The ≈80-kb pksX gene cluster in Bacillus subtilis encodes an unusual hybrid polyketide/nonribosomal peptide synthase that has been linked to the production of the uncharacterized antibiotic bacillaene. Multiple copies of this synthase, each similar in size to the ribosome, assemble into a single organelle-like complex with a mass of tens to hundreds of megadaltons. The resource requirements of the assembled megacomplex suggest that bacillaene has an important biological role. By coupling a differential NMR spectroscopic technique with genetically manipulated strains of B. subtilis, we were able to characterize the structure of this unusual secondary metabolite, which could not be predicted by using bioinformatic analysis. We report that bacillaene is a linear molecule with two amide bonds: the first links an α-hydroxy carboxylic acid to a ω-amino carboxylic acid containing a conjugated hexaene, and the second links the hexaene-containing carboxylic acid to an (ω-1) amino carboxylic acid ...

TY - JOUR. T1 - Rapamycin blocks IL-2-driven T cell cycle progression while preserving T cell survival. AU - Gonzalez, Juana. AU - Harris, Tom. AU - Childs, Geoffrey. AU - Prystowsky, Michael B.. PY - 2001/1/1. Y1 - 2001/1/1. N2 - Effective cellular immune responses require increases in antigen-specific T lymphocytes; IL-2 drives antigen-stimulated T cell proliferation and is largely responsible for the increases observed. We used microarrays containing ∼9000 mouse cDNAs to study IL-2-induced gene expression. IL-2 induces the expression of genes that regulate cell cycle progression, control cell survival, and increase synthetic and metabolic processes during proliferation. IL-2 also suppresses expression of genes that block cell cycle progression and promote cell death. Rapamycin inhibits IL-2-driven proliferation by downregulating the expression of genes required for key processes required for cell cycle progression. Rapamycin also preserves cell survival by keeping intact the IL-2-induced ...

In this work, we have investigated theoretically the structure/hyperpolarizability correlations of push-pull molecules sing a two-form two-state model. For this, we have defined a parameter MIX characterizing the mixing between the two-limiting resonance forms and thus ruling the molecular structure and polarization. Also, we have analyzed the solvent effect on the structure and on the polarizabilities of push-pull molecules using the Onsager reaction field theory. We have shown that the dependences of the linear and nonlinear polarizabilities on the solvent dielectric constant look similar to the structure/(hyper)polarizability correlations. Finally, we have investigated experimentally the solvent effect on a series of push-pull polyenes of increasing length. In particular, we have determined the scalar (mu) g(Beta) (0) product (where (mu) g is the ground-state dipole and (Beta) (0) the vector part of static quadratic hyperpolarizability tensor) in different solvents using the ...
The antifungal polyene macrolide nystatin is produced by Streptomyces noursei ATCC 11455. The nystatin biosynthesis gene cluster of Streptomyces noursei has been cloned and sequenced, and a biosynthesis route has been predicted. In the present work, investigation of genes presumably involved in post-PKS modifications of nystatin is described. The aim of this work was to better understand the nystatin biosynthesis and to further use this information for generation of novel nystatin analogues. Two PKS-modifications of the nystatin molecule were targeted in this study: glycosylation with mycosamine at C-19 and oxidation of the exocyclic methyl group at C-16.. Two genes putatively involved in mycosamine biosynthesis (NysDIII and NysDII) and one in attachment of mycosamine to the nystatin aglycone (nysDI) have been identified in the nystatin gene cluster. Their functions have been suggested, respectively, as a putative mannose dehydratase, aminotransferase and a glycosyltransferase. The deoxysugar ...

TARGET OF RAPAMYCIN (TOR) is an evolutionarily conserved Ser/Thr kinase that has been shown to control a network of growth signaling pathways in yeast and animal cells in response to environmental cues (Dennis et al., 1999; Thomas, 2000; Rohde et al., 2001; Bjornsti and Houghton, 2004; Abraham, 2005). The regulatory function of TOR kinase requires various interacting partners (Abraham, 2002; Jacinto and Hall, 2003). For instance, it has been shown that the activation of TOR kinase is mediated by interaction with the Regulatory-Associated Protein of TOR (RAPTOR), which is believed to recruit substrates for TOR (Hara et al., 2002; Kim et al., 2002; Nojima et al., 2003). On the other hand, TOR has been known to be inactivated by an antifungal immunosuppressant rapamycin, which binds with TOR and FKBP12 to form a toxic complex that mimics stress signals (Heitman et al., 1991; Koltin et al., 1991; Chen et al., 1995). FKBP12 is a peptidyl-prolyl isomerase that was originally identified as the ...

Two crystal forms of unligated FKBP12.6 exhibit multiple conformations in the active site and in the 80s loop, the primary site for known protein-recognition interactions. The previously unreported NMR backbone assignment of FKBP12.6 revealed extensive doubling of amide resonances, which reflects a slow conformational transition centered in the 80s loop. The primary known physiological function of FKBP12.6 involves its role in regulating the RyR2 isoform of ryanodine receptor Ca{sup 2+} channels in cardiac muscle, pancreatic β islets and the central nervous system. With only a single previously reported X-ray structure of FKBP12.6, bound to the immunosuppressant rapamycin, structural inferences for this protein have been drawn from the more extensive studies of the homologous FKBP12. X-ray structures at 1.70 and 1.90 Å resolution from P2{sub 1} and P3{sub 1}21 crystal forms are reported for an unligated cysteine-free variant of FKBP12.6 which exhibit a notable diversity of conformations. Background Toll-like receptor 5 (TLR5) is a potentially target related with immunosuppressant rapamycin. Our aim was to evaluate 131I-anti-TLR5 mAb for non-invasive in vivo graft visualization and quantification in rapamycin treated allogeneic transplantation model compared with nonspecific tracer 18F-FDG. Methods Labeling, binding and stability studies were performed. Allografted BALB/c mice were divided into rapamycin-treated and non-treated group, respectively. In vivo dynamic whole-body phosphor-autoradiography and ex vivo biodistribution studies were conducted. Results Phosphor-autoradiography imaging showed clear graft localization from 12 h onward after 131I-anti-TLR5 mAb injection, significantly higher graft uptake in rapamycin-treated group. Pretreatment with anti-TLR5 mAb blocked graft uptake, which were consistent with ex vivo biodistribution studies. As the nonspecific radiotracer, high 18F-FDG uptake was only observed in non-treated allorejection group, not in treated group. The ...

Fecapentaenes are a class of conjugated ether lipids which have been identified as the major component of human fecal mutagenicity in the Ames Salmonella mutagenesis assay. Human epidemiologic data have indicated that most healthy North American populations eating a western diet do excrete detectable levels of fecapentaenes. Excreted fecapentaene levels seem to reflect levels throughout the colonic lumen, and levels vary characteristically between individuals. Those individuals found to excrete high levels of fecapentaene appear, based on limited data, to be at decreased risk of colorectal neoplasia. Carcinogenicity studies in rats and mice have been predominantly negative, however, increased tumor incidence in mice exposed to fecapentaenes as neonates has recently been reported. Fecapentaenes are direct-acting genotoxins, which may react with DNA through free radical mechanisms, and/or aldehyde formation. Mechanisms by which fecapentaene-induced DNA damage may mediate carcinogenesis are ...

Invasive fungal infections are a significant cause of morbidity and mortality in children. Successful management of these systemic infections requires identification of the causative pathogen, appropriate antifungal selection, and optimisation of its pharmacokinetic and pharmacodynamic properties to maximise its antifungal activity and minimise toxicity and the emergence of resistance. This review highlights salient scientific advancements in paediatric antifungal pharmacotherapies and focuses on pharmacokinetic and pharmacodynamic studies that underpin current clinical decision making. Four classes of drugs are widely used in the treatment of invasive fungal infections in children, including the polyenes, triazoles, pyrimidine analogues and echinocandins. Several lipidic formulations of the polyene amphotericin B have substantially reduced the toxicity associated with the traditional amphotericin B formulation. Monotherapy with the pyrimidine analogue flucytosine rapidly promotes the emergence ...

Based on the enhanced fluorescence of amphotericin B in acid solutions, a quantitative assay for this polyene antibiotic has been developed that is sensitive and linear in the range of 0.1 to 10.0 muM. The binding of amphotericin B to Saccharomyces cerevisiae was assayed under various conditions as the amount bound to cells in a dialysis chamber or after centrifugation. Two types of binding were defined: weak, reversible binding occurred at 0 C or higher temperatures and even in the presence of inhibitors of energy metabolism, whereas strong, irreversible binding did not occur at 0 C and was inhibited when energy metabolism was blocked. Only strong binding was correlated with cell killing. Weak binding probably involves the outer layer of the membrane; strong binding probably requires disruption of hydrophobic regions of the cell membrane.
Resistance to cytotoxic and cytostatic treatments is a common limitation in cancer treatment. Rapamycin resistance was in part attributed to the rapamycin-induced increase in pAkt-Ser473 levels, which was documented in vitro and in vivo on biopsies of patients treated with the rapalog RAD001 (7) and was found to derive from the elimination of the negative feedback loop of the mTOR target p70 S6K on the PI3K pathway. In the present study, we sought to suppress the PI3K pathway upstreams with a somatostatin analogue that was previously shown to target this pathway (14, 15) to restore the control of Akt and therefore induce sensitivity to rapamycin treatment.. The serine/threonine kinase p70 S6K phosphorylates IRS-1 at serine residues, causing its inactivation. Rapamycin, by inhibiting mTOR, decreases p70 S6K activity and reduces serine IRS-1 phosphorylation, leading to its activation. Octreotide was previously reported to inhibit IRS-1 by decreasing its tyrosine phosphorylation levels through the ...
