In immunocompromised patients, human cytomegalovirus (HCMV) infection can lead to severe, life-threatening diseases, such as pneumonitis, hepatitis, gastrointestinal tract disease, and retinitis. We previously reported that a 70% ethanol extract of Elaeocarpus sylvestris leaves (ESE) inhibits human cytomegalovirus (HCMV) replication in vitro. In the present study, we determined the solvent fraction of ESE that inhibits HCMV replication using activity-guided fractionation. Activity-guided fractionation of ESE was performed to determine the solvent fraction that inhibits HCMV replication. Effects of solvent fractions on HCMV lytic gene expression and major immediate-early (MIE) enhancer/promoter activity were further investigated. Among the solvent fractions tested, the EtOAc fraction of ESE markedly reduced HCMV lytic gene expression and viral replication in vitro without exerting significant cytotoxic effects against human foreskin fibroblasts (HFF). Furthermore, the EtOAc fraction negatively affected

Gynostemma is a powerful adaptogen with the special ability of being able to restore homeostatis (balance and equilibrium) to the different body systems. The body systems include the cardiovascular, digestive, immune, nervous and reproductive systems. By restoring balance and equilibrium to these systems the body and mind become more harmonious. The chemical constituents responsible for the adaptogenic characteristic of Gynostemma are saponins (triterpene glycosides or gypenosides). Gynostemma contains the widest range of saponins from all the plants in nature. For example there are approximately four times as many saponins in Gynostemma Pentaphyllum as there are in ginseng. There are more than 100 different gypenosides in Gynostemma, all very similar to the ginsenosides of Panax Ginseng and Siberian Ginseng. Four of Gynostemmas saponins share the identical chemical structure as the saponins found in Ginseng and eleven more are nearly identical. There is a saying that Gynostemma is Ginseng at ...

Oxygen radical absorbance capacity (ORAC) is a method of measuring antioxidant capacities in biological samples in vitro. A wide variety of foods have been tested using this method, with certain spices, berries and legumes rated highly in extensive tables once published by the United States Department of Agriculture (USDA), but withdrawn in 2012 since no correlation between test results and biological activity could be determined, stating that no physiological proof in vivo existed in support of the free-radical theory. Although not all have been unilaterally dismissive, the majority position supports the USDA decision. Alternative measurements include the Folin-Ciocalteu reagent, and the Trolox equivalent antioxidant capacity assay. The assay measures the oxidative degradation of the fluorescent molecule (either beta-phycoerythrin or fluorescein) after being mixed with free radical generators such as azo-initiator compounds. Azo-initiators are considered to produce the peroxyl radical by ...

Natural News) Garlic contains many health benefits that people swear by, but theres also something to be said about aged garlic and its unique therapeutic properties. In fact, aged garlic is better at managing a persons cholesterol levels than raw garlic, as noted by scientists from Nanyang Polytechnic in Singapore. The study, which appeared in the Journal of Medicinal Food, compared the biological effects of both aged and raw garlic extracts using a clinical trial of people with hypercholesterolemia.. Hypercholesterolemia may sound like its straight out a medicine book; however, most people are familiar with its more common term - high cholesterol levels. The condition is marked by elevated levels of total cholesterol, which includes both low-density lipoprotein (LDL)-cholesterol and high-density lipoprotein (HDL)-cholesterol. Its also called dyslipidemia since reduced HDL-cholesterol levels and increased triglycerides accompany it. People with high cholesterol, for the most part, dont ...

Abstract: Antioxidant Actions of Dried Flower Extracts of Hibiscus sabdariffa L. On Sodium Arsenite - Induced Oxidative Stress in Rats
Gentamicin is a widely used antibiotic for the treatment of adverse urinary tract infections (UTI), which in turn causes nephrotoxicity to uroepithelial cells and hence an alternative safe herbal remedy is much desired to compensate these toxic effects. The bacterial adhesion to the uroepithelial cells is the primary step in UTI and it induces various immunogenic reactions leading to the generation of reactive oxygen species (ROS), which are detrimental to the cells survival. Inhibition of bacterial adherence to urinary tract epithelial cells has been assumed to account for the beneficial action ascribed to cystone (an ayurvedic polyherbal formulation) in the prevention of UTI. In this study, we have examined the effect of cystone on the adherence of pathogenic [2-(14)C]-acetate labeled Escherichia coli (MTCC-729) to rat proximal renal tubular cells (NRK-52E cells). Further, the antioxidant property of cystone was studied using hydrogen peroxide (400 μM) as a pro-oxidant in NRK-52E cells. The results

The immunomodulatory effects of the ethanol extract of Gynostemma pentaphyllum (GP-EX) were examined in electric footshock (EFS)-stressed mice. The mice were orally administered various doses of GP-EX for 7 days before exposure to EFS (duration: 3 min, interval: 10 s, intensity: 2 mA) once a day from day 8 for 14 days with continuous daily feeding of GP-EX. Oral administration of GP-EX to mice prevented EFS stress-induced immunosuppression as determined by the lymphoid organ (thymus and spleen) weight and cellularity. In addition, oral administration of GP-EX restored EFS-suppressed functional properties of mature lymphocytes in terms of concanavalin A-induced proliferation of splenocytes and lipopolysaccharide-induced cytokine production (TNF-α, IL-1β). Furthermore, we found that mice that were orally administered with GP-EX generated much more potent ovalbumin-specific cytotoxic T lymphocyte responses upon intravenous ovalbumin injection compared to the untreated controls. Radix salviae miltiorrhizae (Danshen in Chinese), a classic traditional Chinese medicine (TCM) herb, has been used for centuries to treat liver diseases. In this study, the preventive and curative potential of Danshen aqueous extract on acute/chronic alcoholic liver disease (ALD) and non-alcoholic fatty liver disease (NAFLD) was studied. The in vivo results indicated that Danshen could alleviate hepatic inflammation, fatty degeneration, and haptic fibrogenesis in ALD and NAFLD models. In the aspect of mechanism of action, the significant reduction in MDA levels in both ALD and NAFLD models implies the decreased levels of oxidative stress by Danshen. However, Danshen treatment could not activate the internal enzymatic antioxidant system in ALD and NAFLD models. To further explore the hepatoprotective mechanism of Danshen, an in silico-based network pharmacology approach was employed in the present study. The pharmacological network analysis result revealed that six potential active ingredients such as

Butein (3,4,2,4-tetrahydroxychalone), a plant polyphenol, is a major biologically active component of the stems of Rhus verniciflua Stokes. It has long been used as a food additive in Korea and as an herbal medicine throughout Asia. Recently, butein has been shown to suppress the functions of fibroblasts. Because fibroblasts are believed to play an important role in promoting the growth of breast cancer cells, we investigated the ability of butein to inhibit the clonogenic growth of small numbers of breast cancer cells co-cultured with fibroblasts in vitro. We first measured the clonogenic growth of small numbers of the UACC-812 human breast cancer cell line co-cultured on monolayers of serum-activated, human fibroblasts in the presence of butein (2 μg/mL) or various other modulators of fibroblast function (troglitazone-1 μg/mL; GW9662-1 μM; meloxican-1 μM; and 3,4 dehydroproline-10 μg/mL). In a subsequent experiment, we measured the dose-response effect on the clonogenic growth of UACC-812

CIR Safety Review: The cells just below the surface of the aloe plant produce a bitter yellow latex containing a number of anthraquinones. Reported to be phototoxic, anthraquinones are also gastrointestinal irritants responsible for the laxative effects claimed with aloe. An industry established limit for anthraquinones in aloe-derived material for non-medicinal use is 50 ppm or lower.. The CIR Expert Panel concluded that anthraquinone levels in the several Aloe barbadensis extracts are well understood and can conform to the industry-established level of 50 ppm. Although the phototoxicity of anthraquinone components of aloe plants has been demonstrated, several clinical studies of preparations derived from Aloe barbadensis plants demonstrated no phototoxicity, confirming that the concentrations of anthraquinones in such preparations are too low to induce phototoxicity. The characterization of Aloe barbadensis-derived ingredients from other species of aloe is not clear. In aloe-derived ...
