Endocrine side effects of the immunosuppressive drug cyclosporine (CyA) include changes in anterior pituitary hormone secretion. The aim of the present study was to examine the effects of CyA on the responsiveness of in situ and ectopic anterior pituitary prolactin (PRL), growth hormone (GH) and luteinizing hormone (LH) release response to dopamine (DA) and thyrotropin-releasing hormone (TRH) treatment in young female rats, and to evaluate the possible PRL participation in these effects. Thirty day old rats were rendered hyperprolactinemic by transplanting an anterior pituitary gland of a littermate donor, under the kidney capsule, and were then injected with CyA or vehicle for 2 or 8 days. Sham-operated rats were used as controls and treated in the same way. CyA treatment prevented the increase in plasma PRL levels which occurred in controls after pituitary grafting. In vitro basal PRL release of in situ pituitaries from either sham-operated and/or pituitary-grafted animals was decreased by CyA ...

TY - JOUR. T1 - Recent advances in the control and function of the anterior pituitary. AU - Stonesifer, Larry D.. AU - Kohler, Peter O.. PY - 1981/10. Y1 - 1981/10. N2 - Major advances in our understanding of the synthesis and release of anterior pituitary hormones have been made over the past several years. Neurons of the hypothalamus have been found to serve as neuroendocrine transducers in that they have both electrical and secretory functions. Peptidergic neurons respond to appropriate stimuli with a release of hypothalamic factors into the hypophyscal-portal system. These factors or hormones ultimately control the endocrine function of anterior pituitary cells. Three hormones, Thyrotropin Releasing Hormone (TRII), Gonadotropin Releasing Hormone (GnRH or LHRH) and somatostatin have been identified, synthesized and tested for clinical applications. The clinical assessment of pituitary function has been greatly improved by new and improved radioimmunoassays. One of the recent clinical ...

Unexplained weight pick up has been connected to hormonal irregular characteristics. As ladies develop more seasoned, they will probably encounter this issue.. Every hormone in our bodies resembles a key which needs to turn a bolt to have the capacity to do its work. Different irritations can influence the receptors and that is the reason lose weight from midriff few hormones may get to be distinctly insufficient. Estrogen is a particularly essential hormone. When it is not adjusted with the related hormone progesterone, this awkwardness can bring about water maintenance, memory misfortune, and headache cerebral pains. A hormonal awkwardness may likewise bring about fat collection around the weight loss diet that really works and thighs.. At last, it can quicken the ...
The insulin-like growth factor 1 (IGF-1) receptor is a protein found on the surface of human cells. It is a transmembrane receptor that is activated by a hormone called insulin-like growth factor 1 (IGF-1) and by a related hormone called IGF-2. It belongs to the large class of tyrosine kinase receptors. This receptor mediates the effects of IGF-1, which is a polypeptide protein hormone similar in molecular structure to insulin. IGF-1 plays an important role in growth and continues to have anabolic effects in adults - meaning that it can induce hypertrophy of skeletal muscle and other target tissues. Mice lacking the IGF-1 receptor die late in development, and show a dramatic reduction in body mass, testifying to the strong growth-promoting effect of this receptor. Mice carrying only one functional copy of IGF-1R are normal, but exhibit a ~15% decrease in body mass ...
Prostacyclin molecule. Computer model showing the structure of the hormone prostacyclin, or prostaglandin I2 (PGI2). Atoms are colour-coded (carbon: dark grey, hydrogen: light grey, oxygen: red). Prostacyclin acts to widen blood vessels (thus lowering blood pressure) and to prevent aggregation of platelets (preventing blood clotting). These effects are the opposite of those of the related hormone thromboxane, and the balance between the two is important in regulating the functioning of the circulatory system. When taken as a drug it is known as epoprostenol. - Stock Image C003/3311

Although physiological and anatomical evidence had clearly indicated for many years that the secretion of anterior pituitary hormones is under control by the central nervous system, it is only recently that the isolation and determination of structure of three hypo- thalamic hypophysiotropic hormones have been accomplished. This has brought the concept of neurohormonal control of adenohypophyseal function into precise biochemical and chemical terms. The relative ease of synthesis of TRH (thyrotropin-releasing hormone), LH-RH (luteinizing hormone-releasing hormone), somatostatin and their analogues has opened a new era in the field of endocri- nology and has led to a rapid expansion of our knowl- ge of the control of anterior pituitary function. The rapid evolution of fundamental and clinical research on hypothalamic hormones and the many potential clinical applications indicate the importance of inte- grating the knowledge gained in recent years. This is well illustrated in the Proceedings of the
Research is lacking to define precisely the mechanism of this relationship. The effects of a particular hormone may be mediated directly by the hormone or indirectly through effects on other systems such as the HPA axis. Thus, sex hormones can exert their positive or negative feedback on gonadotropin-releasing hormone (GnRH) from the hypothalamus and luteinizing hormone (LH) and follicle-stimulating hormone (FSH) from the anterior pituitary depending on the levels of these hormones in the blood. The hypothalamic and anterior pituitary hormones affect immune responses directly (Whitacre, 1999). Thus, this bidirectional regulation of the immune response exists - there are sex hormone receptors on the surfaces of immune cells, and cytokine receptors (IL-1R, IL-18R) have been discovered on hormone-producing tissues. Additionally, proinflammatory cytokines (TNF-alpha and IL-1B) stimulate the release of glucocorticoids from the HPA axis, which regulates the inflammatory process (Bijlsma, 1999).. The ...
Seabirds often have high loads of contaminants. These contaminants have endocrine disrupting properties but their relationships with some endocrine mechanisms are still poorly investigated in free-living organisms. This is the case for the stress response which shifts energy investment away from reproduction and redirects it towards survival. In birds, this stress response is achieved through a release of corticosterone and is also accompanied by a decrease in circulating prolactin, an anterior pituitary hormone widely involved in regulating parental cares. We measured blood concentrations of some legacy persistent organic pollutants (POPs) andmercury (Hg) and examined their relationshipswith the corticosterone and prolactin responses of known-age (9-46 years old) incubating snow petrels (Pagodroma nivea) to a standardized capture/handling stress protocol. In this Antarctic seabird, we also investigated whether high contaminant burden correlates with a higher occurrence of egg neglect, a ...

Yeast two-hybrid testing and RT-PCR analyses. Interactions between A2 and different glycoprotein hormone subunits were assessed in the yeast two-hybrid system using pGBT9 GAL4-binding domain (BD) and pGADGH GAL4-activation domain (AD) vectors (12). Specific binding of different protein pairs was evaluated based on the activation of GAL1-HIS3 and GAL4-lacZ reporter genes (Clontech Laboratories Inc., Palo Alto, California, USA). The mature region of human A2 cDNA was fused to the GAL4-AD in a yeast shuttle vector, pGADGH. Similarly, cDNAs encoding other glycoprotein hormone subunits without the signal peptides were fused to the GAL-4-BD of the pGBT9 vector. Specific interactions of different protein pairs were evaluated based on the activation of the GAL1-HIS3 reporter gene in medium lacking leucine, tryptophan, and histidine but in the presence of 5 mM 3-aminotriazole (13). At least 10 different colonies expressing each pair of fusion proteins were tested.. For RT-PCR analysis of A2 and B5 mRNA ...

The growth hormone is probably the only anterior pituitary hormone which does not stimulate any other endocrine gland. In man, over secretion or hyper pituitarism of S.T.H, in child hood leads to gigantism. In the adults over secretion leads to elongation of long bones of limbs and lower jaw and a gorilla like appearance [Acromegaly]. Failure of G.H from an early age makes the child dwarf [ Dwarfism]. After ovulation, the pituitary hormones-FSH and LH released from the anterior pituitary cause the remaining parts of the dominant follicle to transform into the corpus luteum. It continues to grow for some time after ovulation and produces significant amounts of hormones, particularly progesterone,[3] and, to a lesser extent, estrogen. Progesterone plays a vital role in making the endometrium receptive to implantation of the blastocyst and supportive of the early pregnancy; it also has the side effect of raising the womans basal body temperature.[4][medical citation needed] The increased progesterone and BBT will cause at least a modest rise in BMR at the start of the luteal phase and remains in an elevated state until the phase ends. Research has discrepant findings on how much of an increase typically occurs and how much it can vary. Early, small sample studies have found various figures, such as; a 6% higher postovulatory sleep metabolism,[5] a 7% to 15% higher 24 hour expenditure following ...
