DVC is developing the recombinant rF1V plague vaccine on behalf of the U.S. Department of Defense (DoD). The vaccine will be used for pre-exposure prophylaxis to combat infection with Yersinia pestis, the causative agent of plague. DVC is a contractor for the Medical Countermeasure Systems Joint Vaccine Acquisition Program (MCS-JVAP) and the sponsor of the associated Investigational New Drug Application for the plague vaccine.. We are excited the FDA has granted Orphan Drug status for rF1V vaccine, said Gary S. Nabors, Ph.D., President of DVC. Orphan Drug Designation provides important incentives to support the development of products for rare diseases. These incentives include the waiving of prescription drug user fees and the ability to market the vaccine exclusively in the U.S. for seven years following approval.. The plague vaccine is being developed as an important component of the DoDs medical countermeasures portfolio against bioterrorism agents. Drugs and biologics developed for ...
Plague, one of the most devastating infectious diseases in human history, is caused by the bacterial species Yersinia pestis. A live attenuated Y. pestis strain (EV76) has been widely used as a plague vaccine in various countries around the world. Here we compared the whole genome sequence of an EV76 strain used in China (EV76-CN) with the genomes of Y. pestis wild isolates to identify genetic variations specific to the EV76 lineage. We identified 6 SNPs and 6 Indels (insertions and deletions) differentiating EV76-CN from its counterparts. Then, we screened these polymorphic sites in 28 other strains of EV76 lineage that were stored in different countries. Based on the profiles of SNPs and Indels, we reconstructed the parsimonious dissemination history of EV76 lineage. This analysis revealed that there have been at least three independent imports of EV76 strains into China. Additionally, we observed that the pyrE gene is a mutation hotspot in EV76 lineages. The fine comparison results based on ...
An effective intranasal (i.n.) vaccine against pneumonic plague was developed. The formulation employed two synthetic lipid A mimetics as adjuvant combined with Yersinia pestis-derived V- and F1-protective antigens. The two nontoxic lipid A mimetics, classed as amino-alkyl glucosaminide 4-phosphates …
Background: F1-V from Yersinia pestis has been shown to be a protective antigen against disease. We modified the F1-V antigen with α-galactose residues and immunized α galactosyl transferase (α-GT) KO mice in order to mimic the same defect in humans that results in circulating antibodies against α-gal epitopes enhancing immune responsiveness. Aim: To demonstrate that α-gal modified F1-V induces a more robust immune response than unmodified F1-V. Methods: α-GT KO mice were injected subcutaneously with different doses of α-gal-F1-V or F1-V. All mice were boosted with F1-V at 36 dpi and necropsied on day 40 in order to evaluate in vitro recall response of lymphocytes and to measure anti-F1-V antibody titers. Results: Mean IgG titer values were 50,000 and 10,000 for mice immunized with the α-gal-F1-V or F1-V, respectively. The relative avidity of the antibody response was also higher for the α-gal-F1-V immunized mice. Moreover, lymphocytes from α-gal-F1-V immunized mice demonstrated ...
An emerging fungal pathogen? New research provides even more evidence that a previously undescribed, cold-loving fungus is associated with white-nose syndrome, a condition linked to the deaths of up to 1,000,000 cave-hibernating bats in the northeastern and mid-Atlantic states. Since the winter of 2006-2007, bat populations plummeted from 80 to 97 percent at surveyed bat-hibernation caves, called hibernacula. USGS microbiologist Dr. David Blehert and his colleagues identified the fungus last year, and have followed up by trying to determine if the fungus may be responsible for the deaths or if it is simply a side effect of another underlying disease. The researchers found that 90 percent of all bats they examined from suspected WNS sites had a severe fungal skin infection that did not just occur on the skin, but below it as well. The growth temperature requirements of the fungus are consistent with the core temperatures of cave-hibernating bat species throughout temperate regions of the world. ...

Staff will go door to door to neighbors near the case to inform them about plague found in the area and educate them on reducing their risk. Health care providers and others close to the patient who have been determined to have been exposed are taking preventive antibiotic therapy. Antibiotics are currently used to treat plague. The National Institute of Allergy and Infectious Diseases is also working with the U.S. Department of Defense, the Centers for Disease Control and Prevention (CDC), and the U.S. Department of Energy to develop a vaccine that protects against inhalationally acquired pneumonic plague. According to the CDC, while plague vaccines are in development, they are not expected to be commercially available in the immediate future.. It is important to note that a rise in plague cases at this time of year is not unusual. According to the CDC, while plague cases can occur in any season of ...
in this report because of uncertainties about the efficacy of the vaccine (see Alternatives to Vaccination and Supplemental Protection). If vaccinees are believed to have been exposed to plague or if their risk for exposure is exceptionally high, prophylactic antibiotic use should be considered as a supplement to vaccination. Vaccination is not intended as a method of controlling plague epidemics because several months are required to complete the primary vaccination series and to develop adequate levels of protective antibodies. Routine vaccination is not necessary for persons living in areas in which plague is enzootic (e.g., the western United States). Vaccination is not indicated for hospital staff or other medical personnel in such areas, nor for most travelers to countries in which cases of plague have been reported, ** particularly if their itineraries are limited to urban areas and modern hotel accommodations. Supply and Storage Plague vaccine is manufactured by Greer Laboratories, Inc., ...
Currently, plague is managed in prairie dogs through manual application of insecticides to burrows to kill the fleas that transmit Y. pestis. However, this process is labor intensive, and recent evidence suggests that fleas can develop resistance to the most frequently used pesticide. The USGS National Wildlife Health Center (NWHC), in conjunction with others, has developed and tested a sylvatic plague vaccine (SPV), deliverable to prairie dogs via palatable bait that offers an additional approach for plague management. From 2013-15, NWHC scientists conducted a large, collaborative field study to test the effectiveness of SPV in reducing mortality from plague in four species of prairie dogs in 7 western states. This study involved state, federal, tribal and non-government agencies organized under the Black-footed Ferret Recovery Implementation Team (BFFRIT), a multi-agency effort led by the US Fish and Wildlife Service. The study found that vaccine treatment increased prairie dog abundance and ...
880-01Yersinia pestis and Francisella tularensis, the causative agents of plague and tularemia, have been labeled as potential biological weapons. In the event of an intentional aerosol release of either pathogen, the only FDA-approved treatment is rigorous antibiotic therapy. The focus of this research was to determine if use of known immune stimulants could provide rapid and sustained vaccine protection against pneumonic infections of Y. pestis or F. tularensis.;880-02The AGPs also functioned effectively as adjuvant in a plague vaccine. Our two-dose intranasal (i.n.) vaccine with Y. pestis -derived Caf1 and V antigens provided mice with 100% protection just 21 days after the priming dose. A single i.n. vaccine protected more than 75% of vaccinates 45-180 days post-vaccine. The protection levels seen from our AGP-based vaccine correlated well with total sera IgG titers and promoted a mixed Th1/Th2 response. Our intranasal vaccine protected 90% of Sprague-Dawley rats challenged with ...

A Study from Thucydides to Justinian by Christine A. Smith Throughout history, humans have been faced with disastrous catastrophes which must be endured in order to survive. One of the most incomprehensible disasters for humanity has been the plague. This term in Greek can refer to any kind of sickness; in Latin, the terms are plaga and pestis. In antiquity, two of the most devastating plagues were the Athenian plague of 430 B.C. and the Justinianic plague of 542 A.D. This paper will discuss these plagues, the manner in which they spread, and their consequences for the survivors. Also, the ways in which ancient writers wrote about these disasters will be discussed, with special reference to the role of the gods. Much of what is conventionally believed about these plagues comes from comparisons with the Black Death, a visitation of bubonic plague during the fourteenth century A.D. Although the sources for the Athenian and Justinianic plagues are insufficient, there is some question as to the ...

The epidemiological use of the term plague is currently applied to bacterial infections that cause buboes, although historically the medical use of the term plague has been applied to pandemic infections in general. Plague is often synonymous with bubonic plague, but this describes just one of its manifestations. Other names have been used to describe this disease, such as The Black Plague and The Black Death; the latter is now used primarily by scholars to describe the second, and most devastating, pandemic of the disease ...
In Plague Inc., playing Parasite on Normal is a little tough at first. Its a slow spreading plague, but it has an Ability that is just amazing and makes it pretty easy to beat, just time consuming.. The key to the beating the Parasite plague is its special ability in the Abilities section called Symbiosis.. With three levels of Symbiosis purchased, youll be able to have any and all Tier 1 and all but one Tier 2 symptoms without your plague being detected. You wont be purchasing much until the end though, youll let a bunch of symptoms mutate for free, and then purchase the deadly symptoms at the end to save some points.. ...

Cultures of Plague opens a new chapter in the history of medicine. Neither the plague nor the ideas it stimulated were static, fixed in a timeless Galenic vacuum over five centuries, as historians and scientists commonly assume. As plague evolved in its pathology, modes of transmission, and the social characteristics of its victims, so too did medical thinking about plague develop.

9:13-21 The sixth angel sounded, and here the power of the Turks seems the subject. Their time is limited. They not only slew in war, but brought a poisonous and ruinous religion. The antichristian generation repented not under these dreadful judgments. From this sixth trumpet learn that God can make one enemy of the church a scourge and a plague to another. The idolatry in the remains of the eastern church and elsewhere, and the sins of professed Christians, render this prophecy and its fulfilment more wonderful. And the attentive reader of Scripture and history, may find his faith and hope strengthened by events, which in other respects fill his heart with anguish and his eyes with tears, while he sees that men who escape these plagues, repent not of their evil works, but go on with idolatries, wickedness, and cruelty, till wrath comes upon them to the utmost. ...

Topping our list of most deadly plagues is one of the most infectious and hardest to treat diseases of all time, and a bane to health workers world wide, Tuberculosis. Having in previous generations been called consumption, TB is very much a sleeping giant in terms of disease. Despite only showing around 10 million cases per year as active, and about 2 million ending up as terminal, it is estimated that nearly a third of the earths population have some latent infection from the Mycobacterium tubercolosis bacteria. Part of the reason TB is very infectious is the nature of the symptoms themselves. Besides sweating, chills, weight loss and fatigue, sneezing and an exceptional amount of coughing may occur, often enough to cause bleeding or scarring in the lungs. Through this coughing, the bacteria can be spread to other healthy individuals who may or may not show positive symptoms there after. If the individual happens to be malnourished or infected with an immune suppressing disease such as HIV, ...

The plague continues to spread among the Markab population as Dr. Franklin confronts Lazarenn and demands to know why he wanted to keep the plague a secret. Lazarenn explains that the plague is not something normally talked about -- it is considered a disease that only occurs to an immoral people, because the first documented case struck an immoral population isolated on the Markab homeworld, killing the entire populace. They assumed that the plague had been wiped out, and therefore was no longer a threat. Lazarenn explains that Drafa, the disease, had been used to frighten children into behaving, and was still believed to only inflict the immoral. Though the first new case had been discovered nearly a year ago, the Markab were still scared to admit that the problem had resurfaced, and since none of them wanted to be considered immoral, it was kept secret, which made the disease spread even faster. In an attempt to escape the disease, the Markab have fled to every Markab colony, including ...

A recombinant strain of attenuated Salmonella enterica serovar Typhi surface-expressing Yersinia pestis F I antigen was generated by transforming strain BRD1116 (aroA aroC htrA) with plasmid pAH34L encoding the Y. pestis caf operon. BRD1116/pAH34L was stable in vitro and in vivo. An immunisation regimen of two intranasal doses of I x 10(8) cfu of BRD1116/pAH34L given intranasally to mice 7 days apart induced the strongest immune response compared to other regimens and protected 13 out of 20 mice from lethal challenge with Y pestis. Intranasal immunisation of mice constitutes a model for oral immunisation with Salmonella vaccines in humans. Thus, the results demonstrate that attenuated strains of S. enterica serovar Typhi which express Y pestis F1 antigen may be developed to provide an oral vaccine against plague suitable for use in humans. Other manifestations include septicaemic plague without bubo, pneumonic plague (primary or secondary to bacteraemia), meningitis, and pharyngitis. Following an incubation period of two to five days, plague has a sudden onset of fever, chills, headache, malaise, myalgia and nausea. In bubonic plague, bacteria move from the site of inoculation to regional lymph nodes that enlarge and become painful swellings known as bubos. The bacteria can spread via the bloodstream resulting in septicaemia that if untreated, can lead to a rapid deterioration and death. In pneumonic plague the bacteria either infiltrate the lungs causing primary pneumonia, or secondary pneumonia occurs after bacteraemic spread in bubonic plague. This causes severe shortness of breath and cough with blood stained sputum, leading to respiratory failure. Pneumonic plague is rapidly fatal if untreated. Treatment ...

The Hebrew words are so used as to give the name plagues to all the wonders God did against Pharaoh. Thus, it appears that the language in the account in Exodus puts forward the wondrous character of these dealings of Yahweh with Pharaoh. The account of the plagues is found in Exodus 7:8-12:31 Psalm 78:42-51; Psalm 105:27-36. These poetical accounts of the plagues have a devotional purpose and do not give a full historical narrative. Psalm 78 omits plagues 4, 6, 9; Psalm 105 omits plagues 5 and 6. Both psalms change the order of the plagues. Account of the preparation which led up to the plagues is found in the narrative of the burning bush (see BUSH, BURNING), the meeting of Aaron with Moses, the gathering together of the elders of Israel for instruction and the preliminary wonders before Pharaoh (Exodus 3; Exodus 4). This preparation contemplated two things important to be kept in view in considering the plague, namely, that the consummation of plagues was contemplated from the beginning ...

A research team from Northern Arizona University has isolated the probable cause of the plague that terrorized tens of millions 1,500 years ago.. NPR reported on Wednesday that, using DNA extracted from dental pulp, they were able to reassemble the genetic code of the bacteria behind the Justinian plague, named after the Byzantine emperor who saw it overwhelm his subjects - and nearly himself - around 541 A.D.. Some of the estimates are that up to 50 million people died, evolutionary biologist David Wagner told NPR.. Though the pandemic predated the Black Plague by 800 years, the team wrote in the journal The Lancet Infectious Diseases that the two plagues not only emerged from China, but both spread from rodents to humans.. We conclude that the Y pestis lineages that caused the Plague of Justinian and the Black Death 800 years later were independent emergences from rodents into human beings, they wrote. These results show that rodent species worldwide represent important reservoirs for the ...
The Plague is far older than previously known and later changed to become much more virulent-so virulent that it may have contributed to the decline of Classical Greece and the Roman and Byzantine empires and later killed off 30 to 50 percent of Europes population, a new study says. The bacteria that causes the Plague, Yersinia pestis , diverged from the less-pathogenic Y. pseudotuberculosis bacterium about 5,783 year ago. That divergence, and therefore the bacterias possibility of infecting humans, is much earlier than scholars previously estimated. While the Plague would go on to kill tens of millions of people in Europe and Asia, researchers have found DNA of Y. pestis in the teeth of a Russian person who died 5,000 years ago. They say that although the plague doesnt appear to have been as prevalent or as virulent in the Bronze Age, even then it may have triggered migrations of populations in Europe and Asia and caused population decreases. Writing the in the journal Cell, biologist Simon ...
