Here we describe a new, robust method for the fabrication of amphiphilic Janus particles based on immobilization of polymethylsilsesquioxane (PMSQ) particles on polycarbonate (PC), a high impact-resistance polymer with superior mechanical properties. The immobilization of the particles on the PC microspores is performed via inverse solvent displacement method. PMSQ particles are added to a PC solution in tetrahydrofuran (THF), a good solvent for PC. The solution is then precipitated by the introduction of aqueous surfactant solution (antisolvent for PC) under an ultrasonic field. The present work describes a selection of strategies for the synthesis of functional aliphatic polycarbonates. Using an end-group functionalization strategy, a series of DNA-binding cationic poly(trimethylene carbonate)s was synthesized for application as vectors for non-viral gene delivery. As the end-group functionality was identical in all polymers, the differences observed in DNA binding and in vitro transfection studies were directly related to the length of the hydrophobic poly(trimethylene carbonate) backbone and the number of functional end-groups. Reaction parameters of importance include a suitable starting molar ratio of diaryl carbonate to dihydroxy aromatic compound, the most appropriate reaction time, reaction temperature and pressure ranges. The target polycarbonate is then prepared using reaction parameters indicated by the process model in a reactor which forms an element of the process model. Characteristics of the reactor which may be included in the process model include agitation rate and reflux ratio during the distillative removal of hydroxy aromatic compound formed as a by-product during the polymerization.
Plasma levels of granulocyte lactoferrin, granulocyte myeloperoxidase and granulocyte elastase in complex with alpha 1-proteinase inhibitor (E-alpha 1PI) were investigated in regular hemodialysis patients dialyzed with hollow-fiber dialyzers made from polycarbonate (FD 100) or cuprophan (GFS 120 H). Plasma levels of all these main granulocyte components increased significantly during hemodialysis. E-alpha 1PI levels were significantly higher in patients dialyzed with the polycarbonate compared with the cuprophan membrane, whereas the increases of myeloperoxidase and lactoferrin were not different for the two dialyzers. On the other hand, plasma C3a levels were higher in patients dialyzed with the cuprophan compared with the polycarbonate dialyzer. Therefore, granulocyte activation during hemodialysis does not necessarily need complement activation.

In order to get a more homogenous fraction, liposomes were extruded through 400 nm and 100 nm polycarbonate membranes. The surplus of free calcein was removed from the calcein-loaded liposomal solution by size exclusion chromatography (SEC). Because this method diluted the sample, the phospholipid concentration of the liposomal sample was determined by Bartlett method. The leakage assay of calcein-loaded liposomes was done by incubating the liposomes in solutions of sodium cholate, sodium taurocholate and sodium deoxycholate of different concentrations during a longer period of time.
