Purpose: : Molecular imprinting has been used to prolong drug release in drug delivery applications. Hyaluronic acid (HA) has been shown to decrease lysozyme sorption and improve wettability in model contact lenses when used as an internal wetting agent. It was of interest to investigate the release of imprinted HA to determine if imprinting prolonged HA release and if this released HA decreased lysozyme sorption. Methods: : Model conventional (pHEMA) and silicone (pHEMA/TRIS) hydrogels were synthesized. HA was added to the reaction mixture prior to UV polymerization. The release of HA into PBS was measured using UV Spectroscopy at 280 nm. Throughout the release, hydrogels were removed from PBS, dried and then incubated with radiolabeled lysozyme for 2 hrs (pHEMA) and 24 hrs (pHEMA/TRIS) to determine if the released HA could decrease sorption. The internal morphologies for a series of heterogeneous PHEMA and P[HEMA-co-MeO-PEGMA] [PHEMA = poly(2-hydroxyethyl methacrylate), MeO-PEGMA = poly(ethylene glycol) methyl ether methacrylate] hydrogels were characterized by scanning electron microscopy (SEM) in conjunction with a sample drying procedure, and by laser scanning confocal microscopy (LSCM) without prior drying. Compared to SEM, LSCM was far simpler and more rapid technique for imaging hydrogels. An optical biosensor for lactate detection is described. By encapsulating enzyme-phosphor sensing molecules within permeable hydrogel materials, lactate-sensitive emission lifetimes were achieved. The relative amount of monomer was varied to compare three homo- and co-polymer materials: poly(2-hydroxyethyl methacrylate) (pHEMA) and two copolymers of pHEMA and poly(acrylamide) (pAam). The effect of 2-hydroxyethyl methacrylate (HEMA) grafting on the structure and physicochemical properties of methylcellulose (MC)-based films was investigated by using gamma (γ) radiation. HEMA (0.1-1%, w/w) was incorporated into MC-based aqueous formulation containing 1% MC, 0.5% vegetable oil, 0.25% glycerol and 0.025% Tween®80. Films were obtained by casting and then exposed to γ-radiation (5-25 kGy). Results revealed that films containing 1% HEMA showed the highest puncture strength (PS) values (282 N/mm) at 10 kGy. Measurements of water vapor permeability (WVP) demonstrated that films exposed to high radiation doses (25 kGy) generated highest barrier properties (5.5 g mm/m2 day kPa) due to a possible grafted complex MC-HEMA. Fourier transform infrared (FTIR) analysis showed a decrease of the vinyl vibration band from HEMA at 1636 cm−1 after grafting, suggesting that graft polymerization occurred via a HEMA reaction from vinylidene into methylene group, combined with HEMA homopolymerization.
