However, the main criticism for the clinical applicability of aPS/PT testing is the lack of reproducibility of the results among laboratories. In this study, we measured IgG and IgM aPS/PT using our original in-house enzyme-linked immunosorbent assays (ELISA) and commercial ELISA kits to assess the assay performance and to evaluate the accuracy of aPS/PT results. Microbubble-assisted ultrasound (sonopermeabilization) results in reversible permeabilization of the plasma membrane of cells. This method is increasingly used in vivo because of its potential to deliver therapeutic molecules with limited cell damage. Nevertheless, the effects of sonopermeabilization on the plasma membrane remain not fully understood. We investigated the influence of sonopermeabilization on the transverse mobility of phospholipids, especially on phosphatidylserine (PS) externalization. We performed studies using optical imaging with Annexin V and FM1-43 probes to monitor PS externalization of rat glioma C6 cells. Sonopermeabilization induced transient membrane permeabilization, which is positively correlated with reversible PS externalization. This membrane disorganization was temporary and not associated with loss of cell viability. Sonopermeabilization did not induce PS externalization via activation of the scramblase. We hypothesize that acoustically induced membrane pores may
6. CONCLUSIONS 1. We found that mast cell stimulation can induce PS externalization in the absence of secretory response. 2. We identified GPI-APs as molecules whose engagement can induce sustained and reversible non-apoptotic PS externalization. 3. GPI-AP-induced PS externalization was determined as non- apoptotic and distinct from the FceRl-induced PS externalization. 4. The effect of multiple triggering on PS externalization was additive and dependent on a tlpe of stimulus and cells engaged. 5. We identifred PLSCR1 as a molecule that becomes tyrosine phosphorylated in mast cells stimulated through GPI-APs. 6. We found that the PLSCR1 tyrosine phosphorylation is not associated with mast cell secretory response' and with GPI-AP- or FceRl-induced non-apoptotic PS externalization. 7. Using confocal microscopy and electron microscopy visualization of PLSCR1 in the course of mast cell activation we found that PLSCR1: (1) is not co-localized with extemalized PS, (2) is not co- localized with ...
Figure 4. Syndapins associate with PS-containing membranes via eF-BAR domains. A, Syndapin II domain structure (gray stars, NPFs) and schematic representation of different syndapin II constructs and the corresponding syndapin I proteins (right column). B, C, Syndapin I and II-l directly bind to PS-containing liposomes. Shown are liposome floatation analyses of GST-syndapin I (B) and II-l (C) with liposomes composed of (1) 80% PE and 20% PC (PE/PC); (2) 90% PE and 10% PS (PE/PS); and (3) 70% PE, 20% PC, and 10% PIP2 (PE/PC/PIP2). Density gradient fractions were immunoblotted with anti-GST antibodies. Liposomes floated into fraction 2, as proven by incorporation of fluorescent lipids and fluorescence measurements (compare with H). Note that both syndapin I as well as syndapin II-l protein material specifically associates with PS-containing liposomes tightly enough to withstand floating through the sucrose gradients and can therefore be detected together with the liposomes in fraction 2. Introduction: The phosphatidylserine receptor (PSR) plays an important role in apoptosis and inflammation. Whether this receptor is expressed in damaged brain cells in the early stages of stroke is unknown.. Hypothesis: The purpose of this study was to determine if this receptor is expressed on ischemic neurovascular endothelial cells and if so, can it bind with PS-containing liposomes (PS-liposomes). The latter would allow for a neurovascular targeting strategy for therapeutic delivery to ischemic brain cells.. Methods: Sprague Dawley rats (n=22) were randomly divided into sham, cerebral ischemia, and subarachnoid hemorrhage (SAH) injury groups. Cerebral ischemia was induced by middle cerebral artery occlusion (MCAO) for 2 hours using an intraluminal suture method. SAH was induced by an endovascular perforation method. Brain tissue was harvested for immunofluorescence staining at day 2 after surgery. Cultured human umbilical vein endothelial cells (HUVEC) were pre-treated with tumor necrosis ...

Scientific organizers: Prof. H. Heerklotz, U Freiburg • Dr. S. Fiedler, U Toronto/CAN Scope Cells and organelles establish different lipid compositions and properties in the outer and inner lipid leaflet. The complex and energy consuming machineries needed to regulate and maintain this state imply that membrane asymmetry is a key property for proper function of the membrane and, in turn, the survival of the cell. Until recently, this apparently crucial property could hardly be addressed in model membrane studies since asymmetric models were available only to a very limited extent. As a consequence, only rather few, pioneering studies have provided insight into the specific functions of membrane asymmetry. Presently, there is encouraging progress in developing easy-to-prepare, asymmetric membrane models in the form of liposomes, free standing bilayers, and solid supported membranes. It can be expected that such models will soon become the standard systems for biophysical membrane studies and

The plasma membrane has a highly asymmetric distribution of lipids and contains dynamic nanodomains many of which are liquid entities surrounded by a second, slightly different, liquid environment. Contributing to the dynamics is a continuous repartitioning of components between the two types of liquids and transient links between lipids and proteins, both to extracellular matrix and cytoplasmic components, that temporarily pin membrane constituents. This make plasma membrane nanodomains exceptionally challenging to study and much of what is known about membrane domains has been deduced from studies on model membranes at equilibrium. However, living cells are by definition not at equilibrium and lipids are distributed asymmetrically with inositol phospholipids, phosphatidylethanolamines and phosphatidylserines confined mostly to the inner leaflet and glyco- and sphingolipids to the outer leaflet. Moreover, each phospholipid group encompasses a wealth of species with different acyl chain combinations

During cell apoptosis, natural phosphatidylserines exposed on the outer membranes of apoptotic cells can prevent the occurrence of unexpected immune activation by directing antigen-presenting cells, such as macrophages and DCs that differentiate into the tolerogenic phenotype, and promoting the release of anti-inflammatory cytokines (28). To examine whether the PS-mimetic polymers can emulate the modulating effect of apoptotic cells and elicit anti-inflammatory responses in activated macrophages, we investigated the potential effect of ZPS, NZPS, and MPC on RAW 264.7 macrophages in the presence of lipopolysaccharide (LPS), an immunostimulatory molecule. For a fair comparison, ZPS, NZPS, and MPC were prepared into nanogels with uniform sizes (~40 nm) and surfaces (table S1). The zeta potential of anionic NZPS nanogel was negative (~−20 mv), while those of zwitterionic ZPS and MPC nanogels were close to neutral. RAW 264.7 macrophages (105 per well) were treated with nanogels composed of MPC, ...
Nonionic and anionic water-soluble amphiphiles were shown to increase strongly the binding of fluorescein isothiocyanate-conjugated annexin V (FITC-annexin V) in human erythrocytes pretreated with the aminophospholipid translocase (APLT) inhibitor n-ethylmaleimide (NEM). At high sublytic amphiphile-concentrations the binding of FITC-annexin V, monitored in a flow cytometer, was time- and temperature-dependent and occurred heterogeneously in the cell population, with 43-81% of cells being stained above background following incubation for 60 minutes at 37 degrees C. The increased FITC-annexin V binding apparently indicates an increased flop rate of phosphatidylserine (PS) to the outer membrane leaflet. When the NEM-pretreatment was omitted, the FITC-annexin V binding was markedly, but not completely, reduced. In erythrocytes incubated with a zwitter-ionic amphiphile, a small increase in FITC-annexin V binding was detected, while cationic amphiphiles did not induce an increased FITC-annexin V ...
This open-label, dose escalation study will be conducted in approximately 24 patients chronically infected with HCV and HIV. Patient cohorts will receive ascending dose levels of bavituximab weekly for up to eight weeks. HCV and HIV viral titers and other biomarkers will be evaluated, although they are not formal study endpoints.. In the United States alone, an estimated 300,000 individuals are co-infected with HCV and HIV, representing up to 30% of all HIV-infected patients. Co-infected patients have been shown to have a lower response to current HCV treatment regimens and the adverse effects of these regimens can be especially problematic for some HIV patients.. Bavituximab is a monoclonal antibody in a new class of anti-phosphotidylserine (PS) immunotherapeutics that targets and binds to cellular components that are normally not present on the outside of cells, but which become exposed on certain virally infected cells and on the surface of enveloped viruses, including both HCV and HIV. ...

When CD47 is ligated with a specific ligand, phosphatidylserine exposure is observed on the surface of the erythrocyte in a process known as eryptosis, which is similar to apoptosis. The aim of this study was to identify new proteins within the CD47 receptor mediated phosphatidylserine pathway and to elucidate the functions of any newly identified proteins. Previous research had suggested a link between protein 4.1R and CD47. Therefore, diseased protein 4.1R deficient erythrocytes were used as a molecular tool to help explore the role of protein 4.1R in the CD47 pathway. To achieve this aim, flow cytometry to measure phosphatidylserine exposure, 1D and 2D protein immunoblotting, 1D and 2D electrophoresis, RS100 ProteinChips coupled to SELDI-TOF, Q-TOF and MALDI-TOF, calcium influx, 2D serine/threonine phosphoblots and protein kinase A and protein kinase C inhibition were employed. Results showed increased phosphatidylserine exposure in 4.1R deficient erythrocytes and protein immunoblots ...
Asthma is associated with airway obstruction, airway inflammation, and bronchoconstriction. Immune cell infiltration and their production of inflammatory cytokines, such as interleukin (IL)-13, IL-17A, and tumor necrosis factor-α (TNF-α), increase airway smooth muscle contractility by increasing the calcium sensitivity of a pathway involving the guanosine triphosphatase RhoA, the kinase ROCK2, myosin light chain phosphatase, and myosin light chain. Mfge8 is an integrin-binding and phosphatidylserine-binding protein that functions in the clearance of apoptotic cells. Comparison of wild-type and mfge8-deficient mice in a mouse model of asthma (induced by ovalbumin) revealed that the mfge8-deficient mice had increased pulmonary resistance, a measure of airway hyperresponsiveness, but the abundance of infiltrated immune cells, the types of cells present, and the abundance of the inflammatory cytokines were the same between the two genotypes of mice. Isolated tracheal rings from the ...

PS 100 (standardized phosphatidylserine) Supports mental function Phosphatidylserine is a phospholipid found in all cells, but is most concentrated in brain cells. This familiarly-called brain nutrient supports cognitive function, emotional well-being and behavioral performance by restoring cell membrane composition. It has also been shown to support memory. In a double blind placebo controlled multicenter study of 425 individuals, daily supplementation of phosphatidylserine over six months resulted in statistically significant support of behavioral and cognitive parameters. In another six-month study of 40 individuals, phosphatidylserine supplementation enhanced cerebral metabolism and outcomes of cognitive training. These findings are consistent with earlier studies. In a placebo- controlled investigation of 149 subjects, the group receiving phosphatidylserine scored higher than placebo in performance tests related to memory tasks of daily life. In a trial of 35 subjects, those receiving

Bavituximab is an investigational monoclonal antibody that targets PS. Signals from PS inhibit the ability of immune cells to recognize and fight tumors. Bavituximab is believed to override PS mediated immunosuppressive signaling by blocking the engagement of PS with its receptors as well as by sending an alternate immune activating signal. Previous studies demonstrated PS-targeting antibodies shift the functions of immune cells in tumors, resulting in multiple signs of immune activation and anti-tumor responses. Peregrines clinical development strategy for bavituximab currently focuses on small, early-stage, proof-of-concept trials evaluating the drug in combination with other cancer treatments. As part of this approach the National Comprehensive Cancer Network (NCCN) has awarded grants to support three different clinical trials of bavituximab treatment combinations. These trials will evaluate novel bavituximab combinations in glioblastoma, head and neck cancer, and hepatocellular carcinoma ...

The surface charge of the plasma membrane is a crucial determinant of the targeting of signaling proteins. Yeung et al. have developed a probe that can reveal the location of the major negatively charged phospholipid, phosphatidylserine (PS), within cell membranes. PS was found in the cytosolic leaflet of the plasma membrane and on endosomes and lysosomes. It appeared to play a role in the targeting of a variety of positively charged proteins within these membranes.. T. Yeung, G. E. Gilbert, J. Shi, J. Silvius, A. Kapus, S. The characteristic features of cancer cells are aberrant (acidic) intracellular pH and elevated levels of phosphatidylserine. The primary focus of cancer research is concentrated on the discovery of biomarkers directed towards early diagnosis and therapy. It has been observed that azoxymethane-treated mice demonstrate an increased expression of calnuc (a multi-domain, Ca2+- and DNA-binding protein) in their colon, suggesting it to be a good biomarker of carcinogenesis. We show that culture supernatants from tumor cells have significantly higher amounts of secreted calnuc compared to non-tumor cells, selectively packaged into exosomes. Exosomal calnuc is causal for epithelial-mesenchymal transition and atypical migration in non-tumor cells, which are key events in tumorigenesis and metastasis. In vitro studies reveal a significant affinity for calnuc towards phosphatidylserine, specifically to its C-terminal region, leading to the formation of molten globule conformation. Similar structural ...
TY - JOUR. N2 - Earlier reports on nonneural cells have shown that the normally inner plasma membrane lipid, phosphatidylserine (PS), flip-flops out during the early stages of apoptosis, whereas DNA laddering and plasma membrane permeabilization occur during the late stages. In this study, the applicability of these parameters to CNS-derived neuronal cells was tested using hippocampal HN2-5, cells that undergo apoptosis under anoxia. Because such insults on unsynchronized cells, e.g., undifferentiated HN2-5 cells, result in both early and late apoptotic cells, we mechanically separated these cells into three fractions containing (a) cells that ...
Metastases are a major cause of cancer mortality. AXL, a receptor tyrosine kinase aberrantly expressed in many tumors, is a potent oncogenic driver of metastatic cell motility and has been identified as broadly relevant in cancer drug resistance. Despite its frequent association with changes in cancer phenotypes, the precise mechanism leading to AXL activation is incompletely understood. In addition to its ligand growth arrest specific-6 (Gas6), activation of AXL requires the lipid moiety phosphatidylserine (PS). Phosphatidylserine is only available to mediate AXL activation when it is externalized on cell membranes, an event that occurs during certain physiologic processes such as apoptosis. Here, it is reported that exposure of cancer cells to phosphatidylserine-containing vesicles, including synthetic liposomes and apoptotic bodies, contributes to enhanced migration of tumor cells via a PS-Gas6-AXL signaling axis. These findings suggest that anticancer treatments that induce fractional cell ...
Two-dimensional crystals of annexin-V bound to lipid layers containing dioleoylphosphatidylserine have been obtained in the presence of Ca2+. The crystals diffract to 20 A resolution and have the symmetry of the plane group p3 (unit cell dimensions: a = b = 94 A, gamma = 120 degrees). Electron image analysis revealed that the crystals are composed of trimers of annexin-V forming triskelion-like motifs. Each annexin-V molecule has a characteristic elongated shape, about 65 A by 20 A, when observed perpendicularly to the crystal plane. It is composed of two staggered domains of similar size, about 40 A by 20 A. Both domains are made of two sub-domains. The present data suggest that the four resolved sub-domains represent the folding units corresponding to the four 70 amino acid repeating segments characteristic of all annexins.

The effect of glucosylceramide (GlcCer) on activated protein C (APC)-phospholipid interactions was examined using fluorescence resonance energy transfer. Human APC, labeled with either fluorescein (Fl-APC) or dansyl (DEGR-APC) donor, bound to phosphatidylcholine/phosphatidylserine (PC/PS, 9:1 w/w) vesicles containing octadecylrhodamine (OR) acceptor with a K(d) (app) = 16 micro g/ml, whereas Fl-APC (or DEGR-APC) bound to PC/PS/GlcCer(OR) (8:1:1) vesicles with a K(d) (app) = 3 micro g/ml. This 5-fold increase in apparent affinity was not species-specific since bovine DEGR-APC also showed a similar GlcCer-dependent enhancement of binding of APC to PC/PS vesicles. From the efficiency of fluorescence resonance energy transfer, distances of closest approach of approximately 63 and approximately 64 A were estimated between the dansyl on DEGR-APC and rhodamine in PC/PS/GlcCer(OR) and PC/PS(OR), respectively, assuming kappa(2) = 2/3. DEGR-APC bound to short chain C8-GlcCer with an apparent K(d) of 460 ...
Approach and Results-TF activity and antigen levels on intact cells were measured using factor Xa generation and TF monoclonal antibody binding assays, respectively. Exposure of phosphatidylserine on the cell surface was analyzed using thrombin generation assay or by binding of a fluorescent dye-conjugated annexin V. 2′,7′-dichlorodihydrofluorescein diacetate was used to detect the generation of reactive oxygen species. Our data showed that HNE increased the procoagulant activity of unperturbed THP-1 cells that express traces of TF antigen, but had no effect on unperturbed endothelial cells that express no measurable TF antigen. HNE increased TF procoagulant activity but not TF antigen of both activated monocytic and endothelial cells. HNE treatment generated reactive oxygen species, activated p38 mitogen-activated protein kinase, and increased the exposure of phosphatidylserine at the outer leaflet in THP-1 cells. Treatment of THP-1 cells with an antioxidant, N-acetyl cysteine, suppressed ...
The present findings demonstrate that the lipid peroxidation product HNE can induce apoptosis in PC12 cells and primary rat hippocampal neurons and suggest that HNE is a mediator of oxidative stress-induced apoptosis. HNE induced loss of plasma membrane phosphatidylserine asymmetry (detected by annexin-V binding) and nuclear condensation and DNA fragmentation in PC12 cells and hippocampal neurons. HNE-induced cell death was prevented by cycloheximide, actinomycin-D, and aurintricarboxylic acid, consistent with a role for macromolecular synthesis and endonuclease activity in the cell death process. Although we recently reported that low concentrations of cycloheximide that do not cause sustained blockade of protein synthesis can protect neurons against excitotoxic and oxidative insults by a mechanism that apparently involves stimulation of antioxidant pathways (Furukawa et al., 1997), the concentration of cycloheximide used in the present study (10 μm) causes an essentially complete and ...
Foamy macrophages, containing myelin degradation products, are abundantly found in active multiple sclerosis (MS) lesions. Recent studies have described an altered phenotype of macrophages after myelin internalization. However, mechanisms by which myelin affects the phenotype of macrophages and how this phenotype influences lesion progression remain unclear. We demonstrate that myelin as well as phosphatidylserine (PS), a phospholipid found in myelin, reduce nitric oxide production by macrophages through activation of peroxisome proliferator-activated receptor β/δ (PPARβ/δ). Furthermore, uptake of PS by macrophages, after intravenous injection of PS-containing liposomes (PSLs), suppresses the production of inflammatory mediators and ameliorates experimental autoimmune encephalomyelitis (EAE), an animal model of MS. The protective effect of PSLs in EAE animals is associated with a reduced immune cell infiltration into the central nervous system and decreased splenic cognate antigen specific
The 14-3-3 family of adaptor proteins regulate diverse cellular functions including cell proliferation, metabolism, adhesion and apoptosis. Platelets express numerous 14-3-3 isoforms, including 14-3- z , which has previously been implicated in regulating GPIb a function. Here we show an important role for 14-3-3 z in regulating arterial thrombosis. Interestingly, this thrombosis defect is not related to alterations in von Willebrand factor (VWF)-GPIb adhesive function or platelet activation, but instead associated with reduced platelet phosphatidylserine (PS) exposure and procoagulant function. Decreased PS exposure in 14-3-3 z -deficient platelets is associated with more sustained levels of metabolic ATP and increased mitochondrial respiratory reserve, independent of alterations in cytosolic calcium flux. Reduced platelet PS exposure in 14-3-3 z -deficient mice does not increase bleeding risk, but results in decreased thrombin generation and protection from pulmonary embolism, leading to ...
There has been a reasonable amount of research done on Phosphatidyl Serine and its affects on athletes. Several studies show no benefits, but the majority show that the phospholipid reduces the exercise-induced increase in cortisol levels, and attenuates disturbances in mood or perception of fatigue. In terms of the science, phosphatidyl serine definitely gets the thumbs-up for athletic recovery.. However, when it comes to memory enhancement or reducing cortisol levels in the general population there has been mixed results in the literature. This is not surprising, as most scientists designing studies prove hopelessly inept in choosing representative sample groups and simply select whoever volunteers (normally students). Whereas all the athletes in the studies are put under stress, this method of testing normal individuals is unsuitable; you get 100 volunteers, of which only 10 may actually have high cortisol levels. A poor way to to test a nutrient that primarily works by bringing cortisol ...
During efferocytosis, phagocytic cells recognize dying cells by receptors binding to ligands specifically exposed on apoptotic cells. Multiple phagocytic receptors and some of their signaling pathways have been identified. However, the downstream pathways of tethering receptors that secure apoptotic cells remain elusive. It is generally assumed that tethering receptors induce signaling to mediate engulfment via interacting with co-receptors or other engulfment receptors located nearby. However, it is poorly understood whether co-receptors for tethering receptors exist during efferocytosis, and, if they do, whether they are indispensable for this process. Here, we address this issue using glycophosphatidylinositol (GPI)-anchored annexin A5 (Anxa5-GPI), an artificial tethering receptor without a putative co-receptor. Phagocytes expressing Anxa5-GPI exhibited enhanced binding of apoptotic cells, resulting in promoted ingestion of apoptotic cells in a phosphatidylserine-dependent manner. ...

MACS technique eliminates apoptotic sperms and may be indicated prior to ICSI, in order to guarantee that the injected spermatozoa are not damaged at a molecular level. Alternatively it could be combined with PICSI.. The externalization of the phospholipid phosphatidylserine (PS) to the sperm plasma membrane is a characteristic feature of the apoptotic phenomenon that occurs early during the process of sperm cell death. This basic knowledge has prompted investigators to develop a magnetic-based selection system for sperm cells that can separate early apoptotic from non-apoptotic germ cells (MACS).. Human sperm quality is defined by the classical parameters, concentration, motility and morphology, according to standard WHO diagnostic semen analysis. Nevertheless, hidden anomalies affecting spermatozoa membranes and causing apoptosis are present. Such features are not routinely detected in ejaculated spermatozoa but they have been proven to have a negative impact on ART outcome. Indeed, successful ...
The Annexin V-Biotin Apoptosis Detection Kit is based on the observation that soon after initiating apoptosis, most cell types translocate the membrane phospholipid phosphatidylserine (PS) from the inner face of the plasma membrane to the cell surface. Once on the cell surface, PS can easily bind to Biotin-conjugated Annexin V, a protein that has a strong natural affinity for PS. Annexin V-Biotin can be detected in conjunction with conventional dye-staining using any streptavidine- or avidin-dye reagents, such as (strept)avidine-fluorescein, -peroxidase, -alkaline phosphatase (AP), and -β-gal, etc. |p>‧ Detection method- Flow cytometry (Ex = 488 nm; Em = 530 nm) using and fluorescence microscopy |br>‧ Sample type- Living cells (suspension and adherant)|br>‧ Species reactivity- Mammalian |br>‧ Applications- Detect early/middle stages of apoptosis; differentiate apoptosis from necrosis.|p>|b>Features and Benefits|/b>|br>‧ Simple one step staining procedure in 30 minutes|br>‧ Fast and

Studies suggest phosphatidylserine has an effect on neurotransmitters that may play a role in cognitive functions. It has been shown that treatment with phosphatidylserine enhances cholinergic neurotransmission [2,3] and even increases the turnover of dopamine and norepinephrine in the brain [4].. In 1986 Delwaide et al. [5] reported that patients suffering from senile dementia improved their cognitive disorder with oral phosphatidylserine supplementation (derived from bovine brain cortex). Bovine brain cortex derived phosphatidylserine has also been shown to shift EEG power more towards the normal level in patients with mild primary degenerative dementia [6]; improved behavioral and cognitive parameters were also observed [7] as well as improved learning and memory in aged rodents [8,10].. However, because of concerns about mad cow disease (bovine spongiform encephalopathy [BSE]) [9], commercially available products are now made from cabbage or soy. Although the fatty acid composition of ...
The (Na+ +K+)-activated, Mg2+-dependent ATPase from rabbit kidney outer medulla was prepared in a partially inactivated, soluble form depleted of endogenous phospholipids, using deoxycholate. This preparation was reactivated 10 to 50-fold by sonicated liposomes of phosphatidylserine, but not by non-sonicated phosphatidylserine liposomes or sonicated phosphatidylcholine liposomes. The reconstituted enzyme resembled native membrane preparations of (Na+ +K+)-ATPase in its pH optimum being around 7.0, showing optimal activity at Mg2+:ATP mol ratios of approximately 1 and a Km value for ATP of 0.4 mM. Arrhenius plots of this reactivated activity at a constant pH of 7.0 and an Mg2+: ATP mol ratio of 1:1 showed a discontinuity (sharp change of slope) at 17 degrees C, with activation energy (Ea) values of 13-15 kcal/mol above this temperature and 30-35 kcal below it. A further discontinuity was also found at 8.0 degrees C and the Ea below this was very high (greater than 100 kcal/mol). Increased Mg2+ ...

The present study provides evidence that the tirucallic acid isomer αATA(8,24) is more potent than tirucallic acid derivatives investigated so far with respect to induction of apoptotic cell death in prostate cancer cells in vitro and in vivo. Furthermore, the differential selectivity of the tirucallic acid derivatives in destroying cancer cells without overt cytotoxicity to normal cells encouraged us to analyze the mechanisms of their toxicity.. Cell death occurs mostly dichotomously as either apoptosis or necrosis. Apoptosis is an active, programmed process of autonomous cellular dismantling that does not elicit inflammation known to be produced by necrosis (Fink and Cookson, 2005). Therefore, drugs leading to apoptotic cell death are of great interest in biomedical and pharmaceutical research. On the basis of several markers of apoptosis, such as loss of membrane phospholipid asymmetry resulting in phosphatidylserine externalization, caspase-3 activation, and the late-occurring DNA ...

PKC is a major intracellular receptor for the mouse skin tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA). PKC represents a large family of phosphatidylserine-dependent serine/threonine kinases. Protein kinase Cε (PKCε) is a member of the novel PKC sub-family. PKCε is a transforming oncogene and a predictive biomarker of various human cancers, including prostate cancer (PCa). We have shown that PKCε expression level correlates with human PCa aggressiveness. PKCε overexpression is sufficient to transform androgen-dependent LNCaP cells into an androgen-independent variant, which rapidly initiates growth in vivo in both intact and castrated male nude mice. PKCε silencing using specific siRNA in DU145 cells inhibits Stat3-regulated gene expression (COX-2) and cell invasion. Also, PKCε deletion (even one allele) in TRAMP mice prevents development of PCa. Taken together, the evidence indicates an association of PKCε to PCa development and aggressiveness. Thus, PKCε may be a potential ...
Acute kidney injury (AKI) is defined by the rapid loss of kidney function due to tissue damage. It affects 10-30 % of hospitalized patients and is independently associated with increased morbidity and mortality. Ischemia-reperfusion injury (IRI) is the most common pathoetiological mechanism of AKI, whereby tissue injury is mediated by reactive oxygen species. Ischemic AKI leads to the rapid upregulation of a transmembrane protein, kidney injury molecule-1 (KIM-1) on the apical membrane of proximal tubular epithelial cells (TECs). Previous work from our group and others demonstrated that the extracellular domain of KIM-1 specifically binds to phosphatidylserine on apoptotic cells, thereby transforming KIM-1-expressing TECs into semi-professional phagocytes for apoptotic corpses. The pathophysiological role of KIM-1 in AKI and relevant signalling mechanisms have not yet been elucidated. Using an in vivo model of AKI in mice genetically deficient in Kim-1, we reveal that Kim-1 expression protects mice from

We have examined intracellular transport and metabolism of the fluorescent analogue of phosphatidylserine, 1-palmitoyl-2-(N-[12[(7-nitrobenz-2-oxa-1,3-diazole-4-yl)amino] dodecanoyl])-phosphatidylserine ([palmitoyl-C12-NBD]-PS) in cultured fibroblasts. When monolayer cultures were incubated with liposomes containing (palmitoyl-C12-NBD)-PS at 37 degrees C, fluorescent PS was transported to the Golgi apparatus. NBD-containing analogues of phosphatidylcholine, phosphatidylethanolamine (PE), or phosphatidic acid did not accumulate in the Golgi apparatus under the same experimental conditions. We suggest that the transport is not due to endocytosis, but is the result of incorporation and trans-bilayer movement of the (palmitoyl-C12-NBD)-PS at the plasma membrane followed by translocation of the lipid from plasma membrane to the Golgi apparatus via nonvesicular mechanisms. Uptake of fluorescent PS was inhibited by depletion of cellular ATP and was blocked by structural analogues of the lipid or by ...
Heralding the charge higher was AcelRx Pharmaceuticals (NASDAQ:ACRX) which advanced 18% on the week after disclosing on Tuesday that its post-operative pain management system, Sufentanil NanoTab PCA, met its primary endpoint in late-stage trials and reconfirmed all previous late-stage study results. This data is strong enough for AcelRx to seek approval from the Food and Drug Administration with a new drug application filing expected next quarter. Id be cautious with AcelRx moving forward, as the possibility of an FDA rejection for opioid-based treatments is always a possibility.. Also gaining double digits on the week was the highly embattled Peregrine Pharmaceuticals (NASDAQ:PPHM) which rallied after the FDA approved its late-stage trial design for its second-line non-small-cell lung cancer immunotherapy, Bavituximab. While trial design approvals are rarely big news, it is in this case because mid-stage results for Bavituximab have been all over the place. At first Bavituximab demonstrated a ...

Isolation stress-induced vocalization and spontaneous activity for 10 min, which were attenuated by intracerebroventricular (i.c.v.) injection of PS. The effect of PS was compared with other phospholipids or l-serine, a constituent of PS. Phosphatidylcholine (PC) had no effect on these behavior, but phosphatidylethanolamine (PE) significantly increased vocalizations and spontaneous activity compared with PS. l-Serine similarly decreased isolation-induced vocalizations and ...
Platelets play a central role in stopping bleeding (haemostasis) and causing blockages in arteries (arterial thrombosis), particularly in coronary artery disease. At sites of blood vessel damage, they rapidly form aggregates or clumps which are stabilized by the local production of an enzyme called thrombin and generation of a clot of fibrin (a type of protein). Platelets coordinate this local clotting process by exposing a certain type of lipid (fat) called phosphatidylserine (PS) on their cell surface, allowing recruitment of the components of the clotting system that lead to the generation of a clot. This is called a procoagulant surface. The procoagulant activity therefore depends upon the surface area of membrane with exposed PS. In a paper published today in Circulation, Professor Alastair Poole of Bristols School of Physiology and Pharmacology and colleagues show that this surface area is enhanced substantially by the formation of balloon structures from platelets, with subsequent ...
The numerous and diverse biological roles of Phosphatidylserine (PtdSer) are featured in this special issue. This review will focus on PtdSer as a cofactor required for stimulating TYRO3, AXL and MERTK - comprising the TAM family of receptor tyrosine kinases by their ligands Protein S (PROS1) and growth-arrest-specific 6 (GAS6) in inflammation and cancer. As PtdSer binding to TAMs is a requirement for their activation, the biological repertoire of PtdSer is now recognized to be broadened to include functions performed by TAMs. These include key homeostatic roles necessary for preserving a healthy steady state in different tissues, controlling inflammation and further additional roles in diseased states and cancer. The impact of PtdSer on inflammation and cancer through TAM signaling is a highly dynamic field of research. This review will focus on PtdSer as a necessary component of the TAM receptor-ligand complex, and for maximal TAM signaling. In particular, interactions between tumor cells and their

ESMO) 2016 Congress. The presentation included interim efficacy and safety outcomes, as well as initial findings from the companys ongoing biomarker analysis of samples collected during the study. The SUNRISE Phase III trial was discontinued earlier this year based on a pre-specified interim analysis although patient treatment and follow-up in the study were allowed to continue. The pre-planned biomarker analysis has been taking place as patient follow-up has continued and available results were evaluated as part of the recent top-line data analysis. The study protocol pre-specified the collection of thousands of patient samples for exploratory analyses over a wide range of possible biomarkers, including pre-treatment levels of beta-2 glycoprotein-1 (β2GP1). Data presented at ESMO demonstrated that patients with pre-treatment β2GP1 levels between 200 and 240 (representing approximately 30% of randomized patients) achieved a statistically significant, 5.5-month improvement (13.2 months vs. 7.7 ...
TY - JOUR. T1 - CD4+, but not CD8+, T cells from mammary tumor-bearing mice have a down-regulated production of IFN-γ. T2 - Role of phosphatidyl serine. AU - Cheng, Xiaofang. AU - Lopez, Diana M.. PY - 1998/3/15. Y1 - 1998/3/15. N2 - IFN-γ production is dramatically reduced in T cells from mice bearing large mammary tumors. This inhibition of IFN-γ gene expression occurs mostly in CD4+ T cells, as determined by ELISA and reverse transcriptase-PCR. The effects of known mammary tumor factors in normal T cells and its subsets were evaluated. Pretreatment with granulocyte-macrophage CSF resulted in increased IFN-γ levels by T cells, while PGE2 pretreatment equally decreased the levels of this cytokine in CD4+ and CD8+ T cells from normal mice. Interestingly, phosphatidyl serine (PS) down-regulated the IFN-γ production of CD4+, but not that of CD8+, T cells. Methylation analysis indicated that the CpG dinucleotide in SnaBI site of the IFN-γ, 5 promoter flank region was hypermethylated in CD4+, ...

Contemporary humans face a major dilema: while we may enjoy longer life-spans than our ancestors, we are also confronted with the unique prospect of physically outlasting our capacities to think and reason. Currently, there are more than 30 million U.S. citizens over the age of 65, and it has been estimated that up to 50% will experience a noticeable decline in their mental abilities when faced with such common daily tasks as manipulating words, recalling names, and maintaining concentration and mental focus. A gradual loss of higher-level cognitive functions can appear in otherwise healthy individuals as young as fifty years of age, and can have a devastating impact on self-esteem, productivity, and general well-being. Much of the current research being conducted in the field of human memory is aimed at slowing down this gradual loss of cognitive capacity and devising protocols for conserving ones mental abilities. Over the last decade investigators have become particularly interested in ...
