The patterned assignment of different cell fates, shortly termed patterning, lies at the basis of growth, development and reproduction of multicellular organisms. The single-layered epidermal tissue of Arabidopsis thaliana is characterized by hair-like cellular protuberances, the root hairs and the leaf trichomes, respectively, emerging from a subset of cells. Specification of hair and non-hair cell fate occurs in a tightly controlled fashion by gene regulatory networks of overlapping components that, however, often have opposite functions in root and shoot. Leaf epidermal pavement cells grow into jigsaw-piece shapes, highly deviating from such classical forms. We investigate the topology of developing Arabidopsis leaves composed solely of pavement cells. Image analysis of around 50,000 cells reveals a clear and unique topological signature, deviating from previously studied epidermal tissues. This topological distribution is established early during leaf development, already before the typical pavement cell shapes emerge, with topological homeostasis maintained throughout growth and unaltered between division and maturation zones. Simulating graph models, we identify a heuristic cellular division rule that reproduces the observed topology. Water relations of tomato fruit and the epidermal and pericarp activities of the putative cell wall loosening and tightening enzymes Xyloglucan endotransglycosylase (XET) and peroxidase were investigated, to determine whether tomato fruit growth is principally regulated in the epidermis or pericarp. Analysis of the fruit water relations and observation of the pattern of expansion of tomato fruit slices in vitro, has shown that the pericarp exerts tissue pressure on the epidermis in tomato fruit, suggesting that the rate of growth of tomato fruit is determined by the physical properties of the epidermal cell walls. The epidermal activities of XET and peroxidase were assayed throughout fruit development. Temporal changes in these enzyme activities were found to correspond well with putative cell wall loosening and stiffening during fruit development. The completion of germination of seeds of Arabidopsis thaliana is marked by the appearance of the radicle through the surrounding endosperm and testa. Using confocal microscopy and green fluorescent protein (GFP)-transformed embryos to highlight the epidermal cell walls it has been possible to conduct time-lapse photography of individual embryos during their germination.
