Furthermore APC has direct and indirect anti-inflammatory actions. It prevents leucocyte rolling, tissue factor exposure and tumor necrosis factor production by monocytes, thrombin-mediated inflammatory actions and apoptosis of endothelial cells [Van de Wouwer et al., 2004]. The latter function is mediated by endothelial protein C/APC receptor (EPCR), another transmembrane protein, which also participates in protein C activation [Van de Wouwer et al., 2004]. Protein C and APC can both bind to EPCR. When APC is bound to EPCR, it changes its substrate specificity and looses its ability to inactivate factors V and VIII, and it acquires the capacity for protease activated receptor- 1 (PAR-1) cleavage, through which, in cooperation with EPCR, it elicits anti-apoptotic effects in the endothelial cells [Van de Wouwer et al., 2004]. APC is aided in its anticoagulant functions by protein S ...

The antimicrobial properties of tulathromycin were investigated for M.haemolytica and P.multocida. Three invitro indices of antimicrobial activity, minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and time-kill curves, were established for six isolates of each organism. Each index was measured in two growth media: Mueller-Hinton broth (MHB) and calf serum. It was shown that MICs and MBCs were markedly lower in serum than in MHB. MHB:serum ratios for MIC were 47:1 (M.haemolytica) and 53:1 (P.multocida). For both serum and MHB, adjustment of pH led to greater potency at alkaline compared to acid pH. Tulathromycin MIC was influenced by size of inoculum count, being 4.0- to 7.7-fold greater for high compared to low initial counts. It was concluded that for the purpose of determining dosages for therapeutic use, pharmacodynamic data for tulathromycin should be derived in biological fluids such as serum. It is hypothesized that invitro measurement of MIC in broth, ...

Genome Announcements. 1(5). DOI: 10.1128/genomeA.00848-13. Interpretive Summary: The bacterium Mannheimia haemolytica is a prominent and costly cause of respiratory disease in domestic cattle. During periods of stress or concurrent respiratory viral infection some serotypes of the bacterium proliferate dramatically, increasing the odds of progression to pneumonic disease. The genomes of two isolates which exhibit dramatic proliferation were sequenced to facilitate molecular characterization of the phenomenon and the potential development of novel disease control methods. Technical Abstract: Here we report two genomes, one complete and one draft, from virulent bovine strains of Mannheimia haemolytica serotype A1 recovered prior to the field usage of modern antimicrobial drugs. ...

Idiopathic inflammatory myopathy is a group of disorders characterized by inflammation of the muscles used for movement (skeletal muscles). Idiopathic inflammatory myopathy usually appears in adults between ages 40 and 60 or in children between ages 5 and 15, though it can occur at any age.The primary symptom of idiopathic inflammatory myopathy is muscle weakness, which develops gradually over a period of weeks to months or even years. Other symptoms include joint pain and general tiredness (fatigue). There are several forms of idiopathic inflammatory myopathy, including polymyositis, dermatomyositis, and sporadic inclusion body myositis.Polymyositis and dermatomyositis involve weakness of the muscles closest to the center of the body (proximal muscles), such as the muscles of the hips and thighs, upper arms, and neck. People with these forms of idiopathic inflammatory myopathy may find it difficult to climb stairs, get up from a seated position, or lift items above their head. In some cases, ...

Mannheimia haemolytica strains isolated from North American cattle have been classified into two genotypes (1 and 2). Although members of both genotypes have been isolated from the upper and lower respiratory tracts of cattle with or without bovine respiratory disease (BRD), genotype 2 strains are much more frequently isolated from diseased lungs than genotype 1 strains. The mechanisms behind the increased association of genotype 2 M. haemolytica with BRD are not fully understood. To address that, and to search for interventions against genotype 2 M. haemolytica, complete, closed chromosome assemblies for 35 genotype 1 and 34 genotype 2 strains were generated and compared. Searches were conducted for the pan genome, core genes shared between the genotypes, and for genes specific to either genotype. Additionally, genes encoding outer membrane proteins (OMPs) specific to genotype 2 M. haemolytica were identified, and the diversity of their protein isoforms was characterized with predominantly unassembled,
Of the two active ingredients evaluated in this study, only DRAXXIN is approved for use in swine to treat SRD. The positive control treatment was penicillin G procaine, which was administered intramuscularly for two consecutive days at the rate of 5 mL per 100 lb body weight. Such off-label use of penicillin G procaine is reportedly widespread in the US, as a national survey conducted in 2006 noted that 43.9 per cent of randomly selected sites administer the antibiotic to nursery pigs and 46.6 per cent of randomly selected sites to grower/finisher pigs. The primary reason provided for giving penicillin G procaine was to treat respiratory disease.18 Thus, the positive control therapy used in this study reflected a commonly implemented strategy for treating naturally occurring cases of SRD. The comparison of DRAXXIN to penicillin G procaine was regarded as being useful in evaluating the efficacy of tulathromycin under typical field conditions from the time of treatment for SRD through harvest. ...

Exeggcute: combination of EGG and EXECUTE. EXECUTE means carry out a task, referring presumably to Exeggcute s efficiency in carrying out its attacks. EXECUTE also has a special meaning to put to death a condemned prisoner, but this is not likely the intended meaning in regard to Exeggcute, since killing is a violation of the Pokemon ethic (see Exeggutor). EXEGGCUTE may also be a combined form of CUTE, since Exeggcute is so cute, but scholars disagree on this point ...
In the present study, recombinant proteins representing the transmembrane domain of M. haemolytica outer membrane protein A (OmpA) from a bovine serotype A1 isolate (rOmpA1) and an ovine serotype A2 isolate (rOmpA2) were over-expressed, purified and used to generate anti-rOmpA1 and anti-rOmpA2 antibodies, respectively. An examination of the binding specificities of these antibodies to M. haemolytica isolates representing different OmpA subclasses revealed that cross-absorbed anti-rOmpA1 antibodies recognised OmpA1-type proteins but not OmpA2-type proteins; conversely, cross-absorbed anti-rOmpA2 antibodies recognised OmpA2-type proteins but not OmpA1 type proteins. This demonstrated that OmpA1 and OmpA2 are surface-exposed and could potentially bind to different receptors in cattle and sheep. The outer membrane subproteomes of seven M. haemolytica isolates and one M. glucosida isolate were also characterised and compared after growth in complex growth medium in order to identify OMPs with ...
The cost of Bovine Respiratory Disease to the beef industry due to death, poorer conversions, and therapy is estimated to cost more than $3 billion per year. Identifying and mitigating Bovine Respiratory Disease in cattle can be difficult due to the increased susceptibility for Bovine Respiratory Disease in high risk cattle. One management option to minimize an outbreak of respiratory disease is the use of metaphylaxis, the mass treatment of a group of calves to reduce the incidence and adverse effects of respiratory disease on high risk animals. Criteria used to determine the necessity of metaphylactic treatment against Bovine Respiratory Disease in feedlots can be based on several factors depending on feedlot preference; however, the primary criteria often considered are: a known history of no previous vaccinations, overall appearance of cattle, source of cattle, Bovine Respiratory Disease in calves received from same source previously, long shipping distance, season of the year, and light arrival
Although bovine herpesvirus-4 (BHV-4), a gammaherpesvirus lacking a clear disease association, has been demonstrated in many tissues during persistent BHV-4 infection, a likely site of virus persistence is in cells of the monocyte/macrophage lineage. To establish an in vitro model of persistent infection potentially useful for examining the molecular mechanisms of BHV-4 persistence/latency, we infected the bovine macrophage cell line BOMAC. Following extensive cell death, surviving cells were found to be persistently infected, maintaining the viral genome over many passages and producing low levels of infectious virus. Although selection was unnecessary for the maintenance of the viral genome, cells persistently infected with recombinant BHV-4 containing a neomycin-resistance gene could be selected with geneticin, thus confirming that persistent BHV-4 infection was compatible with cell survival and replication. Furthermore, persistent BHV-4 infection caused no decrease in the growth rate of BOMAC cells.
Numerous mechanisms have been suggested for how bacterial toxins kill susceptible mammalian cells. Several recent studies demonstrated the importance of mitochondrial targeting of toxins produced by H. pylori, C. difficile, and S. aureus to mitochondria (56). In these cases toxin-mediated cell death was caspase independent and did not result in typical PTPs in the MOM (14, 18).. Previously, we reported that M. haemolytica LKT induces apoptosis of BL-3 cells in a caspase-9-dependent manner and that in mitochondria isolated from LKT-intoxicated BL-3 cells there was gross damage to the MOM (4). Based on these observations, we hypothesized that LKT is transported into the cell and binds directly to mitochondria. In the present study, we first demonstrated that full-length LKT protein could be identified in purified mitochondrial lysates from LKT-treated BL-3 cells (Fig. 1A). Transfection of anti-LKT antibodies into BL-3 cells prevented binding of LKT to mitochondria. Confocal microscopy and flow ...
Bovine respiratory disease (BRD) accounts for up to 70 percent of health disruptions and $500 million in associated medical costs and production losses, thus receiving considerable attention from cattle health personnel in production and research alike. Experiments were conducted in 2 studies to investigate aspects of nutrition and animal health in receiving cattle. Blood chemistry and immune components of sick and healthy cattle, along with feed intake, growth, and feed efficiency were evaluated for methods potentially useful in BRD diagnosis or offsetting performance losses. Intake, blood chemistry analysis, and immune proteins known as acute phase proteins provided patterns with possible complementarity for more accurate objective BRD diagnosis. Feed efficiency was improved by supplementing amino acids to exceed nutrient requirements. Utilizing these findings could provide incremental improvements in the current stalemate against BRD, improving both beef industry profitability and animal ...

The current study demonstrates efficacy of the M hyo and PCV2 combination vaccine in 3 week old piglets in the presence of high PCV2 MDI level. Results of this study are in agreement with previous findings regarding the efficacy of the combination vaccine in the face of average PCV2 MDI levels (9.38 log2) [25].. Definitive diagnosis of EP is based on the demonstration of typical lung lesions [3]. The mean LLS in this study were significantly reduced in the vaccinated pigs, indicating the test products efficacy against M. hyo. In addition, lung lesions were also less severe in vaccinated pigs as nearly two thirds of vaccinated animals had a score , 10 compared to more than half of the controls with a score , 10. This finding further supports the vaccines efficacy against EP.. Moreover, during the finishing period, vaccinated pigs had a significantly better ADWG and the overall gain was 18 g per day greater than in the control pigs. The ADWG results were also in agreement with several previous ...
