsed laser excitation, high speed photodiode detection and time-correlated single-photon counting. The maximal response was at 100 mM glucose with an apparent K(d) of 13 mM (agarose) and 20 mM (polystyrene), and

Great news from the west coast! Last week, the San Francisco County Board of Supervisors unanimously passed a ban on the sale of polystyrene foam. Foam packing, cups and mooring buoys will be prohibited starting January 1, 2017. This is a major win for the health of our ocean and marine life!. As you may already know, the problems associated with expanded polystyrene (foam) products is that they often fragment into small pieces once in the ocean, where fish, sea turtles or seabirds can mistakenly eat the tiny plastic bits. Nearly 425,000 foam cups, plates and food containers were removed from beaches by volunteers during the 2015 International Coastal Cleanup alone. And even more astounding are the more than 950,000 pieces of foam volunteers found on beaches around the globe during the 2015 Cleanup.. The ban in San Francisco is another step towards trash free seas! We continue to see dirty beaches and debris floating on the oceans surface. Thats why my colleagues and I are committed to ...
The Australian Packaging Covenant has recognised EPS as a priority waste stream and mandated to remove all packaging from landfill by 2025 to reduce this strain.. Businesses need to prepare for the packaging ban before they are caught out and left scrambling, said Chris Tangey, General Manager of Expanded Polystyrene recycler Ecycle Solutions.. South Australia has already implemented a landfill ban, with Victoria having confirmed that a similar ban will be introduced in the near future. It is only a matter of time before other states follow this lead.. The responsibility of recycling Expanded Polystyrene falls on businesses to drive change, said Tangey.. By starting with businesses, a large percentage of EPS waste will be recycled, prompting customers who receive goods to follow suit.. In the 2018/19 financial year, Ecycle Solutions helped 299 retail stores recycle their EPS waste. The volume of loose EPS waste collected from these stores equaled the volume that would fill the Melbourne ...

Local lawmakers in New York City have introduced legislation that would effectively ban the use of polystyrene packaging at restaurants and other foodservice outlets in the area.. The legislation, introduced June 12 into the City Council by members Lewis A. Fidler, Vincent J. Gentile and Letitia James in conjunction with Mayor Michael Bloomberg, would prevent operators from using the plastic foam packaging at fullservice and quickservice restaurants, cafes, delicatessens, coffee shops, grocery stores, vending trucks or carts and cafeterias.. Mayor Bloomberg initially proposed the ban during his State of the City address in February.. If passed, the regulation, which would be enforced by the citys departments of health and consumer affairs, would take effect July 1, 2015. According to the proposed bill, operators who do not comply with the rule would face fines of $250 for the first violation, $500 for the second and $1,000 for the third. Manufacturers of polystyrene packaging also would be ...

Nano-sized materials could find multiple applications in medical diagnosis and therapy. One main concern is that engineered nanoparticles, similar to combustion-derived nanoparticles, may cause adverse effects on human health by accumulation of entire particles or their degradation products. Chronic cytotoxicity must therefore be evaluated. In order to perform chronic cytotoxicity testing of plain polystyrene nanoparticles on the endothelial cell line EAhy 926, we established a microcarrier cell culture system for anchorage-dependent cells (BioLevitatorTM). Cells were cultured for four weeks and exposed to doses, which were not cytotoxic upon 24 hours of exposure. For comparison, these particles were also studied in regularly sub-cultured cells, a method that has traditionally been used to assess chronic cellular effects. Culturing on basal membrane coated microcarriers produced very high cell densities. Fluorescent particles were mainly localized in the lysosomes of the exposed cells. After ...

Abstract: A commercial and very hydrophobic styrene-divinylbenzene matrix, MCI GEL CHP20P, has been compared to octyl-Sepharose beads as support to immobilize three different enzymes: lipases from Thermomyces lanuginosus (TLL) and from Rhizomucor miehie (RML) and Lecitase Ultra, a commercial artificial phospholipase. The immobilization mechanism on both supports was similar: interfacial activation of the enzymes versus the hydrophobic surface of the supports. Immobilization rate and loading capacity is much higher using MCI GEL CHP20P compared to octyl-Sepharose (87.2 mg protein/g of support using TLL, 310 mg/g using RML and 180 mg/g using Lecitase Ultra). The thermal stability of all new preparations is much lower than that of the standard octyl-Sepharose immobilized preparations, while the opposite occurs when the inactivations were performed in the presence of organic co-solvents. Regarding the hydrolytic activities, the results were strongly dependent on the substrate and pH of measurement. ...

A majority of all biologically active proteins are glycosylated and various diseases have proven to correlate with alterations in protein glycosylation. Sensitive identification of different glycoprotein glycoforms is therefore of great diagnostic value. Here we describe a method with potential for glycoprotein profiling, based on lectins as capture probes immobilized on particulate substrates in the nm-range. The nanoparticles present high concentrations of attachment sites for specific ligands and cause minimal steric hindrance to binding. In the present model study the mannose-binding lectin ConA has been coupled to polystyrene nanoparticles via a poly(ethyleneoxide) linker which protects the protein conformation and activity and prevents unspecific protein adsorption. The ConA-coated particles are accommodated at different spots on the analytical surface via oligonucleotide linkage. This attachment, which relies on the hybridization of complementary oligonucleotides, allows firm fixation of ...

Ambient inhalable particulate matter acts as an efficient carrier of allergens allowing penetration deep into the distal airways. Epidemiological studies have shown an association between the increased incidence of asthma and levels of ambient particulate matter from air pollutants. Most experimental models investigating the pathogenesis of allergic asthma have used soluble allergens (sAgs) whereas knowledge is limited regarding the responses evoked by particulate allergens (pAgs). In this study, pathological responses to sAgs were compared with the same amount of allergens adsorbed on to the surface of polystyrene particles, in sensitised mice.. Regardless of allergen type, airways hyper-responsiveness and eosinophil infiltration were significantly greater with pAgs compared with sAgs. The response was blunted in mast cell (MC)-deficient mice suggesting that MCs are crucial in this response. MCs were able to distinguish between pAgs and sAgs. A fivefold greater MC IgE receptor-mediated response ...
