The widespread agricultural problem of pre-harvest sprouting (PHS) could potentially be overcome by improving seed dormancy. Here, we report that miR156, an important grain yield regulator, also controls seed dormancy in rice. We found that mutations in one MIR156 subfamily enhance seed dormancy and suppress PHS with negligible effects on shoot architecture and grain size, whereas mutations in another MIR156 subfamily modify shoot architecture and increase grain size but have minimal effects on seed dormancy. Mechanistically, mir156 mutations enhance seed dormancy by suppressing the gibberellin (GA) pathway through de-represssion of the miR156 target gene Ideal Plant Architecture 1 (IPA1), which directly regulates multiple genes in the GA pathway. These results provide an effective method to suppress PHS without compromising productivity, and will facilitate breeding elite crop varieties with ideal plant architectures. Pre-harvest sprouting reduces the yield of agriculturally important crops such as

DELLA repression of Arabidopsis thaliana seed germination can be lifted either through DELLA proteolysis by the ubiquitin-proteasome pathway or through proteolysis-independent GA hormone signaling. GA-binding to the GID1 (GIBBERELLIN-INSENSITIVE DWARF1) GA receptors stimulates GID1-GA-DELLA complex formation which in turn triggers DELLA protein ubiquitination and proteolysis via the SCFSLY1 E3 ubiqutin ligase and 26S proteasome. Although DELLA cannot be destroyed in the sly1-2 F-box mutant, long dry after-ripening and GID1 overexpression can relieve the strong sly1-2 seed dormancy phenotype. It appears that sly1-2 seed dormancy results from ABA signaling downstream of DELLA since dormant sly1-2 seeds accumulate high levels of ABA hormone and loss of ABA sensitivity rescues sly1-2 seed germination. DELLA positively regulates the expression of XERICO, an inducer of ABA biosynthesis. GID1b overexpression rescues sly1-2 germination through proteolysis-independent DELLA down-regulation associated ...
Abstract, The Arabidopsis Book, 2008 Seed dormancy allows seeds to overcome periods that are unfavourable for seedling established and is therefore important for plant ecology and agriculture. Several processes are known to be involved in the induction of dormancy and in the switch from the dormant to the germinating state. The role of plant hormones, the different tissues and genes involved, including newly identified genes in dormancy and germination are described in this chapter, as well as the use transcriptome, proteome and metabolome analyses to study these mechanistically not well understood processes.
Early dormancy involves a number of phenomena: cessation of active growth, formation of terminal buds, formation of abscission layers in leaves, development of cold resistance, development of winter rest (a chilling requirement), and leaf fall. The buds of some tree species (or some races of a species) have a true rest or winter chilling requirement; usually the roots do not; perhaps the cambium does not. There is a wide range of genetic variation within and between species in response to photoperiod, winter chilling, water nutrients, and other environmental factors which affect the dormancy condition of plants.. During the period of winter rest there are many metabolic and developmental processes going on in the buds and twigs. These processes include respiration, photosynthesis, some cell division, enzyme synthesis, production of growth stimulators, and dissipation of growth inhibitors.. Dormancy of trees can be divided arbitrarily into three phases: early rest, winter rest, and after-rest. ...
Fall dormancy is a determinant factor of alfalfa yield in northern climate but selection for that trait is often done to the detriment of freezing tolerance. Selection for low dormancy in the field is a tedious process that depends on unpredictable fall conditions allowing the identification of less dormant genotypes. Here we describe two screening methods entirely performed indoor for the selection for low dormancy in alfalfa without negative impact of freezing tolerance. One method is for semi-dormant cultivars and includes successive freezing stress to simultaneously improve freezing tolerance and the other is for winterhardy dormant cultivars. In both approaches, alfalfa is exposed to a critical photoperiod during four weeks that allows to discriminate between dormant and less-dormant genotypes. These approaches can be used to generate recurrently selected populations that are progressively less dormant.. ...

Changes in seed dormancy were selected during the domestication of crop plants from wild-plant species, because some features of dormancy that provide ecological advantages presented agronomic disadvantages within a farmed system. Seed dormancy varies widely among seed batches, even for seeds of the same genotype, indicating large environmental influences. For genetic studies, it is essential to obtain such quantitative data on seed material without the effects of variation in environmental conditions, including the maternal growing and seed storage conditions. The genetics of seed dormancy is as predicted on Mathers theory of genetical architecture for a character that has been subject to strong directional selection. Many studies on the genetics of seed dormancy regulation have concluded that dormancy is a quantitative trait controlled by multiple loci that affect different seed tissues, and often different aspects of seed physiology. Therefore, in this study discos on some research on seed dormancy

Background and Aims: Several ecologically important plant families in Mediterranean biomes have seeds with morphophysiological dormancy (MPD) but have been poorly studied. The aim of this study was to understand the seed ecology of these species by focusing on the prominent, yet intractably dormant Australian genus Hibbertia. It was hypothesized that the slow germination in species of this genus is caused by a requirement for embryo growth inside the seed before germination, and that initiation of embryo growth is reliant upon a complex sequence of environmental cues including seasonal fluctuations in temperature and moisture, and an interplay with light and smoke. Using the results, the classification of the MPD level in species of Hibbertia is considered. Methods: Four species of Hibbertia in winter rainfall south-western Australia were selected. These species, whilst differing in geographic distributions, are variously sympatric, and all are important understorey components of plant ...

Like most plants that are native to temperate and boreal regions, Paeonia lactiflora blooms in late spring; it is a perennial that requires a long period of chilling. In order to study the biochemical and physiological changes of herbaceous peony in response to low temperature, six indices related to cold tolerance were determined over 20 different periods of natural conditions, and morphological and phytophenological observations were made. A larger amount of soluble sugars and reducing sugars and a smaller amount of starch were found during dormancy in comparison with the growing season. The concentration of soluble and reducing sugars increased in buds when P. lactiflora plants were subjected to low temperature from 14 November to 4 January. Starch content decreased significantly over the same period. Maximum soluble sugar content peaked later than reducing sugar, on 14 January and 24 December, respectively. Sugars appeared to play an important role in bud development and dormancy. Soluble ...

Pre-harvest sprouting (PHS) is one of the most serious quality defects of grains that impacts on the major crops in the world. Sprouting damage reduces the value of the crops throughout the entire supply chain - from growers to seed and grain merchants; millers and maltsters to bakers; and ultimately, the consumers. It is estimated the PHS produces costs to farmers about $1 billion per year. The foreseen climate irregularity with untimely rainfall has added new challenges to the industry about how to manage PHS damage. In modern commercial crops, loss or reduced seed dormancy and quick germination are the two key aspects directly related to PHS. Last September in Perth (Australia), during the 13th International Symposium on PHS in Cereals (ISPHSC), the consensus was reached that significant progress has been made to understand the mechanisms of seed dormancy and germination, which can now be applied to developing technologies to manage PHS. This Research Topic aims to highlight the current understanding
Pre-harvest sprouting frequently occurs in Triticum aestivum (wheat) and Hordeum vulgare (barley) at the end of the maturity period due to high rainfall, particularly in Asian monsoon areas. Seed dormancy is a major mechanism preventing pre-harvest sprouting in these crops. We identified orthologous sequences of the major Hordeum vulgare (barley) seed dormancy gene Qsd1 in hexaploid wheat cv. Chinese Spring by performing genomic clone sequencing, followed by transcript sequencing. We detected 13 non-synonymous amino acid substitutions among the three sub-genomes of wheat and found that the Qsd1 sequence in the B sub-genome is most similar to that in barley. The Qsd1 sequence in A genome diploid wheat is highly similar to that in the hexaploid A sub-genome. Wheat orthologs of Qsd1 showed closer similarities to barley Qsd1 than did those of other accessions in the DNA database. Like barley Qsd1, all three wheat Qsd1s showed embryo-specific gene expression patterns, indicating that barley and wheat Qsd1

OBJECTIVE 1: Wheat mutants, hypersensitive to abscisic acid (ABA) were characterized using ABA dose-response germination experiments, stomatal closure assays, and carbon isotope discrimination. One mutant showed a reproducible stomatal closure in response to ABA application resulting in measured increase in transpiration efficiency, indicating that it may be more drought tolerant. Drought tolerance experiments were established in the field in 2008. The role of the plant hormone, Giberellin in seed germination and plant height was investigated. The DELLA protein, RGA was shown to be controlled by protein-protein interaction with GA receptor GID1 in Arabidopsis. GID1 can deactivate germination-specific DELLA protein RGL2. The effect of GID1 on seed dormancy is similar to the effect of after-ripening suggesting the processes may share underlying mechanisms. OBJECTIVE 2: 105,797 molecular marker datapoints have been provided to wheat and barley researchers as follows: 6,659 (CA), 32,546 (ID), 9,642 ...

Dormancy is typically broken in the laboratory with a stratification period of 3 days this is known as the 3 day pre chill, and the use of a growth promoter such as potassium nitrate KNO3. However this year in many cases this regime is not enough to completely break the dormancy and we are retesting with a 7 day Prechill period and potassium nitrate. This is naturally causing delays with germination, as we are often retesting as many as three times to obtain the best possible result.. We know that you are anxious for results because we understand that good seed is in short supply, but unfortunately this season is shaping up to be a challenge for us all. We expect this phenomenon to persist at least until we get some very cold weather.. Germination tests are taking longer.. If you are experiencing a delay, please go on-line to see the status of your germination test. We are taking care to provide good notes and revised ETAs for your information.. Dr. Susan E. Meyer. Germination and seed testing. Rose seeds are normally dormant at maturity and require some form of pretreatment in order to germinate. Release from dormancy is a complex process that may involve changes at the pericarp, testa, and embryo levels. The degree of dormancy and the principal level of dormancy control varies among species, cultivars, seedlots, and even among hips within a single bush. Because the achenes have a thick, hard pericarp and do not swell when placed in water, it is often assumed that they are water-impermeable. Work by Svejda (1972) and others has shown that this is not the case. The achenes do take up water, although the mechanical restriction presented by the pericarp can sometimes prevent full imbibition. Tincker and Wisley (1935) showed, for 10 rose species, that cracking the pericarp alone did not remove dormancy. The importance of including treatments that weaken the pericarp in efforts to remove rose seed dormancy depends on the species and the ...
Blaauw Jansen G., 1981: Differences in the nature of thermo dormancy and far red dormancy in lettuce lactuca sativa cultivar noran seeds
INTRODUCTION In a broad sense, seed dormancy can be interpreted as an undesirable characteristic for agricultural cropping since rapid germination and growth are required. However, some degree of dormancy could be advantageous, at least during seed development and storage. All seed surfaces are basically composed of lignocellulosics like-structures, which present a fast degradation by organisms which are capable of breaking them down into digestive units. Additionally, the hydroxyl groups, mainly present in the cellulose chain, are polar groups with high reactivity, suggesting a high affinity towards water (Kiguchi, 1996). Water uptake by itself provokes dormancy break and also accelerated growth (Goodwin, 1966), what can be a problem for long-term storage. Moisture migration within grain containers can lead to detrimental changes in grain appearance and quality and leads to undesirable germination. Storage procedures are normally regulated by monitoring temperature and air moisture (Abdalla & ...
Seeds of the Salsoloideae contain fully differentiated embryos. On imbibition, the embryo cells elongate and the spiral embryo uncoils and ruptures the thin seed coat. This can occur in as little as 10 min. Nearly all of the families showing very fast germination have small to very small seeds and little or no endosperm. Most species have soft, thin seed coats that imbibe water readily. All are from high-stress habitats, either arid or saline or from active floodplains, where they can rapidly exploit temporarily favourable conditions for germination. They exhibit one of two contrasting germination behaviours, either having seeds which all germinate within a very short time of wetting or having seed persistence whereby small amounts of rain cause germination of small fractions of seed from a long-living soil seed bank. Serious confusion in the literature in the use of the term opportunistic is pointed out and clarified ...
N the fall of 1940 several instances of delayed germination were I noted in freshly harvested samples of Vicland oats, a new variety recently described by Stanton (IO) .3 Certain samples when tested at room temperature germinated only 65% in two weeks. However, germination was increased to 95% or better when the seed was prechilled. As this variety was about to be released for commercial prod ~ c t i o n , ~ it seemed important to determine the extent and nature of the delayed germination. The immediate need for such a study was occasioned by the necessity of establishing a satisfactory testing procedure for making germination tests in order to judge freshly harvested seed as to requirements for certification.
Dormancy is a key feature of perennial plants. During dormancy the meristem becomes insensitive to growth-promoting signals for a period of time, before it is released and growth resumes [1, 2]. Bud dormancy is a critical developmental process that allows perennial plants to survive extreme seasonal variations in climate. The regulation of dormancy is a complex process that is necessary for plant survival, development, and architecture [3, 4]. A thorough understanding of regulation mechanisms controlling dormancy in woody perennials would have a variety of applications for genetic improvement of woody trees [3, 5, 6]. Considerable progress has been made in understanding the molecular mechanisms and regulatory pathways involved in bud dormancy [2]. However, until recently such studies focused on regulation at the levels of transcription, post-transcription, and translation [1, 7-12]. Despite the importance of dormancy regulation for perennial behavior [3], the roles of post-translational ...
The successful treatment of breast cancer is limited due to a fraction of tumor cells escaping drug-treatment by entering a dormant state, only to relapse years or decades later at distant sites. Host-driven chronic inflammatory cells such as M2 macrophages play an important role in tumorigenesis, but the role of tumor-intrinsic inflammatory signaling involved in tumor dormancy and recurrence is unknown. We sought to determine the role of tumor-intrinsic inflammatory pathways in mouse mammary carcinoma cells (MMC) treated with Adriamycin (ADR), a clinically relevant chemotherapeutic drug. We found that ADR-induced dormant tumor cells autonomously produced pro-inflammatory cytokines, in vitro. MMC treated with Chloroquine (CQ) prior to ADR treatment displayed a delay in relapse, or prolonging of dormancy, when compared to ADR-treated MMC. Additional gene array data showed predicated activation of NF-κB p65 in ADR-treated dormant MMC that eventually relapsed. These data suggest that the anti-inflammatory

Metastases develop from disseminated tumor cells (DTCs) years or even decades after primary tumor treatment. This is thought to be due to the ability of residual DTCs to remain dormant, but the mechanisms controlling this state are poorly understood. We studied the mechanisms of tumor cell dormancy using the HEp3 HNSCC model. When injected in nude mice, HEp3-GFP cells form tumors with 100% efficiency and spontaneously disseminate to lungs (95% incidence) and bone marrow (BM) (30% incidence). Tracking of GFP-tagged DTCs showed that after ~2 weeks the quiescent DTCs in lungs start to grow to form overt metastasis in 95% of mice. In contrast, those in the BM remain at constant numbers suggesting a dormant phenotype. We published that prolonged in vitro culture (,40 generations) of tumorigenic HEp3 (T-HEp3) cells induced a reprogramming into a dormant phenotype (D-HEp3). This behavior is revealed upon reinoculation of D-HEp3 cells in mice or chick embryo CAMs. The loss of malignancy is in part due ...

Sprouts are the cheapest and most convenient source of complete nourishment. They are food for long-term health and wellness. If consumed judiciously and chosen over other foods, they can prevent diseases and lifestyle-related ailments. Sprouting a seed enables it to produce an enormously enhanced bundle of nutrients.A sprout is at the transitional stage between seed and plant. It is, to put it simply, a baby plant. These are essentially pre-digested foods as the seeds own enzymes do most of the work. The nutritional changes that occur during sprouting primarily happen because complex compounds get broken down into simpler forms. This, added to development of some essential nutrients, constituents, and breakdown of anti-nutrients, makes the whole phenomenon of sprouting possible. Metabolic activity in dormant seeds is initiated as soon as they are hydrated during soaking.Sprouted seeds acquire natural increased digestibility and nutrients as compared to unsprouted seeds. The preparation comprises an extract which is obtained from cells or tissue originating in an organism capable of entering a phase of dormancy in at least one of its parts and comprises at least one substance which induces or maintains the state of dormancy in the organism from which the cells or tissue are derived. The DC may be used for a variety of indications including human medicine and cosmetics, plant growth control and food preservation.
This Grand Challenge specifically relates to the study of dormant cells that exist after seemingly successful treatment (i.e. following a latent period where the patient displays no clinical symptoms). This is distinct from the investigation of indolent cells that may persist for an extended period of time before progressing to malignancy, or cancers (such as chronic myeloid leukaemia) that remain dormant whilst on treatment but recur when therapy is withheld.. In addressing this challenge, teams will need to first overcome the current barrier of how to identify, detect and study dormant cells. It is anticipated that this challenge will require consideration of both tumour intrinsic and extrinsic factors in order to understand how dormant cells and/or their behaviour can be effectively targeted. Importantly, findings from relevant experimental models will ultimately also need to be validated in patients.. Examples of the types of questions that could be addressed in this challenge include (but ...
