Hyperbranched, aliphatic polyesters of theoretically calculated molar masses 1200-44 300 were synthesized in the molten state from 2,2-bis(hydroxymethyl)propionic acid (repeating unit of AB(x) type) and 2-ethyl-2-(hydroxymethyl)-1,3-propanediol (core molecule) using acid catalysis. The synthesis procedure was a pseudo-one-step reaction where stoichiometric amounts corresponding to each generation were added successively. The resulting polymers were glassy, slightly yellow solids at room temperature with hydroxyl groups as terminal groups. The degree of branching of the polyesters was determined with the help of model compounds using C-13-NMR and was found to be near 80%. The material exhibited good thermal stability as analyzed with TGA in a nitrogen atmosphere. Glass transition temperatures were determined using DSC and were found to be about 40 degrees C and to be relatively insensitive to variations in molar mass.. ...

There is an increasing need for synthetic bone substitute materials that decrease the need for allografts and autografts. In this study, composites of β-TCP and a biodegradable poly(L-lactide-co-ε-caprolactone) were manufactured using extrusion to form biodegradable composites with high β-TCP contents for osteoconductivity. The hydrolytic degradation of the composites containing 0, 10, 20, 35 and 50% of β-TCP was studied in vitro for 52 weeks. During the study, it was observed that β-TCP did not have an effect on the degradation rate of the polymer matrix. However, the crystallinity of the materials increased throughout the test series and changes in Tgs were also observed as the comonomer ratio of the polymer matrix changed as the degradation proceeded. 摘要:Based on the metabolic analysis, a succinate and polyhydroxybutyrate (PHB) co-production pathway was designed and engineered in Escherichia coli MG1655. Batch cultivation of the engineered E. coli revealed that it was able to accumulate both extracellular succinate and intracellular PHB simultaneously. PHB accumulation not only improved succinate production, but also reduced pyruvate and acetate secretion. With a consumption of 45 g l(-1) glucose, E. coli QZ1112 was shown to accumulate 24.6 g l(-1) succinate and 4.95 g l(-1) PHB in batch fermentation. The PHB content reached 41.3 wt.% of its cell dry weight, which suggested that the cell debris can be used as value added by-product. ...

In vitro biocompatibility and infiltration behaviour of 3T3 GFP dermal fibroblasts, as an exemplar cell type on electrospun PCL scaffolds, were investigated. Stem cell and tissue engineering offer us with a unique opportunity to research and develop new therapies for treating various diseases that are otherwise incurable using traditional medicines. However, development of these new therapies replies upon the establishment of in vitro cell culture and differentiation systems that mimic in vivo microenvironments required for cell-cell and cell-ECM interaction. The development of these cell culture systems depends upon the identification of appropriate biomaterials and cell sources. Biomaterials should be carefully selected and fabricated into scaffolds for supporting cell growth and differentiation. In this study, we explored the fabrication of 3D electrospun nanofiber scaffolds and demonstrated the feasibility of using these scaffolds for supporting cell growth. Hybrid constructs represent substantial progress in tissue engineering (TE) towards producing implants of a clinically relevant size that recapitulate the structure and multicellular complexity of the native tissue. They are created by interlacing printed scaffolds, sacrificial materials, and cell-laden hydrogels. A suitable biomaterial is a polycaprolactone (PCL); however, due to the higher viscosity of this biopolymer, three-dimensional (3D) printing of PCL is slow, so reducing PCL print times remains a challenge. We investigated parameters, such as nozzle shape and size, carriage speed, and print temperature, to find a tradeoff that speeds up the creation of hybrid constructs of controlled porosity. We performed experiments with conical, cylindrical, and cylindrical shortened nozzles and numerical simulations to infer a more comprehensive understanding of PCL flow rate. Thermo-responsive multiblock poly(ester urethane)s comprising poly(ε-caprolactone) (PCL), poly(ethylene glycol) (PEG), and poly(propylene glycol) (PPG) segments were synthesized. The copolymers were characterized by GPC, NMR, FTIR, XRD, DSC and TGA. Water-swelling analysis carried out at different temperatures revealed that the bulk hydrophilicity of the copolymers could be controlled either by adjusting the composition of the copolymer or by changing the temperature of the environment. These thermo-responsive copolymer films formed highly swollen hydrogel-like materials when soaked in cold water and shrank when soaked in warm water. The changes are reversible. The mechanical properties of the copolymer films were assessed by tensile strength measurement. These copolymers were ductile when compared to PCL homopolymers. An Enzyme Cascade Synthesis of ε-Caprolactone and its Oligomers
Poly-ε-caprolactone (PCL) is chemically produced on an industrial scale in spite of the need for hazardous peracetic acid as an oxidation reagent. Although Baeyer-Villiger monooxygenases (BVMO) in principle enable the enzymatic synthesis of ε-caprolactone (ε-CL) directly from cyclohexanone with molecular oxygen, current systems suffer from low productivity and are subject to substrate and product inhibition. The major limitations for such a biocatalytic route to produce this bulk chemical were overcome by combining an alcohol dehydrogenase with a BVMO to enable the efficient oxidation of cyclohexanol to ε-CL. Key to success was a subsequent direct ring-opening oligomerization of in situ formed ε-CL in the aqueous phase by using lipase A from Candida antarctica, thus efficiently solving the product inhibition problem and leading to the formation of oligo-ε-CL at more than 20 g L(-1) when starting from 200 mM cyclohexanol. Polyhydroxybutyrate (PHB) is one of the polyhydroxyalkanoates (PHAs) which has biodegradable and biocompatible properties. They are adopted in the biomedical field, in, for example, medical implants and drug delivery carriers. This study seeks to promote the production of PHB by Vibrio sp. BM-1, isolated from a marine environment by improving constituents of medium and implementing an appropriate fermentation strategy. This study successfully developed a glycerol-yeast extract-tryptone (GYT) medium that can facilitate the growth of Vibrio sp. BM-1 and lead to the production of 1.4 g/L PHB at 20 h cultivation. This study also shows that 1.57 g/L PHB concentration and 16% PHB content were achieved, respectively, when Vibrio sp. BM-1 was cultivated with MS-GYT medium (mineral salts-supplemented GYT medium) for 12 h. Both cell dry weight (CDW) and residual CDW remained constant at around 8.2 g/L and 8.0 g/L after the 12 h of cultivation, until the end of the experiment. However, both 16% of PHB content and
