We have reported for the first time some characteristics of the membrane potential of SMCs of human pial arteries. The main finding of this study was that in these vessels rhythmic contractions were associated with generation of APs during spontaneous or K+-induced depolarization. The resting membrane potential of human pial arteries in vitro varied between −40 and −70 mV. The membrane potential of SMCs in rhythmically constricting arteries was significantly less negative than that of the silent ones. This finding indicates that spontaneous activity may be the result of a depolarized state of SMCs. The incidence of spontaneous activity was higher in those arteries that were obtained from patients ,40 years old. Therefore, we cannot exclude an influence of age-related vascular changes or of disease on the determined properties of human pial arteries. Periodic spontaneous activity has been demonstrated in human coronary arteries22 23 24 and was most frequent in vessels taken from older ...
The major findings in newborn pigs are: (1) treatment with the astrocyte toxin, L-2αAAA, or the HO inhibitor, CrMP, block pial arteriolar dilation to ADP, but not to isoproterenol, bradykinin, or sodium nitroprusside; (2) ADP increases brain CO production and this increase is blocked by the astrocyte toxin or inhibition of HO; and (3) ADP increases CO production by astrocytes and, to a lesser extent, cerebral microvessels. These data, coupled with previous results showing CO dilates pial arterioles in vivo, suggest CO is an astrocyte-derived mediator of ADP-induced pial arteriolar dilation in piglets.. ADP can produce endothelium-dependent cerebral vasodilation,19 which may be mediated in part by NO and endothelium-derived hyperpolarizing factor in adult rats.20,21,30 In endothelium-denuded control arteries from rat brain, ADP also produced dose-dependent relaxation, but this relaxation was lower than that found in intact control arteries.31 In adult rats, ADP-induced pial arteriolar dilation ...
Both the blood brain barrier and the blood CSF barrier tightly regulate the CNS fluid interstitial fluid environment of the CNS. You, however, are talking about a different barrier, the barrier formed by the pia mater, sometimes called the brain/CSF barrier. This barrier is less well studied. It does appear to prevent transport of particulate matter and red blood cells, but allow the passage of immune cells and small molecules. In addition to your professors statement that the composition of CSF and brain interstitial fluid is similar, intrathecal injection directly into the CSF in the subarachnoid space provides an effective way to deliver drugs with CNS action, suggesting the pia mater does not effectively restrict small molecules (see Goodman and Gilmans The Pharmacological Basis of Therapeutics Ch 2).. ...
TY - JOUR. T1 - Interaction between ATP-sensitive K+ channels and nitric oxide on pial arterioles in piglets. AU - Bari, F.. AU - Errico, Robert A.. AU - Louis, Thomas M.. AU - Busija, David W.. PY - 1996. Y1 - 1996. N2 - The interaction between ATP-sensitive K+ channels (K(ATP)) and nitric oxide (NO) was studied in pial arterioles of piglets. We examined the effects of N(ω)-nitro-L-arginine methyl ester (L-NAME), a general inhibitor of nitric oxide synthase (NOS), and 7-nitroindazole (7-NI), a selective inhibitor of neuronal NOS, on aprikalim-induced cerebral vasodilation. Topically applied, aprikalim, a selective activator of K(ATP), dilated arterioles by 11 ± 7% at 10-8 M and 17 ± 6% at 10-6 M. After L-NAME treatment (15 mg/kg, i.v.), the response was reduced (4 ± 4% and 12 ± 7%, respectively; n = 8, p ,0.05). Administration of 7-NI (50 mg/kg, i.p.) did not change pial arteriolar responsiveness to aprikalim. However, both L-NAME and 7-NI reduced the vasodilator responses to 10-4 M ...

There can be little doubt that the vessel of the histological study drained blood from the retina directly to the pia mater. Material was not available to trace the further course of the retinopial vein. Its size, although small compared with the central retinal vein, was large enough to have drained a substantial area of the temporal retina. There is some doubt as to whether the retina was free of pathology, even though none was reported, as deep cupping of a small area of the disc was present. However, the possibility that the retinopial vein represented a pathological development is unlikely. Where development of supplementary venous drainage from the eye at the disc has been reported, drainage is to the choroid via optociliary veins, not to the optic nerve, and is regarded as a response to obstruction of venous blood flow from the retina.3-5 Moreover, it is associated with a lengthy history of blindness.. A recently reported superior hemiretinal retinal vein that appeared ophthalmoscopically ...
The apparatus was transferred to an inverted microscope (Nikon TMS-F, 20× objective, Nikon, Melville, NY). Steady-state measurements of internal diameter at the midpoint of the segment were made using a high resolution CCD video camera (Hitachi KPC503, Hitachi, San Jose, CA) and a video caliper (Living Systems) calibrated using a stage micrometer. The vessel was superfused with PSS at a rate of 6 ml/min, and the chamber was maintained at 37°C. Chamber temperature and pH were monitored continuously using a probe (Oakton, series 35616, Singapore), and samples of the superfusing buffer were periodically drawn from the chamber for gas analysis (AVL Instruments, model 995, Graz, Austria). A plexiglas cover excluded ambient air from the chamber. The reservoir containing the perfusate and superfusate and the vessel chamber itself were bubbled with gas whose composition was adjusted, using separate tanks and regulators for each of oxygen, carbon dioxide, and nitrogen, to achieve Po2and Pco2values in ...

The glia limitans, or the glial limiting membrane, is a thin barrier of astrocyte foot processes associated with the parenchymal basal lamina surrounding the brain and spinal cord. It is the outermost layer of neural tissue, and among its responsibilities is the prevention of the over migration of neurons and neuroglia, the supporting cells of the nervous system, into the meninges. The glia limitans also plays an important role in regulating the movement of small molecules and cells into the brain parenchyma by working in concert with other components of the central nervous system (CNS) such as the blood-brain barrier (BBB). The end foot processes extending from both perivascular and marginal astrocytes form a close association with the basal lamina of the parenchyma, or the functional components of the brain, to create the glia limitans. This membrane lies deep to the pia mater and the subpial space and surrounds the perivascular (Virchow-Robin) spaces. Any substance entering the central ...

Intriguing questions have recently been raised regarding the applicability of direct observations of the pial microcirculation to the behavior of the total cerebral microcirculation. Operating under the assumption that arteriolar tone and, thus, cerebrovascular resistance is, to some extent, directly related to the intrinsic energy metabolism of the arteriolar wall, a comparative histochemical analysis of cerebral microvessels, both pial and parenchymal, was undertaken. Reactions were chosen on the bases of representation of substrate and of enzymes of glycolysis, the hexose monophosphate shunt, beta-oxidation of fat, Krebs cycle, cytochrome system and ATP hydrolysis. Three metabolically distinct segments of the cerebral microvasculature were delineated with the pial vessels showing strong capacities for glycolysis, beta-oxidation of fats and utilization of glucose through the hexose monophosphate shunt. Microvessels of the gray matter have a qualitatively similar metabolic profile but the ...

The neck is part of a long flexible column, known as the spinal column or backbone, which extends through most of the body. The cervical spine (neck region) consists of seven bones (C1-C7 vertebrae), which are separated from one another by intervertebral discs. These discs allow the spine to move freely and act as shock absorbers during activity.. Attached to the back of each vertebral body is an arch of bone that forms a continuous hollow longitudinal space, which runs the whole length of the back. This space, called the spinal canal, is the area through which the spinal cord and nerve bundles pass. The spinal cord is bathed in cerebrospinal fluid (CSF) and surrounded by three protective layers called the meninges (dura, arachnoid, and pia mater).. At each vertebral level, a pair of spinal nerves exit through small openings called foraminae (one to the left and one to the right). These nerves serve the muscles, skin and tissues of the body and thus provide sensation and movement to all parts of ...

The effects of systemic human recombinant interleukin 2 (rIL-2) infusion upon both the vasoconstrictor effect of hypocapnia and the endothelium-dependent vasodilator effect of acetylcholine (Ach) were examined in anesthetized rats equipped with cranial windows. Prior to the functional studies, each of six animals received an i.v. infusion of rIL-2 (6 × 105 IU/kg) every 8 h for 3 days. At the same time, six control animals received infusions of equivalent volumes of sterile water. Eight h after the final infusion, each animal was anesthetized and equipped with a cranial window for the observation of pial arterioles overlying the left frontoparietal cortex. Pial arteriolar diameters were measured before and after the topical application of Ach which in normal cerebral arterioles elicits the release of endothelium-dependent relaxing factor, causing vasodilation. When arteriolar diameters returned to base line, they were measured again both before and during hyperventilation-induced hypocapnia. ...
Series Editor: Arthur T. Skarin, MD, FACP, FCCP Systemic cancer can affect the central nervous system in several different ways, including direct tumor metastasis and indirect remote effects. Intracranial metastasis can involve the skull, dura, and leptomeninges (arachnoid and pia mater), as well as the brain parenchyma. Of these, parenchymal brain metastases are the most common and have been found in as many as 24% of cancer patients in autopsy studies. It has been reported that metastatic brain tumors outnumber primary brain tumors 10 to 1. To read the full article in PDF: Click here ...

Autopsy brain samples from 51 controls and 57 patients who died with encephalopathy of unknown origin were tested for HHV-6 & HHV-7 using immunohistochemistry, nested and real time PCR. Patients with encephalopathy had a greater frequency of HHV-6 & 7 (89.4%) compared to controls (68.6%).. Also, HHV-6 DNA was found at high levels in both the temporal and frontal lobes, and it was found significantly more frequently in the frontal lobe in these patients than in the temporal lobe.. Patients with encephalopathy also had a significantly higher viral load in the olfactory tract as well as the pia mater, the loose vascular coat that encloses the surface of the cerebral cortex. The authors suggest that the viral load may be high in these locations because these tissues may be used as a pathway to enter the CNS.. There was no difference in HHV-6B viral load in the hippocampus between the patients and controls. Only HHV-6B DNA was found; no HHV-6A DNA could be detected.. Read the full paper.. ...

The human brain is a soft, spongy collection of tissues and nerve cells. The main thing that protects the human brain is the bone of the skull encasing it. Then, there are three layers of membrane, called meninges that also aid in covering and protecting the brain. The three layers are the dura mater, arachnoid mater, and pia mater. The dura mater is closest to the bone, the arachnoid is in the middle and loosely surrounds the brain, and the pia mater is closest to the brain and attached to the spinal cord surface. Finally, the cerebrospinal fluid, a clear body fluid, also cushions the brain. ...
The outermost layer of the meninges and therefore closet to the skull is the Dura mater; it is a thick and tough layer composed of dense fibrous tissue which wraps almost entirely around the whole brain and spinal cord. There is little space between the Dura mater layer and the layer below/the bone above it, the exception to this is when (not in the meninges of the spinal cord) the Dura mater splits into two layers forming a gap between them, which is known as the venous sinus. The Dura contains larger blood vessels which split into the capillaries in the pia mater.. In the cranium, the Dura mater also acts as another layer of periosteum, however in the spine there is an epidural space (filled with fat) which separates the Dura mater and periosteum.. ...
The goal of this study was to determine whether responses of cerebral vessels to intravascular administration of serotonin are altered in stroke-prone spontaneously hypertensive rats. We measured pressure in pial arterioles and cerebral blood flow in normotensive and hypertensive rats during intra-atrial infusion of serotonin. In normotensive rats, pial arteriolar pressure was 48 +/- 3 mm Hg (mean +/- SEM) and cerebral blood flow was 48 +/- 5 ml/min/100 g under control conditions. Intra-atrial infusion of serotonin (5 and 50 micrograms/kg/min for 5 minutes) produced only minimal changes in pial arteriolar pressure (-3 +/- 4 and -4 +/- 4 mm Hg, respectively) and did not alter cerebral blood flow. In hypertensive rats, pial arteriolar pressure was 95 +/- 9 mm Hg and cerebral blood flow was 57 +/- 4 ml/min/100 g under control conditions. In contrast to normotensive rats, intra-atrial infusion of serotonin (5 and 50 micrograms/kg/min for 5 minutes) in hypertensive rats profoundly decreased pial ...

BM is usually blood-borne, and most often associated with a systemic infection. From the clinical standpoint, it is important to confirm or exclude sepsis since it substantially compromises the patients prognosis. Initially bacteria accumulate in the lumen of choroid plexus capillaries located in the cerebral ventricles and in the small and medium-sized vessels of the subarachnoid space, where they induce an inflammatory response. Developing vasculitis leads to the inflammation of the arachnoid, dura mater and pia mater.. As a result of disruption to the blood-brain barrier, bacteria and inflammatory mediators (acute-phase proteins, leukotrienes, prostaglandins, among others) penetrate to the cerebrospinal fluid (CSF), thus exacerbating inflammation. In the acute phase cerebral vessels become dilated with increasing blood flow due to the influence of bacterial toxins. In later phases blood flow is reduced, which is associated with impaired vascular autoregulation(4). The cytotoxic activity of ...
For this reason, I use a lot of CranioSacral Therapy and Myofascial Release in my work. CranioSacral Therapy is a gentle and non-invasive modality. The CranioSacral system extends from the occiput (in the area of the poll), down the spine to the coccyx (tailbone) and is comprised of three layers of membranes. The outermost layer is the dura mater which is a tough, waterproof membrane that houses the brain and spinal cord. The next layer is the arachnoid layer, and the inner layer is the pia mater which adheres snugly to the inside of the spinal cord. These membranes are constantly bathed in fluid. This fluid is known as the cerebrospinal fluid. It pulses throughout the life of the horse (also all other animals as well as humans) and influences the movement of the skull bones and the connective tissue (fascia). ...
Subarachnoid hemorrhage (SAH) is bleeding into the subarachnoid space-the area between the arachnoid membrane and the pia mater surrounding the brain. Symptoms may include a severe headache of rapid onset, vomiting, decreased level of consciousness, fever, and sometimes seizures. Neck stiffness or neck pain are also relatively common. In about a quarter of people a small bleed with resolving symptoms occurs within a month of a larger bleed. SAH may occur as a result of a head injury or spontaneously, usually from a ruptured cerebral aneurysm. Risk factors for spontaneous cases included high blood pressure, smoking, family history, alcoholism, and cocaine use. Generally, the diagnosis can be determined by a CT scan of the head if done within six hours of symptom onset. Occasionally a lumbar puncture is also required. After confirmation further tests are usually performed to determine the underlying cause. Treatment is by prompt neurosurgery or radiologically guided interventions. Medications such ...
